Virulence of Xanthomonas translucens pv. poae Isolated from Poa annua

  • Bacterial wilt is a vascular wilt disease caused by Xanthomonas translucens pv. poae that infects Poa annua, a grass that is commonly found on golf course greens throughout the world. Bacterial wilt causes symptoms of etiolation, wilting, and foliar necrosis. The damage is most prevalent during the summer and the pathogen can kill turf under conditions optimal for disease development. Fifteen isolates of X. translucens pv. poae were collected from northern regions in the United States and tested for virulence against P. annua. All 15 isolates were pathogenic on P. annua, but demonstrated variable levels of virulence when inoculated onto P. annua under greenhouse conditions. The isolates were divided into two virulence groups. The first group containing four isolates generally resulted in less than 40% mortality following inoculation. The second group, containing the other eleven isolates, produced between 90 and 100% mortality following inoculation. These results suggest that differences in the virulence of bacterial populations present on a golf course may result in more or less severe amounts of observed disease.

Occurrence of Sclerotium Rot in Allium tuberosum Caused by Sclerotium rolfsii in Korea

  • In this study, we characterized sporadically occurring sclerotium rot caused by Sclerotium rolfsii in Chinese chive (Allium tuberosum Roth.) in farm fields in Sacheon, Korea. The initial symptom of the disease was water-soaked, which progressed to rotting, wilting, blighting, and eventually death. Further, mycelial mats spread over the lesions near the soil line, and sclerotia formed on the scaly stem and leaves. The sclerotia were globoid, 1~3 mm, and white to brown. The optimum temperature for growth and sclerotia formation on potato dextrose agar (PDA) was $30^{\circ}C$. The diameter of the hypae ranged from 4 to 8 ${\mu}m$. Clamp connection was observed on PDA medium after 5 days of incubation. Based on the mycological characteristics, internal transcribed spacer sequence analysis, and pathogenicity test, the causal agent was identified as Sclerotium rolfsii Saccardo. This is the first report of sclerotium rot in Chinese chive caused by S. rolfsii in Korea.

수확시기에 따른 조생온주 밀감의 품질과 수상저장 효과 (Effect of Stroage on the Tree and Quality of Early Variety of Citrus unshiu Produced in Cheju According to Harvest Time)

  • Effect of storage on the tree and quality of early cultivar of Citrus unshiu Marc. produced in Cheju according to harvest time were investigated. Soluble solids, acid content, flesh ratio, firmness and specific cavity of the fruits harvested at 27th of November were 12.0, 1.03%, 79.98%, 0.774kg-force, 0.912, respectively. Soluble solids, total sugar and reducing sugar were increased gradually, but acid content was decreased slightly as delayed in harvest time. Otherwise specific gravity and firmness were not shown in great during harvest time, color index(a/b) was increased sharply till late November and then remained in stationary phase. On these result, we recommend that the harvest time of early cultivar of Citrus unshiu Marc. would be during December because of wilting partly. Fully ripened-fruits harvested late, compared to ordinary harvest time, with cold storage would be kept more quality and freshness of fruits for fresh consumption than the fruits harvested early and stored at room temperature.

Acacia melanoxylon 잎 메탄올 추출액의 생물활성 (The Bioefficacy of Methanol Crude Leaf Extract of Acacia melanoxylon)

  • The methanolic crude leaf extract of Acacia melanoxylon shows inhibitory effects on seed germination, shoot growth, root growth and biomass of black gram (Phaseolus mungo L. Co 1) and weeds (Cassia occidentalis L. and Cyperus rotundus L.). The post emergence treatment was given on the 8th day after sowing and 4-6 days after seedling emergence. After postemergence treatment with 20% extract concentration, wilting symptoms appeared in P. mungo and C. occidentalis while necrosis occurred in C. rotundus. The seedling growth was inhibitory at all concentrations (5%, 10%, 20%) as compared to control plants. This inhibitory effect may be due to the presence of allelochemicals like phenolics in the A. melanoxylon leaves. Different types of phenolic acids were identified.

