The effects of in vitro culture methods on morphological development and infectivity of Strongyloides venezuelensis filariform larvae ($L_3$) to rats were investigated. A significantly higher body length was observed in $L_3$ from filter paper culture ($597.3{\;}{\pm}{\;}32.2{\;}{\mu\textrm{m}}$) than those in fecal (($509.9{\;}{\pm}{\;}35.0{\;}{\mu\textrm{m}}$) and nutrient broth culture (503.3{\;}{\pm}{\;}31.0{\;}{\mu\textrm{m}}) (P<0.05). Larval infectivity was assessed by exposing rats to 1,000 $L_3$ from each culture and worms were recovered from the lungs and small intestines. Recovery rate of these worms did not show any significant difference. A significantly greater body length of adults was recorded in those corresponding to the $L_3$ harvested from filter paper (2,777.5{\;}{\pm}{\;}204.4{\;}{\mu\textrm{m}}$) and nutrient broth culture (($2.732.5{\;}{\pm}{\;}169.8{\;}{\mu\textrm{m}}$) than those corresponding to the $L_3$ obtained from fecal culture (($2.600.5{\;}{\pm}{\;}172.4{\;}{\mu\textrm{m}}$) (P<0.05). Although worm fecundity and EPG counts differed among culture methods but worm burdens and course of infection did not. These findings suggest that the methods of cultures have a significant effect on the morphological development of the larvae to the $L_3$ stage, but do not influence the infectivity to rats.
Eosinohil and IgE responses of interleukin IL(-5 transgenic and normal C3H/HeN mice were studied after experimental infection with Nippostrongylus brasiliensis 9Nb). Intestinal worms were recovered at day 5 post-infection (PI), and numbers of total white blood cells (WBC) and eosinophils, and total serum IgE and anti-hapten (dinitrophenyl)(DNP) specific IgE titers, were measured at days 0,14 and PI. IL-5 mice appeared resistant to Nb infection showing a significantly ower worm recovery rate than normal mice (P<0.05). Total WBC and eosinophil counts (/mm3) were significantly increased in Nb infected normal mice (p<0.05), but unchanged (total WBC) or decreased (eosinophils) in IL-5 mice at day 21 PI. The total serum IgE level remarkably increased in normal mice, but only a little in IL-5 mice at days 14 and 21 PI. Priming with DNP brought about more remarkable increases of the total and anti-DNP specific IgE in normal mice than in IL-5 mice. The results show that IL-5 mice are resistant to Nb infection, and that eosinophil and IgE responses in these mice are not augmented by N infection.
This study aimed to investigate the infection status, worm development, and phylogenetic characteristics of the intestinal trematode, Stellantchasmus falcatus. The metacercariae of S. falcatus were detected only in the half-beak (Dermogenus pusillus) out of the 4 fish species examined. Their prevalence was 90.0%, and the intensity of infection was 919 metacercariae on average. Worms were recovered from 33 (97.1%) of 34 chicks that were experimentally infected with 200 S. falcatus metacercariae each, and the average recovery rate was 43.0%. The body size and inner organs of S. falcatus quickly increased in the experimental chicks over days 1-2 post-infection (PI). In addition, ITS2 sequence data of this parasite were analyzed to examine the phylogenetic relationships with other trematodes using the UPGMA method. The results indicated that the ITS2 sequence data recorded from trematodes in the family Heterophyidae appeared to be monophyletic. This study concluded that D. pusillus serves as a compatible second intermediate host of S. falcatus in Thailand and that S. falcatus can develop rapidly in the experimental chicks. Data collected from this study can help to close the gap in knowledge regarding the epidemiology, biology, and phylogenetic characteristics of S. falcatus in Thailand.
In an attempt to determine the susceptibility of inbred golden hamsters(Mesocricetus auratus) to the experimental infection with Clonorchis sinensis, twenty-five hamsters were divided into 5 groups and administered orally 5, 10, 20, 30 or 50 metacercariae each. The hamsters were killed on the 45th day after infection. The adult flukes were recovered from all hamsters. The overall recovery rate for the fluke was 57.9% with the range of 48.4% to 92.0%. As the number of the metacercariae given increased, a porportionate decrease in the recovery rate was found, the prepatent period of the fluke in the hamster varied from 15 to 17 days, with the average of 16 days. No significant differences in the development of the flukes were observed. Egg production expressed by EPG, increased step by step as the time elapsed. However, a tendency of decreasing egg laying capacity was observed with the increase of worm burden. It is suggested that the hamster is a suitable final host of C. sinensis.
