Synthesis of Glycosides by Transglycosylation of $\alpha$-Amylase from Soluble Starch in Water-Organic Two Phase System (전분을 기질로 한 이상계에서 Amylase의 당전이반응에 의한 배당체의 합성)

  • Benzylalcohol-$\alpha$-glucoside (BG) was synthesized from soluble starch by transglycosylation of $\alpha$-amylase. Transglycosylation in water-organic two phase system containing 1% soluble starch as a glycosyl donor, 90% benzylalcohol as a glycosyl acceplor, 10% citrate buffer solulion (0.1 M, pH 5.0), and 10 unit of $\alpha$-amylase (Aspergilllw oryzae) was showed highcst efficiency. About 4 mg BG was obtained from 10 mg starch in reaction for 80 hrs at $40^{\circ}C$. Initially benzylalcohol-$\alpha$-maltoside Q3M) was major product, but as the reaction proceeded, it was hydrolyzed to glucose and BG. Finally the product of transglycosylation by $\alpha$-amylase was only BG. The both products did not show reducing powcr and hydrolyzed by $\alpha$-glucosidase and $\alpha$-amylase, respectively. The molecular wcights of both were estimated to be 270 and 432 by ES1-Mass, respectively.

Preparation and Characterization of Sulfonated Poly(phthalazinone ether sulfone ketone) (SPPESK)/Silica Hybrid Membranes for Direct Methanol Fuel Cell Applications

  • Sulfonated poly(phthalazinone ether sulfone ketone) (SPPESK) membranes and sol-gel derived SPPESK/silica hybrid membranes have been investigated as potential polymer electrolyte membranes for direct methanol fuel cell (DMFC) applications. In comparison with the SPPESK membrane, the SPPESK/silica membranes exhibited higher water content, improved proton conductivity, and lower methanol permeability. Notably, the silica embedded in the membrane acted as a material for reducing the fraction of free water and as a barrier for methanol transport through the membrane. From the results of proton conductivity and methanol permeability studies, we suggest that the fractions of bound and free water should be optimized to obtain desirable proton conductivities and methanol permeabilities. The highly sulfonated PPESK hybrid membrane (HSP-Si) displayed higher proton conductivity (3.42 ${\times}$ 10$^2$ S/cm) and lower methanol permeability (4.15 ${\times}$ 10$\^$7/ $\textrm{cm}^2$/s) than those of Nafion 117 (2.54 ${\times}$ 10$^2$ S/cm; 2.36 ${\times}$ 10$\^$6/ $\textrm{cm}^2$/s, respectively) at 30$^{\circ}C$. This characteristic of the SPPESK/silica membranes is desirable for future applications related to DMFCs.

Effects on Skin Irritation and Turnover Rate by the Control of Skin Permeability of Alpha-hydroxyacids

  • The effect of a novel delivery system, water in oil emulsion containing chitosan hydrogel as a inner phase (W/O-C) was evaluated, and the relationships between the skin permeation, the skin primary irritation and the skin turnover rate of AHAs were discussed. We selected glycolic acid (GA), lactic acid (LA), malic acid (MA), and tartaric acid (TA) as model AHAs. The steady state fluxes of 4 AHAs across the excised hairless mouse skin increased as the molecular weights of the AHAs decreased. (GA>LA>MA>TA). The skin turnover times were shortened in all AHAs, compared with control. The skin permeation and the skin primary irritation of the LA decreased and the skin turnover time increased, as the pH increased. The maximum therapeutic index was obtained with pH 3.8, 0.5 M LA. It was suggested that the skin permeability of LA might be a main factor for prediction of the skin irritation and the skin turnover time. On the other hand, the W/O-C containing pH 3.8, 0.5 M LA indicated a good sustained release property of LA, compared with water in oil emulsion without chitosan hydrogel (W/O) or oil in water emulsion (O/W). The skin permeability and the skin irritation of AHAs from the W/O-C edcreased, compared with W/O or O/W, however the skin turnover time showed almost the same value as W/O or O/W. In conclusion, we suggest that the control of the skin permeation of AHAs would be an important tool for reducing the skin irritation and for maintaining the positive effect of AHAs, and the W/O-C system could be a potential candidate for future cosmetological application of AHAs.

