Study on Rheological and Phermal Properties of Dioiscorea batatas DECAISNE Starch (마(Dioscorea batatas DECAISNE)전분의 Rheology 및 열적 특성에 관한 연구)

  • The purpose of this study was to investigate the rheological and thermal properties of yam starch. Yam starch had a hydrodynamic volume with the intrinsic viscosity,[$\eta$], of 0.29dl/g deionized water. The values of the intrinsic viscosity of yam starch, determined to pH 2-11, varied between 0.07 to 0.18 dl/g. The highest intrinsic viscosity was obtained at pH 7. At salt concentrations 0-0.2 M NaCl, the intrinsic viscosity of yam starch was decreased up to 0.05 M NaCl concentration then increased to 0.07 M NaCl concentration and remained constant to reach 0.2 M NaCl concentration. The overlap parameter, calculated with the intrinsic vicosity data, was 3.45 g/dl in deionized water. The thermal properties of yam starch were investigated by Differential Scanning Calorimetry. Three endotherms were observed both pH solution and salt concentation. In the presence of pH 9, the onset temperature of gelatinization peak was the lowest temperature of 50.$32^{\circ}C$ and the enthalpy ($\Delta$H) was increased in this solution. The effect of salt on the thermal properties of yam starch was determined at salt concetration of 0-0.2 M NaCl. The enthalpy significantly decreased to salt concentration 0.07 M NaCl and the lowest onset temperature of this concentration was 52.$90^{\circ}C$.

Evaluation of Antimicrobial, Antithrombin, and Antioxidant Activity of Dioscorea batatas Decne (마(Dioscorea batatas Decne)의 항균, 항산화 및 항혈전 활성 평가)

  • Yam (Dioscorea batatas Decne) has been recognized as healthy food due to its various biological activities, such as anti-obesity, anti-constipation, anti-proliferation, anti-mutagenic activity, and decrease of blood glucose and cholesterol level. In this study, the methanol extract and its organic solvent fractions were prepared from dried Dioscorea batatas Decne, and their antimicrobial, antithrombin, and antioxidant activity were evaluated, respectively. The 66.7% of methanol extract was fractioned into water residue, and most of total-polyphenol/total-flavonoids were found in ethylacetate fraction. This ethylacetate fraction at $500\;{\mu}g/disc$ concentration showed strong antibacterial activity against gram-negative and gram-positive bacteria, except Staphylococcus aureus. Antifungal activity was not observed in methanol extract and its fractions. But, strong antithrombin activity was found in methanol extract. At 4.8 mg/mL concentration, thrombin time was 99.5 sec, which is coincides with 30% activity of aspirin, a commercial available antithrombosis agent. The ethylacetate fraction showed strong DPPH scavenging activity ($IC_{50}$ of $80.5\;{\mu}g/mL$), compared to vitamin C ($IC_{50}$ of $15.2\;{\mu}g/mL$). Also, the ethylacetate fraction showed strong SOD-like activity and reducing power, which are coincide with 43% of vitamin C and 82.7% of butylated hydroxytoluene activity, respectively.

Development of Cryopreservation System using Shoot-Apex in Yam (Dioscorea batatas)

  • The goal of this research was to develop an efficient cryopreservation protocol for germplasms of yam (Diosorea batatas), that were cultivated in Korea. Comparative studies with four other cryogenic techniques and subsequent experiments for shoot regrowth were conducted. in vitro-grown shoot-apices of the D. batatas were successfully cryopreserved by encapsulation-dehydration. The maximum survival of shoot-apices could be achieved when the precultured (with 0.3 M of sucrose for one day) and encapsulated (with a 3%(w/v) Na-alginate solution) apices were dehydrated for $3.5{\sim}4\;h$ prior to direct immersion in LN (liquid nitrogen). The frequency of regrowth rate of cryopreserved apices was not decreased during 3-month storage period. The thawing method markedly affected survival of the cryopreserved apices, and thawing at $40^{\circ}C$ for 3 min produced the best results. When cryopreserved apices were post-cultured on the post-culture medium (MS), supplemented with $0.2mgl^{-1}$ of BA ($N_6$-benzyladenine) and $0.2mgl^{-1}$ of kinetin, they showed direct shooting without callusing.

