Ginkgo Terpene의 Candidate albicans로 인한 관절염에 대한 치료효과 (Therapeutic Anti-inflammatory Effect of Ginkgo Terpene on Arthritis due to Candida albicans)

  • Candida albicans, a polymorphic fungus, causes systemic and local infections. Recent reports show that the fungus is a main etiological agent for the arthritis. For trea tment, antifungal drugs and/or rheumatoid drugs are used, but resistance and side effects limit application of the drugs. In search of new sources for treatment of the fungal arthritis, we choose Egb 761 (extract of Ginkgo leaves 761), one of the most popular over-the-counter herbal medicines. The Egb 761 contains two major ingredients such as terpene and flavonoid. In the present study, we examined if the terpene portion of Egb 761 had anti-inflammatory activity against C.albicans-caused arthritis. The terpene was extracted with combination of methanol and water from the Egb 761, followed by gel-permeation chromatography. Presence of terpene was determined by the Salkowski colorimetric method and HPLC analysis. For an animal model of inflammation induction, mice were given an emulsion form of C.albicans cell wall mixed with Complete Freund's Adjuvant (CFA) by footpad-injection. Results showed that intraperitoneal administration of the water-soluble portion that contained terpene and flavonoid reduced the inflammation. Whereas the terpene had anti-inflammatory activity, flavonoid portion had no such activity, For determination of possible mechanism of the activity, the terpene seemed to be suppression of nitric oxide (NO) production from LPS-treated macrophages. Taken together the Ginkgo terpene may have anti-inflammatory effect against C.albicans-caused arthritis, possibly by blocking NO production.

PREDICTING MALTING QUALITY IN WHOLE GRAIN MALT COMPARED TO WHOLE GRAIN BARLEY BY NEAR INFRARED SPECTROSCOPY

  • Predicting quality traits using near infrared (NIR) spectroscopy on whole grain samples has gained wide acceptance as a non-destructive, rapid and cost effective technique. Barley breeding programs throughout southern Australia currently use this technology as a tool for selecting malting quality lines. For the past 3 years whole grain barley calibrations have been developed at VIDA to predict malting quality traits in the early generation selections of the breeding program. More recently calibrations for whole grain malt have been developed and introduced to aid in selecting malted samples at the mid-generation stage for more complex malting quality traits. Using the same population set, barley and malt calibrations were developed to predict hot water extracts (EBC and IoB), diastatic power, free $\alpha$-amino nitrogen, soluble protein, wort $\beta$-glucan and $\beta$-glucanase. The correlation coefficients between NIR predicted values and laboratory methods for malt were all highly significant ($R^2$ > 0.84), whereas the correlation coefficients for the barley calibrations were lower ($R^2$ > 0.57) but still significant. The magnitude of the error in predicting hot water extract, diastatic power and wort $\beta$-glucan using whole grain malt was reduced by 50% when compared with predicting the same trait using whole grain barley. This can be explained by the complex nature of attempting to develop calibrations on whole grain barley utilizing malt data. During malting, the composition of barley is modified by the action of enzymes throughout the steeping and germination stages and by heating during the kilning stage. Predicting malting quality on whole grain malt is a more reliable alternative to predicting whole grain barley, although there is the added expense of micro-malting the samples. The ability to apply barley and malt calibrations to different generations is an advantage to a barley breeding program that requires thousands of samples to be assessed each year.

Newly identified maltol derivatives in Korean Red Ginseng and their biological influence as antioxidant and anti-inflammatory agents

  • Background: Korean Red Ginseng is a major source of bioactive substances such as ginsenosides. Efficacy of red ginseng extract (RGE), which contains not only saponins but also various non-saponins, has long been studied. In the water-soluble component-rich fraction of RGE (WS), a byproduct generated in the process of extracting saponins from the RGE, we identified previously unidentified molecules and confirmed their efficacy. Methods: The RGE was prepared and used to produce WS, whose components were isolated sequentially according to their water affinity. The new compounds from WS were fractionized and structurally analyzed using nuclear magnetic resonance spectroscopy. Physiological applicability was evaluated by verifying the antioxidant and anti-inflammatory efficacies of these compounds in vitro. Results: High-performance liquid chromatography confirmed that the obtained WS comprised 11 phenolic acid and flavonoid substances. Among four major compounds from fractions 1-4 (F1-4) of WS, two compounds from F3 and F4 were newly identified in red ginseng. The analysis results show that these compound molecules are member of the maltol-structure-based glucopyranose series, and F1 and F4 are particularly effective for decreasing oxidative stress levels and inhibiting nitric oxide secretion, interleukin (IL)-1β and IL-6, and tumor necrosis factor-α. Conclusion: Our findings suggest that a few newly identified maltol derivatives, such as red ginseng-derived non-saponin in the WS, exhibit antioxidant and anti-inflammatory effects, making them viable candidates for application to pharmaceutical, cosmetic, and functional food materials.