An experimental study was carried out to observe the susceptibility of several kinds of laboratory animals to Fibricola seoulensis infection, a diplostomatid fluke of mammals. The metacercariae were obtained from the viscera of the snakes, Natrix tigrina lateralis and 50~2,000 in number each was artificially fed to a total of 127 animals; albino rats, mice, dogs, cats, rabbits and chickens. After 3 days to 8 weeks the animals were sacrificed and the recovery rate of worms as well as their maturity was observed. The results are as follows: 1. The overall worm recovery rates throughout the experimental period was highest in albino rats (40.0%) followed by mice (33.9%), cats (20.9%), dogs (11.4%), rabbits (0.05%) and chickens (0%). However, the recovery rates in the same host decreased as infection progressed longer and variable by the amount of metacercariae given. 2. From albino rats and mice, the highest recovery rates were obtained in 1,000 and 200-metacercariae infection groups respectively, and it is considered that such amount should be the optimum dose for experimental infection of these animals. 3. The main location of F. seoulensis in experimental animals was small intestine especially the duodenum. 4. The maturity index (No. mature worms/ No. examined) was 100% in albino rats and mice, while only 22.7% or 0% in dogs or cats respectively. From the results, it is concluded that albino rats and mice are the most susceptible hosts for F. seoulensis infection among six kinds of laboratory animals examined.
The growth and developmental pattern of H. continua was observed after experimental infection of their metacercariae to chicks. The recovery rate of worms from the chicks at 1 to 28 days post-infection(PI) was 12.8% in average. The rate remained fairly high for early 4 days of infection but decreased thereafter rapidly till 28 days PI. Most of the nukes, 91.9%, were recovered from the ileum of the chicks. In metacercariae, genital organs such as the ovary, testes, seminal vesicle, seminal receptacle and genital sucker were recognizable. At one day PI Mehlis'gland appeared, and at 2 days follicular vitellaria were observed. At 3 days PI, eggs were formed in the uterine tubule and increased in number as the worm grew old. The worms reached $2,990{\;}{\mu\textrm{m}}$ in length and $525{\;}{\mu\textrm{m}}$ in width at 28 days PI. Genital organs developed rapidly in early stages of infection but slowly thereafter to 28 days Pl, whereas non-genital organs developed steadily through the infection period. It was proved by this experiment that chicks should be a moderately suitable final host of H. continua.
An experimental study was performed to observe the infectivlty of sparganum (plerocercold of Spirometra erinncel) treated by prazlquantel, gamma-irradiation and mechanical cutting. The spargana were obtained from the naturally Infected European grass snake, Rhabdophis tigrina, or from the expenmentally infected mice. A total of 83 mice (ICR straine were divided Into 3 experimental groups by the source of the damage, fed each UH 5 spargana, and sacnaced 1 month later for worm recovery. In the praziquantel group, the worms were incubated In the concentration of 10㎍/ml (control: Tyrode for 4 hours) for 0.5. 1.2 and 4 hours at 36℃. and fed to mice. The recovery rate from mice in prazlquantel group was not different from that (80%) of control poop and in the range of 76-100%. In the gamma-Irradiation group, the worms were irradiated by 10-1000 Gy with Csl37. The average recovery rates of 69-100% were not different from that of control up to 100 Gy. The rate was 56% under 150 Gy, and 5% by 1000 Gy. In the mechanical cutting group, the worms were cut at 0.5, 1, 2 and 3 m from the anterior end of the scolex. The average recovery rates In each group were 70-90% and that of control was 90%. The present folding suggests that the sparganum be highly resistant to prazlquantel, gamma- irradiation and mechanical cutting. The vitality center of the sparganum must be at the anterior end of Its scolex.
