A Study on Establishment of the Fermentation Process for Traditional Andong Sickhae (전통 안동식혜의 제조공정 확립에 관한 연구)

  • The fermentation process of Andong Sickhae including optimal composition of the product was investigated. Through sensory evaluation and ingredient analysis of ten samples prepared in the laboratory and the samples collected from Andong region, we found that the ratio of ingredient of the product by weigh are as follow : [glutinous rice(80) : malt(50) : radish(100) : water(500) : ginger(8) : red pepper(4)]. The level of nitrogen compound turned out to be low while that of soluble protein and salt soluble protein was high. The content of total sugar and reducing sugar was found to be considerable high and among the free sugar, maltose was the highest(80%), followed by glucose and maltotriose. Activities of acid protease and saccharogenic amylase were 1.55 unit per milliliter and $12.5D^{40}_{30}^{\circ}$ respectively. Results of sensory evaluation showed that the good Andong Sickhae turned out to have well harmonized taste of flavor, sweetness and sourness while the color looked slightly red.

  • Oranges (Citrus unshiu var.) from local orchards at Seogwi, Daejeong, Hanrim, Cheju and Seongsan where are in the southern, southwestern, northwestern, northern and eastern parts of Cheju island respectively were stored in a semi-underground room and an air-circulating refrigerated store. To compare the storage qualities between the growing districts, changes in freshness, acidity and sugar content of oranges were investigated during storage. 1) Oranges from Seogwi and Cheju districts showed remarkably lower Percentage of rot fruits with no relation of storage temperature than those of Hanrim, Daejeong and Seongsan. 2) Browning of oil gland was severe on oranges of northern and southern districts while it was little developed in case of the other districts. 3) Water soluble total sugar content was gradually increased and titratable acidity and reducing sugar content of pulp tissues was decreased during storage. 4) Oranges from the western part showed the higher ratio of sugar content to acidity and better taste as compared with the east oranges.

  • The purpose of this study is to examine material performance of fiber reinforced cement composite for mass production. It is necessary to manufacture SHCC(Strain Hardening Cement Composite) by batch plant for field application and mass production. For the study, a mock-up test of SHCC manufactured in the batch plant was conducted, and the performance was compared with SHCC manufactured in the laboratory. Assessment items were freshness and hardening properties. Specifically, direct tensile test machine was used for performance verification of SHCC. As a result, there was a tendency of less satisfactory fiber dispersion and performance of strain hardening compared with the performance of SHCC manufactured in the laboratory. To address this, dry mixing and mortar mixing time should be increased compared to laboratory mixing, and injection time of an agent such as a water reducing agent should be properly controlled according to mixing combination, or the capacity to secure dispersion and homogeneity of material.

Engineering Characteristics of the Light Weight Soil Using Phosphogypsum and EPS Beads (인산석고-EPS 조각을 활용한 경량혼합토의 공학적 특성)

  • The current study developed light-weighted mixed soil that can solve problems related with soft soil such as ground subsidence, sliding and lateral displacement of ground. By reducing weight of reclaimed soil through mixing phosphogypsum and recycled EPS beads with the weathered granite soil. A series of geotechnical laboratory tests including physical index test, compaction test, CBR test, and direct shear test were performed and engineering properties were reviewed in order to assess applicability of the light-weighted mixed soil for roads and abutment and various back-filling materials at the reclamation area. Based on the laboratory test results, it was found that the maximum dry unit weight of the light-weighted soil ranges $14.32{\sim}15.79kN/m^3$ and the optimum water content ranges 21.91~24.23%, which means there is 11~19.3% weight decrease effect when comparing with general weathered granite soil. Also it was found that the corrected CBR value ranges 10.4~18.4% satisfying the domestic regulations on road subgrade and back-filling material. In addition, as for shear strength parameter, cohesion ranges 10.79~18.64 kPa and internal frictional angle ranges $35.4{\sim}37.2^{\circ}$, which are similar with those of general construction soil and back-filling material used in Korea. So it can be concluded that light-weighted mixed soil with phosphogypsum can be used effectively for soft reclamation ground as actual filling material and back-filling material. From the current study, it was found that light-weighted mixed soil with phosphogypsum has not only weight reduction effect, but also has no special problems in shear strength and bearing capacity. Therefore, it is expected that phosphogypsum can be recycled in bulk as road subgrade and back-filling material at the reclamation area.

