한국산 효모의 분류학적 연구 (Taxonomical studies of yeasts in Korea)

  • The author attempted the taxonomical studies on yeasts in Takju mash. The samples used for the isolation of yeasts were collected from Takju breweries in Seoul. The yeasts obtained from Takju were identified as follows using the methods of Lodder et al. ; Saccaromyces cerevisiae group II, Saccharmyces cerevisiae group III, other group of Saccharomyces cerevisiae, Hansenula anomala and Pichia polymorpha. Saccharomyces cerevisiae II & III and other group Saccharomyces cerevisiae which were considered as wild yeasts have shown their major role in the fermentation process of Takju brewing. It seems that Hansenula anomala has much connection with the flavour of Takju. Other strains which are poor in their acoholic fermentation and lower in activity of acid production are not considered to be important in Takju brewing.

한국산 야생효모에 관한 연구 2 (Studies on the wild yeasts in Korea (II))

  • From the crops Drosophila collected in Mt. Sokni and Mt.Kyeryong, 7 strains were isolated and then 6 species of wild yeast were identified. 1) Of these six species of wild yeasts two were to be of genus Saccharomyces(Ascosporgenous), two Torulopsis and two Trichosporon (both genuses of Asporogenous). 2) It was found that the fermentation of the wild yeasts isolated from Drosophila was much better than that of any others ; in particular, S. florentinus and S. cerevisiae were good in fermenting maltose. 3) After being cultivated in malt extract agar medium at $25^{\circ}C$ for 3 days, the vegetative cells were found to be big but Torulopsis cells small. 4) It was also observed that the species of yeasts used fro food by Drosophila largely depends on genus and species of Drophila. 5) Of the yeasts isolated from the Drosophila, Trichosporon capitatum and Torulopsis dattila, which has not previously been recorded, were identified. 6) It is believed, therfore, that S.florentinus, powerful in fermenting maltose, will be extremely useful in terms of industrial application.

해양효모의 분류 1 (Studies on Yeasts Isolated from Marine Substrates (I))

  • As a part of taxonomical and ecological studies on the yeasts in marine environments, several kinds of yeasts were isolated from Zostera marina several invertebrates (penaeus, Meretrix and Neptunus) and surface sea water, which are collected at the two established sites of estuarine areas ; Dolsan isaland in Youchun district and Baiksu-ri in Youngkwang district. The obtained results can be summarized as follows. 1. Ascosporgenous Yeasts. Hansenula sp I and Hansenula sp II were isolated from Zotera marina and Hanse nular sp I from penaeus. 2. Asporogenous Yeasts. Trichoporon fermentans, Torulopsis, ernobii and Toruopsis dattia were isolated from Zostera marina, Candida krusei from Meretrix and Neptunus, Torulopsis pinus from surface sea waters, and Phodotorula aurantiaca from Penaeus. 3. More notable isolations of several species from Zostera marina than the other sources could be assumed as related to the higher sugar concentration of this plant.

  • BACKGROUND: Yeast biotechnology finds applications in various industries. Hence, we sought to explore the yeasts associated with the flower of Aster spathulifolius Maxim. This study aimed to isolate yeasts from the flower of the plant and screen for biosurfactant-producing yeasts. METHODS AND RESULTS: We collected flowers of Aster spathulifolius Maxim. and performed pure isolation using four types of media. In total, 117 strains belonging to 4 genera, namely, Cryptococcus (75 strains), Aureobasidium pullulans (30 strains), Candida (11 strains), and Rhodotorula (1 strain), were isolated and identified by ITS sequencing. Upon in-depth analysis, Cryptococcus, the most dominant genus (75 strains) was categorized into the 'Unknown group'. Upon in-depth analysis of A. pullulans, we discovered the 'Unknown group I' (27 strains) and the 'Unknown group II' (2 strains), which have not been reported previously. Two A. pullulans isolates with potent surfactant activity were selected via the screening procedure. CONCLUSION: In this study, a total of 117 strains were isolated from the flower of Aster spathulifolius Maxim. In addition, two biosurfactant-producing yeasts were identified from among the isolated yeasts.

간장발효에 관여하는 효모에 관한 연구 (제2보) -간장 덧 중에 생육하는 효모에 대하여- (Studies on the Yeasts for the Brewing of Soy Sauce(2) -Isolation, identification and classification of the yeasts in the soy sauce mash-)

