The Effects of Saururus chinensis Baill Extract Administration on the Blood Glucose, Electrolyte and Lipid Metabolism in STZ-Induced Hyperglycemic Rats (Streptozotocin 유도 당뇨성 흰쥐의 혈당, 전해질 및 지질대사에 삼백초 추출액이 미치는 영향)

  • The purpose of this study was to observe the effects of the feeding physiological activity substance in Saururus chinensis Baill extract on the improvement of the blood glucose, electrolyte (Na, K, CI) concentration and lipid metabolism in the serum of streptozotocin (STZ, 55 mg/kg B.W., I.P. injection)-induced hyperglycemic rats fed the experimental diets for 5 weeks. Concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, triglyceride (TG) and phospholipid (PL) in serum were significantly higher in the hyperglycemic group (group NSW), STZ (I.P.)+Saururus chinensis Baill leaf 3.5 g% extract group (group NSSL) and STZ (I.P.)+Saururus chinensis Baill root 3.5 g% extract group (group NSSR) than those in the control group (group Normal, basal diet+water). But the concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, TG and PL in serum were remarkably lower in the groups NSSL and NSSR than those in the NSW, whereas the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration in the groups NSSL and NSSR were significantly higher than in the group NSW. Electrolyte and creatinine concentrations in serum were significantly lower in the groups NSSL and NSSR than those in the group NSW. The activities of aminotransferase (AST, ALT), creatine phosphokinase (CPK), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum were remarkably lower in the groups NSSL and NSSR than in the hyperglycemic group NSW. However, no significance was found in the comperative effect of the groups NSSL and NSSR. From the above results, it was suggested that Saururus chinensis Baill was effective in the improvement of the blood glucose, electrolyte, glucide and lipid metabolism in serum of STZ-induced hyperglycemic rats.

Effects of the Molecular Weight and Type of Chitosans on Shelf-life of Makkulli (막걸리의 저장성에 미치는 분자량별 및 형태별 키토산의 영향)

  • Effects of the molecular weight and type of chitosans on shelf-life of Makkulli were evaluated during 18 days of storage at $25^{\circ}C$. Two types of chitosans were studied: ${\alpha}$-chitosans with 11 different molecular weights (water-soluble, Mw = 1, 8, 22, 43, 67 and 616 kDa; acid-soluble, Mw = 282, 440, 746, 1,110 and 2,025 kDa) and ${\beta}$-chitosan (acid-soluble, Mw = 577 kDa). Acid-soluble chitosans were applied as a form of chitosan-ascorbate. All chitosans were added to Makkulli at 0.002% concentration, the optimum concentration established in a preliminary test. Among 12 chitosans, the ${\alpha}$-chitosans with 22 and 440 kDa exhibited stronger antimicrobial effects than did other ${\alpha}$- and ${\beta}$-chitosans. The results for pH, acidity, alcohol concentration, viable cell counts, and sensory evaluation suggested that addition of ${\alpha}$-chitosans with 22 and 440 kDa increased the shelf-life of Makkulli by almost 1 week at $25^{\circ}C$ compared with that of control (without chitosan) and other chitosan-added groups. Extension of Makkulli shelf-life by 1 week is fairly significant in view of the magnitude of the total amount of Makkulli produced in Korea.

Effect of Olive Oil Substitution Ratio on Quality Characteristics of Low-fat Pork Patty (올리브유 대체 비율이 저지방 돈육 패티의 품질 특성에 미치는 영향)

  • This study investigated the effect of olive oil substitution ratio on physicochemical quality of pork patty. Five treatments were performed as follows: control (T0, 20% pork fat), T1 (pork fat 15% and olive oil gel 5%), T2 (pork fat 10% and olive oil gel 10%), T3 (pork fat 5% and olive oil gel 15%), and T4 (olive oil gel 20%). The moisture increased and fat amount was decreased in proportion to olive oil substitution ratio. The L* and a* values were highest levels in raw and cooked pork patty of T0, and the b* value was highly enhanced in of cooked pork patty of T4. The water holding capacity, moisture and fat retention were significantly increased, and the cooking loss, diameter reduction and shrinkage ratio were decreased by olive oil substitution ratio. The hardness, springiness and chewiness showed the highest level in T4, and the cohesiveness and gumminess were highest in T0. The reduction ratio of cholesterol in T1, T2, T3 and T4 were 13.8%, 21.6%, 34.5% and 49.0%, respectively, indicating that the content was lowest level in T4. The palmitic acid was the most abundant saturated fatty acid, and the oleic acid was the most abundant unsaturated fatty acid. The unsaturated fatty acid of pork patty was increased in proportion to olive oil substitution ratio. Therefore, the olive oil gel substitution of low-fat pork patty results in a positive effect on the physicochemical qualities due to reduced cholesterol and saturated fatty acids.

