차전초 잎의 물 추출물이 고지방식이로 비만 유도된 C57BL/6 생쥐의 체지방 감소에 미치는 영향 (Effect of Plantaginis asiaticae Folium water extract on body fat loss in high fat-induced obese C57BL/6 mice)

  • Objective : Plantago asiatica L. has been used for many years in Korea, China, and Japan for the treatment of many diseases such as anti-inflammatory, anti-asthma and diuretic. But the anti-obesity effect of Plantaginis asiaticae Folium has not been studied yet. Thus, this study aimed to demonstrate the anti-obesity efficacy of Plantaginis asiaticae Folium water extract (PAF) in high fat diet-induced obesity animal model. Method : Male, five-weeks old C57BL/6 mice were divided into 5 groups; ND (normal diet + 0.9% saline), HFD (high-fat diet + 0.9% saline), PC (high-fat diet + Garcinia cambogia 500 mg/kg), PAF 100, 300 (high-fat diet + PAF 100, 300 mg/kg. Treatments were performed daily for 6 weeks per os. We evaluated the changes of body weight, white adipose tissues weight, hepatic and fecal lipids level, pancreatic lipase activity and lipid profiles in plasma. Result : Body weights, adipose tissue weights and FER were significantly lower in mice fed the high-fat diet with PAF treatment than in mice fed the high-fat diet alone. In plasma, TG, TC, LDL-C, ATS and ALT levels significantly decreased in PAF groups compared with HFD group. Also, hepatic triglyceride (TG) levels decreased and fecal TG increased in PAF groups compared to HFD group. In addition, the activity of pancreatic lipase was decreased in the PAF groups compared to that of HFD group. Conclusion: These results suggest PAF extract is effective in preventing and improvement obesity, so it might be a potential and safe material for anti-obesity agent of weight control or functional supplements.

  • Objective : This study was intended to examine the effects of water extract of Prunellae Spica (PS), which is a herb with 'cold' nature based on hot and cold theory of traditional Korean medicine. Methods : Hyperthyroidism was induced in SD rats by LT4 (0.5 mg/kg, i.p.) daily for four weeks. After 2 weeks of LT4 injection, rats were divided randomly into four groups; normal, LT4-induced hyperthyroid control, PS extract (500 mg/kg, p.o.)-treated group, and propylthiouracil (PTU, 10 mg/kg, s.c.)-treated positive group. After 2 weeks of drug treatment, all rats were sacrificed and harvested blood samples and thyroid tissues. The changes of body weight, food and water intake, and body temperature were measured weekly. Serological markers were analyzed in sera using an enzyme-based assay, and thyroid tissues were stained with Hematoxylin & Eosin (H&E). Brain and dorsal root ganglion (DRG) tissues were isolated and analyzed the expression of transient receptor potential (TRP) channels by Western blot. Results : PS extract administration attenuated the loss of body weight and the increase of body temperature in LT4-induced hyperthyroidism rats. PS extract increased the level of thyroid stimulating hormone (TSH) and decreased tiiodothyronine (T3) and tetraiodothyronine (T4). In action mechanism, PS extract regulated the expression of transient receptor potential channel subfamily V member 1 (TRPV1) and transient Receptor Potential channel subfamily M member 8 (TRPM8), the thermoregulators. Conclusion : To conclude, PS extract can improve the symptoms of hyperthyroidism through regulation of the thyroid hormones imbalance and thermoregulation via TRP channels.

