Effects of 1-tetradecanol and β-sitosterol Isolated from Dendropanax morbifera Lev. on Skin Whitening, Moisturizing and Preventing Hair Loss (미백, 보습 및 탈모방지에 대한 황칠나무(Dendropanax modifera Lev.)에서 분리한 1-tetradecanol, β-sitosterol의 효과)

  • Dendropanax morbifera Leveille (Araliaceae) is an endemic species growing in the south-western part of South Korea and has been used in folk medicine. However, the effects of Dendropanax morbifera Lev. on skin biology remain to be elucidated. In this study, we isolated 1-tetradecanol and ${\beta}$-sitosterol from the n-hexane fraction of Dendropanax mobifera Lev. and To investigate the whitening effect of the fraction, we tested the inhibition of tyrosinase activity of 1-tetradecanol. The results show that the inhibitory effect of the 1-tetradecanol was higher than water extract and n-hexane fraction. And 1-tetradecanol significantly reduced melanin contents of B16F10 cells compared to more than water extract and n-haxane fraction dose-dependantly without cell cytotoxicitiy (below $100{\mu}g/mL$). We also investigated the skin moisturizing effect using HR-1 hairless mice. The transepidermal water loss (TEWL) in the 1-tetradecanol treated group was significantly smaller than that in the other groups. To investigate the effect of the preventing hair loss by ${\beta}$-sitosterol, we observed HR-1 hairless mice through periodic growth feature. The results suggest that hair loss of mice by ${\beta}$-sitosterol was delayed and it's hair density showed the highest. These data provide evidence that Dendropanax morbifera Lev. may be a potent candidate for the improvement of both skin whitening, moisturizing and alopecia from the point of cosmetic industry view.

Cosmetic Application Using Skin Whitening and Anti-microbial effects of Ethyl Acetate and n-Butanol Fractions from Eruca sativa (Eruca sativa 에칠아세테이트와 부탄올 분획물의 미백 및 항균효과를 이용한 화장품 응용연구)

  • Eruca sativa, called arugula, is a perennial plant in the Brassicaceae family, an edible plant commonly used in Italian cuisine. To study as a cosmetic material application E. sativa was extracted with 70% ethanol (ES). Then ES was fractionated using n-hexane, chloroform, ethyl acetate, n-butyl alcohol and water (EHex, EEA, ECHCl3, EBuOH and EDW). EEA showed mushroom tyrosinase inhibitory activity. ES, EEA and EBuOH showed inhibition of tyrosinase activity. As a result, ES is expected to have skin whitening efficacy. ES was applied to 0.05, 0.1% the toner and emulsion formulation to test the stability. The anti-microbial activity of eight bacteria and fungi including Staphylococcus aureus and Propionibacterium acnes which cause dermatitis and acne was evaluated. EEA showed effects in all of microorganisms. The toner and emulsion containing ES with 0.05, 0.1% were passed in the challenge test. At -20, 4, 25, 55 ℃ and daylight, there was no significant change on pH, viscosity for 4 months. However, emulsions had phase separation phenomenon at 55 ℃, so the base formulation needs improvement. In addition, through the skin penetration test, EEA penetrated 0.058% in 6 hr, predicting the clinical efficacy. This means that E. sativa can contribute whitening agent and the synergistic effect of preservatives.

Antioxidative and Whitening Effects of Rubus parvifolius L. Extract on Dermal Cytotoxicity of ZnSO4, Mordant (매염제인 ZnSO4의 피부독성에 대한 멍석딸기 추출물의 항산화 및 미백효과)

