Distribution and diversity of rhizosphere bacteria of mixed halophytes vegetation native to the Goraebul sand dune, Korea : Approaches to coastal dune conservation (한반도 고래불 해안사구에 자생하는 혼합 염생식물군락 근권세균의 분포 및 다양성 고찰 : 해안사구 보전을 위한 접근)

  • Coastal dunes must be conserved. Their native halophytes support coastal geography while their symbiotic microorganisms help vegetation thrive. The Goraebul coast has the largest, well-conserved dune system on the East Sea of the Korean Peninsula due to a climax mixed halophyte (C. soldanella, C. kobomugi, and E. mollis) vegetation support. This study identified rhizobacteria and their diversity in mixed halophyte communities unique to Goraebul. Five phyla, 12 genera, and 21 species were identified based on 16S rDNA sequences from 65 isolates. The phylum Bacillota, class Bacillota, order Bacillales, and family Bacillaceae were identified, with Bacillus as the dominant genus (46.15%). The richness and Shannon's diversity were higher at the species than at the genus level due to the dominance of Bacillus; however, various Bacillus species (7) were identified. Therefore, the climax mixed vegetation adapted to the Goraebul coast may exert natural selection pressure in favor of the common characteristics of Bacillus. However, despite this advantage, the Shannon equitability (0.86), Simpson (0.08), and Shannon diversity (2.79) indexes indicate a stable rhizosphere cluster and the climax mixed vegetation is affected by symbiotic relationships between healthy rhizosphere microbiota.

Unrecorded Fungi Isolated from Rhizosphere Soil of Fallopia sachalinensis in Dokdo Islands (독도 왕호장근 근권 토양에서 분리된 미기록 균류)

  • We isolated fungi from the rhizosphere of Fallopia sachalinensis in Dokdo islands. Morphological and molecular characters, based on the internal transcribed spacer (ITS), and partial large subunit (LSU) or partial beta-tubulin genes, were used to identify the isolated fungi. The results revealed the fungi isolated from the Fallopia rhizosphere to be Penicillium striatisporum and Gongronella sichuanensis. Given that these species have never previously been recorded in Korea, we have described the morphological and molecular characteristics of these fungi in this study.

  • Numerous studies have established a correlation between sociodemographic characteristics and water usage, identifying population as a primary independent variable in mid- to long-term demand forecasting. Recent dramatic sociodemographic changes, including urban concentration-rural depopulation, low birth rates-aging population, and the rise in single-person households, are expected to impact water demand and supply patterns. This underscores the necessity for operational and managerial changes in existing water supply systems. While sociodemographic characteristics are regularly surveyed, the conducted surveys use aggregate units that do not align with the actual system. Consequently, many water demand forecasts have been conducted at the administrative district level without adequately considering the water supply system. This study presents an upward water demand forecasting model that accurately reflects real water facilities and consumers. The model comprises three key steps. Firstly, Statistics Korea's SGIS (Statistical Geological Information System) data was reorganized at the DMA level. Secondly, DMAs were classified using the SOM (Self-Organizing Map) algorithm to consider differences in water facilities and consumer characteristics. Lastly, water demand forecasting employed the PCR (Principal Component Regression) method to address multicollinearity and overfitting issues. The performance evaluation of this model was conducted for DMAs classified as rural areas due to the insufficient number of DMAs. The estimation results indicate that the correlation coefficients exceeded 0.9, and the MAPE remained within approximately 10% for the test dataset. This method is expected to be useful for reorganization plans, such as the expansion and contraction of existing facilities.

On-line Monitoring of the Flocs in Mixing Zone using iPDA in the Drinking Water Treatment Plant (정수장 응집혼화공정에서의 응집플럭 연속 모니터링)

  • This study evaluated the flocs forming characteristics in the mixing zone to increase the coagulation effect in the drinking water plant. As a measuring tool of formed flocs, on-line particle dispersion analyzer (iPDA) was used in Y drinking water plant. To evaluate the forming flocs, many parameters such as poly amine, coagulant dosing amount, raw water turbidity, and pH was applied in this study. During the periods of field test, poly aluminium chloride (PACl) as a coagulant was used. With the increase of the raw water turbidities, poly amine was also added as one of aids for increasing in coagulation efficiency. The turbidity and pH of raw water was ranged from 7 to 9 and from 25 to 140 NTU, respectively. The increasing of raw water turbidity brought the bigger floc sizes accordingly. From a regression analysis, $R^2$ value was 0.8040 as a function of T, raw water turbidity. Floc size index (FSI) was obtained from a correlation equation as follows; FSI = 0.9388logT - 0.3214 Also, polyamine gave the bigger flocs the moment it is added to the coagulated water in the rapid mixing zone. One of parameters influencing the floc sizes was the addition of powdered active carbon(PAC) in the mixing zone. In case of higher turbidity of raw water, $R^2$ value was 0.9050 in the parameters of [PACl] and [PAC]; FSI = $0.0407[T]^{0.324}[PACI]^{0.769}[PAC]^{0.178}$ On-line floc monitor was beneficial to evaluate the flocs sizes depending on the many parameters consisting raw water properties, bring the profitable basic data to control the mixing zone more effectively.