A survey of intestinal helminths was undertaken in riparian people in Xieng Khouang Province, Lao PDR. Fecal specimens were collected from 643 people (289 males and 354 females) residing in 4 districts (Nonghet, Kham, Phoukout, and Pek) and were examined by the Kato-Katz technique. The overall helminth egg positive rate was 41.2%, and hookworms revealed the highest prevalence (32.7%) followed by Trichuris trichiura (7.3%) and Ascaris lumbricoides (5.6%). The positive rate for small trematode eggs (STE), which may include Opisthorchis viverrini, heterophyids, and lecithodendriids, was 4.4%. For recovery of adult helminths, 12 STE or nematode/cestode egg-positive people were treated with 40 mg/kg praziquantel and 15 mg/kg pyrantel pamoate, and then purged. Mixed infections with 2 Haplorchis species (H. pumilio and H. taichui), Centrocestus formosanus, Opisthorchis viverrini, a species of cestode (Taenia saginata), and several species of nematodes including hookworms and Enterobius vermicularis were detected. The worm load for trematodes was the highest for H. pumilio with an average of 283.5 specimens per infected person followed by C. formosanus, H. taichui, and O. viverrini. The worm load for nematodes was the highest for hookworms (21.5/infected case) followed by E. vermicularis (3.2/infected case). The results revealed that the surveyed areas of Xieng Khouang Province, Lao PDR are endemic areas of various species of intestinal helminths. The STE found in the surveyed population were verified to be those of heterophyids, particularly H. pumilio.
The echinostomatid metacercariae encysted in the gill of the fresh water fish, Pseudorasbora larva were identified through obtaining adult worms after eBperimental infection to mice. In addition, a brief course of worm development and maturation was observed in this experimental host. The results were as follows: 1. The echinostomatid metacercariae were elliptical, golden yellow, 0.073∼0.078 mm long and 0.0541∼0.065 mm wide. Their head portions were characterized by the presence of a head crown armed with collar spines of total 24 in number and interrupted at the mid-dorsal side of the oral sucker. 2. The average rate of worm recovery froth 12 mice (on the 1-2lth postinfection days) was 19.4 % and the rate revealed no decrease in accordance with the increase of infection duration. The worms were collected chiefly from the lower part of the small intestine. 3. After the infection, their sexual maturation was attained in 5 days and their growth in size nearly completed in 7 days. The early growth curve of genital organs was S shape while that of nongenital organs was C form. In 5 day old worms, 1 or 2 eggs were found from their uteri and the stools of mice revealed echinostomatid eggs from the 5-6th postinfection day. 4. The 7 day old adult worms were ovoid in shape, 0.54-0.69 mm long and 0.29-0.34 mm wide, and characterized by a well developed head crown with 24 collar spines and vitelline follicles distributed from the acetabular level down to the posterior end of body. Based on these characters they were identified to be Echinochasmus japonicus Tanabe, 1926. From these results, it is verified that p. larva is one of the second intermediate hosts of 5. jatonicus in Korea.
The worm development and egg laying pattern of Echinostoma hortense (Trematoda; Echinosto-matidae) were studied in albino rats and the brief clinical course was observed in human volunteers. A total of 21 rats were infected with 20~69 metacercariae each and two humans were with 7 and 27 metacercariae, which were collected from the loaches. For recovery of worms, the rats were sacrificed at irregular intervals from the 6th to 150th day after infection and the human volunteers were treated with praziquantel and purged with magnesium salt on the 26~27th day. The stools of the rats and humans were examined for the eggs. The results were as follows: 1. The worm recovery rate from the rats was not affected by the increase of infection time but varied individually; 9.1~50.0% (31.1 % in average). From humans, 14.3% and 37.0% (32.4% in average) of challenged were recovered. 2. In the rats, it was revealed that the worms rapidly grew for the first 14 days to become 7.59mm in average length and 1.17mm in average width but the growth became much slower thereafter until the 150th day; 7.95mm in length on the 21 th day, 9.04mm on the 28th day, 10.21mm on the 49th day and 12. 62mm on the 150th day. During the early stage of infection, the growth of genital organs (male or female) was expressed as sigmoid curves whereas non-genital organs (such as suckers) was simply as straight lines. 3. The prepatent period of this fluke was 10~12 days in the rats and 16~17 days in men. After the start of oviposition, the egg production by the worms remarkably increased, reached maximum on the 32~33th day, followed by decrease thereafter. The maximum value of E.P.G./worm was 390. 4. The major subjective symptoms in human volunteers were abdominal pain and diarrhea during the early stage of infection. The results show that human is as susceptible as the rats to E. hortense infection and the amount of egg production in the rats is greatly affected by the age of worms.