  • This study was done to evaluate the dermatoxicity of zinc sulfate (ZnSO4) and the protective effect of Rubus parvifolius L. (RP) extract on cytotoxicity of ZnSO4, mordant in cultured SK-MEL-3 cells. For this study, it was done an antioxidative effect as DPPH-radical scavenging ability as well as the diminutive ability of total melanin with cell viability. ZnSO4 significantly decreased cell viability in dose-dependently, and it was mid-toxic. The ascorbic acid significantly increased cell viability damaged by ZnSO4-induced cytotoxicity. In the protective effect of RP extract on ZnSO4-induced cytotoxicity, RP extract significantly increased cell viability compared with ZnSO4-treated group, and also it showed both the DPPH-radical scavenging ability and the decrease of total amount of melanin. From these findings, the cytotoxicity of ZnSO4 is correlated with oxidative stress, and also RP extract effectively protected ZnSO4-induced cytotoxicity via antioxidative effect such as DPPH-radical scavenging ability with the whitening effect by the decrement of total amount of melanin. Conclusively, the natural ingredients like RP extract may be a useful agent for the improvement of antioxidative and whitening effects

Whitening and Anti-inflammatory Constituents from the Extract of Citrullus lanatus Vines (수박 덩굴 추출물 유래 미백 및 항염 활성 성분)

  • In this study, we investigated whitening and anti-inflammatory constituents from a watermelon (Citrullus lanatus, C. lanatus) vines (leaves and stems). As anti-melanogenesis and anti-inflammatory activities were screened for the ethanol extract and solvent fractions, n-hexane (n-Hex) and ethyl acetate (EtOAc) fractions showed the most potent activities. Three constituents were isolated from the n-Hex and EtOAc fractions of C. lanatus; ${\alpha}-linolenic$ acid (1), sigmast-7-en-O-${\beta}$-D-glucopyranoside (2), 1-feruloyl-${\beta}$-D-glucopyrinoside (3). The chemical structures of the isolated compounds were elucidated based on the spectroscopic data including $^1H$ and $^{13}C$ NMR spectra, as well as comparison of the data to the literature values. Whitening and anti-inflammatory effects were studied for the isolated compounds. Upon the anti-melanogenesis tests using ${\alpha}-MSH$ stimulated B16F10 melanoma cells, the compounds 1 and 3 inhibited the cellular melanogenesis and intracellular tyrosinase activities effectively. For the anti-inflammation tests using lipopolysaccharide (LPS)-induced RAW 264.7 cells, the isolates 1 and 3 were determined to decrease the production of nitric oxide (NO) and pro-inflammatory cytokines ($TNF-{\alpha}$, IL-6). Based on these results, C. lanatus vines extract could be potentially applicable as whitening and anti-inflammatory ingredients in cosmetic formulations.

  • The milling process is considered as causing one of the greatest grain losses among all the processes in rice post-production. Major source of grain losses in the rice milling is considered as the whitening process. This study was attempted to develop an abrasive-type whitener, the whitening chamber of which being supplied by jet-air evenly and continuously. To investigate the milling performance by the new whitener, three kind of emery-stone grit(#36, #41, and #46), and three levels of rotational speed of emery stone roller (950, 1070, and 1200 rpm) were tested. The jet-air abrasive-type whitener was also compared with the conventional abrasive-type having no jet-air blower in terms of their milling performance. In addition, the effect of different combinations of sequential uses of the abrasive- and friction-type whiteners on the milling performance was also experimentally evaluated. The results of this study are summarized as follows; 1. In general, the whitening system combined with the abrasive type whitener with jet-air supply, which was newly designed, and the existing jet-air friction type whiteners produces more milled- and head-rice by about 0.3% points and 2.8% points, respectively than the system combined with the existing abrasive type without the jet-air supply under the same operational conditions. The former also consumed less electricity by 0.024 KwH per 100kg of milled rice production and gave more milling capacity by about 35 kg/hr. As compared with the conventional whitening system consisting of jet-air friction type whiteners only, the former yielded more milled- and head-rice by 1.5% points and 4.4% points, respectively. 2. The abrasive roller having 46 grit emery was better than the roller having 36 grit in aspects of milling performance and machine efficiency, in general. 3. With regard to the effect of combination method of abrasive type and friction type whiteners, one pass in abrasive type plus three passes in friction type gave better milling performance and energy efficiency than the two passes in abrasive type plus two passes in friction type regardless of the designs of the emery stone rollers. 4. The increase in rotational speed of the emery stone roller from 950 rpm to 1200 rpm presented negative effects on milled and head-rice yields and machine efficiency, but slightly positive effect on milling capacity.

