Development of Kinetic Models Describing Kinetic Behavior of Bacillus cereus and Staphylococcus aureus in Milk

  • This study developed predictive models to evaluate the kinetic behaviors of Bacillus cereus and Staphylococcus aureus in milk during storage at various temperatures. B. cereus and S. aureus (3 Log CFU/mL) were inoculated into milk and stored at $10^{\circ}C$, $15^{\circ}C$, $20^{\circ}C$, and $30^{\circ}C$, as well as $5^{\circ}C$, $15^{\circ}C$, $25^{\circ}C$, and $35^{\circ}C$, respectively, while bacterial populations were enumerated. The growth data were fitted to the modified Gompertz model to estimate kinetic parameters, including the maximum specific growth rate (${\mu}_{max}$; Log CFU/[$mL{\cdot}h$]), lag phase duration (LPD; h), lower asymptote ($N_0$; Log CFU/mL), and upper asymptote ($N_{max}$; Log CFU/mL). To describe the kinetic behavior of B. cereus and S. aureus, the parameters were fitted to the square root model as a function of storage temperature. Finally, the developed models were validated with the observed data, and Bias (B) and Accuracy (A) factors were calculated. Cell counts of both bacteria increased with storage time. Primary modeling yielded the following parameters; ${\mu}_{max}$: 0.14-0.75 and 0.06-0.51 Log CFU/mL/h; LPD: 1.78-14.03 and 0.00-1.44 h, $N_0$: 3.10-3.37 and 2.09-3.07 Log CFU/mL, and $N_{max}$: 7.59-8.87 and 8.60-9.32 Log CFU/mL for B. cereus and S. aureus, respectively. Secondary modeling yielded a determination of coefficient ($R^2$) of 0.926.0.996. B factors were 1.20 and 0.94, and A factors were 1.16 and 1.08 for B. cereus and S. aureus, respectively. Thus, the mathematical models developed here should be useful in describing the kinetic behaviors of B. cereus and S. aureus in milk during storage.

미국삼(Panan quinquefolium)에서 분리한 뿌리썩음병균 Cylindrocarpon destructans의 후막포자 생성 및 분리 (Production and Isolation of Chlamydospores in Cylindrocaupon destructans Causing Root Rot of Panax quinquefolium)

  • Incubation condition affecting the chlamydospore formation and isolation from mycelia and conidia of Cylindrocarpon destructanse (isolate ACY-9701), isolated from the root rot lesion of the American ginseng (Panax quinquefolium) was investigated. Chlamydospores were formed from mycilia but not from conidia on the Czapek-Dox agar without carbon or nitrogen source after 20 days incubation at 2$0^{\circ}C$. In the medium added with nitrogen and carbon sources, immatured chlamy-dospore-like cells were formed from microconidia and mycelia as well. Immatured chlamydospore-like cells were formed from mycelia as well as microconidia In corn, kidney bean, and pea root extracts after 20 days incubation at 20"C, while typical chlamydospores were formed from both of them in the root extract of Panax quinquefolium. The 3.6 log chlamydospore/mm" was converted from microconidia in the medium, which was equal to 2.5% conidia formed. Under the light condition (251.1 pmol/m" sec, 12 hrs dark and light cycle), 4.2 log/mm" of chlamydospores were converted from interracially or terminal cells of macroconidia, which was 4.0% of macroconidia produced on Potato dextrose agar (PDA). When mycelia and microconidia were stored at -7$0^{\circ}C$ for 32 days and incubated on PDA after thawing at room temperature to isolate chlamydospores from them, microconidia and mycelia were still alive. Meanwhile, microconidial lysis was found after heating them at 32$^{\circ}C$ for 7 days, but the chlamydospores converted from macroconidia were not lysed up to 13 days at 32"C. to 13 days at 32"C.ot;C.

  • Stratigraphic subdivision, correlation and depositional environments of Oligocene strata in the Cheju basin are studied using microfossils, wireline-log, trace elements from Geobuk-1, Okdom-1 and JDZ V-2 exploratory wells as well as seismic attributes. Application of these Integrated approaches is becoming increasingly an important tool in sedimentary and petroleum geology. The purpose of this study is to provide an integrated sequence-stratigraphic interpretation for petroleum system in the Cheju basin. During the last several years the Cheju basin has been studied for petroleum potential in terms of sequence stratigraphy. The strata assigned to be of the Early Miocene in the previous studies are reinterpreted to be of Oligocene in age. Depositional environments of these strata are also reinterpreted to have been influenced significantly by marine floodings. This interpretation is supported by the lines of evidence such as occurrences of abundant dinoflagellates and calcareous nannofossils, higher sulfur (1000-10000 ppm), TOC contents $(>1{\~}3{\%})$ and hydrogen index, and specific biomarkers such as dinostrane and $C_{30}$ 4-methyl steranes. Seismic facies, determined as a fairly continuous amplitude reflection, relatively parallel uniform strata, also provides a clue for recognizing marine transgressions during the Oligocene. Two 2nd--order stratigraphic cycles are observed in Oligocene strata based on well-log responses and the presence of microfossils. Each sequence comprises shallow marine deposits in the lower part and inner-outer neritic deposits in the upper part, thus showing a upward-deepening trend. In petroleum exploration point of view, the presence of marine strata would provide a better exploration potential for source rock and reservoir quality in the Cheju basin, offshore Korea.

