The Development of Treatment System for Removing the Low Concentrated Nitrogen and Phosphorus Using Phototrophic Bacteria and Media (광합성 박테리아 및 담체를 이용한 하천의 저농도 질소, 인 처리 시스템 개발)

  • We used phototrophic bacteria to remove low concentrated organic materials (CODCr), nitrogen and phosphorus. We applied $COD_{Cr}$ 37.3 mg/L, $NH_3-N$ 4.0 mg/L, and $PO_4^{3-}-P$ 1.0 mg/L (C:N:P=100:10:1) in the batch test, and the removal efficiencies were shown as follow: $COD_{Cr}$ 87.4%, $NH_3-N$ 46.3%, $PO_4^{3-}-P$ 79.7%. The aerobic process with mixed phototrophic bacteria, ceramic media, and media KSP01 showed the removal efficiencies of $COD_{Cr}$, $NH_3-N$, and $PO_4^{3-}-P$, each as 72.7% and 79.2%, respectively in the lab-scale reactor. The maximum $PO_4^{3-}-P$ removal efficiency reached 92.6% by adjusting pH. There were three conditions used to remove $NH_3-N$. The highest removal efficiency was 98.5% with 10.2 L/min of aeration in 1-2 reactors, and the result of applying river-water showed the high removal efficiency of $NH_3-N$ (82.8%). Therefore, this purification system may be useful to control nitrogen and phosphorus at low concentration in field.

Distribution of Biota and Removal Efficiency of Organic Matter in Natural Wetland (농촌배수처리용 습지의 생물상 및 유기물 제거율)

  • Distribution of biota and removal efficiency of organic matter in natural wetland systems were examined from June to October 1998. The aquatic macrophyte communities were consisted of 13 families and 22 species. The composition of occurrence species were as follows: Phragmites communis Trinius are 35%, Persicaria thunbergii Nakai are 19%, Typha angustifolia L. are 17%, Bidens tripartita L. are 16%, Echinochloa crus-galli (L.) Beauv. are 12% and others are l%. The fauna collected from 5 sites consisted of total 6 classes of macroinvertebrates. The composition of occurrence classes were as follows: Insecta are 76%, O1igochaeta are 19%, Gastrapoda are 4% and others are 1%. These included 18 families, 7 orders of Insects. The larve and mosquito and midges were found in wetlands. Preventive strategies are needed for the suppression of mosquito at the stage of wetland planning and design. In wetland, removal efficiency of dissolved organic carbon is low because of primary production and the background level of DOC in the wetland discharge seems to be 5 mgC/1, but those of BOD was ca. 50%. Wetlands receiving water of low concentration can not work as purification field.

Purification and Characteriztion of an Antifungal Antibiotic from Bacillus megaterium KL 39, a Biocontrol Agent of Red-Papper Phytophtora Blight Disease. (고추역병균 Phytophthora capsici를 방제하는 길항균주 Bacillus megaterium KL39의 선발과 길항물질)

  • For the biological control of Phytophthora blight of red-pepper caused by Phytophthora capsici, an antibiotic-producing plant growth promoting rhizobacteria (PGPR) Bacillus sp. KL 39 was selected from a local soil of Kyongbuk, Korea. The strain KL 39 was identified as Bacillus megaterium by various cultural, biochemical test and API and Microlog system. B. megaterium KL 39 could produce the highest antifungal antibiotic after 40 h of incubation under the optimal medium which was 0.4% fructose, 0.3% yeast extract, and 5 mM KCl at 30 C with initial pH 8.0. The antifungal antibiotic KL 39 was purified by Diaion HP-20 column, silica gel column, Sephadex LH-20 column, and HPLC. Its RF value was confirmed 0.32 by thin-layer chromatography with Ethanol:Ammonia:Water = 8:1:1. The crude antibiotic KL39 was active against a broad range of plant pathogenic fungi, Rhizoctonia solani, Pyricularia oryzae, Monilinia fructicola, Botrytis cinenea, Alteranria kikuchiana, Fusarium oxysporum and Fusarium solani. The purified antifungal antibiotic KL39 had a powerful biocontrol activity against red-pepper phytophthora blight disease with in vivo pot test as well as the strain B. megaterium KL 39.

Photodegradation of some Organophosphorous Pesticides (일부 유기인계 농약의 광분해성)

  • The present study was performed to investigate photodegradation rate constants and degradation products of dichlorvos and methidathion by the USEPA method. The two pesticides were very stable in sunlight for 16 days from September 2 to 18, 1998 and humic acid had no sensitizing effect on the photolysis of each pesticide in sunlight. The photolysis rate was fast-est for methidathion, followed by dichlorvos in the presence of UV irradiation. Photodegradation rate constant and half-life of dichlorvos were 0.0208 and 33.3 min, respectively. Photodegradation rate constant and half-life of methidathion were 0.6789 and 1.0min, respectively. The two pesticides were degraded completely in the presence of UV irradiation and UV irradiation with TiO$_2$in about 3 hours. Therefore, it is suggested that UV treatment will be effective for the degradation of pesticides in the process of drinking water purification. In case of dichlorvos and methidathion, UV irradiation with TiO$_2$was more effective for degradation than W irradiation. In order to identify photolysis products, the extracts of degradation products were analyzed by GC/ MS. The mass spectrum of photolysis products of dichlorvos was at m/z 153, those of the photolysis of methidathion were at m/z 198 and 214, respectively. Photolysis products of dichlorvos was Ο, Ο-dimethyl phosphate(DMP), those of methidathion were Ο, Ο-dimethyl phosphorothioate(DMTP) and Ο, Ο-dimethyl phosphorodithioate (DMDTP).