Stem Rot of Garlic (Allium sativum) Caused by Sclerotium rolfsii

  • Stem rot disease was found in garlic (Allium sativum L.) cultivated from 2008 to 2010 in the vegetable gardens of some farmers in Geumsan-myon, Jinju City, Gyeongnam province in Korea. The initial symptoms of the disease were typical water-soaked spots, which progressed to rotting, wilting, blighting, and eventually death. White mycelial mats had spread over the lesions near the soil line, and sclerotia had formed over the mycelial mats on the stem. The sclerotia were globoid in shape, 1~3 mm in size, and tan to brown in color. The optimum temperature for growth and sclerotia formation on potato dextrose agar (PDA) medium was $30^{\circ}C$. The diameter of the hyphae ranged from approximately 4 to $8\;{\mu}m$. Typical clamp connection structures were observed in the hyphae of the fungus, which was grown on PDA medium for 4 days. On the basis of the mycological characteristics and pathogenicity of the fungus on the host plants, the causal agent was identified as Sclerotium rolfsii Saccardo. This is the first report of stem rot disease in garlic caused by S. rolfsii in Korea.

Extracellular Polymeric Substances of Pseudomonas chlororaphis O6 Induce Systemic Drought Tolerance in Plants

  • Pseudomonas chlororaphis O6 induces systemic tolerance in plants against drought stress. A volatile, 2R, 3R-butanediol, produced by the bacterium causes partial stomatal closure, thus, limiting water loss from the plant. In this study, we report that applications of extracellular polymeric substances (EPS) from P. chlororaphis O6 to epidermal peels of leaves of Arabidopsis thaliana also reduce the size of stomatal openings. Growth of A. thaliana seedlings with applications of the EPS from P. chlororaphis O6 reduced the extent of wilting when water was withheld from the plants. Fluorescence measurements showed photosystem II was protected in the A. thaliana leaves in the water stressed EPS-exposed plants. These findings indicate that P. chlororaphis O6 has redundancy in traits associated with induction of mechanisms to limit water stress in plants.

First Report of Macrophomina phaseolina Causing Charcoal Rot in Bottle Gourd in Korea

  • Stem blight symptoms were observed in greenhouse-grown bottle gourds (Lagenaria siceraria) in Wanju, Korea. Infected plants showed leaf chlorosis and wilting; the plants eventually died with the infected leaves remaining attached, and black sclerotia formed on the diseased stem. Based on the morphological characteristics and pathogenicity results and sequence analyses of the internal transcribed spacer, translation elongation factor 1-α, β-tubulin, and calmodulin regions, the isolated fungus was Macrophomina phaseolina. This is the first report of charcoal rot in Lagenaria siceraria caused by Macrophomina phaseolina in Korea.

Occurrence of Fusarium Wilt in Basil Caused by Fusarium oxysporum in Korea

  • Wilt symptoms were observed in basil (Ocimum basilicum) plants grown in a vinyl greenhouse located in Gokseong, Korea, during crop disease surveys conducted in August 2022. The symptoms appeared as wilting of the plants and brown to dark brown longitudinal streaks on the stems at or above the soil line. The disease incidence among the plants in the vinyl greenhouse was 5-20%. Six isolates of Fusarium sp. were obtained from stem lesions and identified as Fusarium oxysporum species complex based on their morphological characteristics. Among the isolates, two were used for phylogenetic analysis and pathogenicity test. Phylogenetic analysis revealed that these isolates belonged to F. oxysporum. Pathogenicity of the isolates was confirmed through artificial inoculation test. The symptoms induced by the isolates were similar to those observed in basil plants in the investigated vinyl greenhouse. This is the first report of F. oxysporum causing Fusarium wilt in basil in Korea.