Evaluations of Antioxidative Activity and Whitening Effect of Extracts from Different Parts of Cosmos bipinnatus (코스모스 부위별 추출물의 항산화 활성과 미백효능평가)

  • In this study, the antioxidative effect, cellular protective effect and inhibitory effect on tyrosinase of Cosmos bipinnatus extracts were investigated. The ethyl acetate fraction of Cosmos bipinnatus flower extract ($11.48\;{\mu}g$/mL) showed more excellent free radical (1,1-diphenyl-2-picrylhydrazyl radical, DPPH) scavenging activity (FSC50) than those of leaf and stem extracts ($17.45\;{\mu}g$/mL). Reactive oxygen species (ROS) scavenging activity (OSC50) of Cosmos bipinnatus extracts on ROS generated in $Fe^{3+}$-EDTA/H2O2 system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction of Cosmos bipinnatus flower extract ($0.56\;{\mu}g$/mL) showed 3 times more excellent ROS scavenging activity than L-ascorbic acid ($1.50\;{\mu}g$/mL). The protective effects of the ethyl acetate fractions of extracts from different parts of Cosmos bipinnatus on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fractions of leaf and stem extract and flower extracts suppressed photohemolysis in a concentration dependent manner ($10\sim50\;{\mu}g$/mL). The inhibitory effect of ethyl acetate fraction of Cosmos bipinnatus flower extract ($62.75\;{\mu}g$/mL) on tyrosinase was investigated to assess the whitening efficacy. The ethyl acetate fraction of Cosmos bipinnatus flower extract showed 3.5 times higher tyrosinase inhibitory effect than arbutin ($226.88\;{\mu}g$/mL) known as an effective whitening agent. These results indicate that fractions of Cosmos bipinnatus extracts can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O2$ and other ROS, and protect cellular membranes against ROS. Fractions of Cosmos bipinnatus extracts can be applicable to new functional cosmetics for antioxidant and whitening.

Anti-Inflammatory and Whitening Effect of the Lyophilized Powder of Oriental Plant Extracts Fermented with Streptococcus thermophilus (Streptococcus thermophilus로 발효한 한약재 발효분말의 항염증 및 미백 효과)

  • To identify new anti-inflammatory and whitening material, this study investigated the whitening and anti-inflammatory effects of the lyophilized powder from 6 oriental plant extracts (OPE; Citrus junos Tanaka, Mori cortex Radicis, Schisandra chinensis Baillon, Coix lachrymajobi var. mayuen, Angelica gigas NAKAI, and Sophora japonica L.) fermented with Streptococcus thermophilus by assessment of cytotoxicity on human dermal fibroblast, inhibitory effect of nitric oxide (NO) prodction, tyrosinase activity and melanin formation. The OPE was fermented with Streptococcus thermophilus at $37^{\circ}C$ for 2 days and the lyophilized powder was manufactured by freezing-dryer. OPE didn't show cytotoxicity at concentration of $500{\mu}g/mL$ using a cytopathic effect reduction method. OPE also exhibited inhibitory effect on nitric oxide (NO) prodction by Griess reagent system. Furthermore, OPE showed inhibitory effect on tyrosinase activity with dose dependent manner, and exhibited significant inhibition of melanin formation by measurement of melanin from culture media (p < 0.05). From these results, 6 OPE extracts showed anti-inflammatory and whitening effect and may be used as an active ingredient for cosmetics.

