Mucosal mast cell (MMC) and goblet cell (GC) responses were observed in the small intestine of two strains of mice (BALBfc and C3H) infected with Weodiplostomum seouLehTe, and their roles in the host defense and worm expulsion were studied. From day 3 to 28 post-infection (Pl) with 200 metacercariae, the worm recovery rate from BALB/c mice was consistently and remarkably higher than that from C3H mice. In the duodenum of both strains of mice, the main habitat of the flukes, mastocytosis was pronounced on day 7 Pl but quickly diminished thereafter. Similar kinetics were observed in the jejunum and ileum, although the extent of mastocytosis was lesser in the ileum than other two areas. These MMC kinetics were not different between the ko strains of mice. Moreover, the extent of mastocytosis was stronger in BALB/C mice than in C3H mice. GC hyperplasia was remarkable in the duodenum of BALBlc mice throughout the course of infection except day 14 Pl, whereas it was recognizable only in the jejunum and ileum of C3H mice on day 7 Pl. Mucin activation was evidently demonstrated in both strains of mice throughout the course of infection, but more marked in BALB/c than in C3H mice. The results strongly suggest that mastocytosis and GC hyperplasia are local immune responses against N. seoulense, however, they play a minor role in the host defense and worm expulsion.
Neodiplostomum seoulensis, one of the human intestinal trematodes , was reinfected to albino rats, and worm recovery rates, histopathology and activity changes of the intestinal brush border membrane bound enzymes were observed. The experimental groups were three: uninfected, primary infection and reinfection. The worm recovery rate in the reinfection group was much lower than in the primary infection group 14 days after infection. The duodenal histopathology showed villous atrophy during the first and second week in the primary infection group. In the reinfection group, however. villous changes occurred as early as 3 days after the infection, and the lesion was found healed 7 days after infection. The activities of alkaline phosphatase and sucrase in the duodenum of primary infection rats decreased nearly half of the controls 2 weeks after infection, whereas the activities were unchanged in the reinfection group. However, no changes in the activities were observed in the proximal jejunum between the experimental groups. These findings suggested that a secondary infection of N. seouLensis in rats should make less damage on the intestinal mucosa than a primary infection. Susceptibilities of 5 different mice strains, including C3H/HeN, BALB/c, C57BL6, FvB and ICR, to Echinostoma hortense infection, was evaluated. The worm expulsion rate, worm size and egg production were observed from 1 to 8 weeks after infection with 30 metacercariae. C3H/HeN and ICR mice showed the highest worm maturation rates. The worm recovery rate and the number of eggs per gram (EPG) of feces was also higher in C3H/HeN and ICR mice than in BALB/c, C57BL6, and FvB mice. It is suggested that E. hortense is highly infectious to ICR and C3H/HeN mice, but not to the other strains of mice. Based on the results obtained, we believe that the susceptibility of different mouse strains to E. hortense infection is dependent on the genetic and immunologic back-ground of mice.
To determine the infectivlty and maturity of nletacercanae of Parqgonimur westermani after keeping at low temperature for a long period, 45 mice and 45 rats were each infected with 20 metacercariae which were kept at $4^{\circ}C$ for 8 to 234 days. The worm recovery in mice increased with age of worm and reached a peak of 32% at 41-50 days and then decreased with age. The rate In rats first decreased to a lowest point of 6% at 71-100 days and then Increased with age. In 42 infected mice and 41 infected rats, 187 immature worms (183 tiny and 4 Juvenile ones) and 190 worms (164 tiny, 19 Juvenile and 7 mature ones) were recovered respectively. Two wormcysts with eggs only and 8 empty wormcysts were also found In the rats. In addition, the frozen metacercanae can still develop to mature worms in SD rats.