  • 간장덧중에 생육하는 효모를 경시적으로 계수 및 분리동정 하고 TTC 정색에 의하여 유별한 결과는 다음과 같다. (1) 간장덧 1ml중에 생육하는 일반효모의 수는 담금직후 $127{\times}10^3$, 1개월후 $86{\times}10^3$, 3개월후 $356{\times}10^3$, 6개월후 $1250{\times}10^3$, 2년후 $65{\times}10^3$ 개로 나타났고 내염성효모수는 담금직후 0, 1개월후 $40{\times}10^3$, 3개월후 $81{\times}10^3$, 6개월후 $358{\times}10^3$, 2년후 $23{\times}10^3$개 로서 일반효모와 내염성효모 다 같이 담금초에서 숙성기 까지 상승하였다가 후숙기에서 감소하는 경향을 보였다. (2) 간장덧으로 부터 경시적으로 임의 분리한 50주의 효모를 동정한 결과 Saccharomyces rouxii 10주, Saccharomyces marxianus 1주, Saccharomyces rosei 3주, Saccharomyces fermentati 1주, Saccharomyces mellis 6주, Saccharomyces acidifaciens 1주, Saccharomyces pastori 1주, Pichia polymorpha 3주, Hansenula anomala 2주, Hansenula saturnus 1주, Hansenula suaveolens 2주, Nadosonia fulvescens 5주, Debaryomyces hansenii 8주, Debaryomyces nicotianae 1주, Debaryomyces kloeckeri 1주, Torulopsis sake 2주, Torulopsis holmii 1주, Candida pelliculosa 1주등 7속 18종으로 동정 되었다. (3) 담금기간에 의한 효모의 분포는 담금초에 Saccharomyces rouxii, Saccharomyces marxianus, Saccharomyces rosei, Pichia polymorpha, Hansenula suaveolens, Hansenula saturnus, Debaryomyces hansenii, Torulopsis sake, Candida pelliculosa, Debaryomyces nicotianae, Nadsonia fulvescens 등이 생육하고 숙성기에 Saccharomyces rouxii, Saccharvmyces rosei, Saccharomyces fermentati, Saccharomyces mellis, Saccharomyces pastori, Hansenula anomala, Saccharomyces acidifaciens, Debaryomyces hansenii 등이 생육하고 후숙기에는 Saccharomyces rouxii, Saccharomyces mellis, Nadsonia fulvescens, Debaryomyces hansenii, Debaryomyces kloeckeri, Torulopsis sake, Torulopsis holmii 등이 생육하였다. (4) 담금초기에는 TTC white 효모가 많이 나타났고 발효기 와 후숙기에는 TTC red 효모가 50% 이상을 차지 하였다. (5) 동정한 이들 효모를 TTC 정색에 따라 유별한 결과 Saccharomyces mellis, Saccharomyces pastori는 TIC red로, Saccharomyces acidifaciens는 red pink. Hansenula saturnus, Debaryomyces kloeckeri, Torulopsis holmii는 pink, Saccharomyces marxianus, Nadsonia fulvescens는 white로 각각 나타났고 기타는 전보의 결과와 같았다. Saccharomyces rouxii대부분이 red였으나 red pink로 나타난 것도 있었다, (6) 간장덧중에 생육하는 효모의 종류는 국중에 생육하는 효모의 종류와 거의 비슷하게 나타났으나 국효모는 대부분이 내염성이 없었고 간장덧중의 효모는 발효기간의 경과에 따라 내염성 효모수가 증가 되었다.

대전과 충남 지역에서 채취한 야생화로부터 효모의 분리 및 동정 (Isolation and Identification of Yeasts from Wild Flowers of Daejeon City and Chungcheongnam-do in Korea)

  • 자연환경에서 분포하고 있는 효모의 mycoflora를 확인하고 궁극적으로 이들을 고부가가치의 산업에 응용하기 위한 연구의 일환으로 대전과 인근 충남지역 일부의 야생화들을 채집하여 효모들을 분리, 동정하였다. 대전지역 야생화 42점에서 32종의 효모들을 54균주 분리하였고 이들 중 Cryptococcus 속균과 Phodotonula 속균 등이 우점균이었으며 Metschnikowia 속균과 같이 희귀 효모들을 20여종 분리하였다. 대전 인근의 충남 금산군과 논산군 일부지역의 야생화 28점에서 모두 18종의 효모들을 26균주 분리하였다. 이들 균 중에서 Candida 속균과 Pseudozyma 속균 등이 우점균으로 분리되었다.

Mycological Characteristics of Nine Unrecorded Yeasts from Flowers in the Orchard of Yesan-gun, Chungcheongnam-do and Hanbat Arboretum in Daejeon City, Korea