  • A muti-lamellar emulsion containing a pseudoceramide, N-Ethanol-2-myristyl/ palmityl-3-oxostearmide/arachidamide(PC-9) has been prepared and its efficacy evaluation has been investigated. In order to prepare a muti-lamellar emulsion, first, the gram ratios of PC-9, fatty acid and cholesterol on the phase diagram to be capable of forming their lamellar liquid crystal structures were determined and secondly, the multi-lamellar emulsion was preprared using glyceryl monostearate and polyoxyethylene glyceryl monosteartate as emulsifers together with above mentioned pseudo-stratum corneum lipid components. Besides natural oils such as olive oil had a tendency to build up the multi-lamellar emulsion. And according as the amount of oil increased in the emulsion, it was observed that the optical anisotropy of “Maltese Cross” which was a typical configuration of multi-lamella mesophase texture diminished. In the dried state of the multi-lamella emulsion, it was examined to transform its emulsion phase into a lamella liquid crystal one. And finally, when the emulsion was applied into a human skin, it was investigated that it had effectiveness in reducing transepidermal water loss (TEWL) of the skin.

Characteristics of Ice Wine Fermentation of Freeze-Concentrated Campbell Early Grape Juice by S. cerevisiae S13 and D8 Isolated from Korean Grapes (포도로부터 분리한 S. cerevisiae S13 및 D8에 의한 캠벨 얼리 동결농축 과즙의 아이스와인 발효 특성)

  • Cryoextraction (a freeze concentration using an instrument) can increase the sugar concentration in grape juice by reducing its water content, similar to the natural freezing of grapes for natural ice wine. In this study, fermentation of freeze-concentrated Campbell Early grape (Vitis labruscana) juice to 36 $^{\circ}Bx$ was carried out using Saccharomyces cerevisiae strains D8 and S13 isolated from Korean grapes. During the fermentation, strains S13 and D8 showed rapid sugar reduction and alcohol production compared with S. cerevisiae Fermivin$^{(R)}$ used as a control. After nine-day fermentation, the residual sugar contents were lower in W13(9.77%) and D8 wine(9.07) than that in Fermivin$^{(R)}$ wine(14.0%). Total acid content was high in the D8>S13>Fermivin$^{(R)}$ wine, in that order. The acetaldehyde content was highest in the D8 wine and lowest in the Fermivin$^{(R)}$ wine, among the three. The methanol content was slightly higher in the S13 and D8 wines than in the Fermivin$^{(R)}$ wine. In the sensory evaluation, the S13 wine exhibited the highest score in flavor and taste among the three wines. Both the two S13 and D8 wines exhibited higher scores than Fermivin$^{(R)}$ wine in overall preference.

Inflammatory mediator regulation of the Zizyphus jujube leaf fractions in the LPS-stimulated Raw264.7 mouse machrophage (LPS로 염증이 유도된 Raw 264.7 대식세포에서 대추(Zizyphus jujube) 잎 분획물의 염증매개물질 억제)

  • Zizyphus jujube leaf fractions (ZLFs) showed no cytotoxic effects of up to $100{\mu}g/mL$, while the anti-inflammatory effects of ZLFs were analyzed by checking the productions of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), cyclooxygenase-2 (COX-2), and inflammatory cytokines TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 in the lipopolysaccharide (LPS)-stimulated Raw264.7 macrophage up to the concentration of $100{\mu}g/mL$. ZLFs ($100{\mu}g/mL$) demonstrated a strong anti-inflammatory activity that reduced 61~85% of NO and 71~100% of $PGE_2$ production in the LPS-stimulated Raw264.7 macrophage. Even the low ZLFs concentration of $1{\mu}g/mL$ have reduced NO and $PGE_2$ production by 34~64%. Expressions of COX-2 protein were also effectively inhibited by the ZLFs. Furthermore, the TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 production were significantly suppressed through the treatment of ZLFs at concentrations of 1, 10, and $100{\mu}g/mL$. In the order of the Zizyphus jujube leaf water fraction (ZLWF) < buthanol fraction (ZLBF) < ethyl acetate fraction (ZLEF) showed anti-inflammatory activity. In particular, the ethyl acetate fraction ZLEF at $100{\mu}g/mL$ showed an excellent anti-inflammatory activity by reducing the production of NO, $PGE_2$, COX-2, and inflammatory cytokines (TNF-${\alpha}$, IL-$1{\beta}$, IL-6) in the level of Raw264.7 macrophage without LPS-stimulation or even better. The results of our study suggest the potential of ZLEF for use as an excellent ant-inflammatory inhibiting mediator and may be used as a therapeutic approach to various inflammatory diseases.