Intestinal Immune Modulating Polysaccharides of Atractylodes lancea DC. Rhizomes

  • A kind of traditional herbal prescription, Sip-Jeon-Dae-Bo-Tang (TJ-48), has been reported to improve the general condition of cancer patients receiving chemotherapy and /or radiation therapy, and to accelerate hematopoietic recovery from bone marrow injury by mitomycin C. In the present studies, we found that hot-water extract from Atractylodes lancea DC. rhizomes contributed mainly to intestinal immune modulating activity of TJ-48 on Peyer's patch cells mediated-hematopoietic response. After the fractionation, ALR-5 II a-1-1, 5 II b-2-2 and 5 II c-3-1 were further purified from crude polysaccharide fraction. Chemical analyses of each fraction indicated that ALR-5 II a-1-1 mainly contained arabinogalactan fraction whereas ALR-5 II b-2-2 and 5 II c-3-1 mostly comprised pectic polysaccharide fractions as the active polysaccharide ingredients. In order to analyze the essential structure of the activity, ALR-5 II a-1-1 was treated by sequential enzymatic digestion using exo-${\alpha}$-L-arabinofuranosidase and exo-${\beta}$-D-(1\longrightarrow3)-galactanase. Based upon the results of chemical and MALDI-TOF-MS analyses and activity on the digested fractions, the galactosyl side chains consisting of 6-linked Galf and Galp over tetrasaccharide in ALR-5 II a-1-1 might be responsible for the potent intestinal immune modulating activity. To characterize moiety of ALR-5 II c-3-1 for the expression of activity, endo-${\alpha}$-D-(1\longrightarrow4)-polygal acturonase (GL-PGase) purified from dried leaves of Panax ginseng digested ALR-5 II c-3-1. The results of structural analyses and activity on the digested fractions showed that PG-2, which structurally resembles to rhamnogalacturonan II (RG II), and PG-3 (galacturono-oligosaccharides) contained potent intestinal immune modulating activity. Further purification of the other acidic fraction (ALR-5 II b-2-2) indicated that ALR-5 II b-2-2Bb showed that the most potent activity. ALR-5 II b-2-2Bb also contained the unusual component sugars characteristics in RG- II as well as PG-2 derived from ALR-5 II c-3-1, but it could not be digested with GL-PGase. The present studies of relationship between structures and intestinal immune modulating activity of the active polysaccharides purified from A. lancea DC. rhizomes suggested that neutral galactosyl chains consisting mainly of (1\longrightarrow6)-linked Galf and Galp, and RG- II -like moiety with unique component sugars, such as 2-Me-Xyl, 2-Me-Fuc, Api, AceA, Kdo and Dha should play an important role in the potent intestinal immune modulating action of A. lancea DC. rhizomes.

Removal of Bromate by Iron, Copper and Silver Impregnated Activated Carbon (철, 구리, 은 첨착활성탄을 이용한 브롬산염의 제거)

  • The purpose of this research is to remove the bromate that is a disinfection by-poduct of water purification by ozone. I achieved a high rate of removal with iron, copper, or silver impregnated activated carbon by using both the adsorbing power of granular activated carbon and the oxidizing power of metal ions as deoxidizing agents. In the removal test of bromate with the quantity of activated carbon input I injected each activated carbon by 0.1, 0.3, 0.1, and 1.0 g and let them react for 240 minutes. I found the quantity of removed bromate was in proportion to the amount of input. The removal rate of bromate increased about 20% when I used acid treated activated carbon. The metal impregnated activated carbon had a higher removal rate of bromate than that of general activated carbon by about $30{\sim}50%$. Iron impregnated activated carbon showed a 92% removal rate of bromate. Iron, copper, or silver impregnated activated carbon removed about $0.9{\sim}1.5mg\;{BrO_3}^-/g$ while general activated carbon removed about $0.02{\sim}0.45mg\;{BrO_3}^-/g$. In the continuous column reaction, there were breakthrough phenomena at 96, 180, and 252 hours when I tested EBCT by 1, 2 and 3 minutes while I was changing the flux rate of bromate from 15.6 to 46.8 mL/min.