New Analytical Method to Identify Chromium Species, Cr(III) and Cr(VI), and Characteristic Distribution of Chromium Species in the Han River (한강수계해서의 크롬(III,VI) 종(species) 분포 및 분석방법 정립)

  • An adequate method to identify chromium separation, Cr(III) and Cr(VI), in water samples were studied by using High Performance Liquid Chromatography(HPLC) coupled with Inductively Coupled Plasma Mass Spectometer(ICP-MS) equipped with Dynamic Reaction Cell(DRC). The characteristic distribution of Cr(III) and Cr(VI) in the raw water taken at the six water intake stations in Seoul, was analyzed by the method developed by the authors. The chromium species separated by HPLC was isocratically conducted by using tetrabutylammonium phosphate monobasic(1.0 mM TBAP), ethylenediaminetetraacetic acid(0.6 mM EDTA) and 2% v/v methanol as the mobile phase. 5% v/v methanol was used as flushing solvent. A reactive ammonia($NH_3$) gas was used to eliminate the potential interference of $ArC^+$. Several Parameters such as solvent ratio, pH, flow rate and sample injection volume were optimized for the successful separation and reproducibility. Although it has been reported thai the separation sensitivity of Cr(III) is superior to that of Cr(VI), the authors observed Cr(VI) was more sensitive than Cr(III) when ammonia($NH_3$) gas was used as the reaction gas. It took less than 3 minutes to analyze chromium species with this method and the estimated detection limits were $0.061\;{\mu}g/L$ for Cr(III) and $0.052\;{\mu}g/L$, for Cr(VI). According to the results from the analysis on chromium species in the raw water of the six intake stations, the concentrations of Cr(III) ranged from 0.048 to $0.064\;{\mu}g/L$(ave. $0.054\;{\mu}g/L$) while that of Cr(VI) ranged from 0.014 to $0.023\;{\mu}g/L$(ave. $0.019\;{\mu}g/L$). Recovery ratio was very high($90.1{\sim}94.1%$). There were two or three times more Cr(III) than Cr(VI) in the raw water.

Effects of Activated Carbon Types and Service Life on Removal of Odorous Compounds: Geosmin and 2-MIB (활성탄 재질과 사용연수에 따른 Geosmin과 MIB 흡착특성)

  • Adsorption performance of odorous compounds such as geosmin and 2-MIB on granular activated carbon were evaluated in this study. The coal-based activated carbon was found more effective than other carbons in adsorption of geosmin and 2-MIB. The wood-based virgin activated carbon was less effective than coconut- and coal-based carbon in adsorption nevertheless having larger pore volume and specific surface area than others carbons. The maximum adsorption capacity(X/M) of coal-based activated carbon for geosmin and 2-MIB was $1.2\sim1.9$ and $2.1\sim2.6$ times larger than coconut- and wood-based virgin activated carbon, respectively. Carbon usage rate (CUR) of coal-, coconut- and wood-based virgin activated carbons for geosmin and 2-MIB were 1.72 and 1.44 g/day, 1.72 and 2.05 g/day and 2.12 and 1.90 g/day, respectively. In the evaluation of adsorption isotherm of geosmin and 2-MIB for coal-, coconut- and wood-based virgin activated carbons, k value of 2-MIB was lower than geosmin, It menas 2-MIB is more difficult to remove by activated carbon adsorption than geosmin. The relationship of max. adsorption versus total pore volume of coconut- and wood-based virgin and used activated carbon for geosmin and 2-MIB were $y=264,459\times-79,047(R^2=0.95)$, $y=319,650\times-101,762(R^2=0.93)$.

A Comparative Study on Thermal and Belt Press Dewatering for Waterworks Sludge Rduction (열 탈수와 벨트프레스 탈수장치의 현장적용에 따른 탈수성 비교연구)

  • The water content of dewatered cake produced from belt press dewatering equipment was about 75 wt% which was some high to handle it, so the equipment contained a limit at the economical and environmental aspect. The thermal dewatering equipment built as an alternative to overcome several problems was set up at the sludge treatment field and estimated some feasibility as comparison with the dewatering performance of belt press. First, dewatering properties of waterworks sludge was analyzed by monthly. The sludge of a water shortage season contained a high organic content which led to be difficult to dewater the cake, the other side the sludge of rainwater season was ease to dewater because of low organic content. According to the results to analysis the water content of dewatered cake produced from two equipments on the base of the seasonal dewatering properties, the water content of dewatered cake produced from thermal dewatering for sludge of water shortage season was $41.6{\sim}48.3$ wt% and $71{\sim}84$ wt% from belt press. In the case of rainwater season, the water content of dewatered cake produced from thermal dewatering was $34{\sim}37.7$ wt% and $57{\sim}70$ wt% from belt press. It was understood that thereduction of water content of cake by thermal dewatering was larger than belt press. The economical aspect for two equipments was evaluated on considering the reduction of cake treatment amount as the decrease of water content of cake. When putting the cost index of thermal dewatering into 100, belt press was 121. This meant that thermal dewater was more economical than belt press by about 20% in the side of construction and operation. In conclusion, thermal dewatering equipment was estimated by producing the low water content dewatered cake as well as being operated with low coat.