Antioxidant Activities and Monacolin K Production on Solid-State Fermentation of Diverse Yam by Aspergillus Species Strain (Aspergillus 속 균주를 이용한 마 품종별 고체발효시 Monacolin K 생산과 항산화 활성)

  • This study was conducted to investigate the characteristics between non-fermented chinese yam and rice (non-FCYR) and fermented chinese yam and rice (FCYR). 1.1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, total polyphenol, flavonoid contents and reducing power were investigated to evaluate the anti-oxidation activities of FCYR. Also, productivity of monacolin K, major medicinal ingredient of FCYR, was investigated, using Aspergillus terreus KCCM 12225. In case of non-FCYR, DPPH radical scavenging activities for three kinds of yam (Dioscorea japonica, Dioscorea batatas, Dioscorea opposita) and rice were estimated about 51.8, 66.4, 42.2 and 7.5%, while in case of FCYR, those for three kinds of yam and rice were estimated to increased about 64.7, 74.7, 52.8, and 32.3%, respectively. Total polyphenol contents of non-FCYR were estimated to 196.9, 265.7, 160.1, and 91.6 mg/kg, while total polyphenol contents of FCYR were estimated to increase about 530.7, 708.3, 427.2, and 265.9 mg/kg, respectively. Total flavonoid contents of FCYR increased to 2.8-3.3 times higher than those of non-FCYR. Reducing power of non-FCYR was about 0.97, 1.28, 0.64, and 0.17 (OD at 700 nm), while, that of FCYR increased about 1.75, 2.38, 1.24, and 0.46. Remakable increase in monacolin K productivity of FCYR was observed. Monacolin K productivity of FCYR was estimated to 467.1, 514.8, 339.2, and 272.5 mg/kg, respectively. In this study, fermented chinese yam (Dioscorea batatas) was estimated to be effective biological activity material.

Occurrence and Vertical Distribution of Meloidogyne incognita in Chinese Yam (Dioscorea batatas) (마 재배지의 뿌리혹선충의 발생 실태 및 수직 분포)

  • The study was conducted to investigate the infestation and distribution of plant-parasitic nematodes on Chinese yam (Dioscorea batatas) in Andong, Korea. Root-knot nematodes, Meloidogyne incognita was the most prevalent nematode species which is found from 43 yam fields (81.1%) with high population densities (average of 450 juveniles/$300cm^3$). Other nematodes, root-lesion nematode (Pratylenchus spp.) and pin nematode (Paratylenchus spp.) were less problematic. Density of juveniles (J2) of M. incognita was $10\;J2/300cm^3$ by August, then increased to 274 in September and 624 in October. The highest J2 density was found at the soil depth between 40 and 50 cm ($1,840\;J2/300cm^3$). M. incognita was able to infest yam tuber down to a depth of 70 cm and developed galls outside and brown spots inside. The highest number of females were found at 40-50 cm (79 females/10 cm piece) tuber from the top.

Effects of Yam (Dioscorea batatas Dence) Extracts on the Growth and Nucleus-DNA Damage of the Plant Cells Treated with $\gamma$-Radiation (마 추출물이 방사선처리 식물세포의 생장과 핵 DNA 손상에 미치는 영향)

  • This study was carried out to evaluate the effects of yam (Dioscorea batatas Dence) extracts on the cell viability, growth and nucleus-DNA damage of tobacco cells which were exposed to $\gamma$-radiation stress. The viability and growth of tobacco cells exposed to 20 Gy of radiation stress were effectively recovered by pretreatment of 10 mg/L ethylacetate (EtOAc) yam extract. Pretreatment of EtOAc extract showed 20% higher cell viability and fresh weight growth than that of cells without pretreatment in 20 Gy radiation treated tobacco cells. Nucleus-DNA damage was measured as the ratio of tail length (T) to head length (H) in individual comet image isolated from tobacco cells. The T/H ratio of control-cells and treated-cells at 20 Gy were 1.05 and 1.68, and % head DNA of those cell were 86.7 and 71.3%, respectively, suggesting that nuclei of tobacco cells were severely damaged in the integrity of DNA by the treatment of $\gamma$-radiation. However, pretreatment of MeOH, EtOAc and n-BuOH extracts decreased radiation induced DNA-damage in the tobacco cells, showing T/H ratio of 1.37, 1.01 and 1.10 and % head DNA of 81.5, 87.6 and 88.7%, respectively.