  • 실내 배양체를 이용한 최근의 연구들은 혼합영양성 와편모류 Dinophysis와 종속영양성 와편모류 Oxyphysis oxytoxoides가 둘 다 혼합영양성 섬모류인 Mesodinium rubrum을 먹이생물로 이용하여 잘 성장한다는 점을 보고한 바 있다. 본 연구는 자연 생태계에서 포식자인 D. acuminata 및 O. oxytoxoides와 그들의 먹이인 M. rubrum사이의 상호작용을 연구할 목적으로 2011년 7월말부터 8월말까지 한 달간에 걸쳐 마산만의 한 고정 정점에서 하루 2회씩 개체군 변동을 모니터링 하였다. 본 연구동안 염분은 강수량이 높게 나타난 시기에 최소 5까지 급격하게 낮아지다가 강수량이 감소하는 8월말로 갈수록 약 28까지 점차적으로 증가하는 경향을 나타냈고, 수온은 8월 20일까지는 평균적으로 $26.5^{\circ}C$를 유지하였으나, 이후에는 평균 $21^{\circ}C$를 유지하였다. M. rubrum은 연구기간 동안 지속적으로 출현하였으나 그 변동 폭($13-492\;cells\;mL^{-1}$)이 매우 컸다. D. acuminata와 O. oxytoxoides의 개체수는 각각 $n.d.-19,833\;cells\;L^{-1}$$n.d.-100,333\;cells\;L^{-1}$의 범위를 나타냈다. 전반적으로 D. acuminata와 먹이생물인 M. rubrum의 개체수 출현 양상은 시간차를 두고 서로 밀접한 관계를 보이기도 하였으나, M. rubrum이 높은 밀도로 출현함에도 불구하고 D. acuminata의 대번식이 일어나지 않는 경우가 존재하였는데, 이러한 이유는 극심한 염분의 변화 및 포식 등의 결과로 판단된다. 한편, O. oxytoxoides는 M. rubrum이 지속적으로 출현함에도 불구하고 $19-24^{\circ}C$의 수온 범위에서만 출현한 결과로 볼 때, 먹이생물의 부족보다는 수온 등의 환경요인이 O. oxytoxoides의 밀도 변화에 큰 영향을 미쳤을 것으로 판단된다.

  • Situated close to Heathrow Airport, and adjacent to the M4 and M25 Motorways, the site at Axis Park is considered a prime location for business in the UK. In consequnce two of the UK's major property development companies, MEPC and Redrew Homes sought the expertise of Intergeo to remediate the contaminated former industrial site prior to its development. Industrial use of the twenty-six hectare site, started in 1936, when Hawker Aircraft commence aircraft manufacture. In 1963 the Firestone Tyre and Rubber Company purchased part of the site. Ford commenced vehicle production at the site in the mid-1970's and production was continued by Iveco Ford from 1986 to the plant's decommissioning in 1997. Geologically the site is underlain by sand and gravel, deposited in prehistory by the River Thames, with London Clay at around 6m depth. The level of groundwater fluctuates seasonally at around 2.5m depth, moving slowly southwest towards local streams and watercourses. A phased investigation of the site was undertaken, which culminated in the extensive site investigation undertaken by Intergeo in 1998. In total 50 boreholes, 90 probeholes and 60 trial pits were used to investigate the site and around 4000 solid and 1300 liquid samples were tested in the laboratory for chemical substances. The investigations identified total petroleum hydrocarbons in the soil up to 25, 000mg/kg. Diesel oil, with some lubricating oil were the main components. Volatile organic compounds were identified in the groundwater in excess of 10mg/l. Specific substances included trichloromethane, trichloromethane and tetrachloroethene. Both the oil and volatile compounds were widely spread across the site, The specific substances identified could be traced back to industrial processes used at one or other dates in the sites history Slightly elevated levels of toxic metals and polycyclic aromatic hydrocarbons were also identified locally. Prior to remediation of the site and throughout its progress, extensive liaison with the regulatory authorities and the client's professional representatives was required. In addition to meetings, numerous technical documents detailing methods and health and safety issues were required in order to comply with UK environmental and safety legislation. After initially considering a range of options to undertake remediation, the following three main techniques were selected: ex-situ bioremediation of hydrocarbon contaminated soils, skimming of free floating hydrocarbon product from the water surface at wells and excavations and air stripping of volatile organic compounds from groundwater recovered from wells. The achievements were as follows: 1) 350, 000m3 of soil was excavated and 112, 000m3 of sand and gravel was processed to remove gravel and cobble sized particles; 2) 53, 000m3 of hydrocarbon contaminated soil was bioremediated in windrows ; 3) 7000m3 of groundwater was processed by skimming to remove free floating Product; 4) 196, 000m3 of groundwater was Processed by air stripping to remove volatile organic compounds. Only 1000m3 of soil left the site for disposal in licensed waste facilities Given the costs of disposal in the UK, the selected methods represented a considerable cost saving to the Clients. All other soil was engineered back into the ground to a precise geotechnical specification. The following objective levels were achieved across the site 1) By a Risk Based Corrective Action (RBCA) methodology it was demonstrated that soil with less that 1000mg/kg total petroleum hydrocarbons did not pose a hazard to health or water resources and therefore, could remain insitu; 2) Soils destined for the residential areas of the site were remediated to 250mg/kg total petroleum hydrocarbons; in the industrial areas 500mg/kg was proven acceptable. 3) Hydrocarbons in groundwater were remediated to below the Dutch Intervegtion Level of 0.6mg/1; 4) Volatile organic compounds/BTEX group substances were reduced to below the Dutch Intervention Levels; 5) Polycyclic aromatic hydrocarbons and metals were below Inter-departmental Committee for the Redevelopment of Contaminated Land guideline levels for intended enduse. In order to verify the qualify of the work 1500 chemical test results were submitted for the purpose of validation. Quality assurance checks were undertaken by independent consultants and at an independent laboratory selected by Intergeo. Long term monitoring of water quality was undertaken for a period of one year after remediation work had been completed. Both the regulatory authorities and Clients representatives endorsed the quality of remediation now completed at the site. Subsequent to completion of the remediation work Redrew Homes constructed a prestige housing development. The properties at "Belvedere Place" retailed at premium prices. On the MEPC site the Post Office, amongst others, has located a major sorting office for the London area. Exceptionally high standards of remediation, control and documentation were a requirement for the work undertaken here.aken here.

