. Mediator of transcriptional regulation is the evolutionary conserved coactivator complex that plays He central role in the integration and recruitment of diverse regulatory signals and transcription machinery to certain promoters. In yeast, each Mediator subunit is required for transcriptional regulation of a distinct group of genes. In order to decipher the mechanistic roles of Mediator proteins in regulating developmental specific gene expression, we isolated, and analyzed a multiprotein complex containing Drosophila Mediate. homologs (dMediato.). dMediato. interacts with several sequence-sperific transcription factors and basal transcription machinery, and is critical for activated transcription in response to diverse transcriptional activators. In order to elucidate the function of Mediator in metazoan development, we isolated mutants of a conserved Mediate. subunit, Drosophila Med6 (dMed6). dMed6 null homozygotes failed to pupate and died in the third larval instar. Larval mitotic cells and most imaginal discs showed severe defects in proliferation, but no apparent morphological defect was observed in other larval tissues. Clonal analysis of dMed6 mutant cells revealed that dMed6 is essential for cell viability and proliferation of most adult cell types. Drosophila cDNA microarray, quantitative RT-PCR, and in situ expression analyses of developmentally regulated genes in dMed6 mutants showed that transcriptional activation of a subset of genes involved in neuroblast proliferation in the larval brain were most affected. Our results suggest that dMed6 is required in most for transcriptional regulation of a subset of genes important for cell proliferation and metabolism.
The Potexvirus Alternanthera mosaic virus (AltMV) has multifunctional triple gene block (TGB) proteins, among which our studies have focused on the properties of the TGB1 protein. The TGB1 of AltMV has functions including RNA binding, RNA silencing suppression, and cell-to-cell movement, and is known to form homologous interactions. The helicase domains of AltMV TGB1 were separately mutated to identify which regions are involved in homologous TGB1 interactions. The yeast two hybrid system and Bimolecular Fluorescence Complementation (BiFC) in planta were utilized to examine homologous interactions of the mutants. Helicase motif I of AltMV TGB1 was found to be critical to maintain homologous interactions. Mutations in the remaining helicase motifs did not inhibit TGB1 homologous interactions. In the absence of homologous interaction of TGB1, subcellular localization of helicase domain I mutants showed distinctively different patterns from that of WT TGB1. These results provide important information to study viral movement and replication of AltMV.
Glucoamylase of Saccharomyces diastaticus is produced as a large precursor composed of signal peptide (21 amino acid residues), Thr and Ser-rich region and functional glucoamylase. To evaluate the utility of the glucoamylase signal peptide (GSP) for the secretion of foreign proteins, four types of GSP mutants (ml : Pro-18 longrightarrowLeu-18, m2 : Tyr-13 longrightarrowLeu, m3 : Ser-9longrightarrowLeu-9, m4 : Asn-5 longrightarrowPro-5) were constructed and secretion efficiency of each mutant was compared with that of native GSP by the expression and secretion of Bacillus subtilis CMCase under the control of GAP in N-terminal domain and hydrophobic domain. n mutant 4, a polar amino acid was replaced by a helix - breaking Pro residue. CMCase activity assay and Western blot analysis revealed that CMCase secretion by GSP mutants replaced by Leu were increased compared with native GSP. In the case of m2 and m3, the substitution of Leu for Tyr-13 and Ser-9 in the hydrophobic region resulted in a twofold increase in the extracellular CMCase activity.