Anti-coagulation and Platelet Aggregation Inhibitory Activities of the Ethanol Extract of Aerial Bulbils of Dioscorea alata L. (열대마 영여자 추출물의 항응고 및 혈소판 응집저해 활성)

  • Dioscorea alata L, commonly known as "tropical yam" is the most widely consumed yam species among the 650 yam species belonging to the Dioscoreacea family. It is extensively cultivated in tropical and subtropical regions and is a major food source in Africa and India. Also, it is used for medicinal purposes, particularly in China and Taiwan, for its anti-inflammatory properties. In comparison to other yam varieties such as D. batatas or D. opposita, the tropical yam has gained popularity in Korea due to its higher yield per unit area. In this study, the nutritional characteristics and anti-thrombosis activity of the aerial bulbils of D. alata L. tropical yam were compared to those of D. opposita. The results showed that the aerial bulbils of tropical yam exhibited nutritional characteristics and potent anticoagulant activity compared to those of domestic yam varieties. The bulbils extract of tropical yam showed superior anticoagulant activities against thrombin, prothrombin and blood coagulation factors. Furthermore, the bulbils extract of tropical yam exhibited strong platelet aggregation inhibition at 0.25 mg/ml and showed no hemolytic activity up to a concentration of 2.5 mg/ml. These findings suggest the potential development of high-value anti-thrombosis agents utilizing the aerial bulbils of tropical yam.

Antioxidant Activities and Monacolin K Production of Fermented Chinese Yam by Monascus sp. MK805 (Monascus sp. MK805 균주를 이용한 발효마의 Monacolin K 생산과 항산화 활성)

  • This study was carried out to investigate the characteristics of fermented chinese yam and rice (Dioscorea japonica, Dioscorea batatas, Dioscorea opposita, Rice, FCYR) using Monascus sp. MK805. The extracts from FCYR were measured to examine pigments, antioxidant activities were investigated through DPPH radical scavenging activity, total polyphenol and flavonoid contents, reducing power. Also it was investigated monacolin K productivity by FCYR. Pigments productivity (yellow, orange and red) were 26.2, 13.9, 17.3 at Dioscorea japonica, 41.9, 22.6, 53.2 at Dioscorea batatas, 12.5, 7.5, 9.7 at Dioscorea opposita and 10.1, 7.7, 10.2 at rice, respectively. DPPH radical scavenging activity of FCYR was about 69.7, 79.6, 57.8, and 42.3%, total polyphenol contents of FCYR was about 480.6, 658.7, 379.3, and 212.9 mg/kg, total flavonoid contents of FCYR was about 342.5, 448.4, 235.2, and 168.7 mg/kg, reducing power of FCYR was about 1.57, 2.14, 1.14, and 0.35 (OD at 700 nm), respectively. And then monacolin K productivity of FCYR was about 453.8, 509.5, 332.2, and 263.2 mg/kg, respectively.

Inhibitory Effect of Yam (Dioscorea batatas DECENE) Extracts on the Mutagenicity (돌연 변이원에 대한 마(Dioscorea batatas DECENE)추출물의 억제 효과)

  • The inhibitory effects of methanol, ethanol, chloroform/ methanol and water extracts from natural and cultural yams on the mutagenicity in the cooked pork (broiled and panfried) and the chemically induced mutagen, sodium azide, benzo(a)pyrene and 2-aminofluorene were investigated using salmolla typhimurium TA 100. In the presence of the S9 mixture, ethanol extract from natural yam showed high inhibitory effect on the mutagenicity of the cooked pork. But benzo(a)pyrene, supposed to be produced in mutagen during cooking, did not show high inhibitory effect in same extract. Besids, the yam extract on the mutagenicity of the sodium azide without S9 mixture showed low inhibitory effect. However 2-aminoflourene with S9 mixture showed high inhibitory effect, 91.5%.