The effects of anti-allergic drugs on intestinal mastocytosis and the expulsion of Neodiplostomum seoulense were observed in Sprague-Dawley rats, after oral infection with 500 metacercariae. The drugs used were hydroxyzine (a histamine receptor H$_1$ blocker), cimetidine (a H$_2$ blocker), cyclosporin-A (a helper T-cell suppressant), and prednisolone (a T- and B-cell suppressant). Infected, but untreated controls, and uninfected controls, were prepared. Worm recovery rate and intestinal mastocytosis were measured on weeks 1, 2, 3, 5, and 7 post-infection. Compared with the infected controls, worm expulsion was significantly (P < 0.05) delayed in hydroxyzine- and cimetidine-treated rats, despite mastocytosis being equally marked in the duodenum of all three groups. In the cyclosporin-A- and prednisolone-treated groups, mastocytosis was suppressed, but worm expulsion was only slightly delayed, without statistical significance. Our results suggest that binding of histamine to its receptors on intestinal smooth muscles is more important in terms of the expulsion of N. seoulense from rats than the levels of histamine alone, or mastocytosis.
The metacercariae of Echinostoma cineterchis (Trematoda: Echinostomatidae) were found infected in the loaches (Misgurnus anguillicaudatus) purchased from two local markets in Korea. Identification of the metacercariae was done after obtaining adult worms from experimentally infected albino rats. Brief course of worm development in rats was also observed up to 28th post-infection day. The results are as follows: 1. The metacercariae of E. cinetorchis were detected from 5 (10.9%) out of 46 loaches examined and the average metacercarial burden per infected loach was 34.4. They were always found from the posterior abdominal wall. 2. The worm recovery rate from 8 rats was 24.6% in average. It was observed that the rate decreased according to age of infection; from 40.7% after 6 days to 15.0% after 28 days. 3. The adult worms were morphologically characterized by the presence of head crown with derfally uninterrupted collar spines, 37-38 in number, and the tendency of migration or disappearance of testes. So that they were identified as Echinostoma cinetorchis Ando et Ozaki, 1923. 4. The development of worm was very rapid during 6-10 days after infection and became much slower thereafter. During the rapid worm growth the development of genital organs was more prominent than that of non-genital organs. From the results it is concluded that the loach is involved in the life cycle of E. cinetorchis in this country and may take a role for infection source to animals and man. It is also confirmed that migration or loss of testes is the distinct specific character of this echinostomatid fluke.
The growth and development of Gymnophalloides seoi were studied in C3H/HeN mice and effects of immunosuppression of the host on the worm development were observed. Two hundred metacercariae of G. seoi were orally administered to each mouse, and worms were recovered on days 1, 3, 5, 7, 14 and 21 post-infection (PI). The worm recovery rate was significantly higher in immunosuppressed (ImSP) mice than in immunocompetent (ImCT) mice except on days 1 and 3 PI. The worms attained sexual maturity by day 3 PI with eggs in the uterus, and worm dimensions and the number of uterine eggs continuously increased until day 14 PI in ImSP mice. Worms recovered from ImSP mice were significantly larger in size than those from ImCT mice on days 1 and 3 PI, and the number of uterine eggs was significantly larger in ImSP mice on days 5 and 7 PI. Genital organs such as the ovary, testes, and vitellaria, that were already developed in the metacercarial stage. grew a little in size until day 14 PI. The results show that the C3H/HeN mouse is, though not excellent, a suitable laboratory host for G. seoi.