육미지황탕박의 사료가치에 관한 연구 (Studies on the Feed Value of Yukmijihwangtang Meal)

  • 본 시험은 육미지황탕박의 사료가치를 측정하기 위한 기초 자료를 제공하기 위하여 두가지 실험을 실시하었다. 실험 1에서는 육미지황탕 단미재료의 열탕 추출 전후의 화학적 조성을 조사하고, 소실률을 알아보기 위하여 육미지황탕박으로 TMR의 0, 10 및 20%를 대체하여 in vitro 실험을 수행하였다. 실험 2에서는 당밀의 첨가가 육미지황탕박 사일리지의 기호성과 반추위내 발효성상을 조사하기 위하여 육미지황탕박에 당밀 첨가수준을 0, 0.5 및 1.0%로 하여 사일리지를 제조하고, 우리나라 재래산양에 대한 기호성과 반추위내 발효성상을 조사하였다. 실험 1의 결과로서, 단백질의 조성에 있어 택사는 열탕침출 후 12.5%로서 높은 편이며 복령은 0.7%로서 매우 적은 편이었다. 조지방은 산수유의 경우 추출 전 후가 각각 7.6 및 8.9%로 높은 편이지만, 나머지 재료는 1.4~4.3%의 범위였다. NDF 함량은 추출 후 기준으로 목단피, 복령, 택사 순으로 높고 숙지황이 가장 낮았다. 그리고 추출 후의 ADF 함량은 산수유, 복령, 목단피의 순으로 많고 산약이 가장 낮았다. 육미지황탕박의 TMR에 대한 대체 비율에 따른 건물 소실률은 육미지황탕박의 첨가비율이 높은 시험구에서 많이 감소하였는데 10% 대체 시에는 유의차가 인정되지 않는 범위 내에서 감소하였지만 20% 대체 시에는 0 및 10% 대체 시에 비하여 유의하게 감소하였다. 실험 2의 결과에 있어, 30분 동안 및 6시간동안의 섭취량은 당밀 첨가구가 무첨가구에 비하여 섭취량이 유의하게 많았으며, 당밀 첨가수준이 높아짐에 따라 pH 및 초산의 수준에는 일정한 경향을 관찰할 수 없었고, 총산 프로피온산 발레린산의 수준은 높아지는 경향이었다. 그리고 이소-낙산 낙산 이소-발레린산의 농도는 낮아지는 경향이었다. A/P 비율은 당밀 1% 첨가구에서 가장 낮게 나타났으나 일정한 경향을 나타내지는 않았다. 이상의 결과에서 보면, 육미지황탕박은 TMR 사료 등에서 낮은 범위내에서 활용이 가능한 것으로 판단된다. 육미지황탕박은 열탕처리를 거침으로 인하여 수용성 탄수화물이 부족하므로, 당밀 등의 수용성탄수화물을 첨가하여 사일리지를 제조하는 것은 기호성을 향상시키고, 반추위내 발효성상에도 부정적인 영향이 없어 유용한 방법 이라고 생각된다. 첨가 수준에 있어서는 1.0% 수준이면 좋은 품질을 유지할 수 있을 것으로 생각된다.

