Effects of Rumex axetosella, Sonchus oleraceus and Euphoibia jolkini Extracts on Melanin Synthesis in Melanoma Cells (애기수영, 방가지똥 및 암대극 추출물이 Melanoma Cell에서 멜라닌 합성에 미치는 영향)

  • In this study, we investigated the effect of Rumex axetosella, Sonchus oleraceus and Euphoibia jolkini extracts on tyrosinase activity and melanin production as natural products of whitening functional cosmetics. To measure the melanin production, 50, 100, $200{\mu}g/mL$ of Rumex axetosella, Sonchus oleraceus and Euphoibia jolkini extracts were treated on ${\alpha}-MSH$ treated B16F10 melanoma cells, respectively. Melanin contents in ${\alpha}-MSH$ treated B16F10 melanoma cells were decreased by 41.5, 51.11, and 61% in $200{\mu}g/mL$ treatment compared to none treatment, respectively. In addition, the intracellular tyrosinase activity was decreased after treatments with all extracts. Furthermore, $100{\mu}g/mL$ of Euphoibia jolkini extract was decreased 81.5% of melanin production in B16F10 melanoma cells. When the three extracts were compared, Euphoibia jolkini extract was considered to be the most functional material for whitening effect.

Antimelanogenic Effect of Taurine in Murine Melanoma B16F10 Cells (B16F10 Murine Melanoma 세포에서 멜라닌생성억제에 대한 타우린의 효과)

  • Taurine has been shown to be tissue-protective against oxidant-induced injury and is a powerful regulator of the immune system. However, there is no study on the antimelanogenic effect of taurine. In this study, we investigated the whitening effect of taurine in B16F10 mouse melanoma cells. Cell viability was measured by MTT assay. We examined melanin contents and tyrosinase activity according to time and concentration. Extracellular signal regulated kinase (ERK) is an important regulator of melanogenesis. It has been reported that activated ERK induced microphthalmia associated transcription factor (MITF) phosphorylation and its subsequent degradation and thus reduced melanin synthesis. In our B16F10 cell culture system, taurine led to decrease melanin contents by 21% at 48 hr. We then observed taurine effects on ERK-P, MITF and tyrosinase by Western blot. ERK was activated at 18 hr and 24 hr, whereas MITF reduced. We could not observe any differences in the levels of tyrosinase. These results suggested that taurine inhibited melanogenesis by ERK signal pathway via MITF degradation. We expect that taurine has potential skin whitening agents in cosmetics.

Modulation of Melanin Synthesis by Amaranthus spp. L Seed Extract in Melan-a Cells

  • Anti-melanogenic effects of amaranth (AT), one of the key source of squalene, were investigated in melanocytes. Amaranth seed powder was extracted with water and melan-a cells were treated with various concentrations of AT. By using HPLC, content of myo-inositol, one of potential active components, was measured in the crude extract of AT.AT reduced the melanin content in melan-a melanocytes and down-regulated melanogenic enzyme activity such as tyrosinase, TRP-1 and TRP-2. By regulating melanogenic enzyme activity, AT may be a potential natural source for whitening agent. Myo-inositol was detected in AT by HPLC and may be one of the active compounds from AT involved in the regulation of anti-melanogenesis. In this study, we demonstrated that AT has anti-melanogenesis properties. This new function of amaranth may be useful in the development of new skin-whitening products and its value as food.

The Study on Pharmacological Activation as Cosmetic Material of Rosa rugosa Thunb. Flowers Extract (해당화꽃 추출물의 화장품소재로서 약리활성에 관한 연구)

  • Objectives : In this study, we investigated the effect of ethanol extract from Rosa rugosa Thunb. flowers(ERF) on the activities of antioxidant, antiwrinkle and whitening. Methods : We measured antioxidant efficacy of ERF by using 1,1-diphenyl-2-picrylhydrazyl(DPPH) assay. Also we confirmed the inhibitory effect of ERF on collagenase activity and melanin synthesis by using collagenase assay kit and dihydroxiphenylalanine staining, respectively. To evaluate the anti-inflammatory effects of ERF, we examined the inflammatory mediator IL-6. Results : ERF showed highly efficacy in DPPH radical scavenging activity. ERF dose-dependently suppressed collagenase activity. Ultraviolet-induced production of IL-6 decreased by ERF treatment. In B16F10 cell, ERF significantly reduced tyrosinase activity and melanin synthesis. Conclusions : From the above results, it was indicated that ERF could be utilized as anti-aging and whitening cosmetic ingredients.

Inhibitory Effects of Root Extracts on Melanin Biosynthesis in Rodgersia podophylla A. Gray (도깨비부채 뿌리 추출물의 멜라닌 생성억제효과)

  • Rodgersia podophylla was a native medicinal plant cultivated in Korea. During the search for a new whitening natural herb, we found that underground part of Rodgersia podophylla showed inhibitory activity against melanin biosynthesis in melan-a cells by brocking tyrosinase activity. In addition, this plant exhibited protective effect on UV-B region. These results suggest that underground part of Rodgersia podophylla might be used as whitening agent for the skin.

  • This study was carried out to investigate the melanin synthesis inhibitory effect of Aspergillus fumigatus fermented extract. In this study, we revealed the effects of A. fumigatus fermented extract on melanin contents, mushroom tyrosinase activity, and expression levels of mRNA and proteins of melanogenesis-related gene in B16F10 melanoma cells. A. fumigatus fermented extract inhibited both melanin contents and tyrosinase activity. In addition, the expression level of mRNA or proteins of melanogenesis was down-regulated in the A. fumigatus fermented extract treated B16F10 cells with dose-dependent manner. Moreover, when the clinical test was conducted, it was confirmed that the use of the fermented extract of A. fumigatus for 8 weeks improved skin brightness 1.586 times brighter and skin melanin 1.331 times better compared to the control product. Taken together, our results suggest that A. fumigatus fermented extract has melanogenesis inhibitory effect and whitening activity, thus it showed the possibility for using as a functional whitening cosmetic resource.

  • Hwang Chil (Dendropanax morbifera Lev.) belongs to Aralia elata group. This plant is only found in Korea and is considered to be along with ginseng. The physiological activities of it have been reported as having scavenging effect on oxygen free radicals and anti-carcinogenic effect. As most of the anti-oxidants were effective on protecting hepatotoxicity from alcohol, hard tissues restoring and skin whitening, the effectiveness of Hwang Chil extracts was screened. (omitted)

  • The inhibitory effect of extract of Atractylodis Rhizoma alba on melanin biogenesis was studied by using B16/F10 melanoma in culture. Atractylodis Rhizoma alba significantly inhibited tyrosinase activity, and melanin contents with or without $\alpha$-MSH and forskolin in vitro. Melanin contents and tyro-sinase activity have decreased in a dose-dependent manner. These results show that extract of Atractylodis Rhizoma alba could be developed as skin whitening components of cosmetics.
