For the development of a new antidementia functional food or alternative drug using agricultural products, Job's tears (Coix lachrymajobi L.), which shows high acetylcholinesterase (AChE) inhibitory activity (55.1%) was selected and the extraction conditions of AChE inhibitor were optimized. AChE inhibitor of Job's tears was maximally extracted when it was treated with 60% methanol at $40^{\circ}C$ for 6 h. The AChE inhibitor of the methanol extracts was partially purified by systematic solvent extraction, thin layer chromatography, silica gel chromatography and reverse-phase HPLC and the partial purified AChE inhibitor with inhibitory activity ($IC_{50}$) of $0.608\;{\mu}g$ was obtained. The partial purified AChE inhibitor was soluble in methanol and hexane, and insoluble in water. Its maximum absorption spectra was 230 nm and also it was stable in the range of $30^{\circ}C$ and $70^{\circ}C$ and pH 4.0-8.0 for 1 h.

Paclitaxel (PTX) is a effectively chemotherapeutic agent which is extensively able to treat the non-small cell lung, pancreatic, breast and other cancers. But it is a practically insoluble drug with water solubility less than $1{\mu}g/mL$, which restricts its therapeutic application. To overcome the problem, hyaluronic acid-complexed paclitaxel nanoemulsions (HPNs) were prepared by ionic complexation of paclitaxel (PTX) nanoemulsions and hyaluronic acid (HA) to specifically target non-small cell lung cancer. HPNs were composed of ${\small{DL}}-{\alpha}$-tocopheryl acetate, soybean oil, polysorbate 80, ferric chloride, and HA and fabricated by high-pressure homogenization. The HPNs were $85.2{\pm}7.55nm$ in diameter and had a zeta potential of $-35.7{\pm}0.25mV$. The encapsulation efficiency was almost 100%, and the PTX content was 3.0 mg/mL. We assessed the in vivo antitumor efficacy of the HPNs by measuring changes in tumor volume and body weight in nude mice transplanted with CD44-overexpressing NCI-H460 xenografts and treated with a bolus dose of saline, $Taxol^{(R)}$, PTX nanoemulsions (PNs), or HPNs at a dose of 25 mg/kg. Suppression of cancer cell growth was higher in the PN- and HPN-treated groups than in the $Taxol^{(R)}$ group. In particular, HPN treatment dramatically inhibited tumor growth, likely because of the specific tumor-targeting affinity of HA for CD44-overexpressed cancer cells. The loss of body weight and organ weight did not vary significantly between the groups. It is suggest that HPNs should be used to effective nanocarrier system for targeting delivery of non-small cell lung cancer overexpressing CD44 and high solubilization of poorly soluble drug.
To increase the dissolution rate of practically insoluble biphenyl dimethyl dicarboxylate (DDB), various solid dispersions were prepared with water soluble carriers, such as povidone (PVP K-30), poloxamer 407, sodium deoxycholate (SDC) and polyethylene glycol (PEG) 6000, at drug to carrier ratios of 1:3, 1:5 and 1:10 (w/w) by solvent or fusion method. Dissolution test was performed by the paddle method. The dissolution rate of DDB tablets (25 mg) on market was found to be very low (11.44, 9.02 and 6.42% at pH 1.2, 4.0 and 6.5 after 120 min, respectively). However, dissolution rates of DDB from various solid dispersions were very fast and reached supersaturation within 10 min. DDB-PEG 6000 solid dispersion appeared to be better in enhancing the in vitro dissolution rate than others. Furthermore, the incorporation of DDB and phosphatidylcholine (PC) into ${\beta}-cyclodextrin$ at ratios of 1:2:20, 1:5:20 and 1:10:20 resulted in a 4.9-, 11.2- and 19.6-fold increase in DDB dissolution after 120 min as compared with the pure drug, respectively. This might be attributed to the formation of lipid vesicles which entrapped a certain concentration of DDB during dissolution. On the other hand, the permeation of DDB through rabbit duodenal mucosa was examined using some enhancers such as SDC, sod. glycocholate (SGC) and glycyrrhizic acid ammonium salt (GAA). Only trace amounts of DDB were found to permeate through deuodenal mucosa in the absence of enhancer. SDC was found to markedly decrease the permeation flux of DDB, however, SGC and GAA (5 mM) enhanced the flux of DDB 1.6 and 2.4 times higher as compared with no additive, respectively.