Quality characteristics of Doenjang prepared with sweet potato (고구마를 이용한 된장의 품질 특성)

  • The effect of sweet potato on the quality of Doenjang was investigated during fermentation. Viable cells of yeast decreased gradually after 4 weeks of fermentation, but those of aerobic bacteria increased in the late stage. Amylase activity of Doenjang was higher in the late stage of fermentation, while neutral protease maintained high activity during fermentation. Hunter L and b values of Doenjang decreased gradually during fermentation, while a value was increased. The pH of Doenjang decreased gradually until 10 weeks of fermentation, and the titratable acidity was low in the sweet potato added groups. The acid value was low in the Shinyulmi sweet potato added Doenjang. Water activity and oxidation-reduction potential of Doenjang decreased during fermentation. Reducing sugar of Doenjang decreased in the middle stage of fermentation, and it was low in sweet potato added groups. The alcohol content of Doenjang decreased after 2 weeks of fermentation. Amino and ammonia-type nitrogen of Doenjang increased during fermentation and reached the maximum after 10 and 12 weeks of fermentation, respectively. After 12 weeks fermentation, 8% of Shinyulmi sweet potato added Doenjang was more favorable taste, flavor and overall acceptability (p<0.05) than the control or the Yeonwhangmi sweet potato added groups.

Physicochemical Properties and Nutritional Components of Fermented Black Jujube (발효 흑대추의 이화학적 특성 및 영양성분)

  • The physicochemical properties and nutritional components of fermented black jujube (FBJ) were compared with those of dried jujube (DJ) to improve their utilization as food materials. The moisture contents and brix of the DJ and FBJ were 22.6% and 6.07 brix, and 10.36% and 7.23 brix, respectively. Their L values were 34.78 and 31.82, and the a and b values of DJ were higher than those of FBJ. DJ had a lower water activity level (0.75) than FBJ (0.45), but FBJ had a higher amount of reducing sugar and soluble protein than DJ. The major free sugars in the two types of jujube were fructose and glucose, and their potassium content was highest (about 90%) among minerals. Their unsaturated fatty acid contents were 51.0% and 59.83%, respectively, and the major fatty acids myristoleic acid, palmitic acid, and palmitoleic aid were detected in them.

Soju brewing characteristics of yeast strains N4 and N9 isolated from Korean traditional Nuruk (재래누룩에서 분리한 N4와 N9 효모의 증류식 소주 양조특성)

  • Soju is a Korean traditional distilled alcoholic beverage produced from mashes various crops and Nuruk which is cultured with wild microorganisms. This study was conducted to investigate rice-Soju brewing characteristics of yeasts isolated from Korean traditional Nuruk. The general components of rice (Hanarumbyeo) raw materials were 14.7 g of water, 6.8 g of crude protein, 0.9 g of crude lipid, 0.4 g of crude ash, and 76.5 g of carbohydrate in 100 g. Saccharifying and proteolytic activities in Hanarumbyeo ipguk (solid-state culture of Aspergillus luchuensis) were also determined. The alcohol content of the fermented wash from isolates was 15.37-16.58% (v/v), which is 16.7-36.0% higher than that of industrial yeasts (12.33-13.19%). Reducing sugar contents were 2.04-3.92 and 7.92-8.78 g/100 mL in the isolates and industrial yeasts, respectively. The isolated yeasts showed 25.2-52.7% higher yield of distillates (41% alcohol) compared to industrial yeasts. Forty-one components were detected in the rice distillated Soju (25% alcohol) and principal component analysis revealed differences between the isolated and industrial yeasts with respect to the contents of i-BuOH, isobutanal diethyl acetal, ethyl caprate, and tetradecanoic acid.