Hericium erinaceum 액체배양 생성물의 Acetyl-cholinesterase 저해 활성과 항산화 활성 (AChE Inhibitory Effect and Antioxidative Activity of Submerged Cultured Products from Hericium erinaceum)

  • H. erinaceum은 현재 산업적으로 매우 주목되고 있는 기능성 소재들 중의 하나이다. 본 연구에서는 식용은 물론 각종 약리효과를 나타내는 H. erinaeum의 기능성 식품의 제품화 연구 일환으로, 지금까지 체계적으로 연구된 바 없는 H. erinaceum의 액체배양을 시도하고, 이 버섯의 액체배양 생성물의 acetylcholinestrase (AChE) 저해활성과 항산활성의 효과를 자실체와 비교하면서 평가, 규명하고자 하였다. Electrophorous electricus 유래 AChE에 대한 저해 활성을 조사한 결과, 자실체 및 배양 생성물 (균사체와 배양여액)은 10 mg/ml 농도에서 75$\sim$85%의 저해활성을 보였고, 저해기작은 모두 일반적 비경쟁적 저해 (general non-competitive inhibition)에 의하였다. 특히, 배양여액의 ethanol 침전 상징액은 10 mg/ml의 농도에서 약 94%의 매우 높은 저해활성을 보였다. 한편, linoleic acid를 기질로 한 지질과산화에 대한 노루궁뎅이 버섯 유래 시료들 (자실체와 균사체 추출물, 균사체 추출물의 ethanol 침전물 및 배양여액)의 억제능을 Rhodan-Fe (FTC)법으로 조사한 결과, 균사체와 자실체 추출물은 0.1 mg/ml의 농도에서 지질의 초기 과산화를 완전히 억제하였다. 또 TBA-RS로 조사한 경우도 항산화 표준품으로 사용한 tocopherol, vitamin C 및 rutin보다 우수하였고, 시료들 간의 큰 차이 없이 약 95%의 매우 우수한 지질과산화 억제율을 나타내었다.

오미자의 Schizandrin과 기름성분 및 추출수율 변화 (Schizandrin, Oil Compounds, and Their Extraction Yield in Fruits of Schizandra chinensis Baillon)

  • 오미자 인제수집종의 과실부위별 schizandrin과 기름함량은 종자에서 가장 높았는데 각각 1.01%와 27.6%로 나타났다. 국내 수집종 오미자 55계통에 대한 성분분석 결과, 과실에 함유된 schizandrin의 함량은 평균 0.84%이었고, 종자에 함유된 기름함량은 평균 27.9%이었으며 지방산조성은 palmitic acid 3.6%, stearic acid 0.6%, oleic acid 19.7%, linoleic acid 73.1%, linolenic acid 3.0%로 불포화지방산조성이 95.8%로 높게 나타났다. 인제수집종 종자에서 용매추출한 기름은 4.29%의 schizandrin을 함유하여 오미자의 schizandrin은 종자기름에 대부분 함유되었다. 분쇄하지 않은 종자와 과실을 끓는 물로 추출하였을 경우 schizandrin수율은 각각 23.8%와 17.3%로 낮게 나타났다. 그리고 4개월 동안 물에 침지된 종자내 schizandrin과 기름성분은 상온 저장된 종자와 비슷한 함량인 1.18%와 25.2%를 나타내, 오미자 음료제조 후 부산물인 종자는 기름이나 지용성 약리성분을 추출할 수 있는 원료로 이용될 가능성이 있었다.

손바닥 선인장의 항산화활성 및 분말 첨가 쿠키의 품질특성 연구 (The Antioxidant Activity of Korean Cactus (Opuntia humifusa) and the Quality characteristics of Cookies with Cactus Powder added)