A self-microemulsifying drug delivery system(SMEDDS) composed of Cremophor $EL^{\circledR},\;Labrasol^{circledR}$, and Lauroglycol $FCC^{circledR}$ was prepared for the enhancement of solubility, dissolution rate and bioavailability of ibuprofen(IBP), which is water-insoluble but soluble in oils and surfactants. Phase diagram with various regions including microemulsion area was depicted. The SMEDDS was encapsulated in soft gelatin capsules and their dissolution characteristics in various media were observed in comparison to the generic products commercially available in the market. Soft capsules of SMEDDS formulation showed better dissolution profiles, especially in acidic condition, than the others. For the period of 1 hr dissolution in pH 1.2 medium, it reached over 70% dissolution from soft capsules, compared to less than 40% dissolution from commercial reference tablets. On the other hand, in vivo pharmacokinetic parameters were obtained after oral administrations of different IBP preparations to Sprague Dawley rats. SMEDDS formulation showed higher $C_{max}$ and greater $AUC_{0-5hr}$ than the suspension of reference tablet or IBP powder. Therefore, it is possible to conclude that a newly developed soft capsules employing SMEDDS provides an alternative preparation to improve oral bioavailability of IBP.
일반적으로 cyclodextrin (CD)은 liposome의 구조를 불안정하게 만드는 것으로 알려져 있다. 본 연구에서는 양친매성 고분자전해질을 리포좀에 도입하여 CD에 대한 리포좀의 안정도를 증진시키는 연구를 수행하였다. Transmission electron microscopy와 photocorrelation spectroscopy 결과들로부터 ${\beta}-CD$ (${\beta}CD$)와 hydroxypropyl-${\beta}CD$ ($HP{\beta}CD$)를 함유하는 리포좀에 고분자가 도입되었을 때 CD를 함유하는 phosphatidylcholine (PC)-cholesterol (Chol) 리포좀보다 우수한 구조적 안정성을 나타내었다. 또한, rhaponticin (Rh)을 $HP{\beta}CD$에 포접시키고 이를 함유하는 PC-Chol 리포좀과 고분자가 도입된 리포좀의 안정도를 비교해 보았을 경우도 마찬가지로 고분자가 도입된 리포좀이 월등히 향상된 구조적인 안정성을 나타내는 것을 확인하였다. 이와 더불어 guinea pig의 피부조직을 사용하고 franz-cell을 통한 in vitro 피부흡수실험을 수행한 결과, $HP{\beta}CD$에 의해 가용화된 Rh의 피부흡수가 고분자가 도입된 리포좀에 의해 증진됨을 확인할 수 있었다. 상기 결과들은 양친매성 고분자 전해질의 도입에 따라 CD에 의해 가용화된 특정한 활성성분을 함유하는 리포좀의 구조적인 안정성을 효과적으로 증진시킬 수 있음을 확인시켜 주었으며, 이렇게 향상된 리포좀의 구조적인 안정성을 통해 약물전달시스템 측면에서 많은 응용이 가능할 것으로 생각된다.
Although lovastatin (LS) is widely used in the treatment of hypercholesterolemia, its bioavailability is known to be around 5%. This study was aimed to increase the solubility and dissolution-permeation rates of LS using solid dispersions (SDs) with bile salts. The solubilities of LS in water, aqueous bile salt solutions and non-aqueous vehicles were determined, and effects of bile salts on the cellulose or duodenal permeation of LS from SDs were evaluated using a horizontal permeation system. SDs were prepared at various ratios of LS to carriers, such as sodium deoxycholate (SDC), sodium glycocholate (SGC) and/or 2-hydroxypropyl-$\beta$-cyclodextrin (HPCD). The addition of bile salts (25 mM) in water increased markedly the solubility of LS by the micellar solubilization. Some non-aqueous vehicles were effective in solubilizing LS. From differential scanning calorimetric studies, it was found that the crystallinity of LS in SDs disappeared, indicating a formation of amorphous state. The SDs showed markedly enhanced dissolution compared with those of their physical mixtures (PMs) and drug alone. In the dissolution-permeation studies using a cellulose membrane, the donor and receptor solutions were maintained as a sink condition using pH 7.0 phosphate buffer containing 0.05% sodium lauryl sulfate (SLS). The flux of LS alone was nearly same as that of LS-SDC-HPCD (1:3:6) PM. However, the flux of LS-SDC-HPCD (1:3:6) SD slightly increased compared with drug alone and PM, suggesting that entrapment of LS in micelles does not significantly hinder the permeation across cellulose membrane. In the dissolution-duodenal permeation studies using a LS-HPCD-SDC (1:3:6) SD, the addition of various bile salts in donor solutions (25 mM) enhanced the permeation of LS markedly, and the fluxes were found to be $0.69{\pm}0.41$, $0.87{\pm}0.51$, $0.84{\pm}0.46$, $0.47{\pm}0.17$ and $0.68{\pm}0.32{\mu}g/cm^2/hr$ for sodium cholate (SC), SDC, SGC, sodium taurodeoxycholate (STDC) and sodium taurocholate (STC), respectively. The stepwise increase of donor SGC concentration increased the flux dose-dependently. From the relationship of donor SGC concentration and flux, the concentration of SGC initiating the permeation across the duodenal mucosa was calculated to be 11.1 mM, which is nearly same as the critical micelle concentration (CMC, 11.6 mM) of SGC. However, with no addition of bile salts and below CMC, the permeation was very limited and irratic, indicating that LS itself is very poor permeable. Higher protions of bile salt in SD such as LS-SDC or LS-SGC (1 : 49 and 1 : 69) showed highly promoted fluxes. In conclusion, SD systems with bile salts, which may form their micelles in intestinal fluids, might be a promising means for providing enhanced dissolution and intestinal permeation of practically insoluble and non-absorbable LS.