Synthesizing Behavior of Calcium Chloroaluminate with using MSWI Fly Ash in the Different Sintering Conditions (생활폐기물(生活廢棄物) 소각(燒却) 비산(飛散)재의 소성(燒成) 조건(條件)에 따른 Calcium Chloroaluminate 합성(合成) 거동(擧動)에 관한 연구(硏究))

  • The aim of this study is to investigate the behaviors of synthesis of Calcium Chloroaluminte($11CaO{\cdot}7Al_2O_3{\cdot}CaCl_2$) under the melting temperature of municipal solid waste incineration fly ash(MSWI fly ash); $900^{\circ}C-1300^{\circ}C$ of sintering temperature. A sludge generated from a water purification plant (SW) was also used to fill up $Al_2O_3$ source, which might be deficient in MSWI fly ash. MF and SW were mixed at mixing ratio of 10 to 7, which is the mole ratio of CaO to $Al_2O_3$ of calcium Chloroaluminte. Mixed samples were sintered in the opened sintering reactor and the closed sintering reactor, respectively. The results showed that calcium chloroaluminte was formed at $1,000^{\circ}C$ in both reactors, but the temperature of decomposition of calcium chloroaluminte depended upon sintering reactor type; an opened sintering reactor and a closed sintering reactor, owing to the vaporization velocity of Cl.

Oxidation of Organic Compounds Using $TiO_2$ Photocatalytic Membrane Reactors ($TiO_2$ 광촉매 막반응기를 이용한 유기물의 산화)

  • The photodegradation efficiency of formic acid on $TiO_2$ photocatalytic membranes was investigated. A new titania membrane reactors for purification of water combining microfiltration with photocatalytic degradation of organic compounds were developed. Titania membrane tubes(average pore size of $0.2\mu m$) were prepared by the slip casting, and porous thin films of $TiO_2$ were formed on the tube surface by the sol-gel process to increase the surface area, and consequently to increase photodegradation efficiency of organic compounds. The UV light with the wavelength of 365 nm was used as a light source for photocatalytic reactions. The photodegradation efficiency of the organic compounds was strongly dependent on the flux of the solution, the microstructure of the membrane (sol pH), and the amount of $O_2$ supplied. The effects of the primary oxidant such as $H_2O_2$ and dopants such as $Nb_2O_5$ on the photodegradation efficiency were also investigated. The results showed that more than 80% of formic acid could be degraded using membrane coated with a $TiO_2$ sol of pH 1.45. The photodegradation efficiency could be improved by about 20% when adding $H_2O_2$ in feed solution or doping $TiO_2$ membranes with $Fe_2O_3$.

Study on the Relationship between Aggregation Structure and Flow Rate Depending on Extrusion Temperature at Complex Mold of (Activated Carbon/Polymer Binder) ((활성탄/고분자바인더)복합성형체의 압출온도에 따르는 응집구조 및 유량 상관성에 관한 연구)

  • As the need for miniaturization in water purification filter increases, the development of filter media for single filtration with multiple function was strongly required. In this study, the molded activated carbon composed of single unit was manufactured by extrusion-sintering process, and then the flow rate, density and porosity were investigated using the molded activated carbon manufactured at various extrusion temperature. We confirmed that it was possible to manufacture the single unit-molded activated carbon when the extrusion temperature was $140{\sim}230^{\circ}C$ more than $133^{\circ}C$ being of polymer binder melting point, and the optimal extrusion temperature for the molded activated carbon with maximum flow rate was $170^{\circ}C$ since the molded activated carbon had low density and high through porosity. Also we confirmed that the flow rate through the molded activated carbon was strongly dependent upon through pore porosity compared to total porosity for the molded activated carbon.

Verification of Antibacterial Activities of Oriental Herbal Medicine Extracts (한약재 추출물의 항균활성 검증)

  • The antibacterial activities of nine oriental herbal medicine extracts were measured against seven kinds of bacteria known to cause food poisoning and disease. The extracts were prepared with distilled water using an autoclave ($121^{\circ}C$ for 30 min). The extraction yields of nine oriental herbal medicines were in the rage of 0.5%-33.4%. The antibacterial activities were evaluated using the paper-disc method. Extracts of Hwangryeon, Jiyu, and Ohbaeja showed antibacterial activities while the others did not. Extracts of Coptis japonica, Hwangryeon, and Sanguisorba officinalis, Jiyu showed antibacterial activities only against Clostridium perfringens and Listeria monocytogenes. Extracts of Rhus javanica, Ohbaeja showed the strongest antibacterial activities against all the bacteria tested. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of R. javanica extracts were in the range of 2.4~20 mg/ml for the bacteria tested. In conclusion, the extract of R. javanica showed superior antibacterial activities against the seven bacterial strains tested. The MIC and MBC values for each bacterium were represented as strain names (MIC, MBC), which were Bacillus cereus (11.2, 20), Clostridium perfringens (2.4, 4.7), Escherichia coli (9.1, 9.1), Listeria monocytogenes (11.1, 20), Salmonella typhi (4.7, 9.1), Staphylococcus aureus (2.4, 4.7), and Vibrio parahaemolyticus (2.4, 5.3). Further purification and study of this antibacterial material would be helpful for developing antibiotics or promoting synergistic effects with known antibiotics, producing antibacterial material for lengthening the shelf-life of food, and so on.
