This study was carried out to investigate a point of difference between normal and inferior Korean red ginseng (Naeback red ginseng = red ginseng with white part of clear boundary in phloem and/or xylem of ginseng body, saengnaeback red ginseng red ginseng with white part of indistinct boundary). White part with clear or indistinct boundary in center of ginseng body was observed in inferior red ginseng (naeback and saengnaeback red ginseng), and the differences in the internal color intensity was also found with naked eye. In hunter color values of normal and inferior parts of red ginseng in accordance with particle size, L value was increased with a diminishment in particle size, while a and b value were decreased. Absorbance at visible spectrum did not differ from water and 70% ethanol extract from normal and inferior parts of red ginseng, but absorbance in UV spectrum of extract from naeback part showed higher than those of normal and saengnaeback part. In comparison of intrastructure by electron microscope, the horizontal and vertical section of cortex and pith layer from normal part showed the very dense state, but small holes were found in naeback part of red ginseng by naked eye and electron microscope. The specific surface area of normal, naeback and saengnaeback part appeared 3.02, 3.33 and 6.55 ㎡/g, respectively. From above results, we consider saengnaeback red ginseng is red ginseng in the intermediate process which normal red ginseng changes to naeback red ginseng.
In order to investigate the inferior factor of red ginseng quality, the contents of various chemical components, physico-chemical properties and arrangement state of ginseng cells were observed. Contents of total reducing sugar, reducing sugar, crude protein, crude fibre and specific gravity of inside white part of red ginseng were less than those of normal part. But differences in content of crude saponin, HPLC pattern of ginsenosides and reducing ability for DP P H(1,1-dipheny 1-2-picrylhydrazyl) between normal and inside white part of red ginseng were not found. The optical density of 1 water extract of normal part of red ginseng did not differ from that of inside white 1 part of red ginseng, but the visible and UV absorbance of acid hydrolyzate of normal red ginseng showed higher than those of inside white part of red ginseng. The differences in the internal color and tissue of normal and inside white part of red ginseng were easily found with naked eye, and by the microscopic fractography, the orangement state of ginseng cell in the inside white part of red ginseng was less dense than that in normal red ginseng.
This study was carried out to evaluate the preventive effect of three forms of Korean ginseng roots (fresh, white and red) against bisphenol A (BPA) toxicity in mouse male germ cells (GC-2spd, TM3, TM4). ROS (reactive oxygen species) generation were measured by DCF-DA (2',7'-dichlorohydrofluorescein diacetate) assay. Also, semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR) was performed to quantify the mRNA expression levels of apoptosis-related genes, Bax (pro-apoptotic gene) and Bcl2 (anti-apoptotic gene). ROS generation was increased by $50{\mu}M$ BPA, but definitely decreased by treatment with Korean ginseng extracts (fresh, white and red) in mouse male germ cells. In especial, Korean fresh ginseng extract reduced significantly ROS production to normal control. In addition, Korean fresh and white ginseng extracts suppressed the apoptosis of mouse male germ cells by fine-tuning mRNA levels of apoptotic genes changed by BPA. In general, Korean fresh ginseng extract was more effective than white ginseng extract for reducing BPA-induced oxidative stress and apoptosis in mouse male germ cells. Therefore, Korean fresh and white ginseng may help to alleviate biphenol A toxicity in mouse male germ cells.
Korean ginseng (Panax ginseng Meyer) is a traditional herb used across the world to treat various diseases. Although, red ginseng is this herb's most famous product and has demonstrated diverse pharmacological activities, white ginseng (WG) is another ginseng product that is made fresh and individually regulated in Eastern Asia. Red and white ginseng show different characteristics due to distinct processing steps despite originating from the same plant, and the drug interactions induced by WG have not been well documented. Selegiline is a selective monoamine oxidase (MAO) inhibitor used as an antidyskinetic and antiparkinsonian agent. Here we developed a quantification method for selegiline in mouse plasma using a C8 stationary phase in triple quadrupole-mass spectrometry (LC-MS/MS) with multiple reaction monitoring (MRM). The validated LC-MS/MS method was successfully applied to determine the potential interaction with WG extract (0.1 g/kg/day) pre-administered for 4 weeks. The $AUC_{0-240min}$ of selegiline was altered due to a decrease in the absorption of selegiline with repeated administration of WG extract.
This study was carried out to investigate the effect of water extracts from 4-, 5- and 6-year old red white ginseng roots on the inhibition of lipolysis in fat cells induced by toxohormone-L which has been known as lipolytic and anorexigenic factors. Toxohormone-L was obtained by partial purification of the ascites fluid from mice which had been inoculated with sarcoma-180. Each water extract of ginseng was effective in vitro against the inhibition of lipolysis induced by the toxohormone-L at the concentrations over 10~100 $\mu\textrm{g}$/ml. At the concentration of 1,000 $\mu\textrm{g}$/ml, the inhibition ratio of lipolysis by the water extracts of 4-, 5- and 6-year old white ginseng roots were 56.3, 59.7 and 59.4%, and those of red ginseng roots were 78.6, 79.1 and 82.5%, respectively, indicating that the 6-year old red ginseng was the most effective in the inhibition of the lipolysis. The total inhibitory activity per gram of ginseng sample was higher in the 4-year and 5-year old white ginseng roots at the concentrations of 100 $\mu\textrm{g}$/ml and 1,000 $\mu\textrm{g}$/ml, respectively, while for the red ginseng it was higher in the 6-year old ginseng than other ages regardless of the reaction concentration. Only once injection of the ginseng extracts(300 mg/ml in saline solution) with 10 ${mu}ell$/g of weight to the mice bearing sarcoma-180 was not effective against the extension of their life spans. On the contrary, the life spans of the mice were rather shortened by the injection of 5-year old white ginseng and 6-year old red ginseng, as compared to that of control.

