진범(Aconitum pesudo-laeve var. erectum) 추출물의 항노화 및 항당뇨 효과 (Anti-aging and Anti-diabetes Effects of Aconitum pesudo-laeve var. erectum Extracts)

  • 진범은 열독을 제거하고 발한을 유도하여 몸의 수분을 조절하는 효과를 가져 한방에서 사용되어 왔으며, 항염증 활성과 장기의 면연체계를 조절하는 효과가 있는 것으로 알려졌다. 본 연구에서는 진범 추출물의 항노화와 항당뇨 활성을 조사하였다. 물과 에탄올을 이용하여 진범을 추출하고 그 추출물이 가지는 DPPH, ABTS 라디칼 소거활성, PF 및 TBARs와 같은 항산화 활성과 ${\alpha}$-amylase와 ${\alpha}$-glucosidase 저해활성을 측정하였다. 진범 물과 에탄올 추출물의 농도 $100{\mu}g/ml$, $50{\mu}g/ml$ 처리군부터 50% 이상의 DPPH 라디칼 소거활성이 나타났고, ABTS 라디칼 소거활성은 에탄올 추출물의 농도 $1,000{\mu}g/ml$ 처리군에서 $99.8{\pm}0.1$%로 가장 높게 나타났다. 베타카로텐리놀레이트 모델 시스템을 이용한 항산화 활성 측정결과 $500{\mu}g/ml$ 이상의 농도로 첨가하였을 때 물 추출물에서 1.27 PF, 1.33 PF값을, 에탄올 추출물에서는 1.40 PF, 1.49 PF로 나타났으며, 에탄올 추출물의 농도 $1,000{\mu}g/ml$ 처리군에서 $0.16{\pm}0.03{\mu}M$로 가장 낮은 TBARs값을 나타내었다. 물 추출물의 농도 $1,000{\mu}g/ml$ 처리군와 에탄올 추출물의 100~$1,000{\mu}g/ml$ 처리군에서 90% 이상의 ${\alpha}$-amylase 저해 활성과 60% 이상의 ${\alpha}$-glucosidase 저해 활성을 나타내었다. 이러한 결과를 종합하여 볼 때 진범의 생리활성을 이용하여 항노화 화장품 원료와 항산화 및 항당뇨 예방물질 소재로의 활용이 가능할 것으로 판단된다.

옥광(Castanea crenata) 밤송이 추출물의 생리활성 효과 (Biological Activities of Extracts from Okkwang (Castanea crenata) Chestnut Bur)

  • 옥광(Okkwang; Castanea crenata) 밤송이 고형분의 phenolic 화합물의 함량은 물, 40% 에탄올 추출물에서 각각 11.24, $10.28{\mu}g/50{\mu}g$ solid로 측정되었다. DPPH 라디칼 소거능은 물, 에탄올 추출물 $50{\mu}g/mL$ solid 농도에서 각각 85, 87%의 전자공여능을 나타내었고, ABTS 라디칼 소거능은 물, 에탄올 추출물에서 각각 100, 86%의 전자공여능을 나타내었다. PF에서는 물, 에탄올 추출물 $200{\mu}g/mL$ solid 농도에서 각각 1.22, 1.45 PF를 나타내었고, TBARS는 물 추출물에서 83%, 에탄올 추출물에서는 73%의 효능을 나타내었다. XOase 저해 효과는 물, 에탄올 추출물 $200{\mu}g/mL$ solid 농도에서 각각 54, 43%를 나타내었다. ${\alpha}$-Glucosidase 저해 효과는 $50{\mu}g/mL$ solid 농도에서 물 추출물은 95%, 에탄올 추출물에서는 96%의 저해 효과를 나타내었다. 미백을 나타내는 tyrosinase 저해 효과는 $200{\mu}g/mL$ solid 농도에서 에탄올 추출물은 27%의 효능을 나타내었다. 주름개선을 보여주는 collagenase 저해 효과는 $200{\mu}g/mL$ solid 농도에서 물, 에탄올 추출물 각각 93, 94%의 우수한 주름개선 효과를 나타내었으며, elastase 저해 효과는 $200{\mu}g/mL$ solid 농도에서 에탄올 추출물은 56%의 효능을 나타내었다. 항염증 효과를 나타내는 HAase 저해 효과를 측정한 결과 $200{\mu}g/mL$ solid 농도에서 물, 에탄올 추출물에서 각각 96, 52%의 항염증 효과를 나타내었다. 이러한 결과로 보아 OCS 추출물은 항산화 활성, 통풍 억제, 항당뇨, 미백, 주름개선, 항염증 효과가 우수하므로 기능성 소재로서 사용이 가능할 것으로 기대된다.