The complete life cycle of Echinostoma hertense has been maintained in the laboratory, using Lymnaea persia snails and Rana nigromaculata tadpoles as the first and second intermediate hosts. ICR mice was used as the definitive host. Within the egg of 5. hotense, the miracidium was fully matured in 13 days of incubation at $29~30^{\circ}C$. The miracidium was $93.8{\times}53.6{\;}{\mu\textrm{m}}$ in average size, covered with numerous cilia of $7~11{\;}{\mu\textrm{m}}$ length. The epidermal plates were arranged in 6-8-4-2 formula. The first generation rediae ($1.19{\times}0.27{\;}mm$ in average size) were observed in 14 days after miracidial challenge to the snails, and the second generation rediae ($1.40{\times}0.26{\;}mm$ in average size) in 30 days. The average sixte of the cercaria was $295.5{\times}145.0{\;}{\mu\textrm{m}}$. Their head crown was poorly developed, and collar spines were not yet observed. After a cercarial challenge to the tadpoles, all of the tadpoles became infected and the average worm recovery rate was 88.5%. The majority of the metacercariae (75.5%) were recovered from the muscle of the tadpole's posterior body and the rest (24.3%) from their gills. The metacercariae from the tadpoles were elliptical, and $167.7{\times}129.9{\;}{\mu\textrm{m}}$ in average size. The recovery rate of adults from the mice was difFerent by the age of the metacercariae grown in the tadpoles. The metacercariae younger than 5 hrs could not infect mice whereas those older than 6 hrs could infect mice. The recovery rate became higher as the metacercaria matured, with the peak recovery rate of 90.0 % at the metacercarial age of 9 days. Thereafter the recovery rate decreased to 55.0% at the age of 50 days. As shown by the above results, the whole life cycle of E. hcrtense has been completed in the laboratory. At least 55~58 days were required to maintain one egg-to-egg cycle of E. hortense.
We investigated the induction of resistance to Clonorchis sinensis infection by prior infection in rat and hamster models. Animals were challenged with C. sinensis metacercariae, then treated with praziquantel and reinfected. Worm recovery rate in reinfected animals was used to estimate resistance to reinfection. The determined resistance rates to reinfection in rats and hamsters were 97.7% and 10.3%, respectively. In rats, cure from the primary infection of C. sinensis increased resistant to reinfection, and the greatert the worm burden and the longer the duration of primary infection, the higher was the resistance rate. For primary infection doses of 10, 40 and 100 metacercariae per rat, the resistance rates were 87.4%, 93.8% and 98.4%, respectively. The resistance rates in rats after 2 or 8-week primary infection were 78.7% and 95.3%, respectively. All worms recovered from reinfected rats were immature. When cured rats were administered with methylprednisolone, resistance to reinfection became impaired. These findings indicate that rats develop a high degree of resistance to reinfection by C. sinensis after cure. The growths and maturations of reinfected worms were also impaired.
Worm recovery rates and pathologic changes in small intestine of albino rats were observed after concurrent and challenge infections with metacercariae (MCI of Fibricota seouLensis and MetaBonimus vokogaupci, and compared with those of single Infection groups. Albino rats In concurrent Infection group were killed 20 days after feeding with 1,000 MC of each fluke. Rats in challenge. infection group were fed with 1.000 MC of f seouleni,s and challenged by 1,000 MC of M. yokogawai 10 days after primary infection, then killed 10 days thereafter. In concurrent infection group, mean number of F. seoulensis and M. yokogawai recovered, 250 and 118 respectively, were similar to those of single infection groups. However, more flukes were collected from the duodenum and less flukes were from the ileum than from single infection group. In challenge Infection group, the recovery rate of F. seoulensis was similar to that of sin91e infection group and the distribution of thIn flukes was similar to that of concurrent infection group. Mean number of M. yokogculat, 69, was signiflcantly lower than that of single infection group. Its distribution, however, extended to the duodenum and most of the flukes were recovered from the jejunum. In concurrent infection group, villi of the duodenum were more markely thickened, fused and shortened than those in F. seoulensis sing1e Infection group. The crypt epithelium appeared to be hyperplastic and inflammatory cell infiltration into the villous stroma was mild. Villous atrophy in the jrlunum and ileum was ndlder than in M. yokogawai single infection group. In challenge Infection group, the fhldlngs were simuar to those of concurrent Infection group. Crypt epithelium hyperplasla was not severe in duodenum. Inflammatory reaction was observed in submucosa of the Jrjunum and ileum. From the above results, it Is considered that F. seoulenis ikabiting in upper part of small intestine affect the settlement of M. yokoguwai introduced later In lower part of the intestine.
