포공영(浦公英)의 항산화 및 화장품약리학적 활성 연구 (Study on the Anti-oxidant and Cosmeceutical Activity of Taraxacum platycarpum)

  • Objectives : In this study, anti-oxidant activity of Taraxacum platycarpum is confirmed to investigate cosmeceutical activity for utilization as a cosmetic ingredients. Methods : In the anti-oxidant and whitening effect experiment, the electron donating ability of Taraxacum platycarpum extracts, their SOD-like activity, and the inhibitory activity of xanthine oxidase, tyrosinase were examined. Results : Radical-scavenging activity of water and ethanol extract was examined using ${\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl$ (DPPH). Water and ethanol extract from Taraxacum platycarpum showed 62.9% and 54.7% at 500 ppm concentration in DPPH radical inhibition, respectively. In the test of superoxide dismutase (SOD)-like activity, water extract showed 63.4% at 1,000 ppm concentration, while ethanol extract showed 49% at 1,000 ppm concentration. In the test of xanthine oxidase inhibition, ethanol extract showed 66.8% at 1,000 ppm concentration. Tyrosinase inhibition effect related to whitening effect showed 36.3% and 54.2% in ethanol extract and water extract at 1,000 ppm concentration, respectively. Conclusion : According to these results, it is possible that the extract of Taraxacum platycarpum can be used as a new natural material of cosmetic industry.

도화(桃花, Pruni persicae Flos) 분획물의 항산화 및 항염증 효과 (Antioxidant and Anti-inflammatory Effects of Fractions from Pruni persicae Flos)

  • Objectives : The solvent extracts of Pruni persicae Flos were investigated for the activities of anti-oxidant and anti-inflammatory effects to apply as a functional ingredient for cosmetic products. Methods : In this study, the fractions of P. persicae Flos were extracted with 70.0% acetone and purified using Sephadex LH-20 column chromatography. As a result, eight fractions were isolated. We performed MTT assay, total polyphenol contents, DPPH free radical scavenging assay, SOD-like activity, xanthine oxidase inhibition assay, astringent activity assay, hyaluronidase inhibition assay and the production of nitric oxide. Results : For anti-oxidant effects, the electron donating ability of fraction (Fr.) 2-5, Fr.-8 isolated from P. persicae Flos was above 90.0% at 100 ppm respectively. The superoxide dismutase (SOD) - like activity of Fr.-5 isolated from P. persicae Flos was 92.1% at 1,000 ppm. The xanthine oxidase inhibitory effect of Fr.-6 isolated from P. persicae Flos was about 83.3% at 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 94.0% for Fr.-4 isolated from P. persicae Flos at 500 ppm. In the anti-inflammation effect, the Fr.-4 isolated from P. persicae Flos inhibited the generation of nitric oxide. Conclusions : All these findings suggested that the fractions of P. persicae Flos has a great potential as a cosmeceutical ingredient with a anti-oxidant and anti-inflammatory effects.

율피의 생리활성 몇 미백효과를 이용한 화장품신소재에 관한 연구 (A Study on the Application of New Cosmetic Materials of Whitening Effect and the Physiological Activities of Chestnut Inner Shell)

  • Objectives : This is the study of the application as the ingredients of cosmetics through the examination of the function for physiological activity of Chestnut inner shell. Methods : Chestnut inner shell, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect, nitric oxide inhibition have been investigated in the physiological activity measurement of function experiment. Results : We used BHA and kogic acid for the comparative. As a result of testing electron donating ability, at over 100ppm of water extract and ethanol extract, BHA showed relatively high donating ability by more than 90%. And as a result of measuring SOD like activity, 1000ppm of water extract showed an effect of 30% and ethanol extract showed an effect of 40%, BHA showed an effect of 30%. In the xanthine oxidase inhibition test, 1000ppm of water extract showed an effect of 70% and ethanol extract showed an effect of 63%, BHA showed an effect of 100%. In the tyrosinase inhibition test, 1000ppm of water extract showed an effect of 55% and ethanol extract showed an effect of 87%, Kogic acid showed an effect of 98%. In the anti-inflammatory test, the water extract and ethanol extract inhibited the generation of nitric oxide. Conclusions : The results indicated that extract of Chestnut inner shell can be used as a natural ingredients with biological function in cosmetics ingredients.