고랭지 배추재배를 위한 뿌리혹병 저항성 품종선발 (Screening of Resistant Chinese Cabbage Cultivar against Clubroot (Plasmodiophora brassicae) for Cultivation in Highland)

  • 본 연구는 53의 배추품종 및 계통에 대한 고랭지재배에 적합한 품종을 선발하기 위하여 원예적 특성이 우수하면서 십자화과 작물 재배지역에서 심각하게 발생하는 배추 뿌리혹병에 저항성인 배추를 선발하기 위하여 실험을 수행하였다. 34개 품종의 배추와 19개 계통의 배추를 배추뿌리혹병원균으로 오염된 고랭지 포장에 정식하였다. 정식 30일 후에 내서성 및 지상부 뿌리혹병 발생에 의한 시들음 조사를 하였고, 정식 60일 후에 뿌리흑병 지하부 발생조사, 품종 및 계통 별 수량성을 알아보고자 생체중을 측정하였다. 정식 30일 후 내서성 및 뿌리혹병에 대한 지상부 시들음 조사에서 '불암플러스', '강력여름배추' 품종과 'SP-02', 'SP-05', 'CR DB 29', 'CR DB 50' 계통이 저항성이 있는 것으로 나타났다. 정식 60일 후 뿌리혹병 발병률에 대한 지하부 조사에서는 '대통 품종', 'Super CR Kimi 85 품종', 'super CR Hiraki' 품종이 뿌리혹병에 대한 저항성이 높은 것으로 조사되었다. 정식 60일 후 품종별 생체중 조사에서는 'Super CR Hiroki' 품종이 생체중 2,500 g이상으로 다른 품종에 비하여 월등히 좋았다. 배추정식 초기인, 정식 후 30일 지상부의 시들음 조사에서 저항성품종으로 조사된 성적과 정식 후 60일 뿌리혹병에 대한 지하부 저항성 조사 성적은 다소 차이가 있었다. 배추품종으로 'Super CR Hiraki', ' Super CR Kimi 85', 'CR 장군'이 배추 계통으로 'CR DB 29', 'CR DB 50'이 내서성 및 뿌리혹병 발생에도 저항성을 보였으며 아울러 원예학적 특성 중 생체중에 대해서도 윌등히 우수하여 강원도 고랭지 재배에 적합할 것으로 사료된다.

주요 사료작물의 곤포 Silage 조제이용에 관한 연구 II. 생육단계별 건물축적형태화 곤포사일리지 조제이용 (Study on Baled Silage Making of Selected Forage Crop and Pesture Grasses II. Yield performance and nutritieve evaluation of baled silage as affected by stage of growth)

  • 본 연구는 1991년부터 1993년까지 사료작물과 목초류 7종(호맥, 대맥, 연맥, Italian ryegrass, alfalfa, orchardgrass 및 혼과목초)을 공시재료로 하여 곤포사일리지(BS=baled silage making)조제이용을 위한 수확시기와 이때의 BS 생산성 및 품질을 평가하였다. 시험방법에 있어서 작물재배는 농촌진흥청의 사료작물 및 목초 표준경종방법에 준하여 실시하였다. 사일리지 조제작업중 BS는 자체적으로 제작한 각형 베일러를 이용 개체당 크기 90cm$\times$60cm$\times$50cm, 중량은 건물기준 20kg 내외, 곤포압착 21~23mm를 유지하여 조제하였다. 한편 고수분 재료는 수분정도에 따라 예건처리(무처리, 0.5일 및 1.0일)와 formic acid(무처리, 0.3% 및 0.6%)를 병행 처리하여 BS 를 조제하였다. 맨류작물의 BS 생산을 위한 수확적기는 대맥, 황숙기, 호맥과 연맥은 유숙기 이었으며 이때의 BS 수량은 각각 11.92톤(대맥), 12.64톤(호맥) 및 8.41톤/ha(연맥)이었다. Italian ryegrass 및 목초의 수확적기는 각 초종 모두 개화기로서 년간 BS 생산성은 Italian 13.81톤(2회 예취), 혼파목초 11.46톤(3회), alfalfa 10.62톤/ha(3회)이었다. 한편 조기수확에 따른 고수분 재료의 BS 품질은 무처리시의 35점에서 예건 및 FA 동시처리시에는 84점으로 향상되었다. Italian tyegrass의 BS품질도 무처리시의 61점에서 예건 및 FA 처리시에는 88점으로 개선되는 효과가 있었다.