고체분산체는 난용성약물의 용해도를 향상시키는 기술로 용출특성을 고분자의 성질, 고분자의 함량 및 물리화학적 성질에 의하여 변화한다. 본 연구에서는 이부프로펜과 셀룰로오스 아세테이트를 사용하여 분무건조와 로터리 증발법을 사용하여 고체분산체를 제조하였다. 제조한 고체분산체의 표면성질을 주사전자현미경(SEM)을 사용하여 분석하였으며, X선 회절기(X-RD)와 사치주사열량계(DSC)를 사용하여 결정성 변화를 관찰하였다. 고체분산체의 화학적 변화를 관찰하기 위하여 적외선 분광기(FTIR) 분석을 하였으며, 수분 친화도를 측정하기 위하여 수분접촉각 측정을 하였다. 용출 특성은 인공정액(pH 6.8) 및 인공위액 (pH 1.2)에서 관찰하였다. 실험결과 약물의 결정성은 고분자의 비율이 증가하면 결정성이 감소하였으며, 화학적 변화는 없었다. 수분 친화도는 고분자비율과 비례하였으며, 분무건조로 제조된 고체분산체의 경우가 조금 높은 수분 친화도를 가졌다. 용출실험 결과 인공위액에서는 고분자의 비율이 높아질수록 용출률이 증가하였으며, 인공장액에서의 용출속도는 고분자의 비율과 반비례하는 것을 볼 수 있었다.
본 연구에서는 초임계 이산화탄소를 이용한 SAS 공정을 난용성 약물인 이트라코나졸과 친수성 물질인 HP-$\beta$-CD의 포접복합체 미세입자의 제조에 적용하기 위한 기초 연구를 수행하였다. 이트라코나졸과 HP-$\beta$-CD를 1:2의 몰비로 혼합한 용액을 사용하여 35∼$65^{\circ}C$의 온도범위와 83∼140 bar의 압력범위에서 SAS 공정으로 이트라코나졸/HP-$\beta$-CD 포접복합체 미세입자를 제조하였으며, 이트라코나졸 및 HP-$\beta$-CD 원재료의 열적 특성과 비교함으로써 초임계 유체 공정에 의해 포접복합체를 제조할 수 있음을 확인하였다. 또한, SAS 공정에 의해 제조된 이트라코나졸/HP-$\beta$-CD 포접복합체의 인공위액에 대한 이트라코나졸의 용출시험을 수행한 결과 이트라코나졸 원재료와 이트라코나졸을 함유한 대표적 시판 제제인 스포라녹스 캡슐제와 비교해 투입된 이트라코나졸의 50∼80%에 해당하는 양이 용출개시 5분 만에 방출되는 매우 빠른 초기 용출특성을 보임을 확인할 수 있었다. 이트라코나졸 /HP-$\beta$-CD 포접복합체의 제조시 SAS 공정 조건이 35$^{\circ}C$에서 $65^{\circ}C$로 높아짐에 따라 이트라코나졸의 용출률이 감소하였을 뿐만 아니라 열분석 결과 이트라코나졸의 용융 피크의 세기도 점차로 증가하게 된다는 결과로부터 포접복합체의 형성이 이루어지는 주변 매질의 온도가 높아짐에 따라 초임계 이산화탄소 분자의 활동도가 증가하게 되어 이트라코나졸과 HP-$\beta$-CD의 포접복합체 형성에 필요한 결합력이 점차로 약해져서 포접 효율이 저하하게 됨을 확인할 수 있었다.