How effective has the Wairau River erodible embankment been in removing sediment from the Lower Wairau River?

  • The district of Marlborough has had more than its share of river management projects over the past 150 years, each one uniquely affecting the geomorphology and flood hazard of the Wairau Plains. A major early project was to block the Opawa distributary channel at Conders Bend. The Opawa distributary channel took a third and more of Wairau River floodwaters and was a major increasing threat to Blenheim. The blocking of the Opawa required the Wairau and Lower Wairau rivers to carry greater flood flows more often. Consequently the Lower Wairau River was breaking out of its stopbanks approximately every seven years. The idea of diverting flood waters at Tuamarina by providing a direct diversion to the sea through the beach ridges was conceptualised back around the 1920s however, limits on resources and machinery meant the mission of excavating this diversion didn't become feasible until the 1960s. In 1964 a 10 m wide pilot channel was cut from the sea to Tuamarina with an initial capacity of $700m^3/s$. It was expected that floods would eventually scour this 'Wairau Diversion' to its design channel width of 150 m. This did take many more years than initially thought but after approximately 50 years with a little mechanical assistance the Wairau Diversion reached an adequate capacity. Using the power of the river to erode the channel out to its design width and depth was a brilliant idea that saved many thousands of dollars in construction costs and it is somewhat ironic that it is that very same concept that is now being used to deal with the aggradation problem that the Wairau Diversion has caused. The introduction of the Wairau Diversion did provide some flood relief to the lower reaches of the river but unfortunately as the Diversion channel was eroding and enlarging the Lower Wairau River was aggrading and reducing in capacity due to its inability to pass its sediment load with reduced flood flows. It is estimated that approximately $2,000,000m^3$ of sediment was deposited on the bed of the Lower Wairau River in the time between the Diversion's introduction in 1964 and 2010, raising the Lower Wairau's bed upwards of 1.5m in some locations. A numerical morphological model (MIKE-11 ST) was used to assess a number of options which led to the decision and resource consent to construct an erodible (fuse plug) bank at the head of the Wairau Diversion to divert more frequent scouring-flows ($+400m^3/s$)down the Lower Wairau River. Full control gates were ruled out on the grounds of expense. The initial construction of the erodible bank followed in late 2009 with the bank's level at the fuse location set to overtop and begin washing out at a combined Wairau flow of $1,400m^3/s$ which avoids berm flooding in the Lower Wairau. In the three years since the erodible bank was first constructed the Wairau River has sustained 14 events with recorded flows at Tuamarina above $1,000m^3/s$ and three of events in excess of $2,500m^3/s$. These freshes and floods have resulted in washout and rebuild of the erodible bank eight times with a combined rebuild expenditure of $80,000. Marlborough District Council's Rivers & Drainage Department maintains a regular monitoring program for the bed of the Lower Wairau River, which consists of recurrently surveying a series of standard cross sections and estimating the mean bed level (MBL) at each section as well as an overall MBL change over time. A survey was carried out just prior to the installation of the erodible bank and another survey was carried out earlier this year. The results from this latest survey show for the first time since construction of the Wairau Diversion the Lower Wairau River is enlarging. It is estimated that the entire bed of the Lower Wairau has eroded down by an overall average of 60 mm since the introduction of the erodible bank which equates to a total volume of $260,000m^3$. At a cost of $$0.30/m^3$ this represents excellent value compared to mechanical dredging which would likely be in excess of $$10/m^3$. This confirms that the idea of using the river to enlarge the channel is again working for the Wairau River system and that in time nature's "excavator" will provide a channel capacity that will continue to meet design requirements.