한국 및 중국산 약용버섯류의 추출용매에 따른 생리활성 성분 비교 (Comparison of physiological activity of medicinal mushrooms produced in Korea and China using different extraction solvents)

  • 한국과 중국에서 재배한 약용버섯인 노루궁뎅이, 동충하초, 영지의 열수추출물과 70% ethanol 추출물에 대해서 DPPH 라디컬 소거능, 아질산 소거능, 총 폴리페놀 함량, 베타글루칸 함량을 비교하기 위하여 측정하였다. DPPH 라디컬 소거능은 열수추출물과 70% ethanol추출물에서 한국산 노루궁뎅이와 동충하초가 월등히 높았으며, 추출용매별로 비교하였을 경우, 노루궁뎅이와 동충하초는 열수추출물의 항산화효능이 높았다. 아질산염 소거능의 경우, 한국산과 중국산 영지의 열수추출물에서 각각 41%와 39%로 나타나 70% ethanol 추출물에 비하여 높았다. 총 폴리페놀 함량은 한국산 노루궁뎅이와 동충하초는 열수추출물에서 높은 함량을 보였으며, 영지의 경우는 70% ethanol 추출물에서 더 높은 함량치를 나타냈다. 베타글루칸 함량은 한국산 동충하초가 45.11%를 보이며 가장 높았으며, 영지 역시 중국에서 재배한 것에 비하여 높은 함량을 나타냈다. 노루궁뎅이는 중국산이 한국산에 비하여 높은 베타글루칸 함량을 보였다. 위와 같은 결과로부터 각각의 버섯류에 맞는 최적의 용매를 사용해야 최대의 유효성분을 추출할 수 있다고 사료된다. 본 연구에서 사용한 노루궁뎅이, 동충하초, 영지가 한국 및 중국에서 재배되는 버섯의 대표성을 나타내기에는 부족하나 이러한 연구결과를 토대로 더 많은 비교샘플을 확보하여 약용버섯류에 대한 기능성 평가를 위한 기초자료로 활용하고자 한다.

Screening for Antioxidative and Antimutagenic Capacities in 7 Common Vegetables Taken by Korean

  • This study was performed to investigate the antioxidative effect as the inhibition of MDA and BSA conjugation reaction, inhibition of lipid peroxidation and the scavenging effect on DPPH radical, and antimutagenic capacities as the Ames test in 7 common vegetables taken by Korean for suggestion of prevention and dietetic treatment of chronic diseases and development of antioxidative and antimutagenic functional food. The water extracted fractions of perilla leaves were most effective in the inhibition of MDA and BSA conjugation reaction showing 77.2% of inhibition rate among 7 vegetables. The inhibition rates of ethanol extracted fractions of sedum and wild water dropwort on the lipid peroxidation were 67.1% and 61.5%, respectively. The ethanol extracted fractions of crown daisy and wild water dropwort showed the most effective results among 7 vegetables in the DPPH radical scavenging capacities showing inhibition rate of 78.8% and 73.6%, respectively. The indirect and direct antimutagenic effects of ethanol extract of 7 vegetables were examined by Ames test using Salmonella typimurium TA98 and TA100. Inhibitory effects of wild water dropwort was superior to the other vegetables on the Ames test. These results suggest that common 7 vegetables taken by Korean are believed to be a possible antioxidative and antimutagenic capacities, although the resultswere different, some or less, according to the assay method and vegetables used.