Validation of the correlation-based aerosol model in the ISFRA sodium-cooled fast reactor safety analysis code

  • ISFRA (Integrated SFR Analysis Program for PSA) computer program has been developed for simulating the response of the PGSFR pool design with metal fuel during a severe accident. This paper describes validation of the ISFRA aerosol model against the Aerosol Behavior Code Validation and Evaluation (ABCOVE) experiments undertaken in 1980s for radionuclide transport within a SFR containment. ABCOVE AB5, AB6, and AB7 tests are simulated using the ISFRA aerosol model and the results are compared against the measured data as well as with the simulation results of the MELCOR severe accident code. It is revealed that the ISFRA prediction of single-component aerosols inside a vessel (AB5) is in good agreement with the experimental data as well as with the results of the aerosol model in MELCOR. Moreover, the ISFRA aerosol model can predict the "washout" phenomenon due to the interaction between two aerosol species (AB6) and two-component aerosols without strong mutual interference (AB7). Based on the theory review of the aerosol correlation technique, it is concluded that the ISFRA aerosol model can provide fast, stable calculations with reasonable accuracy for most of the cases unless the aerosol size distribution is strongly deformed from log-normal distribution.

탄성파 기록으로부터 기본 파형 추출에 관한 연구 (Waveform Estimation from Seismic Records)

  • 탄성파 파형은 탄성파트레이스가 탄성파의 고유파형과 매질의 반사계수시계열과의 1차 콘볼루션이라는 가정 아래 위너적합필터(Wiener match filter)에 의하여 산출될 수 있다. 이때 매질의 반사계수 시계열은 음파 및 밀도검층 기록으로부터 도출한다. 본 논문에서는 파형 산출의 한 방법으로 탄성파 트레이스 및 반사계수 시계열을 보정하는 전처리과정의 효과를 검증하는 실험을 실시하였다. 이 전처리과정은 시간 측정은 탄성파 트레이스로부터, 그리고 반사계수는 검층 기록으로부터 잘 측정될 수 있다는 가정에 근거한다. 따라서 전처리과정은 1) 음파검층 기록의 깊이로부터 시간으로의 변환을 탄성파 트레이스에 잘 적합하도록 시작시각과 구간별 시간 길이를 조절하고, 2)탄성파 트레이스의 진폭을 검층기록에서 도출된 반사계수와 잘 적합하도록 조절하는데 이때 탄성파와 검층의 저주파 성분만을 비교한다.

Microbiological Quality and Safety Assessment of Commercial Ready-to-Eat Side Dishes Sold in Gyeonggi-do

  • 2019년 경기도내 전통시장, 대형마트, 반찬전문점에서 수거한 반찬류의 미생물 품질을 조사하였다. 반찬류 108건의 식중독 원인균을 검사하였고, 그 중 75건에 대해서는 구매 장소별, 조리 방법별 위생세균 검사를 진행하였다. 14건(12.9%)에서 Bacillus cereus가 검출되었으며, 나머지 94건에서는 식중독 원인균이 검출되지 않았다. 위생세균 검사에서 일반세균의 평균 검출량(범위)은 전통시장이 5.8 log CFU/g (3.0-8.2 log CFU/g), 대형마트는 4.3 log CFU/g (2.3-7.8 log CFU/g), 반찬전문점에서는 3.8 log CFU/g (0.0-6.9 log CFU/g)로 나타났으며, 구입 장소에 따른 유의적인 차이가 있었다(Ρ <0.05). 전통시장의 일반세균수와 대장균군은 대형마트, 반찬점과 통계적으로 유의한 차이가 있었고, 생채류, 나물류, 볶음류, 젓갈류, 조림류 순으로 일반세균수와 대장균군이 높게 검출되었다. 콩나물 무침의 보관온도별 일반세균수의 변화는 냉장보관(4℃)에서는 72시간 경과에서도 큰 변화가 없었으나, 상온보관(20℃) 및 고온보관(35℃) 시 구입 후 각각 9시간, 6시간 경과 시 부패의 가능성이 제기되어 구입 즉시 냉장보관 할 것을 권장하며, 제품을 판매하는 시설에서도 냉장보관을 하여야할 것이다.