Antioxidative capacity of hydrolyzed rapeseed cake extract and oxidative stability of fish oil-in-water emulsion added with the extract

  • Rapeseed cake was extracted with 80% ethanol and then fractionated with $H_2O$ (fraction I) as well as with 30% (II), 50% (III), 70% (IV), and 100% ethanol (V). Total phenolic content (TPC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, ferric-reducing antioxidant potential, and Trolox equivalent antioxidant capacity were in the order of fractions II > III > I > IV > V. The three fractions with high antioxidant activities and TPC (I, II, and III) were pooled and hydrolyzed by NaOH solution, resulting in 18.97 mg sinapic acid/g hydrolyzed extract and 21- and 2.2-fold increases in TPC and DPPH radical scavenging activity, respectively. Hydrolyzed rapeseed cake extracts (200, 500, and 1,000 ppm) and catechin (200 ppm) as a comparison were added to 10% fish oil-in-water emulsion, and their effects on oxidative stability were investigated by measuring hydroperoxide values (PV) during refrigerated storage. PVs were significantly lower in the emulsions with added hydrolyzed extract as compared to the control (p<0.05) and significantly decreased with increasing extract concentration (p<0.05) over a period of 29 days. The emulsion added with hydrolyzed extract showed higher PV than that added catechin at the same concentration (200 ppm) during 13-22 days (p<0.05), but after then, the PV was not significantly different (p>0.05). This study indicates that hydrolyzed rapeseed cake extract rich in sinapic acid may inhibit oxidation in a fish oil-in-water emulsion in a concentration-dependent manner.

Analysis of Flood Inundation Using LiDAR and LISFLOOD Model (LiDAR 고도자료와 LISFLOOD 모형을 이용한 홍수범람해석)

  • Great loss of life and property has been occurred by the severe flood globally. In Korea, a flood inundation map is used as one of the non-structural measures for reducing flood damage, and various inundation models have been studied for flood inundation analysis. This study applies LiDAR data and LISFLOOD model for flood inundation analysis and discusses the the modeling results from levee breaching scenarios for evaluating the applicability of the model to stream inundation modeling. In the results of LISFLOOD modeling, maximum inundation area was similar to the inundation map by HEC-RAS model just less than 4%. The inundation area by each levee breaching scenario showed the difference from 0.2% to 6.5%. Inundation processes were different each other according to the position of levee breach point, and maximum inundation area and depth were changed by the flow direction of stream and flood plain. This study shows that LISFLOOD model can be applied properly to stream inundation analysis using various inundation scenarios.

A clinical effect of desensitizing agents applied on the root surface after periodontal treatment (치주 치료 후 적용된 상아질 지각과민 억제제의 임상 효과에 대한 연구)

  • The purpose of this study was to compare the effect of desensitizing agents applied on hypersensitive root surface following periodontal treatment. This study included 21 subjects(168 vital teeth). To evaluate dentin sensitivity, three clinical tests(tactile, air stream, cold water) were tried and three different densensitizing agents(MS coat, Elmex gel. Superseal) were individually applied. After application, reassessment was done at 1 minute, 1 week, 1 month and 3 months. The results were as follows : 1. The degree of dentin sensitivity was measured highly in the sequence of cold water, air stream and tactile and significantly decreased in all four groups with lapse of time(p<0.05). 2. There was no significant difference between all four groups in the tactile test with lapse of time. 3. There was no significant difference between three experimental groups in the air stream test with lapse of time. however, one minute later, it was measured highly in the sequence of Superseal, MS coat and Elmex 4. There was no significant difference between three experimental groups in the cold water test with lapse of time. As a result of this study, all of three agents were significantly effective in reducing dentin hypersensitivity and these agents could be positively employed to patients complaining of dentin hypersensitivity following periodontal treatment.

Moisture Absorption Characteristics of Pt/Nafion Membrane for PEMFC Prepared by a Drying Process (건식법에 의해 제조한 PEMFC용 Pt/나피온 막의 흡습 특성)

  • A simple drying process was developed for the preparation of a Pt/Nafion self-humidifying membrane to be used for a proton-exchange membrane fuel cell (PEMFC). Platinum (II) bis (acetylacetonate), $Pt(acac)_2$ was sublimed, penetrated into the surface of a Nafion film and then reduced to Pt nanoparticles simultaneously without any support of a reducing agent in a glass reactor at $180^{\circ}C$ for 15 min. The process was carried out in $N_2$ atmosphere to prevent the oxidation of Pt nanoparticles at high temperature. The morphology and distribution of the Pt nanoparticles were observed by transmission electron microscopy (TEM) and energy dispersive spectroscopy (EDS), and we found that the average Pt particle size was ca. 3.7 nm, the penetration depth was ca. $17{\mu}m$. Almost all Pt nanoparticles were formed just beneath the surface and the number density decreased rapidly as the penetration depth increased. To estimate water absorption characteristics of the Nafion membranes, water uptake at an isothermal condition was measured by dynamic vapor sorption (DVS), and it was found that water uptake of the Pt/Nafion membrane was higher than that of the neat Nafion membrane.