골질 보호 작용이 있는 수종 생약 추출액의 래디칼 소거능 및 DNA 보호 효과 (Radical Scavenging Activities and Protective Effects against Oxidative Damage to DNA of Extracts from Medicinal Plants with known Osteoprotective Effects)

  • Among aging diseases, the most basic problem is a decrease in bone mineral density. Patients with osteoporosis are steadily increasing in the world. This study was investigated to show the relationship between osteoporotic effects and oxidative damage. Water extracts of 15 medicinal plants and ethanol extracts of 14 medicinal plants with known anti-osteoporotic effects, were tested for their radical scavenging activity using DPPH, ABTS, SRSA and FRAP assay. Water extract of Cornus officinalis, Rubus coreanus and ethanol extract of Rubus coreanus, Viscium album var. coloratum, Cimicifuga heracleifolia showed about 15-20 mg/g of total phenolic contents. Water extract of Cornus officinalis, Rubus coreanus and Epimedium koreanum showed high radical scavenging activity. Ethanol extract of Drynaria fortunei, Cornus officinalis, Rubus coreanus, Gentiana scabra and Astragalus membranaceus showed high radical scavenging activity. Water extract of Drynaria fortunei, Cornus officinalis, Nelumbo nucifera, Epimedium koreanum, and Gentiana scabra showed very strong protective effect against oxidative damage to DNA. These results suggest the correlation between anti-osteoporotic effects and antioxidative effects.

추출용매에 따른 자소 색소의 염색성 및 기능성 (Characteristics and Dyeability of Perilla Frutescens L. Britt Extracts with Different Solvents)

  • In this study, we examined the influence of the pigment characteristic and dyeing condition on dyeing properties and functionality by using Perilla Frutescens L. Britt extracts, in which ethanol, distilled water and NaOH solution were used as 3 different solvents. Changes in dyeing conditions include variations in dye concentration, dyeing temperature, time and pH on dye uptake, and K/S values were compared according to these changes. Additionally, color changes were observed according to the use and types of mordant. Ultraviolet-visible spectrum was utilized to investigate the pigment characteristic, and as a result, chlorophyll was identified in ethanol extract, whereas tannin was identified both in distilled water extract and NaOH solution extract. By using FT-IR analysis, these tannins in distilled-water-extract and NaOH solution extract were verified to be hydrolyzable tannin. When dyeing silk, dye uptake increased as dye concentration, dyeing temperature and time increased, while it decreased as pH of the extract increased. Fabrics dyed without a mordant produced Y-series colors, and fabrics dyed with mordants showed various colors depending on the mordant types. Even though color fastness to washing and light was unsatisfactory, fastness to rubbing and perspiration showed relatively high grade. Moreover, deodorant ability of dyed fabric improved.

Antioxidant Properties of Water and Aqueous Ethanol Extracts and Their Crude Saponin Fractions from a Far-eastern Sea Cucumber, Stichopus japonicus

  • Water and 70% ethanol extracts obtained from a sea cucumber (Stichopus japonicus) body wall by heat reflux or pressurized solvent extraction showed 2,2-diphenyl-1-picrylhydrazyl ($DPPH^{\cdot}$) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical cation ($ABTS^{{\cdot}+}$) scavenging activities comparable to those of fruits and vegetables. The highest activities were observed for the water extract from heat reflux extraction. Crude saponins exhibited higher radical scavenging activities than the soluble matters in the extracts. However, they were responsible for only about 3 to 15% of the scavenging activities of the extracts. Total phenolic contents showed a significant correlation with $DPPH^{\cdot}$ scavenging activities, suggesting a significant contribution of phenolic constituents to the antioxidant properties of the extracts. However, total flavonoid contents showed little correlation with the radical scavenging activities. The results suggest that the water or 70% ethanol extract obtained from sea cucumber body wall by simple heat reflux extraction could provide considerable antioxidant benefits.