  • Six unrecorded yeasts, Cryptococcus festucosus 41-3, Cryptococcus heveanensis 56-4, Debaryomyces nepalensis 95-4, Issatchenkia occidentalis 142-1, Dioszegia zsoltii 39-1, and Kwoniella europala 47-2 were screened from 108 yeasts isolated from flowers and fruits in orchards of Yesan-gun, Chungcheongnam-do, Korea. The morphological and cultural characteristics of these unrecorded yeasts were investigated. They had various shapes, including ellipsoidal, globose, and oval, and also had budding mode in vegetable reproduction, except I. occidentalis 142-1 (fission mode). K. europaea 47-2 only formed pseudomycelium. D. zsoliti 39-1 did not grow in yeast extract-malt extract medium, potato dextrose medium, and vitamin-free medium. C. festucosus 41-3 grew well in 5% NaCl-containing yeast extract-peptone-dextrose medium and had a growth pH range of 7.0~10.0. Three unrecorded yeasts Ogataea polymorpha HB45-1, Rhodotonula hinnulla HB62-2, and Cryptococcus rajasthanensis HB80-4 were screened from 51 yeasts isolated from flowers in Hanbat arboretum in Daejeon city, Korea. They were globose in shape and did not form pseudomycelium. In addition, O. polymorpha HB45-1 and C. rajasthanensis HB80-4 had budding mode in vegetable reproduction. All of them grew well in vitamin-free medium and C. rajasthanesis HB80-4 also grew in 50% glucose and 5% NaCl-containing YPD medium.

효모의 종류에 따른 탁주의 품질특성 변화 (Quality Characteristics of Takju by Yeast Strain Type)

  • Investigation of the quality characteristics of Takju brewing mashes prepared with several different yeasts was carried out during the 42-day brewing process, including examination of titratable acidity, amino acid content, sugar contents, alcohol contents, pH, reducing sugar, yeast population, lactic acid bacteria, and sensory evaluation. Fermivin yielded the highest titratable acidity, while la parisienne had the lowest. Fermivin also had the highest value of amino acid content (14.33%), while other yeasts shows period tended to increase over exclude Instaferm and Song chun. The amount of sugars in instaferm ($11.33^{\circ}Bx$) and instaferm red ($10.53^{\circ}Bx$) were higher than those in La parisienne red and Song chun ($9.57^{\circ}Bx$). The alcohol contents of instaferm was the highest value (15.20%) while the alchol content of the other yeasts tended to increase the fementation period. Fermivin had the highest pH at $12^{th}$ day among the yeasts(4.02), Instaferm red had the lowest pH among the yeasts (3.87) while safbrew wb-06 had the highest reducing sugar content(117.60 mg/mL). Instaferm obtained the highest preference in the sensory evaluation for all items.

Inhibitory Effects of Na-Hypochlorite and Heating on the Mycobiota Associated with Fruits or Juice of Passion (Passiflora edulis Sims) in Uganda

  • A total of 34 species belonging to 21 genera of fungi were recorded on passion fruits of both pure and hybrid origin in Uganda, however, the pure type exhibited wider spectrum (28 species and 16 genera) than the hybrid type (21 & 15). Also, yeasts (unidentified and Rhodotorula mucilaginosa) were also encountered in high numbers. Moreover, the mean count of all mycobiota obtained from the pure type was higher than that of hybrid, despite the bigger size of the later. Members of yeasts and Cladosporium followed by Phoma, Penicillium, Fusarium and Alternaria species dominated on passion fruits of pure origin, while only C. cladosporioides, F. solani and yeasts dominated on the hybrid type. Treatment with Na-hypochlorite exhibited inhibitory effects on the total mycobiotic propagules as well as the dominant species from fruits of both types. The current results, therefore, suggest the use of Na-hypochlorite to control the post-harvest mycobiota associated with passion fruits. Regarding the mycobiota contaminating passion juice, yeasts were found to be the major contaminants with Candida parapsilosis being the most common. Moulds constituted only a minor proportion with Acremonium strictum followed by Fusarium chlamydosporum, F. moniliforme, F. acuminatum and F. solani as the most dominant species. In the heat-treated juice samples, the counts of the most commonly encountered mycobiota (both yeasts and molds) were significantly inhibited or completely eliminated. Some unidentified Bacillus species were also recovered from the juice, however, their counts in the heated samples were increased but insignificantly.

Microbiological Characteristics and Physiological Functionality of New Records of Yeasts from Wild Flowers in Yokjido, Korea

  • Two new yeast records, Cryptococcus adeliensis YJ19-2 and Cryptococcus uzbekistanensis YJ10-4 were screened from 60 yeasts strains that were isolated and identified from wild flowers in Yokjido, Gyeongsangnam-do, Korea. The morphological and cultural characteristics of the newly recorded yeasts and the physiological functionalities of the supernatants and cell-free extracts obtained from their cultures were investigated. The two newly recorded yeasts did not form ascospores and pseudomycelia. They also grew well in yeast extract-peptone-dextrose broth. C. uzbekistanensis YJ10-4 grew in a vitamin-free medium and was also tolerant to sugar and salt. Antihypertensive angiotensin I-converting enzyme inhibitory activity of the supernatant from C. adeliensis YJ19-2 was high (71.8%) and its cell-free extract also showed very high (81.2%) antidiabetic $\acute{a}$-glucosidase inhibitory activity.
