Plant factory can control artificially the environments for crop cultivation, so they can produce high quality agricultural products all year round. This study was carried to select suitable kohlrabi cultivar for hydroponics in a closed-type plant factory system. We used three cultivars of red kohlrabi, 'Asac kohl', 'Kolibri', and 'Purple king' as plant materials. The artificial light source was LED light, light intensity and photoperiod were $249{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ and 12/12 hours (day/night period), respectively. Hydroponic cultivation type was used circulating deep flow technique. At 43 days after transplanting, fresh weight of whole plant and tuber and leaf area were not significantly different among cultivars. Shoot dry weight and tuber dry weight were highest in 'Asac kohl' cultivar, and number of leaves was highest in 'Purple king' cultivar. Sugar content and yield were highest in 'Asac kohl' cultivar. Considering the growth and marketable yields, 'Asac kohl' was the optimal kohlrabi cultivar for hydroponic cultivation in a closed-type plant factory system.
To identify potential gastrointestinal prokinetic agents, water and 70% ethanol extracts and isolated compounds from 41 different traditional medicinal herbs were evaluated for the stimulation of gastrointestinal (GI) motility in vivo. Of the 41 water and 70% ethanol extracts, 12 extracts were found to enhance GI motility activity in mice by more than 10%. The 12 extracts are as follows: Atractylodes japonica (root), Crataegus pinnatifida (flower), Aucklandia lappa (root), Inula helenium (root), Cynanchum wilfordii (root), Chinese Liriope platyphylla (root), Codonopsis pilosula (root), Glehnia littoralis (root), Pinellia ternate (tuber), Agastache rugosa (aerial part), Angelica decursiva (whole plant), and Peucedanum praeruptorum (whole plant). In particular, the extracts from Atractylodes japonica (root), Cynanchum wilfordii (root) and Angelica decursiva (whole plant) have demonstrated the highest GI motility activity. In addition, 26 isolated compounds from the medicinal herbs were tested, and 8 isolated compounds were found to be active. They are ${\alpha}$-ionone, ${\beta}$-ionone, trans-caryophyllene, cedrol, methyl-3,5-di-O-E-caffeoyl-quinate, lobetyolin, oleoyllinoleoylolein and cis-jasmone. ${\beta}$-ionone from Aucklandia lappa (root) showed the most potent GI motility activity. The active traditional medicinal herbs and isolated compounds might be therapeutically advantageous in the treatment of GI motility disorders.
Late blight of potato caused by Phtophthora infestans is one of the most destructive disease in organic farming as well as in the conventional cultivation in Korea. Incidence and progress of the disease largely depend on climatic conditions around the fields. However, the disease completely destroys whole leaves of the plant within two weeks in organic farming fields. While, potato leaves in conventional farming fields remain healthy for 5 weeks after the first symptom appearance, because fungicides were applied four times in average to control the disease. Area under disease progress curve (AUDPC) of organic farming fields ranged from 2314 to 2421 and quickly expanded compare to that of conventional farming fields. The tuber yield in the organic farming fields ranged from 0.96 ton per 10a to 1.5 ton per 10a, while it was 2.7 ton per 10a in conventional farming fields. Tuber yields were reduced to $42\~63\%$ by the infection of late blight and the reduction rate was closely related with the time of first occurrence of the disease in organic farming fields. Physiological races of the pathogen were diversified with R0,1,3,4,5,7,10,11 in organic farming fields at Hongchun area. Pathogen races were distributed with similar pattern at different areas of organic farming fields.

The interstitial cells of Cajal (ICC) are pacemaking cells required for gastrointestinal motility. The possibility of whether DA-9701, a novel prokinetic agent formulated with Pharbitis Semen and Corydalis Tuber, modulates pacemaker activities in the ICC was tested using the whole cell patch clamp technique. DA-9701 produced membrane depolarization and increased tonic inward pacemaker currents in the voltage-clamp mode. The application of flufenamic acid, a non-selective cation channel blocker, but not niflumic acid, abolished the generation of pacemaker currents induced by DA-9701. Pretreatment with a $Ca^{2+}$-free solution and thapsigargin, a $Ca^{2+}$-ATPase inhibitor in the endoplasmic reticulum, abolished the generation of pacemaker currents. In addition, the tonic inward currents were inhibited by U-73122, an active phospholipase C inhibitor, but not by $GDP-{\beta}-S$, which permanently binds G-binding proteins. Furthermore, the protein kinase C inhibitors, chelerythrine and calphostin C, did not block the DA-9701-induced pacemaker currents. These results suggest that DA-9701 might affect gastrointestinal motility by the modulation of pacemaker activity in the ICC, and the activation is associated with the non-selective cationic channels via external $Ca^{2+}$ influx, phospholipase C activation, and $Ca^{2+}$ release from internal storage in a G protein-independent and protein kinase C-independent manner.