Oxidative stability of extracts from red ginseng and puffed red ginseng in bulk oil or oil-in-water emulsion matrix

  • Background: Explosive puffing can induce changes in the chemical, nutritional, and sensory quality of red ginseng. The antioxidant properties of ethanolic extracts of red ginseng and puffed red ginseng were determined in bulk oil and oil-in-water (O/W) emulsions. Methods: Bulk oils were heated at $60^{\circ}C$ and $100^{\circ}C$ and O/W emulsions were treated under riboflavin photosensitization. In vitro antioxidant assays, including 2,2-diphenyl-1-picrylhudrazyl, 2,2'-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid, ferric reducing antioxidant power, total phenolic content, and total flavonoid content, were also performed. Results: The total ginsenoside contents of ethanolic extract from red ginseng and puffed red ginseng were 42.33 mg/g and 49.22 mg/g, respectively. All results from above in vitro antioxidant assays revealed that extracts of puffed red ginseng had significantly higher antioxidant capacities than those of red ginseng (p < 0.05). Generally, extracts of puffed red and red ginseng had high antioxidant properties in riboflavin photosensitized O/W emulsions. However, in bulk oil systems, extracts of puffed red and red ginseng inhibited or accelerated rates of lipid oxidation, depending on treatment temperature and the type of assay used. Conclusion: Although ethanolic extracts of puffed red ginseng showed stronger antioxidant capacities than those of red ginseng when in vitro assays were used, more pro-oxidant properties were observed in bulk oils and O/W emulsions.

A Study on Effect of Environmental Characteristics by Intake Mixture Temperature in Scrubber EGR System Diesel Engines

  • The effects of intake mixture temperature on performance and exhaust emissions under four kinds of engine loads were experimentally investigated by using a four-cycle, four-cylinder, swirl chamber type, water-cooled diesel engine with scrubber EGR system operating at three kinds of engine speeds. The purpose of this study is to develop the scrubber exhaust gas recirculation(EGR) control system for reducing $NO_x$ and soot emissions simultaneously in diesel engines. The EGR system is used to reduce $NO_x$ emissions. And a novel diesel soot-removal device of cylinder-type scrubber with five water injection nozzles is specially designed and manufactured to reduce soot contents in the recirculated exhaust gas to the intake system of the engine. The influences of cooled EGR and water injection, however, would be included within those of scrubber EGR system. In order to survey the effect of intake mixture temperature on performance and exhaust emissions, the intake mixtures of fresh air and recirculated exhaust gas are heated by a heating device with five heating coils made of a steel drum. It is found that the specific fuel consumption rate is considerably elevated by the increase of intake mixture temperature, and that $NO_x$ emissions are markedly decreased as EGR rates are increased and intake mixture temperature is dropped, while soot emissions are increased with increasing EGR rates and intake mixture temperature. Thus one can conclude that the performance and exhaust emissions are considerably influenced by the cooled EGR.

Physicochemical Characteristics and Antioxidant Activity of Makgeolli (막걸리의 이화학적 특성과 항산화활성)

  • Makgeolli is a liquor that represents Korea and is made by mixing yeast and water with starch. Physicochemical Characteristics and Antioxidant activities of rice, nuruk, water and Fermentation time different to Makgeolli. Alcohol content was 6.76~14.56%, Reducing sugar content 1.17~1.68g/L and M5~M6 samples was higher 1.68g/L~2.43g/L. M1~M9 samples pH 3.56±0.02 and total acid 0.45~0.83%. Makgeolli total phenol content was average 966.38mg/L and ABTS 0.77mM. The Antioxidant activity measurements of FRAP and ABTS showed a similar correlation with the total phenol content. Makgeolli showed a high correlation cofficient of r GAE / PRequiv=0.8582 and r GAE / TEAC=0.9769. We intend to make rice, yeast, brewing water, fermentation period, etc. different conditions of makgeolli and analyze the status of fermentation progress and use it as basic data of quality indicators of the fermented rice wine process.
