Inhibitive Effects of Edible Mushrooms Extracts on Pathogenic Bacteria and Proliferation of Cancer Cells (식용버섯 추출물의 식중독균 및 암세포 증식에 대한 저해 효과)

  • The antibacterial effect of methanol and water extracts from edible mushrooms on the growth of pathogenic bacteria (Bacillus cereus, Bacillus subtilis, Staphylococcus aureus, Echerichia coli O-157, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella typhi) were investigated. The Lyophyllum cinerascens and Pleurotus ostreatus2 methanol extracts treated with 5.0 mg/disc showed the highest antimicrobial activity against 7 kinds of pathogenic bacteria. And methanol extracts of edible mushrooms showed higher antimicrobial activity against gram positive and gram negative microorganisms than water extracts. The methanol extracts of mushrooms revealed high inhibitive activites in cytotoxicity on human cancer HepG2 and HT-29 cells. The growth of cancer HepG2 and HT 29 cells which treated with 1 mg/mL of Cordyceps militaris and Sarcodon aspratus methanol extracts were strongly inhibited to $67\%$ and $81\%$ respectively. And most of the methanol extracts exhibited the stronger effects against these cells, at the same concentration, comparing with water extracts. Particularly, the methanol and water extracts of Cordyceps militaris, Agaricus blazei, Lyophyllum ulmarium, Ganoderma lucidum and Sarcodon aspratus have the strongest antitumoral effects on HepG2 and HT-29 cells. From these results, it is considered that wild mushrooms have stronger antimicrobial and in vitro cytotoxic effects.

Studies on the Lipids Composition of Korean Edible Mushrooms (한국산 식용버섯의 지방질 성분에 관한 연구)

  • Three fractions-neutral lipids, glycolipids and phospholipids were obtained from total lipids in cultivated mushrooms(Pleurotus ostreatus and Pleurotus sajor-caju) and wild mushrooms (Ramaria botrytis and Lactarius volemus), and their lipid compositions and fatty acid compositions were determined by means of silicic acid column chromatography, thin layer chromatography and gas liquid chromatography. The total lipid contents in mushrooms were $0.32{\sim}0.39%$(wet basis). It was found that the contents of phospholipids$(19.4{\sim}47.4%)$ and neutral lipids$(32.1{\sim}51.9%)$ were high, while that of glycolipids$(14.8{\sim}28.7%)$ were low. The main lipid in neutral lipids was triglyceride$(21.2{\sim}38.2%)$ followed by free sterol$(21.0{\sim}21.9%)$, sterol ester$(10.3{\sim}18.6%)$, but the main lipid in neutral lipid of Pleurotus sajor-caju was free fatty acid(34.1%). The main lipid in glycolipids was steryl glycoside$(15.6{\sim}29.4%)$ followed by esterified steryl glycoside$(13.4{\sim}23.9%)$, monogalactosyl diglyceride$(15.6{\sim}24.6%)$. Among the phospholipids, phosphatidyl choline$(36.7{\sim}49.5%)$, phosphatidyl ethanolamine$(20.9{\sim}29.7%)$, phosphatidyl inositol$(18.4{\sim}26.1%)$ were the major components. The major fatty acid of total lipids was linoleic acid followed by palmitic acid, oleic acid. Fatty acid composition was not significantly different among total lipids, neutral lipids, glycolipids and phospholipids contained in tested mushrooms but the main fatty acid in neutral lipid of Ramaria botrytis was oleic acid(48.7%).

Distribution of mushrooms spontaneously growing in Naejangsan National Park (내장산국립공원의 자생버섯 분포상)

  • Mushrooms in Naejangsan National Park between May and September of 2021 have been surveyed. In this period, a total of 4 divisions, 9 classes, 25 orders, 72 families, 171 genera, and 381 species, including 3 climate-sensitive biological indicator species were found. The order in which the most diverse array of species was observed is Agaricales, which includes 24 families, 64 genera, and 170 species. Among these, the genus Russula was dominant, with 30 species, followed by the genus Amanita with 27 species. Among the 12 grids we investigated, species diversity was greatest in grid F5, in which 56 species of mushrooms were found. In particular, a large number of ectomycorrhizal mushrooms, including Russula spp. and Lactarius spp. were recognized. We presume that the gentle slopes and the low occurrence of Sasa borealis in this area may create a favorable environment for wild mushrooms. In corroboration, some grids (e.g. F6, F8, and F10) covering steep slopes and harboring large numbers of Sasa borealis contained only 19 species. Based on DNA sequence analysis, the NJ21064 was identified as Chlorophyllum hortense, which is newly recorded in Korea.