In this study, we investigated the microbial flora changes in Gugija-Liriope tuber Makgeolli during fermentation and storage periods. We brewed Gugija-Liriope tuber Makgeolli for a week through twostage fermentations and stored the fermentation broth for a month at $4^{\circ}C$ or $20^{\circ}C$. We collected the samples periodically and analyzed microbial flora changes using viable cell counts and PCR-denaturing gradient gel electrophoresis (DGGE). Yeast viable cells were seen to have decreased to 13% of pre-storage levels after storage for 15 days at $20^{\circ}C$; however significant changes were not observed during storage at $4^{\circ}C$. Prolongation of storage time dramatically decreased the availability of viable cells. Yeast viable cell numbers had decreased to 38% of pre-storage levels at $4^{\circ}C$ and 4.8% at $20^{\circ}C$ after storage for 30 days. The results of the DGGE profile for yeast showed that Saccharomyces cerevisiae and Saccharomyces sp. were the predominant strains at the beginning of fermentation and throughout the whole period of storage. Viable cell counts for total bacteria had decreased to 36% of pre-storage levels after storage for 15 days but did not significantly change for the full 30 days of storage at $4^{\circ}C$. Similarly, viable cell counts for bacteria had decreased to 5% while viable cell numbers did not significantly change for the full 30 days at $20^{\circ}C$. Viable cell counts for lactic acid bacteria were performed and the results were similar to those for total bacteria. The results of the DGGE profile for bacteria showed that Weissella cibaria was the predominant strain at the beginning of fermentation. However it had disappeared by the end of fermentation, and Lactobacillus fermentum and Pediococcus acidilactici became the predominant species during storage.
Appropriate soil temperature and early planting of potato is very important for the successful potato-soybean cropping system in central region of South Korea. This experiment was carried out to determine the effect of mulching materials on the growth and yield of potato (Solanum tuberosum L.). Five different mulch treatments were had been applied on an upland soil as follows ; no mulch (NM), transparent film (TF), transparent film + additional transparent film (TF + ATF), black film (BF), and black film + additional transparent film (ATF). In the period of sowing time to removing additional films, mean soil temperature of the treatments was in the order of TF+ATF > TF > BR+ATF > BF as $20.3^{\circ}C$ > $18.5^{\circ}C$ > $16.1^{\circ}C$ > $15.4^{\circ}C$, respectively and that of NM was $13.8^{\circ}C$. The accumulated soil temperature was TF > NM > BF during the removing additional films to earthing at inter-tillage. On the changes in the soil temperature during a whole day, the temperature in the BF was lower than NM during around 18:00 PM to 12:00 NM, while NM was higher than BF in the time period of 10:00AM to 21:00PM. The sequence of potato sprout emergence was 15 > 18 > 20 > 22 days of TF+ATF, TF, BF+ATF, and BF, respectively and that of NM was 24 days. Comparing to the NM, potato sprout emergence was observed on the TF+ATF treated plot as early as 9 days. At 10 days before harvest, the significant difference in the tuber dry weight had been observed and the sequence tuber weight was in the order of TF > TF+ATF > BF+ATF > BF > NM. The potato yields of TF, TF+ATF, and BF+ATF were increased of 40.7, 37.3, and 22% as compared to NM ($2,805kg\;10a^{-1}$), but almost same yield in the BF. The differences of tuber dry weight and potato yields was co-related with the temperature rise of soil by the application of mulching materials on soil. Based on these results, application of mulching film had been very effective to increase the tuber size and the yield of potato by the temperature rise during seedling stage of potato. Transparent mulching was better than black mulching especially for the emergence of sprout of potato in relation to minimizing cooling injury.
This study was carried out to investigate the expression patterns of foreign gene that controlled by tuber-specific patatin promoter in transgenic potatoes. Potato leaf disc cultured in vitro were transformed by the Agrobacterium strain LBA4404 containing pBl121 or pATGUS from potato cv. Desiree. In order to select the transgenic lines, gene-specific primers deduced from the NPTII were synthesized and used for polymerase chain reaction. The down part of the putative transgenic potatoes was transplanted weekly onto sucrose-enriched medium to accelerate the microtuber formation. RNA gel blot analysis was performed on the total RNAs obtained from tuber that had been harvested at a week interval. Also, histochemical assay was observed in the explants transformed with either pBI121 or pATGUS. Results showed that the transgenic plant containing pATGUS expressed GUS transcripts mainly at the tuber, not in stem, with the highest expression level in 5 weeks-grown microtubers. In contrast to pATGUS plants, the transformed plants with pBI121 showed an equal expression pattern throughout the whole developing stages. Consistent with RNA gel blot analysis, histochemical GUS staining and enzyme activity exhibited pATGUS transcripts were at the highest level in 5 weeks cultures. From these results, we suggest that the best stage to analyze the foreign gene introduced by patatin promoter into potato plants is at 5 weeks cultures after tuber formation.
