  • Oryeong-san which was first recorded in Shanghanrun describing the treatments of acute febrile disease is one of the frequently used oriental medicines. Oryeong-san has been prescribed for the treatment of symptoms accompanied by edema. The purpose of this study was to examine the diuretic effects of Oryeong-san by different routes of administration. Oryeong-san (100 mg/kg body weight) was administrated by three different routes in Sprague-Dawley rats: intravenous infusion, intraperitoneal injection and oral intake. Oral intake of Oryeong-san significantly increased urinary volume and excretion of $Na^+$, $Cl^-$, and $K^+$ compared to vehicle-treated control group. The effects were concentration-dependent. Intravenously administrated Oryeong-san increased urinary volume and electrolyte excretion but without significance in hydrated (0.02 ml/min/rat for 90 min) anesthetized rats. Similarly, intraperitoneally injected Oryeong-san had no effects on water and urine electrolyte excretion compared with saline control group. These findings suggest that Oryeong-san has different effects on water and electrolyte balance by routes of administration.

  • The effects of the mixed two solvents, Dimethylacetamide (DMAc) and Dimethylformamide (DMF), and Polyethylene glycol (PEG) and Polyvinylpyrrolidone (PVP) as additives on performance of Polyvinylidene fluoride (PVDF) membranes were studied. Initially, PEG200 was used as a primary additive at fixed percentage of 5% wt. PVP was then blended with PEG200 in different concentrations. PVDF and DMAc were used as polymer and solvent in the casting solutions, respectively. To control the diffusion rate of PVP in the presence of PEG200 and PVP blend, mixtures of DMAc and DMF were used as the mixed solvent in the casting solutions. Asymmetric PVDF membranes were prepared via phase inversion process in a water bath and the effects of two additives and two solvents on the membrane morphology, pure water flux (PWF), hydrophilicity and rejection (R) were investigated. Attenuated Total Reflection Fourier Transform Infrared Spectra (ATR-FTIR) analysis was used to show the residual PVP on the surface of the membranes. Atomic Force Microscopy (AFM) was utilized to determine roughness of membrane surface. The use of mixed solvents in the casting solution resulted in reduction of PVP diffusion rate and increment of PEG diffusion rate. Eventually, PWF and R values reduced, while porosity and hydrophilicity increased.

  • Plant leaf surface is an important niche for diverse epiphytic microbes, including bacteria and fungi. Plant leaf surface plays a critical frontline defense against pathogen infections. The objective of our study was to evaluate the effectiveness of a starch-based super-absorbent polymer(SAP) combination, which enhances water potential and nutrient availability to plant leaves. We evaluated the effect of SAP on the maintenance of bacterial populations. In order to monitor bacterial populations in situ, a SAP mixture containing Pseudomonas syringae pv. tabaci that expressed recombinant green fluorescent protein(GFPuv) was spray-challenged onto whole leaves of Nicotiana benthamiana. The SAP combination treatment enhanced bacterial robustness, as indicated by disease severity and incidence. Unexpectedly, bacterial numbers were not significantly different between leaves treated with the SAP combination and those treated with water alone. Furthermore, young leaves treated with the SAP combination had more severe symptoms and a greater number of bacterial spots caused by primary and secondary infections compared to young leaves treated with the water control. In contrast, bacterial cell numbers did not statistically differ between the two groups, which indicated that measurement of viable GFP-based bacterial spots may provide a more sensitive methodology for assessing virulence of bacterial pathogens than methods that require dilution plating following maceration of bacterial-inoculated leaf tissue. Our study suggests that the SAP combination successfully increased bacterial aggressiveness, which could either be used to promote the ability of biological agents to control weedy plants or increase the robustness of saprophytic epiphytes against competition from potentially harmful microbes.

  • This study was carried out to evaluate the three months subacute intravenous toxicity of water soluble dimethyl dimethoxy biphenylate derivative (DDB-S), a newly formulated therapeutic agent for hepatitis, in Beagle dogs. Groups of 12 male and 12 female dogs were given different dosage of DDB-S, 10 mg/kg/day (high dose group), 5 mg/kg/day (middle dose group), 2.5 mg/kg/day (low dose group) and 0 mg/kg/day (control group) for three months by intravenous route. 1n the three months intravenous toxicity study, there were neither dead animals nor significant changes of body weights during the experimental period. 1n addition to, no significant DDB-S related changes were found in clinical signs, urinalysis and other findings. Statistical changes were observed in hematological. biochemical, partial thromboplastin time (PIT) and organ weight parameters of treated groups. However, these alteration had no relationship with dosage. No histopathological lesions were observed in both control and treated animals. Above data suggest that no observed adverse effect level of test materials in Beagle dogs might be over 10 mg/kg/day in this study.

  • In order to utilize cinder melting slag as a filter media to control the quality of early rainwater, its environmental stability was verified by heavy metal elution experiment and improved by pre-treatment. Possibilities of improving its function as an absorbent was considered. Absorption characteristics of melting slag before and after the pre-treatment were analyzed by heavy metal equilibrium and stationary-phase column experiments, which in turn were analyzed by comparison experiment with activated carbon. As a result of heavy metal elution experiment, every metal item existed in a much lower amount than the criteria or was not detected, implying that there is no problem recycling it. Absorption equilibrium experiment showed that the time for pre-treatment melting slag to reach the equilibrium was reduced, while the absorbed amount was greatly increased. Stationary-phase column experiment assures us that the elimination rate was not changed much by influx rate, pH and the change in packing volume rate, indicating that this melting slag can be used not only as a filter media to control the quality of early rainwater but also in many areas of water-processing.

  • A laboratory jar fermenter was modified to measure the duration for cooling water supply and the temperatures of the coolant at the inlet and outlet of water jacket. Successful operation of temperature control and on-line measurement was achieved by adjusting optimum parameters of the Proportion-Integral-Derivative temperature controller. The variables measured on-line were used to estimate cooling rates from empirical equations comprised of the time period of cooling water supply and the temperatures of coolant. The measured cooling rate showed a good correlation to the specific growth rate during batch cultivation of E. coli. Cooling rate was measured and applied to programmed cell growth in a fed-batch cultivations. Three fed-batch cultivations were demonstrated by feeding substrate to follow the programmed cooling rates increasing exponentially.

  • Effects of washing and additives on the texture of squid surimi gel which has been known to hard to gelation due to high protease activities and many water solubles were studied by SDS-PAGE, compression test, jelly strength and transmission electron microscopy analysis (TEM). Myosin (205 kDa) heavy chain was the major protein in water soluble fractions. It was impossible to make a gel after washing of the minced squid meat. These results suggested that squid (Todarodes pacificus) minced meat does not need a washing for good jelly products. $3.0\%$ of bovine plasma protein (BPP) produced the hardest gel ($16\%$ harder than the control) among the additives including egg white (EW), potato extracts (PE) and transglutaminase-K (TG-K) by compression test (P>0.05). Microstructure of control, $2\%$ EW and $4\%$ TG-K treated gels showed a sponge-like structure with more vacant space. Gels containing $3\%$ BPP formed the most rigid and arranged networks. Those results indicates that poor gel-network formation Was due to the degradation of myofibrillar proteins by proteases contained in the minced meat, which result in non-interlinkage.