  • 유전가열을 이용한 내부가열로서 어육연제품을 조리${\codt}$살균함과 동시에 내부수분을 확산${\codt}$이탈시켜 제품의 수분활성을 조절하여 상온보장이 가능한 보장성 어묵을 가공하는 방법과 제품개발을 위한 원료첨가물의 배합 및 가공조건을 검토한 실험결과를 요약하면 다음과 같다. (1) 고등어 연육의 첨가물 배합조건은 고기풀 100에 대하여 전분(옥수수 전분) $10\%$, 소금 $1.5%$, MSG $0.6\%$, sorbitol $3.0\%$, 설탕 $2.0\%$일 때가 가장 적당하였다. (2) 연제품의 형상과 크기는 두께 0.8cm, 직경 8cm의 원반형이 균일한 가열과 팽화 및 표면경화를 막는데 가장 적당하였고, 이것은 연육을 8cm 직경의 원주형으로 충전하여 이것을 열탕중에서 $2{\sim}3$분간 가열처리하여 gel시킨 후 0.8cm 두께로 절단하여 만들수 있으므로 조작이 매우 편리하였다. 동시에 열탕처리는 유전 가열시간을 단축시키는 효과도 있었다. (3) 유전가열은 $5{\sim}6$분간을 2분, $1{\codt}5$분, $1{\codt}5$분 및 1분씩으로 간헐적으로 행하는 것이 제품의 성상과 수분활성 조절 및 살균효과가 좋았다. 1단계 가열에 의하여 확산${\codt}$이탈된 수분은 $60^{\circ}C$, 3m/sec의 열풍으로 2분간 건조하고 계속적으로 600 W 전열기로 $5{\sim}6$분간 가열 표면배소와 함께 최종적으로 수분활성을 조절할 수 있었다. (4) (3)과 같은 조건으로 가공한 연제품은 수분활성 0.84{\sim}0.86, 생균수 $3{\times}10^2/g$이하, TI함량 27.6mg/g였고 texture 시험성적은 hardness 42, cohesiveness 0.53, toughness 4.6, elasticity 0.8, folding test AA였다. (5) 연제품의 일반성분 조성은 수분 $40.1\%$, 단백질 $20.8\%$, 지방 $17.4\%$, 탄수화물 $16.2\%$ 및 회분$5.5\%$였다.

Determination of the Nutritive Value of Tropical Biomass Products as Dietary Ingredients for Monogastrics Using Rats: 1. Comparison of Eight Forage Species at Two Levels of Inclusion in Relation to a Casein Diet

  • In balance experiments with rats either 25 or 50% of the casein protein in the control diet was replaced with one of the following eight sun-dried tropical biomass products: water spinach plants (WS) (Ipomoea aquatica Forsk), leucaena leaves (LL) (Leuceana leucocephala), duckweed plants (DW) (Lemma minor L.), groundnut foliage (OF) (Arachis hypogaea L.), trichantera leaves (Tric) (Trichantera gigantea), indicago leaves (Ind) (Indigofera hirsuta), mungbean foliage (Mb) (Phaseolus aureus), and cassava leaves (CL) (Manihot esculenta Crantz). The experiment included 102 rats with six individuals per treatment group. In three of the 16 biomass treatment groups, feed intake and weight gain of the rats were unacceptably low, and therefore they were excluded from the statistical evaluation, The crude protein (CP) content of the biomass products varied between 20.9% (Tric) and 33.2% (DW), whereas the content of NDF varied between 18.5% (Ind) and 32.2% (DW) of dry matter (DM). The total content of essential amino acids (g/16 g N) was comparable with that of alfalfa meal, except for GF and Tric, which were inferior. Between plant species, differences in dietary digestibility of organic matter (dOM) and CP (dCP) were observed (p<0.001). Also, the replacement level negatively influenced dOM and dCP (p<0.001). The lowest values for dOM (p<0.001) were observed for diets including biomass products with the highest content of NDF (OF, Tric, Mb, LL). Digestibility of CP was negatively affected by level of protein replacement. Significant (p<0.001) differences were found in N-retention and biological value among diets with different biomass products. The most favourable overall results were obtained for DW, WS and CL. The main factors affecting the nutritive value of the diets tested were their NDF content, dCP and AA profile of the biomass. Also antinutritive component(s) may have influenced the process of digestion and metabolism of some of the biomass products.