Anti-hepatotoxic Activity of Chrysanthemum coronarium L. var. spatiosum Extract (쑥갓의 간독성 보호작용)

  • Total methanolic extract of Chrysanthemum coronarium L. var. spatiosum (Compositae) was revealed to have anti-hepatotoxic activity against galactosamine-induced toxicity on primary cultured rat hepatocytes. After successive partitioning with chloroform, n-butanol, and water, the chloroform fraction showed a significant inhibition activity of 51% at 50 ppm, compared with that of silybin, 45.9% at $100\;{\mu}M$. The chloroform fraction was subjected to silica gel column chromatography and yielded active CH-II, CH-V and CH-VI subfractions, and the anti-hepatotoxic activity of these subfractions were 47.6, 56.3, and 23.2%, respectively, at 50 ppm. Total glutathione contents of CH-II, CH-V, and CH-VI increased by 49.8, 43.9, and 47.5% of the control, respectively at 50 ppm, whereas that of silymarin was, 59.7% at $100\;{\mu}M$ after challenged with galactosamine. The ratio of (reduced glutathione) / (total glutathione) in CH-II, CH-V and CH-VI subfraction showed similar values of $0.86{\sim}0.87$ at 50 ppm, whereas that of silymarin was, 0.85 at $100\;{\mu}M$. The incorporation of $[^3H]-uridine$ uptake into RNA was not affected by these active subfractions.

Effects of Rice Hull Cover for Seed Germination, Types of Tray and Soil, Shading Conditions for Seedling Growth of Codonopsis pilosuala (왕겨 피복에 따른 만삼 종자발아와 육묘를 위한 트레이, 토양 및 차광처리 효과)

  • This study was conducted to determine the effects of rice hull cover on seed germination and tray types, soil types, shading conditions for seedling growth of Codonopsis pilosuala. We also examined the feasibility of cultivation of small seedlings transplanted in the Gyeonggi-do area for a month. Under control condition, the seed germination was 8% whereas it dramatically increased to 78% under rice hull cover treatment. Under the different conditions (tray types, soil types, and shading conditions), young seedlings showed the best quality without shading in TKS soil of 50 plug cell tray, with the growth characteristics of plant height (11.9 cm), number of leaves (71), leaf width (3.1 cm), leaf length (2.6 cm), and root length (14.3 cm). Seedling quality was the best without any shading in TKS+perlite, with the physiological characteristics of evaporation (3.9 $mmol{\cdot}m^{-2}s^{-1}$), carbon assimilation (9.1 ${\mu}mol{\cdot}m^{-2}s^{-1}$), and water use efficiency (2.2 ${\mu}mol{\cdot}m^{-2}s^{-1}$). Considering the economical, morphological, physiological and survival rate of the seedlings, it was an ideal method for transplanting seedlings in the field after they have been grown for 30-45 days in TKS and TKS+perlite of 200 plug cell tray in 0% or 30% shaded conditions. As the results of 5 months examination on the possibility to cultivate Codonopsis pilosuala in the Gyeonggi-do area, 88% to 96% of survival rate was observed with normal induced flowers. Therefore, cultivation of Codonopsis pilosuala was possible in the Gyeonggi-do area. While there were symptoms of etiolation and wilting under no shading condition, they did not appear in 30% and 70% shading conditions. Therefore, cultivation of Codonopsis pilosuala is considered to be necessary 30% or 70% shading.

Antidiabetic Effect of Korean Red Ginseng by Puffing Process in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐에서 팽화가공 처리한 홍삼의 항당뇨 효과)

  • Antidiabetic effect of Korean red ginseng (RG) processed by puffing in streptozotocin (STZ)-induced diabetic (DM) rats was investigated. Five week-old SD rats were divided into four groups; normal control (NC) group, DM group, red ginseng (RG) group and puffed red ginseng (PG) group. The RG and PG groups were orally provided with RG or PG dissolved in water (500 mg/kg) respectively for seven weeks after single injection of STZ (50 mg/kg, i.v.) followed by identification of DM. NC group received saline vehicle instead of STZ. At the end of feeding of RG or PG, the changes of fasting blood glucose, serum insulin and amylase level and serum lipid profiles were evaluated. Also, oral glucose tolerance test (OGTT), comet assay and histopathological examination were performed. At 7th week, the fasting blood glucose levels of the RG and PG groups were reduced compared to the DM group by 11.54% and 20.22%, respectively. The result of OGTT did not show significant differences among DM and two red ginseng groups. While serum insulin and TG levels were predominantly improved in PG group (p<0.05), serum amylase level was increased in RG group. Alkaline comet assay for checking the oxidative damage of DNA showed that TL (tail length, ${\mu}m$) and TM (tail moment) in the blood lymphocyte of PG group significantly decreased in contrast with DM group. Histopathological results of pancreas showed that destruction of exocrine as well as endocrine might be cured by the administration of RG and PG. These results suggest that PG could exert more protection against STZ-induced toxicity than RG group.