공진단(拱辰丹)의 성분 분석 및 갱산화(坑酸化) 작용에 미치는 영향 (An analysis of the Gongjindan's ingredients and its efficacy on anti-oxidation)

  • Objectives : For the congenital feeble, the Gongjindan is useful medicine. The experiment is to estimate the value of the Gongjindan as therapeutic agent preventing against aging with an analysis of the ingredients and the bio-activating effects by enzymologic methods. Methods : General ingredients' of the Gongjindan's extract were analyzed first and the quantitative analysis of a reducing sugar, a soluble protein, an amino acid and minerals was made. The Gongjindan, which is extracted, concentrated, and freeze drying with water, ethanol and chloroform, have got applied for the experiment. The effects on electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, whitening effect have been investigated in the physiological activity measurement of function experiment. Results : The contained amino acid, in order of high amount, is Arginine, Alanine, Glutamic acid, Proline and the contained free amino acid is Glutamic acid, Leucine, Lysine, Phenylalanine. The derivative of free amino acid is ${\gamma}-Aminoisobutyric$ acid, Phosphoserine, Taurine. And the Gongjindan is containing 13 species of minerals in order of Ca>K>Na>Mg>Fe>AI>Mn. Then, to assure of the Gongjindan's capability of anti-oxidation, these following subjects-polyphenol, electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, tyrosinase inhibition- are analyzed and show high activity especially the most in ethanol extracts. Conclusion : With this analysis of ingredients, the Gongjindan is containing many materials functioning as anti-oxidation, anti-aging, anti-fatigue, neurotransmitter and immune agent. Moreover, In every water, ethanol, chloroform extracts, the Gongjindan's capability of anti-oxidation is confirmed so that we can apply to patients' treatment clinically.

판람근(Isatis tinctoria L., 板藍根)추출물의 항산화활성 및 화장품약리활성에 관한 연구 (Study on anti-oxidant and cosmeceutical activities of Isatis tinctoria L.)

  • Objective : In this study, anti-oxidant and cosmeceutical activities of Isatis tinctoria L. extracted from water, ethanol and supercritical fluid condition were confirmed to investigate cosmeceutical activities for utilization as cosmetic ingredient. Methods: Anti-oxidant and cosmeceutical activities were investigated by using electron donating ability, xanthine oxidase, tyrosinase, astringent effect. Result : Isatis tinctoria L. extracts by supercritical fluid, water and ethanol showed good electron donating ability which were 82.7%, 62.6% and 44.8% at the concentration at 1,000ppm, respectively. Xanthine oxidase activity related with purine metabolism was inhibited by ethanol extract about 52.3% at the concentration at 1,000ppm. Tyrosinase inhibition effects, by supercritical fluid extract, ethanol extract and water extract, were 83.3%, 52.9% and 41.2% respectively at 1,000ppm. In the measurement of astringent effect, supercritical fluid extract at the concentration to 5,000ppm showed 85.7% in related activity. The water extract showed 95.9% nitrite scavenging activity at 5,000ppm. Conclusion : According to these results, it is possible that the extract of Isatis tinctoria L. can be used as a new natural material of cosmetic industry.

원기생맥산(元氣生脈散)의 추출물별 항산화작용과 쥐의 운동피로 회복효과 (Effect of Wongisaengmaek-san Extract on Antioxidative and Anti-Fatigue Activity)

  • Objectives : This study aimed to evaluate the effect of Wongisaengmaek-san (extracted with water or 50% ethanol) on antioxidative activity and recovery of fatigue induced by weight-loaded forced swimming exercise. Methods : Antioxidant activity of Wongisaengmaek-san was evaluated in terms of total amount of polyphenol, 1.1-diphenyl-2-picrylhvdroxyl (DPPH) radical scavenging activity and xanthine oxidase inhibition. The anti-fatigue effect of Wongisaengmaek-san was investigated using an acute weight-loaded forced swimming test by monitoring swimming test times and blood biochemical parameters creatinine. BUN, lactic acid dehydrogenase (LDH), free fatty acid (FFA) and lactic acid (LA). Results : 1. 50% EE (ethanol extract) had 2.3 times higher amount of total polyphenol compared to water extract. 2. It was identified that 50% ethanol extract showed enhancement of xanthine oxidase inhibition and DPPH scavenging effect in vitro. 3. According to anti-fatigue effect in the scale of 5% weight loaded mouse swimming test, both water and 50% ethanol extract showed significant improvement of swimming time elongation respectively, and 50% ethanol extract induced a more positive result. 4. After swimming for 1% weight-loaded mouse. water extract and 50% ethanol showed significant anti-fatigue effect with manifestation of blood sampling among both of the intervention: 50% ethanol extract showed a greater result. Conclusions : 50% ethanol extract of Wongisaengmaek-san has more effective antioxidant activity and anti-fatigue than water extract.