Water Quality Management of Simple Piped Water Supply System and Its Satisfaction of the Residents (簡易上水道의 水質管理實態와 給水利用住民의 滿足度)

  • For the purpose of inquiring into the actual conditions of water quality management of Simple Piped Water Supply(SPWS) and the satisfaction and attitudes of inhabitants about utilizing water-supply, this research was done through the questionnaires, the investigation of surrounding environment and water quality analysis in the Sangju city, Kyungsang-pookdo during 4 months from March to June, 1998. The fountainhead of SPES using ground water accounted for 65.3%, which was the highest rate in the group. Most of them was at least 10 years in the number of utilization year. 79.6% of them were exposed to many sorts of surrounding pollution origins. The examination of water was performed only through a test about water purification and 24.5% of them disinfection by chlorine also no residual chlorine was detected at all. All the waterworks did not have any education to the managers of water quality and 81.5% of them held physical examination. As a result of the water examination about SPWS, 65.3% of them were found incongruity and the valley and springing water accounted for higher than the underground water in the rate of incongruity. Looking into the details of the result in the water examination, a category of colon bacilli was ranked in the highest rate and the next one was general bacilli, nitric acid nitrogen and turbidity in order. In the satisfaction degree of the water quality, the satisfied accounted for 44.6%, and 29.2% each. Over the state of satisfaction about the ways of water examination, satisfaction accounted for 44.6%, and unsatisfaction 28.3%. Summarizing the result of the above-stated, the state of Water Quality Management of the SPWS was in a poor condition. Thus for the water Quality Management, systematic and scientific water Quality Management mainly by the administrative organization other than voluntary management by the village should be done beyond doubt. Additionally the opinions and demands of inhabitants utilizing the water supply have to be positively reflected in the affairs of water Quality Management so that the distrust of inhabitants to the SPWS should be settled.

Removal Characteristics of Chloral Hydrate by Activated Carbons and Biofiltration (활성탄과 생물여과 공정에서의 Chloral hydrate 제거 특성)

  • Coal-, coconut- and wood-based activated carbons and anthracite were tested to evaluate adsorption and biodegradation performances of chloral hydrate. In the early stage of the operation, the adsorption was the main mechanism for the removal of chloral hydrate, however as increasing populations of attached bacteria, the bacteria played a major role in removing chloral hydrate in the activated carbon and anthracite biofilter. It was also investigated that chloral hydrate was readily subjected to biodegrade. The coal- and coconut-based activated carbons were found to be most effective adsorbents in adsorption of chloral hydrate. Highest populations and activity of attached bacteria were shown in the coal-based activated carbon. The populations and activity of attached bacteria decreased in the order: coconut-based activated carbon > wood-based activated carbon > anthracite. The attached bacteria was inhibited in the removal of chloral hydrate at temperatures below 10$^{\circ}C$. It was more active at higher water temperatures(20$^{\circ}C$ <) but less active at lower water temperature(10$^{\circ}C$>). The removal efficiencies of chloral hydrate obtained by using four different adsorbents were directly related to the water temperatures. Water temperature was the most important factor for removal of chloral hydrate in the anthracite biofilter because the removal of chloral hydrate depended mainly on biodegradation. Therefore, the main removal mechanism of chloral hydrate by applying activated carbon was both adsorption and biodegradation by the attached bacteria. The observation suggests that the application of coalbased activated carbon to the water treatment should be the best for the removal of chloral hydrate.

Analysis of Bacterial Community Structure of Biological Activated Carbon Process in Drinking Water Treatment Plant Using FISH (FISH법을 이용한 정수처리장 내 생물활성탄 공정의 세균군집 구조 분석)

  • The bacterial community structure in biological activated carbon (BAC) process in drinking water treatment plant was investigated by Fluorescent in situ Hybridization (FISH) with rRNA-targeted oligonucleotide probe. Samples were collected at different three points in BAC process every month for one year. They were hybridized with a probe specific for the alpha, beta, gamma subclass of the class Proteobacteria, Cytophaga-Flavobacteria group and Gram-positive high G+C content (HGC) group. Total numbers of bacteria in BAC process counted by 4',6-diamidino-2-phenylindole (DAPI) staining were $5.4{\times}10^{10}$ (top), $4.0{\times}10^{10}$ (middle) and $2.8{\times}10^{10}$ cells/ml (bottom). The number of the culturable bacteria was from $1.0{\times}10^7$ to $3.6{\times}10^7$ cells/ml and the culturability was about 0.05%. The faction of bacteria detectable by FISH with the probe EUB338 was about 83% of DAPI counts. Gamma and alpha subclass of the class Proteobacteria were predominant in BAC process and their ratios were over 20% respectively. In top and middle, alpha, beta and gamma subclass of the class Proteobacteria competed with each other and their percentages was changed according to the season. In bottom, gamma subclass of the class Proteobacteria was predominant all through the year. It could be successfully observed the seasonal distribution of bacterial community in biological activated carbon process using FISH.