Bar-code (tag) microarrays of yeast gene-deletion collections facilitate the systematic identification of genes required for growth in any condition of interest. Anti-sense strands of amplified bar-codes hybridize with ~10,000 (5,000 each for up-and down-tags) different kinds of sense-strand probes on an array. In this study, we optimized the hybridization processes of an array for fission yeast. Compared to the first version of the array (11 ㎛, 100K) consisting of three sectors with probe pairs (perfect match and mismatch), the second version (11 ㎛, 48K) could represent ~10,000 up-/ down-tags in quadruplicate along with 1,508 negative controls in quadruplicate and a single set of 1,000 unique negative controls at random dispersed positions without mismatch pairs. For PCR, the optimal annealing temperature (maximizing yield and minimizing extra bands) was 58℃ for both tags. Intriguingly, up-tags required 3× higher amounts of blocking oligonucleotides than down-tags. A 1:1 mix ratio between up- and down-tags was satisfactory. A lower temperature (25℃) was optimal for cultivation instead of a normal temperature (30℃) because of extra temperature-sensitive mutants in a subset of the deletion library. Activation of frozen pooled cells for >1 day showed better resolution of intensity than no activation. A tag intensity analysis showed that tag(s) of 4,316 of the 4,526 strains tested were represented at least once; 3,706 strains were represented by both tags, 4,072 strains by up-tags only, and 3,950 strains by down-tags only. The results indicate that this microarray will be a powerful analytical platform for elucidating currently unknown gene functions.
출아 효모에서의 Paf1 복합체는 총5개의 단백질로 구성되어있고, 구성성분들은 출아효모, 초파리, 식물들, 그리고 인간에 이르기까지 구조적으로, 기능적으로 잘 보존되어 있다. RNA 중합효소 II와 결합한 상태로 전사 개시부위부터 종결부위까지 함께 이동하며, 여러 전사인자들의 유입을 위한 매개체로 작용하여, 유전자 발현 조절의 핵심적인 역할을 수행한다. Paf1 복합체는 H2BK123 monoubiquitination에 기여하고, histone crosstalk에 의해 간접적으로 H3K4의 di-, tri-methylation에 기여하는 것이 알려져 있다. 하지지만, Paf1 복합체 구성요소들의 개별적인 기능에 대해서는 연구가 되어있지 않다. 이 연구에서는, Paf1 복합체 구성요소들의 단일 결핍 돌연변이 균주를 만든 후, 이들의 H2BK123 monoubiquitination 및 H3K4 mono-, di-, tri-methylation에 미치는 영향을 관찰했다. 놀랍게도, ${\Delta}paf1$, ${\Delta}rtf1$, ${\Delta}ctr9$ 돌연변이 균주에서는 H2Bub에 영향을 받는 H3K4me2와 H3K4me3뿐 아니라, H2B monoubiquitination에 영향을 받지 않는 H3K4 monomethylation의 심각한 감소를 관찰했다. 그러나, methyl기 전달 효소인 Set1의 발현 정도는 이 돌연변이 균주들에서 변하지 않았다. 이러한 결과로부터, Paf1 복합체가 Set1의 활성이나 Set1 복합체의 안정성을 직접 조절함으로써 H3K4 methylation을 조절할 수 있음을 제시한다.
C. magnoliae JH를 이용하여 높은 수융의 erythritol을 생산 하기 위한 삼투압 내성의 변이균주 개발과 탄소원 및 질소원 의 최적 농도결정에 대한 연구를 수행하였다. 꿀 벌집에서부 터 분리한 야생균주인 C. magnoliae JH를 이용한 baffled R flask 배양에서 당농도 100 밍L인 경우 erythritol 수율은 2 20.3% 이고 생산성은 0.23 g/L-h이었다. 균주의 수율 향상을 위하여 야생균주의 포자에 EMS를 처리한 후 2차에 결친 선 별과정을 통해 높은 농도의 염에서 세포성장이 우수하변서 동시 에 erythritol의 수울과 생산성 이 가장 우수한 삼투압 내 성 변이균주 M26을 최종적으로 선발하였다. 선발한 변이균 주 M26을 이용한 baffled flask 배양에서 100 g/L의 glucose 인 경우에 erythritol의 수율과 생산성이 각각 25.0%와 0.30 g/L-h로서 야생균주에 비하여 증가한 반면에 glycerol의 생성은 오히려 감소하였다. 변이균주 M26을 이용한 탄소원 의 선별에 대한 실험에서는 glucose가 erythritol 생산에 가장 적합하였다. 유기질소원인 yeast extract는 5 g/L의 농도에서 erythritol 수율이 가장 우수하였다. 발효조를 이용한 glucose 농도 결정에 대한 실험에서 세포농도는 glucose의 농도 증가 에 따라 감소하였으나, eηthritol 농도는 당 농도의 증가에 따라 증가하여 250 밍L glucose의 농도에서 최대 127.5 g/L가 생산되었으며, 발효말기에는 glycerol이 존재하지 않았다. 또 한 수율과 생산성도 glucose의 농도 증가에 따라 250 g/L까 지 증가하여 최대 51.0%의 수율과 0.63 g/L-h의 생산성을 나타내었다.