Physiological Activities of Roots Extracts from Calystegia japonica

  • This study was investigated to evaluate the contents of polyphenols and flavonoids, and physiological activities of various extracts from Calystegia japonica roots for making good use of their functional materials. The roots of C. japonica were extracted with water (WE), ethanol (EE) and hot water (HWE) by different methods. Among these extracts, the highest extracting yield was 30.30% of HWE, while the highest contents of total polyphenols and flavonoids were 40.85% and 6.40% of WE, respectively. The nitrite scavenging abilities were ranged from 31.31% (HWE) to 37.46% (EE) at pH 1.2 and 1.0 mg/ml concentration. In the measurements of electron donating abilities, EE showed the highest effect as 91.83% at 0.3 mg/ml assay concentration, and the electron donating ability was decreased as the extract concentration was increased. In the results of superoxide dismutase (SOD)-like activity, HWE showed the highest effect as 7.15% at 1.0 mg/ml. The tyrosinase inhibition activities of WE and EE were 15.28% and 14.97%, respectively. The xanthine oxidase inhibitory effects were ranged from 97.50 to 99.28% at 1.0 mg/ml. These results indicate that C. japonica extract has a good antioxidant effects and could be useful for developing functional products.

Anti-inflammatory Activity of Flavonoids from Populus davidiana

  • An in vitro bioassay-guide revealed that the methanol (MeOH) extract of the stem bark of Populus davidiana showed considerable inhibitory activity against cyclooxygenase (COX-1, COX-2). Continuous phytochemical study of the MeOH extract of this plant led to the isolation of ten flavonoids; sakuranetin (1), rhamnocitrin (2), 7-O-methylaromadendrin (3), naringenin (4), eriodictyol (5), aromadendrin (6), kaempferol (7), neosakuranin (8), sakuranin (9) and sakurenetin-5,4'-di-${\beta}$-D-glucopyranoside (10). Their structures were identified on the basis of their physicochemical and spectroscopic analyses. The isolated compounds, 1-10, were tested for their inhibitory activities against COX-1 and COX-2. Compound 7 was found to have potent inhibitory effect on COX-1 and a moderate effect on COX-2, meanwhile, compounds 1-6 showed moderate inhibition against COX-1 only. Moreover, compounds 5-8 exhibited suppressive effects on xanthine oxidase (XO). These results may explain, in part, the traditional uses of P. davidiana in ethnomedicine.

The derivative of free amino acid is Phosphoserine, Carnocine, ${\gammer}$-Aminoisobutyric acid. And the Gyungokgo contains 14 species of minerals, K>Na>Ca>Mg>Fe>Al>Mn. Then, to assure of the Gyungokgo's anti-oxidation, these following subjects -polyphenol, electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, tyrosinase inhibation- are analyzed and show high activity especially the most in chloroform extracts, (every ingredients written by the order of high amount) Conclusions : The Gyungokgo contains many materials functioning as anti-oxidation, neurotransmitter, anti-fatigue and immune agent.

제주 자생 식물들의 항산화 활성 능력 검색 (Screening for Antioxidative Activity of Jeju Native Plants)

  • 본 연구는 제주 자생 식물들의 항산화 활성을 측정함으로써 천연항산화 소재로의 활용 가능성이 있는 유망 자원을 발굴하고자 하였다. 제주도에서 자생하는 식물 11종을 대상으로 총 폴리페놀 화합물 함량 측정 및 항산화 활성을 측정하였다. 항산화 기능이 좋은 식물 8종을 우선 선별하여 유기용매 분획을 실시하고 같은 실험을 농도별로 진행하여 가능성이 있는 유망 후보를 선별 하였다. DPPH free radical 소거 활성은 구실잣밤나무의 ethyl acetate 층과 애기달맞이의 buthanol 층에서 각각 IC50값이 1.6 ㎍/㎖, 2.4 ㎍/㎖로 좋은 활성을 나타내었다. Nitric oxide 생성 저해 활성은 밤나무의 ethyl acetate 층에서 강한 저해율 보였다. Xanthine oxidase의 억제 효과는 밤나무의 ethyl acetate 층에서 IC50 값이 16 ㎍/㎖로 우수한 활성을 나타내었다. Superoxide radical 소거 효과는 구실잣밤나무의 ethyl acetate 층에서 IC50 7 ㎍/㎖로 좋은 활성을 나타내었고, hydroxyl radical 소거 활성은 구실잣밤나무의 buthanol 층에서 76%로 우수한 활성을 나타냄을 확인 할 수 있었다. 밤나무를 비롯한 항산화 효과가 우수한 식물종에 대해 단일 물질에 대한 분리 작업과 함께 항염증이나 항암, 항노화 등의 실험이 더 깊이 있게 진행된다면 새로운 천연물 유래 생리 활성 물질로서 학술 및 산업적 활용이 가능할 것으로 기대된다.