Biological Activities of Water and Ethanolic Extracts from Allium victorialis L. Mature Leaves

  • Allium victorialis L. (A. victorialis) is a very popular vegetable in Korea. The most commonly used parts of this vegetable are the bulbs and young leaves. To determine if the mature leaves have any beneficial properties, we investigated antioxidant, anti-${\alpha}$-glucosidase, anti-inflammatory, and anticancer activities of water and ethanol extracts from A. victorialis. Antioxidant activity was evaluated by measuring total phenolic content, DPPH and superoxide radicals scavenging activities. The water extract from A. victorialis ($W{\cdot}A$. victorialis) exhibited higher antioxidant ability than the ethanol extract ($E{\cdot}A$. victorialis). Moreover, the water extract showed strong inhibitory effect on ${\alpha}$-glucosidase. On the other hand, the ethanol extract had greater anti-inflammatory activity on murine macrophage cells (RAW 264.7) and greater anticancer activities against human colon cancer cells (HT-29). These results suggest that mature leaves from E·A. victorialis may have health-enhancing effects.

진달래꽃(Rhododendron mucronulatum Flos.) 추출물로부터 Helicobacter pylori 억제 효과를 가지는 phenol성 물질의 정제 및 동정 (Purification and Identification of Phenol Compounds with Inhibitory Activity on Helicobacter pylori from Rhododendron mucronulatum Flos. Extracts)

  • 진달래꽃을 60% ethanol로 추출하여 추출물의 H. pylori에 대한 저해활성의 측정과 저해물질의 분리 및 동정을 시행하였다. 또한 H. pylori균에 대한 항균활성은 60% ethanol 추출물에서 phenol 화합물을 200${\mu}g/ml$ 이상 주입하였을 때부터 22 mm이상의 clear zone이 관찰되어 H. pylori균에 대한 높은 항균력을 나타내었다. 저해물질의 분리는 Sephadex LH-20 column을 이용하여 분리한 결과 6개의 fraction으로 분리되었으며, H. pylori 저해활성을 나타낸 fraction D를 MCI-gel CHP-20 column을 이용하여 normal phase type인 ethanol 100%에서 0%로 농도를 감소시키며 용출한 결과 5개의 단일 물질 분획을 얻을 수 있었다. 그 중 세 가지의 물질에 대해 H. pylori저해활성이 나타나 FAB-MS, $^1H$$^{13}C$ NMR 및 IR spectrum을 사용하여 구조 동정한 결과 quercitrin (quercetin-3-O-rhamnopyranoside), myricitrin (myricetin-3-O-rhamnopyranoside) 및 quercetin인 것으로 확인되었다.

백련초 추출물의 김치발효 젖산균과 식중독균에 대한 항균효과 (The Antibacterial Effects of Backryeoncho(Opuntia ficus-indica var. saboten) Extracts as Applied to Kimchi Fermentation with Lactic Acid Bacteria and Food Poisoning Bacteria)

  • This research evaluated antibacterial and growth inhibition effects on three kinds of lactic acid bacteria and five kinds of food poisoning bacteria using Backryencho powder, hot water, 70% ethanol, and 95% ethanol extracts. Antibacterial activity was shown against Leu. mensenteroides for 8 and 10 mg/disc of the 95% ethanol extract solution and strong proliferation inhibition effects were displayed against B. subtilis, Stap. aureus, E. coli, and S. typhimurium. High antibacterial activity according to certain clear zone formations was shown especially for the 10 mg/disc. A 3% concentration of the 95% ethanol extract showed high growth inhibition effects against lactic acid bacteria, L. brevis, L. plantarum, and Leu. mesenteroids. The measurement of viable cell counts of S. aureus, E. coli, B. subtilis, and S. typhimurium indicated suppression effects by the 3% concentration of the 95% ethanol extract, at 49.60%, 41.54%, 35.95%, 28.82%, and 26.60% respectively. The antibacterial activities of the hot water, 70% ethanol, 95% ethanol extract of Backryencho against food poisoning bacteria and Kimchi fermentation lactic acid bacteria were confirmed through various methods of antibiotic measurement. Based on these results, Backryencho extract is considered a good source for a range of applications as a natural anti-bacterial agent for the storage ability of Kimchi and as a possible food preservative.