A Study on the Antioxidant, Whitening and Anti-Inflammatory Activities of Trichosanthis Cucumeroidis Radix Extract (쥐참외뿌리 추출물의 항산화, 미백, 항염증 활성 연구)

  • In this study, the physiological activity effect of trichosanthis cucumeroidis radix extract on antioxidant activity, whitening, and anti-inflammatory activity was checked, and the possibility of its use as a functional material was checked. The purpose of this study was to confirm the antioxidant activity through DPPH radical scavenging activity of trichosanthis cucumeroidis radix extract, whitening activity effect through melanin production inhibition ability for melanin cell B16F10 melanoma cell, and anti-inflammatory activity effect through NO production inhibition ability for macrophage RAW 264.7 cell. As a result of the study, the concentration-dependent DPPH radical scavenging activity of the trichosanthis cucumeroidis radix extract was confirmed, and DPPH radical scavenging activity similar to that of the positive control Ascorbic acid was confirmed. It was confirmed that the melanin production inhibitory activity induced by 100 nM 𝛼-MSH and the NO production inhibitory ability induced by LPS 1 ㎍/mL were significantly suppressed. Accordingly, it is considered that the trichosanthis cucumeroidis radix extract may be used as a functional material having antioxidant, whitening, and anti-inflammatory effects.

Extraction and effect of whitening agents from chinese plants (중국산 천연물에서 미백성분의 추출 및 효과)

  • In this work, extraction and purification of the possible whitening agents from the Chinese plants; Chrysanthemum morifolium Ramat (xizang cai ju hua), Rhodiola sachalinensis, and Terminalia chebula Retzius have been described. The chopped leaves of Chrysanthemum morifolium Ramat and Terminalia chebula Retzius were added to water and ethyl ether, respectively. Components were separated on a GS310 column ($21.5{\times}500mm$ i.d., $10-15{\mu}m$) and concentrated into four or three portions. The chopped leaves of Rhodiola salientness were added to methanol and separated and concentrated on a column ($C_{18}$ column, $3.9^{\circ}$�F8;300 mm i.d., $15{\mu}m$) into two parts. The whitening effects of extracts were examined by in-vitro melanin production assay, in melana and B16 cells at a concentration of $10{\mu}g/ml$. The ethyl acetate layer of Chrysanthemum morifolium Ramat showed 92% melanin inhibitory at $200{\mu}g/ml$, the extract of Rhodiola sachalinensis showed a whitening effect of about 60% melanin inhibitory, which was more efficient than the whitening effect of arbutin (45.6%). The methanol extract of Terminalia chebula Retzius inhibited melanin expression by 90% at $100{\mu}g/ml$; however, it was toxic to B16 melanoma cells.

Whitening Effect and Antioxidant Activity of Horseradish Subcritical Water Extracts (겨자무 아임계수 추출물의 항산화활성 및 미백효과)

  • Melanin is a natural pigment found in most plants and animals, and it is involved in determining the color of the skin and hair. Melanogenesis is a reactive occurrence in melanocytes aiming to protect the skin from external stimuli, such as ultraviolet rays. Tyrosine is involved in the biosynthesis of the substrate tyrosine into melanin. However, melanin overproduction can lead to skin diseases, such as melasma, blotching, hyperpigmentation, and skin cancer. Although many studies have been conducted on whitening substances, such as kojic acid and arbutin, some countries have banned or refrained from using them due to their side effects. Therefore, this study assessed the potential of horseradish (HR) as a new whitening agent in cosmetic products. For efficient extraction, subcritical water extraction was conducted. The results showed that the horseradish subcritical water 200℃ (SW 200) extract showed high DPPH radical scavenging ability, total phenolic contents (TPC), inhibiting tyrosinase activity and inhibiting melanin production of B16-F10 melanoma cell lines. To investigate its cytotoxicity to the B16-F10 melanoma cell lines, MTT reduction assay and morphological changes were observed. No cytotoxicity was found in horseradish methanol extract and SW 200. In conclusion, this research suggests the possibility of horseradish subcritical water may be useful as a natural whitening ingredient to be used in cosmetic products.