IN VITRO STUDY OF DEMINERALIZATION INHIBITION EFFECT AND FLUORIDE UPTAKE INTO ADJACENT TEETH OF LIGHT-CURED FLUORIDE-RELEASING RESTORATIVES (수종의 불소방출 수복재의 탈회억제 효과 및 불소침투에 관한 연구)

  • The purpose of this study was to evaluate the effect of light cured fluoride-releasing materials on the inhibition of demineralization. In addition, the pattern of fluoride uptake of adjacent tooth structure was analyzed with EPMA. Eighty intact premolars were restored with $Filtek^{TM}$ Z250(control group, composite), Fuji Filling $LC^{TM}$(RMGI), Dyract $AP^{(R)}$ (compomer) and Beautifil II(giomer). Restored teeth were stored in distilled water for 30 days. Then sixty teeth(n=15) were exposed to demineralizing solution(pH 4.3). Demineralized teeth were bisected and polished. The specimens were observed with confocal laser scanning microscope. The depth of outer lesion and the thickness of inhibition zone were measured. Remained twenty teeth(n=5) were bisected for fluoride uptake analysis. The fluoride analysis were taken at enamel-restoration interface and dentin-restoration interface by electron probe micro-analyzer. The results are as follows: 1. The depth of outer lesion of Fuji Filling $LC^{TM}$ Dyract AP, Beautifil II was shallower than that of $Filtek^{TM}$ Z250 at the margin of restoration(p<0.05). 2. The thickness of caries inhibition zone of Fuji Filling $LC^{TM}$, Dyract AP, Beautifil II was greater than that of $Filtek^{TM}$ Z250 at the margin of restoration(p<0.05). 3. Fuji Filling $LC^{TM}$, Dyract AP, Beautifil II groups showed the greater fluoride uptake into enamel and dentine around restoration than $Filtek^{TM}$ Z250 group. 4. In dentin the difference of fluoride concentration were greater than in enamel, and Dyract AP showed the greatest fluoride concentration in dentin.

Distribution of Zooplankton by ADCP's Echo Intensity in the Coastal Water used Yellow Loess (다층 도플러 유속계(ADCP)를 이용한 황토 살포 해역의 플랑크톤 평가)

  • Harmful algal blooms (HABs), commonly known as red tides, are aquatic phenomena caused by the rapid growth and accumulation of certain microalgae, which can lead to marked discoloration of surface waters, and severe impacts on public health, commerce, and the environment. In South Korea, the red tides have been a serious and recurrent problem, especially along the south coast. Plenty of yellow loess was used to control an outbreak of the red tides for 15 years from 1996 until now. Yellow loess was almost sprayed in the vicinity of a large fish farming industry. In this research, the distribution characteristics and density distribution of zooplankton were investigated in autumn (Oct. 2008) and spring (Apr. 2009) using volume backscattering strength (SV) calculated by the zooplankton collected with north pacific standard (NORPAC) net and the echo intensity measured with ADCP at stations on the study area in the spraying ocean of yellow loess (SOYL), and the non-spraying ocean of yellow ocean (NOYL) by the red tide generating every year. The species number and the individuals per unit volume of the zooplankton collected in NOYL was high and it which was collected in SOYL was low. As a result of comparing the volume backscattering strength ($SV_c$) calculated by species and length of the zooplankton collected with NORPAC net with the volume backscattering strength ($SV_m$) calculated by the echo intensity measured with ADCP at stations on the study area, although $SV_c$ and $SV_m$ of NOYL were generally in agreement, $SV_m$ of SOYL was higher than $SV_c$ 4.3dB, i.e. ADCP is greatly influenced by suspended solid in SOYL. The horizontal distribution map of $SV_m$ at the study area in autumn (Oct. 2008) and spring (Apr. 2009) was drawn. $SV_m$ of SOYL is higher than NOYL and autumn is higher than spring. $SV_m$ can suppress the overestimate or underestimate of $SV_c$.