Effect of Dietary Calcium on Spinal Bone Fusion in an Ovariectomized Rat Model

  • Objective : To evaluate the effect of calcium supplementation on spinal bone fusion in ovariectomized (OVX) rats. Methods : Sixteen female Sprague Dawley rats underwent bilateral ovariectomy at 12 weeks of age to induce osteoporosis and were randomly assigned to two groups : control group (n=8) and calcium-supplemented group (OVX-Ca, n=8). Autologous spinal bone fusion surgery was performed on both groups 8 weeks later. After fusion surgery, the OVX-Ca group was supplemented with calcium in drinking water for 8 weeks. Blood was obtained 4 and 8 weeks after fusion surgery. Eight weeks after fusion surgery, the rats were euthanized and the L4-5 spine removed. Bone fusion status and fusion volume were evaluated by manual palpation and three-dimensional computed tomography. Results : The mean fusion volume in the L4-5 spine was significantly greater in the OVX-Ca group ($71.80{\pm}8.06mm^3$) than in controls ($35.34{\pm}8.24mm^3$) (p<0.01). The level of osteocalcin, a bone formation marker, was higher in OVX-Ca rats than in controls 4 weeks ($610.08{\pm}10.41$ vs. $551.61{\pm}12.34$ ng/mL) and 8 weeks ($552.05{\pm}19.67$ vs. $502.98{\pm}22.76$ ng/mL) after fusion surgery (p<0.05). The level of C-terminal telopeptide fragment of type I collagen, a bone resorption marker, was significantly lower in OVX-Ca rats than in controls 4 weeks ($77.07{\pm}12.57$ vs. $101.75{\pm}7.20$ ng/mL) and 8 weeks ($69.58{\pm}2.45$ vs. $77.15{\pm}4.10$ ng/mL) after fusion surgery (p<0.05). A mechanical strength test showed that the L4-5 vertebrae in the OVX-Ca group withstood a 50% higher maximal load compared with the controls (p<0.01). Conclusion : Dietary calcium given to OVX rats after lumbar fusion surgery improved fusion volume and mechanical strength in an ovariectomized rat model.

Skate cartilage extracts containing chondroitin sulfate ameliorates hyperlipidemia-induced inflammation and oxidative stress in high cholesterol diet-fed LDL receptor knockout mice in comparison with shark chondroitin sulfate

  • BACKGROUND/OBJECTIVES: In this study, we investigated the beneficial effects of skate cartilage extracts containing chondroitin sulfate (SCS) on hyperlipidemia-induced inflammation and oxidative stress in high cholesterol diet (HCD)-fed mice in comparison with the effects of shark cartilage-derived chondroitin sulfate (CS). MATERIALS/METHODS: Low-density lipoprotein receptor knockout (LDLR-KO) mice were fed HCD with an oral administration of CS (50 and 100 mg/kg BW/day), SCS (100 and 200 mg/kg BW/day), or water, respectively, for ten weeks. RESULTS: The administration of CS or SCS reduced the levels of serum triglyceride (TG), total cholesterol (TC), and LDL cholesterol and elevated the levels of high-density lipoprotein cholesterol, compared with those of the control group (P < 0.05). Furthermore, CS or SCS significantly attenuated inflammation by reducing the serum levels of interleukin (IL)-1β and hepatic protein expression levels of nuclear factor kappa B, inducible nitric oxide synthase, cyclooxygenase-2, and IL-1beta (P < 0.05). In particular, the serum level of tumor necrosis factor-alpha was reduced only in the 100 mg/kg BW/day of SCS-fed group, whereas the IL-6 level was reduced in the 100 and 200 mg/kg BW/day of SCS-fed groups (P < 0.05). In addition, lipid peroxidation and nitric oxide production were attenuated in the livers of the CS and SCS groups mediated by the upregulation of hepatic proteins of antioxidant enzymes, such as superoxide dismutase, catalase, and glutathione peroxidase (P < 0.05). CONCLUSIONS: These results suggest that the biological effects of SCS, similar to those of CS, are attributed to improved lipid profiles as well as suppressed inflammation and oxidative stress induced by the intake of HCD.