Studies on the Hemolytic Activities of Korean Wild Mushrooms (III) - Hemolytic Characteristics and in vivo Toxicity of Hemolysin of Hebelma crustuliniforme - (한국산 야생버섯의 용혈작용에 대한 연구 - 제 3보 : 무우자갈버섯(Hebeloma crustuliniforme) 용혈독소의 용혈특성 및 in vivo 독성 -)

  • In the previous studies, we surveyed 68 Korean wild mushrooms for their hemolytic activity and found that cold-water extract of Hebeloma crustuliniforme contained heat-resistant hemolysin. In this study, partially purified hemolysin of the mushroom was obtained by cold-water extraction followed by precipitation with ammonium sulfate, solubility fractionation and then dialysis. The hemolysin was found to be > 12,000 in molecular weight and its optimal hemolytic temperature was $37^{\circ}C$ and it's hemolytic activity, on washed erythrocytes and unwashed erythrocytes, respectively, was in the order of sheep > rat > human ${\geq}$ mouse > chicken and sheep > mouse > human ${\geq}$ rat > chicken. When ip injected into ICR mice at 1.38 mg/kg, it incurred prompt hemolysis as well as severe renal toxicity and hepatotoxicity. These results strongly suggest that the toxicity of Hebeloma crustuliniforme, which had been well-known as a toxic wild mushroom, may be at least partly due to its hemolysin.

Status of Radiation Dose and Radioactive Contamination due to the Fukushima Accident

  • Backgrounds: The accident at Fukushima Daiichi Nuclear Power Plant (NPP), March 2011, caused serious radioactive contamination over wide area in east Japan. Therefore, it is important to know the effect of the accident and the status of NPP. Materials and Methods: This paper provides a review on the status of radiation dose and radioactive contamination caused by the accident on the basis of publicized information. Results and Discussion: Monitoring of radiation dose and exposure dose of residents has been conducted extensively by the governments and various organizations. The effective dose of general residents due to the accident proved to be less than a mSv both for external and internal dose. The equivalent committed dose of thyroid was evaluated to be a few mSv in mean value and less than 50 mSv even for children. Monitoring of radioactivity concentration has been carried out on food ingredients, milk and tap water, and actual meal. These studies indicated the percentage of foods above the regulation standard was over 10% in 2011 but decreasing steadily with time. The internal dose due to foods proved to be tens of ${\mu}Sv$ and much less than that due to natural $^{40}K$ even in the Fukushima area and decreasing steadily, although high level concentration is still observed in wild plants, wild mushrooms, animals and some kind of fishes. Conclusion: According to extensive studies, not only the effect of the accident but also the pathway and countermeasures against radioactive contamination have been revealed, and they are applied very effectively for restoration of environment and reconstruction of the area.

Fibrinolytic and α-Glucosidase Inhibitory Activities of Wild Mushroom Methanol Extracts (야생버섯 메탄올추출물의 혈전용해 활성과 α-Glucosidase 저해 활성)

  • The aim of this study was to investigate the physiological functionalities of 60 wild mushroom methanol extracts. We determined their fibrinolytic and ${\alpha}-glucosidase$ inhibitory activities. Among them, 11 mushrooms showed fibrinolytic activity. In particular, Armillaria sp. showed the greatest enzyme activity (4.2 plasmin units/ml) in a fibrin plate assay. The fibrinolytic activities of Amanita virosa. Gymnophilus sp. and Lepista nuda were 2.3, and the activities of Amanita citrina and Cortinarius alboviolaceus were 2.1 and 2.0 plasmin units, respectively. In a ${\alpha}-glucosidase$ inhibitory activity test, Naematoloma fasciculare showed the greatest inhibitory activity at 98.5%. The ${\alpha}-glucosidase$ inhibitory activities of Suillus luteus and Gomphidius maculatus were 66.6% and 61.5%, respectively. This study suggests that both Armillaria sp. and N. fasciculare can be used as candidates for the development of new medicine effective to cardiovascular diseases and biofunctional food.