Background: Ginseng (Panax ginseng) is a widely used traditional herbal supplement that possesses various health-enhancing efficacies. Various ginseng products are available in market, especially in the Korean peninsula, in the form of drinks, tablets, and capsules. The different ginseng types include the traditional red ginseng extract (RGE), white ginseng, and black red ginseng extract (BRGE). Their fermented and enzyme-treated products are also available. Different treatment regimens alter the bioavailability of certain compounds present in the respective ginseng extracts. Therefore, in this study, we aimed to compare the antioxidant and immune-stimulating activities of RGE, BRGE, and fermented red ginseng extract (FRGE). Methods: We used an acetaminophen-induced oxidative stress model for investigating the reduction of oxidative stress by RGE, BRGE, and FRGE in Sprague Dawley rats. A cyclophosphamide-induced immunosuppression model was used to evaluate the immune-stimulating activities of these ginseng extracts in BALB/c mice. Results: Our results showed that most prominently, RGE (in almost all experiments) exhibited excellent antioxidant effects via increasing superoxide dismutase, catalase, and glutathione peroxidase activities in the liver and decreasing serum 8-hydroxy-2'-deoxyguanosine, aspartate aminotransferase, and lactate dehydrogenase levels compared with the groups treated with FRGE and BRGE. Moreover, RGE significantly increased the number of white blood cells, especially T and B lymphocytes, and antibody-forming cells in the spleen and thymus, and it also activated a number of immune cell subtypes. Conclusion: Taken together, these results indicate that RGE is the best supplement for consumption in everyday life for overall health-enhancing properties.
수증기로 찐 다음 뜨거운 물로 추출한 인삼 추출물과 뜨거운 물로만 추출한 인삼 추출물을 대상으로 갈변화 정도와 라디칼 소거활성을 검토하였다. 그결과 수증기로 찌는 것이 물속에서 가열하는 것보다 갈변화 반응이 더 많이 진행되며, 그 추출물의 항산화작용도 더 크다는 것을 알 수 있었다. 이러한 차이는 수삼보다 백삼을 원료로 사용한 경우가 더 큰 것으로 나타났다.
This study was carried out to investigate the optimum conditions for extraction of soluble acidic polysaccharides from ginseng marc. Method of carbazole-sulfuric acid was applied to determine the amount of acidic polysaccharides in ginseng marc. The amounts of soluble acidic polysaccharides in water extract of ginseng marc were increased with increasing extraction temperature. The contents of acidic polysaccharides were not significantly different despite the extraction time increasing from 0.5 hours to 6 hours. To estimate the rehydration rate of the freeze dried polysaccharide, the extracted acidic polysaccharide fraction powder was determined the amount of soluble acidic polysaccharides by carbazole-sulfuric acid method again. The rehydration rate of acidic polysaccharides from water-extract of red ginseng marc at room temperature was 100%. On the other hand, the rehydration rate of acidic polysaccharide of red ginseng marc at boiling temperature was about 50%. The rehydration rate of acidic polysaccharides from water-extract of white ginseng marc at room temperature was 50%. The rehydration rate of acidic polysaccharide of red ginseng marc at boiling temperature was about 40%. The rate of soluble acidic polysaccharide of Red Ginseng is higher than that of White Ginseng. We can find out the maximum extraction method of soluble acidic polysaccharide from ginseng marc.

The purpose of this study is to develop a new preparation process of ginseng extracts having high concentrations of ginsenoside $Rg_3$, $Rg_5$ and $Rk_1$, a special component of Red ginseng. Chemical transformation from ginseng saponin glycosides to prosapogenin was analyzed by the HPLC. Extracts of White ginseng (Panax ginseng) and Fine White ginseng were processed under several treatment conditions including microwave and vinegar (about 14% acidity) treatments. Results of those treatments showed that the quantity of ginsenoside $Rg_3$ increased by over 0.6% at 4 minutes of pH 2~4 vinegar and microwave treatments. The results of processing with MWG-4 indicate that the Microwave and vinegar processed white ginseng extracts (about 14% acidity) that had gone through 4-minute treatments were found to contain the largest amount of ginsenoside $Rg_3$ (0.626%), $Rg_5$ (0.514%) and $Rk_1$ (0.220%). Results of treatments with MFWG-5 showed that the Fine White ginseng extracts that had been processed with microwave and vinegar (about 14% acidity) for 5 minutes were found to contain the largest amount of ginsenoside $Rg_3$ (4.484%), $Rg_5$ (3.192%) and $Rk_1$ (1.684%). It is thought that such results provide basic information in preparing White ginseng and Fine White ginseng extracts with functionality enhanced.