Alpha-glucosidase Inhibition Activity of Methanol Extracts Obtained from Nine Pteridophyte Species Native to Korea (자생 양치식물 9종의 성엽 및 근경 추출물의 α-glucosidase 억제 활성)

  • This study was conducted not only to analyze ${\alpha}$-glucosidase inhibition activity with fronds and rhizomes of nine Pteridophyte species, but also to select the plant materials suitable for natural ${\alpha}$-glucosidase inhibitor. Harvested rhizomes and fronds were washed, freeze-dried and grinded. After conducting ultrasonification extraction for 30 minutes in ultrasonic water tank with 100% methanol solvent, and vacuum filtration, ${\alpha}$-glucosidase inhibition activity was measured. Acarbose was used as the positive control. After mixing $100{\mu}L$ of 0.7 unit ${\alpha}$-glucosidase enzyme solution into $50{\mu}L$ of extract and reacting them at $37^{\circ}C$ for 10 minutes, $50{\mu}L$ of 1.5 mM ${\rho}$-NPG solution was taken and reacted at $37^{\circ}C$ for 20 minutes. The reaction was stopped with 1 mL of 1 M $Na_2CO_3$ and absorbance was measured in 405 nm. With the regression analysis, the content of solubility solids (the value of $IC_{50}$) which can inhibit 50% of 0.7 unit ${\alpha}$-glucosidase solution's activity was investigated. The frond ($IC_{50}=14.00{\sim}913.33{\mu}g{\cdot}mL^{-1}$) and rhizome extracts ($IC_{50}=12.93{\sim}205.84{\mu}g{\cdot}mL^{-1}$) of nine Pteridophyte species showed higher ${\alpha}$-glucosidase inhibition activity in comparison with acarbose ($IC_{50}=1413.70{\mu}g{\cdot}mL^{-1}$). The extracts of fronds and rhizomes showed higher value than acarbose by 1.55~100.98 and 6.87~109.33 times each. Especially, ${\alpha}$-glucosidase inhibition activities of Pyrrosia lingua in fronds and Osmunda cinnamomea var. fokiensis in rhizomes were the highest. The necessary biomass of fronds and rhizomes for inhibiting 50% of ${\alpha}$-glucosidase activity showed the lowest value, 0.35, 0.27 mg each, in O. cinnamomea var. fokiensis. $IC_{50}$ value of P. lingua was the highest among fronds of nine Pteridophyte species, but content of soluble solids was 2.4 times less than O. cinnamomea var. fokiensis. So frond of O. cinnamomea var. fokiensis is more economic in comparison with P. lingua. As the result of this study, O. cinnamomea var. fokiensis showed high ${\alpha}$-glucosidase inhibition activity even with small biomass. Therefore it was considered to be high-valued economic material as natural ${\alpha}$-glucosidase inhibitor.

Effect of Topical Application of Intercellular Lipids on Sodium Lauryl Sulphate-Damaged Skin Barrier Function in Dogs (개에서 sodium lauryl sulphate에 의한 손상 피부에 대한 각질세포간 지질의 국소적용 효과)

  • Ceramide, cholesterol and free fatty acids are the major intercellular lipids, maintaining the integrity of the skin barrier. However, the roles of these lipids in canine skin barrier function are little known. The aim of this study was to evaluate the repairing effects of 2% ceramide (CER), 2% cholesterol (CHO), 2% linoleic acid (LIN) and 2% intercellular lipid mixture (ILM) on damaged canine skin barrier by 1.25% sodium lauryl sulphate (SLS). Transepidermal water loss (TEWL), skin hydration, skin pH and skin thickness were assessed. Histological profiles and transmission electron microscopic (TEM) profiles were assessed on day 12. SLS effectively induced the canine skin barrier damage. TEWL was significantly decreased by topical application of CER and ILM in SLS and vehicle-treated skin on day 8 and 12, respectively (p < 0.05, p < 0.0 I). By end of the experiment all lipids significantly decreased the TEWL as compared with SLS and vehicle control, but CER and ILM more significantly decreased the TEWL than UN and CHO, respectively (p < 0.01). Skin hydration was significantly increased by CER and ILM during experimental periods (p < 0.01). Skin pH was significantly decreased by CER, LIN and ILM. In histological profiles, the thickness of the stratum corneum (SC) was significantly increased by the SC lipids as compared with vehicle and SLS (p < 0.01). Especially, CER and ILM showed more prominent improvement of barrier recovery. In TEM of the SC, SLS induced exfoliations of corneodesmosomes in the SC, and CER and ILM effectively protected exfoliations of corneodesmosomes on SLS-damaged canine skin. These results indicated that topical application of CER and ILM dramatically improved damaged-skin barrier function by SLS. Also, it was considered that the use of CER or ILM was recommended for the management of skin barrier dysfunction by irritant and inflammatory skin disorders such as atopic dermatitis.