Genotyping of the Wild Mushroom Lentinula edodes from Mt. Jungwang and Mt. Gariwang in Korea

  • Lentinula edodes is an edible wild mushroom that can be found in mountainous regions of the Korean peninsula. Wild oak mushrooms were collected from Quercus mongolica at an elevation of more than 1,000 m on Mt. Jungwang and Mt. Gariwang in Gangwon province. We examined 10 oak mushroom strains to evaluate the genetic similarity among strains. Genetic similarity was determined based on the analysis of microsatellite markers (Led A2, Led A8, Led B2, Led B6, and Led D6) registered in the National Center for Biotechnology Information. We also performed dual culture tests on potato dextrose agar for 2 months at $25^{\circ}C$. The observed heterozygosity across all microsatellites ranged from 0.00 and 0.60 among 5 microsatellite markers, and the polymorphism information content values of Led A2, Led A8, Led B2, Led B6, and Led D6 were 0.0000, 0.8144, 0.6194, 0.4892, and 0.5702, respectively (mean value = 0.4987). Confrontation lines between strains were formed for almost all combinations. In conclusion, the oak mushroom populations of Mt. Jungwang and Mt. Gariwang have mixed gene pools. However, further studies are needed to identify genetic similarities and variations among these populations.

Chemical Constituents of Gymnopilus spectabilis and Their Antioxidant Activity

  • Gymnopilus spectabilis, a hallucinogenic mushroom belonging to the family Cortinariaceae, is found growing in dense clusters on stumps and logs of hardwoods and conifers. It contains the hallucinogenic alkaloid psilocybin and its strongly bitter taste makes it undesirable as an edible. In an effort to identify chemical constituents of Korean native wild mushrooms, 4,6-decadiyne-1,3,8-triol (1), ergosta-4,6,8(14), 22-tetraen-3-one (2), bisnoryangonin (3), and hispidin (4) were isolated from the methanolic extract of the fruiting bodies of G. spectabilis. Their structures were assigned on the basis of various spectroscopic studies. Compounds 3 and 4 displayed significant scavenging activity against the ABTS radical cation, DPPH radical, and superoxide radical anion, while 1 and 2 exhibited no antioxidant activity.

Antitumor and Immunomodulatory Activity of Lycoperdon pedicellatum (긴꼬리말불버섯 (Lycoperdon pedicellatum)의 항암 면역활성)

  • Protein-polysaccharide fractions separated from nine Korean wild mushrooms were subjected to an in vitro screening test for lymphoblastogenic activity. Of these, PPLP, the protein-polysaccharide fraction of Lycoperdon pedicellatum, showed the most potent activity and were further investigated for its antitumor activity. When intraperitoneally injected into ICR mice once daily for six days at a dose of 30 mg/kg, PPLP strongly inhibited the growth of sarcoma 180 tumor cells, showing the inhibition ratio of 97.6%. PPLP also showed in vitro inhibitory activity on sarcoma 180 or leukemia L1210 at the concentration of 500 $\mu\textrm{g}$/$m\ell$ or higher. These results strongly suggest that PPLP might exert its antitumor activity through immunostimulation as well as inhibitory activity on the tumor cells.

Morphological Characteristics of the Fruiting Bodies of Wild Lentinula edodes Strains Cultivated on Sawdust Blocks in Korea (국내에서 수집한 야생 표고의 사각 톱밥배지 재배에서 자실체의 형태적 특성)

  • Wild Lentinula edodes (shiitake) strains are an important breeding material for developing new varieties suitable for various consumer preferences and environmental conditions. The morphological characteristics of 100 wild shiitake strains collected from 14 regions were investigated in a sawdust block cultivation. The characteristics of their fruiting bodies were diverse. Among them, 38% of the caps was convex, 33% was broadly umbonate, 27% was applanate, and 2% was shallowly depressed. In terms of their stipes, 55, 23, and 22% were the column, clavate, and funnel type, respectively. The shape of the cap was the most diverse on Mt. Hwaak, whereas the shape of the stipe showed various phenotypes from various regions. Even among strains collected from the same forest, different types of mushrooms were observed. In conclusion, wild shiitake strains that have adapted to various environmental stress can be used to develop new varieties. Thus, securing various genetic resources and studying their traits are important for researching breeding technology.