테프론 열형광선량계(熱螢光線量計)의 특성(特性) (Some Characteristics of Teflon-Thermoluminescent Dosimeters)

  • 방사선(放射線)의 선질(線質) 및 고선량(高線量)의 변화(變化)에 따른 테프론 열형광선량계(熱螢光線量計)의 열형광감응특성(熱螢光感應特性)을 연구(硏究)하였다. 상이(相異)한 방사선(放射線)의 선질(線質)에 대한 TLD감도(感度)의 변화(變化)는 $^{60}Co$ 감마선(線), 유효(有效)에너지가 44keV, 69keV, 108keV인 X선(線) 및 0.044eV의 열중성자(熱中性子)에 조사(照射)시킴으로서, 광자(光子)에너지 의존성(依存性) 뿐만 아니라 이에 따른 초선형특성(超線形特性)의 변화(變化)도 조사하였다. 그 결과(結果)는 다음과 같다. $^{60}Co$감마선(線)에 대해서 흡수선량함수(吸收線量函數)로서의 $T-CaSO_4$:Dy 의 TL감응(感應)은 5Gy 까지는 선형적(線形的)이고, 5Gy 이상에 대해서 이 감응(感應)은 초선형적(超線形的)이었다. T-LiF-7 선량계(線量計)의 초선형특성(超線形特性)이 $T-CaSO_4$:Dy에 비해 현저(顯著)하였으며, 방사선(放射線)의 LET가 낮으면 낮을수록 초선형효과(超線形效果)가 더욱더 커짐을 알 수 있었다. $^{60}Co$ 감마선(線) 10Gy 에 등가(等價)하는 열중성자(熱中性子) 조사(照射)로 부터는 초선형특성(超線形特性)이 관찰(觀察)되지 아니하였다. TLD, T-LiF-7 및 $T-CaSO_4$:Dy 의 $^{60}Co$ 감마선(線)의 선량(線量)에 따른 상대감도(相對感度)(Rs)는 최소자승법(最小自乘法)으로 다음과 같은 실험식(實驗式). 즉(卽), $$R_{LiF}=1.021-0.04581\;logD+0.402(logD)^2-0.405(logD)^3,\;\;5{\times}10^3{\geq}D{\geq}1(Gy)$$$$R_{CaSO_4}=0.976-0.3241\;logD+0.262(logD)^2-0.298(logD)^3,\;5{\times}10^3{\geq}D{\geq}1(Gy)$$으로 각각(各各) 근사(近似)시킬 수 있었다.

  • The classical Geometric Brownian motion (GBM) model for the price of a risky asset, from which the huge financial derivatives industry has developed, stipulates that the log returns are iid Gaussian. however, typical log returns data show a distribution with much higher peaks and heavier tails than the Gaussian as well as evidence of strong and persistent dependence. In this paper we describe a simple replacement for GBM, a fractal activity time Geometric Brownian motion (FATGBM) model based on fractal activity time which readily explains these observed features in the data. Consequences of the model are explained, and examples are given to illustrate how the self-similar scaling properties of the activity time check out in practice.

  • A systematic methodology is developed for the electrofacies determination from wireline log data using multivariate statistical analysis. To consider corresponding contribution of each log and reduce the computational dimension, multivariate logs are transformed into a single variable through principal components analysis. Resultant principal components logs are segmented using the statistical zonation method to enhance the efficiency and quality of the interpreted results. Hierarchical cluster analysis is then used to group the segments into electrofacies. Optimal number of groups is determined on the basis of the ratio of within-group variance to total variance and core data. This technique is applied to the wells in the Korea Continental Shelf. The results of field application demonstrate that the prediction of lithology based on the electrofacies classification matches well to the core and the cutting data with high reliability This methodology for electrofacies classification can be used to define the reservoir characteristics which are helpful to the reservoir management.

다차원 임의 분할표 생성 (Generating Multidimensional Random Tables)

  • 로그선형모형에 기반을 둔 다차원 임의 분할표를 생성하는 방법을 제안하였다. 이를 위해 Lee(1997)가 제안한 선형 결합에 의한 결합분포 생성 방법을 적용하였으며, Pearson 통계량을 연관성 측도로 사용하는 것을 제안하였다. 세 변수가 서로 완전한 연관을 갖는 삼차원 결합분포를 생성할 수 있으므로 본 연구에서 제안한 방법은 사차원 이상 다차원 임의 분할표를 생성하는 문제로 확장될 수 있다.