To measure the soil-to-plant transfer factors ($TF_a,\;m^2\;kg^{-1}$-fresh) of radionuclides deposited during the growing season of potato, a radioactive solution containing $^{54}Mn,\;^{60}Co,\;^{85}Sr$ and $^{137}Cs$ was applied to the soil surfaces in soil boxes 2 d before seeding and three different times during the plant growth. For the pre-seeding application (PSA), radionuclides were mixed with the topsoil (loamy sand and 5.2 in pH). The plant parts investigated were leaves, stems, tuber skin and tuber flesh. The $TF_a$ values of $^{54}Mn,\;^{60}Co,\;^{85}Sr$ and $^{137}Cs$ from the PSA were in the ranges of $1.9{\times}10^{-4}{\sim}1.5{\times}10^{-2}$, $1.8{\times}10^{-4}{\sim}7.5{\times}10^{-4}$, $4.0{\times}10^{-4}{\sim}1.6{\times}10^{-2}$, $1.5{\times}10^{-4}{\sim}3.9{\times}10^{-4}$ respectively, for different plant parts. The TFa values from the growing-time applications were on the whole a few times lower than those from the PSA. For $^{54}Mn,\;^{85}Sr$ and $^{137}Cs$, the $TF_a$ values from the early- or middle-growth-stage application were higher than those from the late-growth-stage application, whereas the opposite was true for $^{60}Co$. Leaves and tuber flesh had the highest and lowest $TF_a$ values, respectively, in most cases. The total uptake from soil by the four plant parts was in the range of $0.05{\sim}3.16%$. In the third year following the PSA, the $TF_a$ values of $^{54}Mn,\;^{60}Co$ and $^{137}Cs$ were $11{\sim}25%$, $21{\sim}25%$ and $38{\sim}67%$ of those in the first year, respectively, depending on the plant parts. The present results can be used for estimating the radiological impact of an acute radioactive deposition during the growing season of potato and for testing the validity of relevant food-chain models.
Objective : For the purpose of developing Convoivulaceae plants recorded in Korean Herbology in Korea, the literatures of the successive generations have been thoroughly investigated to prepare this article. Methods : Books and research papers about Herbology which published at Korea and other countries were examined. Results : A list was made about Convoivulaceae plant which were cultivated or grew naturally in Korea, after investigated the data on domestic and foreign Convoivulaceae plants. Convoivulaceae(grew in Korea) were divided into 6 classes with 18 species. Out of those, 6 classes with 13 species were found serviceable which indicates approximately 72% of all. Out of all 18 species of Convoivulaceae family, Calystegia genus were found 6 species, which were shown the most. Among 4 genus of Calystegia species, medicinal plants were all one species each. Out of all serviceable parts in Convoivulaceae, Herb·Radix·Tuber·Semen·Flower·Stem and Folium parts took first place as 8 species. There are 2 species toxic, the survey said. 6 Classifying medicinal plants among the Convoivulaceae plants by part, whole Herbs are found to be 8 separate species each. When it comes to the nature of taste, 10 kinds of taste without action and 7 kinds of sweet taste are the most common. For channel tropism, kidney channel has the largest number of 7 types. Conclusion : A total of 6 genera and 18 species of Convoivulaceae was found in the Korean among which 6 genera and 13 species(approximately 72%) were medicinal plants. Two species toxic drugs were detected.

Viruses diagnosed on crops including rice plants from farmers or agricultural extension agencies cover the country were 11 species including Broad bean wilt virus 2 (BBWV2) in 2008. Tomato spotted wilt virus (TSWV) was the most important virus having the detection rate of 22.9%. Two viruses of Tomato yellow leaf curl virus (TYLCV) and Tobacco leaf curl virus (TLCV) inducing leaf yellow and curl diseases on tomatoes were occurred newly with the detection rate of 12.2% and 4.0%, respectively, in 2008. Rice stripe virus (RSV) was occurred on 869.5 ha mainly at Jindo and Haenam areas in Jeollanamdo province. At Jindo area, 12 plots were damaged severely with the infected hill rate of 83.8%. At the main production area of oriental melon at Seongju, almost all fruits from whole sale market at Seongju were infected with Cucumber green mottle mosaic virus (CGMMV) as the detection rate of 87%. The areas occurred TSWV in Korea were 25 totally from 2003 including 7 areas newly reported in 2008 including Naju in Jeoallanamdo. TSWV could be reduced as 0.1 % from 5.3% by covering insect proof net in vinyl house after chemical soil sterilization. Tomato yellow leaf curl disease was occurred on April in 2008 at Tongyoung area in Kyeongsangnamdo, and detected continuously at 13 areas, 7 in Kyeongsangnamdo, 4 in Jeollabukdo and 2 in Jejudo. Potato spindle tuber viroid (PSTVd) was occurred abruptly in a confined space of a civil breeding greenhouse and a cultivar evaluation field followed by disuse 17.4 M/T of potato tubers. No PSTVd was detected at 17 fields cultivated the related potatoes to the bred company by RT-PCR.