  • In order to better understand the quality of Korean cactus, Opuntia humifusa, as the functional foodstuffs, it's seeds, stems and fruits were analyzed. In addition, we performed quality evaluation on cookies manufactured with cactus powder added. The results showed the soluble nitrogen-free component levels in the cactus to be very high. The highest component levels were crude fiber(48.75%) and crude fat(7.38%) in the seeds, crude ash(15.39%) in the stems, and moisture(11.26%) and crude protein(8.79%) in the fruits. The antioxidant activities of the cactus were evaluated based on peroxide values(POV), thiobarbituric acid reactive substances(TBARS value), and electron donating ability(EDA) using the DPPH method. The POVs for soybean oil containing a 0.05% concentrated water extract from each part of the cactus revealed a remarkable antioxidant effects on peroxidation as storage time increased. The TBA values were 63.65% in BHT, 60.67% in the stems, 57.31% in the fruits and 49.16% in the seeds. The EDAs were 30.26% in the stems, 25.89% in the fruits, and 25.65% in the seeds. The total polyphenol contents of the water soluble materials from the cactus were 3.14 g/100 g in the fruits, 2.93 g in the stems and 2.11 g in the seeds. For the cookies, as the amounts of fruit and stem powder increased, the spreadness was significantly lowered, but moisture content was increased compared to the control. The color degree of the cookies changed from dim to intense as more fruit powder was added, resulting from a decrease in the L- and b-values and a significantly increased a-value. With the addition of stem powder, the L- and a-values revealed no significant differences, while the b-value was lowered compared to the control as more stem powder added. For texture changes of the fruit cookies, there were significant differences in hardness, gumminess, cohesiveness, springiness and brittleness. The stem cookies, however, showed only differences in hardness, gumminess, and brittleness, and not in cohesiveness and springiness. Finally, an overall sensory preference was found with the 1% fruit powder and 3% stem powder additions.

  • 건조굴 가공시 생굴의 수세${\cdot}$자숙과정에서 아황산 소오다로서 처리하여 건조굴은 저장할 때 일어나는 갈변에 대한 영향을 검토하고 항산화제를 첨가한 효과를 측정한 결과를 요약하면 다음과 같다. 1. 아황산소오다의 침지농도별, 침지시간별 처리효과를 비교하면 전보에서 얻은 생굴중의 환원당과 tyrosine의 제거효과와 같이 건조굴의 저장후에도 0.5M 용액에서 6분간 침지한 후, 60분간 수세한 것의 효과가 가장 좋았다. 침지용액을 산성으로 조절하는 것도 도움이 되었다. 2. 저장중의 TBA치, 지방의 산화적 갈변 및 당아미노 반응에 의한 갈변도의 추세를 검토해 보면 지방산화적 갈변이 주도적이 였고 당-아미노반응 또한 완만하게 특속적으로 진행되고 있으며 두 반응은 서로 관련하고 있어 아황산염의 처리가 지방의 산화적 갈변에도 영향을 미치고 있다. 3. 황산화제의 첨가는 주도적인 갈변요인인 지방의 산화를 억제함은 물론, 아황산염의 처리와 항산화제처리를 병행하였을 때 효과는 상승적으로 나타나 0.5M 아황산소오다 처리와 항산화제를 병용했을 때가 가장 효과가 좋았다.

Anti-thrombic Properties of the Oriental Herbal Medicine, Daejowhan

  • The anti-thrombic properties of the oriental herbal medicine Daejowhan(DJW, 大造丸) which consists of 11 kinds of herbs (indicated as ratio) of Rehmanniae Radix 24%, Hominis Placenta 5%, Testudinis Carapax 9%, Eucommiae Cortex 9%, Asparagi Radix 9%, Phellodendri Cortex 9%, Achyranthis Radix 7%, Liriopis Tuber 7%, Angelicae Sinensis Radix 7%, Ginseng Radix 5% and Schizandrae Fructus 3% were investigated. The water extracts from DJW inhibited Platelet-activating factor(PAF) induced platelet aggregation. DJW was extracted with methanol and further fractionated by ethylacetate. A 70% methanol extract showed a strong inhibition against PAF-induced aggregation in vitro and in vivo assays. The ethylacetate soluble fraction was shown to have inhibitory effect on PAF-induced platelet aggregation in vitro assay. The ethylacetate soluble fraction specially protected against the lethality of PAF, while verapamil did not afford any protection. These results indicate that the water extracts and alcoholic-fractions inhibit the action of PAF in vivo by an antagonistic effect on PAF, so that it may be useful in treating disorders caused by PAF, such as acute allergy, inflammation, asthma, gastrointestinal ulceration, toxic shock and so forth. DJW was investigated regarding its assumed anti-thrombic action on human platelets which was deduced from its ability to suppress Arachidonic acid(AA)-induced aggregation, exocytosis of ATP, and inhibition of Cyclooxygenase(COX) and Thromboxane synthase(TXS) activity. The latter two effects were estimated from the generation of Prostaglandin $E_2(PGE_2)$ and Thromboxane $A_2(TXA_2)$ respectively. Exogenously applied AA ($100{\mu}mol/{\ell}$) provoked a $89\%$ aggregation of platelets, the release of 14 pmol ATP, and the formation of either 225 pg $TXA_2$ or 45 pg $PGE_2$, each parameter being related to 106 platelets. An application of DJW 5 min before AA dose-dependently diminished aggregation, ATP-release and the synthesis of $TXA_2$ and $PGE_2$ with $IC_{50}$ values of 74, 108, 65, $72{\mu}g/m{\ell}$, respectively. The similarity of the $IC_{50}$ values suggest an inhibition of COX by DJW as primary target, thus suppressing the generation of $TXA_2$ which induces aggregation of platelets and exocytosis of ATP by its binding on $TXA_2$-receptors.