EHT1 and EEB1 are the key Saccharomyces cerevisiae genes involved in the synthesis of ethyl esters during wine fermentation. We constructed single (Δeht1, Δeeb1) and double (Δeht1Δeeb1) heterogenous mutant strains of the industrial diploid wine yeast EC1118 by disrupting one allele of EHT1 and/or EEB1. In addition, the aromatic profile of wine produced during fermentation of simulated grape juice by these mutant strains was also analyzed. The expression levels of EHT1 and/or EEB1 in the relevant mutants were less than 50% of the wild-type strain when grown in YPD medium and simulated grape juice medium. Compared to the wild-type strain, all mutants produced lower amounts of ethyl esters in the fermented grape juice and also resulted in distinct ethyl ester profiles. ATF2, a gene involved in acetate ester synthesis, was expressed at higher levels in the EEB1 downregulation mutants compared to the wild-type and Δeht1 strains during fermentation, which was consistent with the content of acetate esters. In addition, the production of higher alcohols was also markedly affected by the decrease in EEB1 levels. Compared to EHT1, EEB1 downregulation had a greater impact on the production of acetate esters and higher alcohols, suggesting that controlling EEB1 expression could be an effective means to regulate the content of these aromatic metabolites in wine. Taken together, the synthesis of ethyl esters can be decreased by deleting one allele of EHT1 and EEB1 in the diploid EC1118 strain, which may modify the ester profile of wine more subtly compared to the complete deletion of target genes.
진핵세포 유전자의 기초대사발현의 조절계를 밝히기 위한 일환으로, 효모의 histidine생합성계 효소의 구조유전자 HIS5를 이용하였다. HIS5 유전자는 충분한 아미노산 조건하에서는 발현이 억제되어 비교적 높은 기초발현만을 하나, 어떤 아미노산이 결핍되면 탈억제되어 높은 발현량을 보이며 탈억제는 cis의 작용인자인 promoter상의 5'-TGACTC-3' 및 trans 작용인자 GCN4와 GCD17 GCN2등이 관여한다. trans 작용인자들에 의한 HIS5 유전자의 발현량의 변화를 간단하게 측정하기 위하여, HIS5 promoter와 repressible acid phoshates(APase)의 구조유전자중 promoter를 제거한 DNA단편을 연결시켜 HIS5-PHO5 융합유전자를 이용하였다. gcn2 및 gcn4 변이주의 APase 활성은 야생주와 비교하여 3내지 4배 낮았으며, gcn2변이주와 gcn2 gcn4 이중변이주의 APase 활성은 유사하였다.
Bacillus subtilis의 생균 및 포자액을 자외선 조사와 diethylsulfate 처리로서 adenine 요구변이주 총 62주를 분리하였다. 이들 변이주는 발효배지에 자외선흡수 물질을 축적하고 있음을 확인하였으며 이 축적물은 thin-layer chromatogram 자외선흡수 곡선등으로 hypozanthine, uracil 임을 동정하였다. 이 중에서 hypoxanthine만을 축적하는 변이주 BS-137의 배지성분을 검토한 결과 탄소원소로서는 glucose가 좋았으며 질소원으로서는 yeastext와 NaNO$_3$ 가 적당하였다. 아울러 배양액으로부터 hypoxanthine을 분리 정제하는 방법을 확립하였다.