창출 성분의 장관면역 자극을 통한 골수세포 증식활성 (Bone Marrow Cell Proliferation Activity through Intestinal Immune System by the Components of Atractylodes lancea DC.)

  • 십전대보탕의 10가지 구성 천연물을 열수추출하여 장관면역 활성을 비교 검토한 결과, 창출(Atractylodes lancea DC., ALR)과 인삼(Panax ginseng C.A. Meyer, PG)에서는 높은 활성을 나타내었으나 약한 활성을 갖는 황기(Astragalus membranacues, ASR)와 당귀(Angelica aculiloba Kitagawa, AR)를 제외하고는 어떤 생약에서도 거의 활성을 갖지 않았다. 한편 이러한 활성을 가진 생약(ALR, PG와 ASR) 중에서도 특히 ALR만이 종류, 생산지 및 육종조건에 관계없이 항상 일정하게 높은 활성을 보여주어 창출의 열수추출물 획분(ALR-0)이 십전대보탕의 장관면역 활성에 중요하게 관여하는 시료로 선정되었다. ALR-0 획분은 다시 용매별로 분획되어 메탄올-가용성 획분(ALR-1), 메탄올-불용성/에탄올-가용성 획분(ALR-2) 및 조다당 획분(ALR-3)으로 조제되었으며 이들의 활성을 측정한 결과, 단지 ALR-3 획분만이 Peyer's patch를 통한 골수세포의 증식을 활성화시키는 장관면역 활성을 시료농도에 의존적으로 나타내었으며 그 외의 획분에는 활성이 거의 없었다. 한편 ALR-3 획분을 $NaIO_4,\;NaClO_2$ 및 pronase로 처리한 후 활성을 측정한 결과, 모든 처리군에서 활성이 감소되었으며 특히 periodate 산화에 의해 심한 활성감소를 보였다. 이러한 결과를 토대로 십전대보탕의 장관면역 활성은 10종의 생약 중에서도 특히 창출이 중요하게 활성에 관여하는 것으로 보이며 다양한 용매별 분획을 통해 저분자보다는 에탄올에 침전된 고분자가 주요 함유물질인 조다당 획분이 장관면역 활성에 관여하는 것으로 추정되었다. 또한 periodate 산화에 의해 활성이 크게 감소되는 것으로 보아 주로 다당류가 창출의 장관면역에 중요한 활성물질인 것으로 생각된다.

백삼의 건조와 가용성 성분 추출에 미치는 pulsed electric fields(PEF)의 효과 (Pulsed Electric Fields Effects on Drying of White Ginseng and Extraction of Soluble Components)

  • 식품의 가공공정에서 투입되는 에너지의 절감을 위한 방안으로 대표적이 농산물 건조제품인 인삼의 건조과정에서 PEF를 처리함으로써 건조효율에 미치는 영향과 PEF를 처리하여 건조한 인삼인 백삼의 열수 추출과정에서의 추출효율에 미치는 영향을 분석하였다. 4년근 수삼을 2 kV/cm, 200 Hz, 7초의 조건으로 PEF를 처리하여 $55{\pm}1^{\circ}C$에서 건조한 결과 건조시간을 약 38% 단축시킬 수 있는 결과를 얻었다. 건조한 백삼을 $95{\pm}1^{\circ}C$로 추출한 결과 무처리에 비해 2 kV/cm, 25 Hz, 7초간 PEF 처리한 건조 백삼, 2 kV/cm, 200 Hz, 7초간 PEF 처리하여 건조한 백삼의 열수 추출물의 고형분 함량, 환원당 함량, 유리당 함량 등에서는 유의적인 차이를 나타내었으나 당도와 진세노사이드 함량에서는 부분적인 유의적 차이만을 나타내었다. 이러한 결과를 감안할 때 수삼에 PEF를 처리함으로써 건조시간을 효과적으로 단축시킬 수 있고 추출시에도 가용성 고형분의 추출시간을 단축할 수 있는 유용한 수단이 될 수 있을 것으로 판단된다.
