The Effect of Jeju Wild Ginseng Extracts on Skin Barrier via Serine-Palmitoyltransferase (제주산양산삼이 세린-팔미토일 전이효소(Serine-Palmitoyltransferase)를 통해 피부 장벽에 미치는 효과에 대한 연구)

  • Skin is the largest organ that protects the body from the external environmental factors such as smog, cigarette smoke, UV. Protective skin barrier is composed with keratinizational keratinocytes and intercellular lipids such as ceramides, cholesterols and fatty acids combined by the lamellar liquid crystal structure. In this research, we confirmed that the Jeju wild ginseng (JWG) extracts dose-dependently increased the expression of serine-palmitoyltransferase (SPT) protein which is associated with ceramide biosynthesis. In addition, emulsion containing 5% JWG extract was applied on skin of human volunteers for 2 weeks and then significantly reduced transepidermal water loss (TEWL) compared to that of control group. As a results, JWG extract increased the biosynthesis of ceramides that is the key components of the skin lipid through enhancing expression of SPT. In addition, JWG extract reduced TEWL resulting in improvement of skin barrier function. In this context, we suggest that JWG extract could be used as a skin barrier enhancer and moisturing agents in cosmetic fileds.

  • Zaltoprofen, a propionic acid derivative non-steroidal anti-inflammatory drug (NSAID), is known to have powerful inhibitory effects on acute, subacute and chronic inflammation. We developed poly(lactide-co-glycolide)(PLGA) microspheres loaded with zaltoprofen for sustained controlled delivery using an oil-water solvent evaporation methods by varying PLGA molecular weight and cosurfactant contents. Physicochemical properties and morphology of zaltoprofen-loaded PLGA microspheres were investigated by scanning electron microscope, X-ray diffraction and differential scanning calorimeter. The size of microspheres increased with the molecular weight of PLGA and the content of cosurfactants. The increase of PLGA molecular weight and cosurfactant content decreased the porosity of microspheres, subsequently resulting in the slow drug release. The results demonstrated that the adjustment of PLGA molecular weight and the cosurfactant content allowed us to control the drug release profiles of drug-loaded microspheres.

Effects of Cattail Pollen Powders on the Rheology of Dough and Processing Adaptability of White Pan Bread (부들화분을 첨가한 밀가루 반죽의 물성과 제빵 적성)

  • To explore cattail pollen powder as a functional food ingredient, we analyzed the general components of pollen powder, tested changes in the physical properties of dough containing the powder, and investigated the process ability of powder-containing dough in bread making by adding 3%, 6%, or 9% by weight of pollen powder to wheat flour. Cattail pollen powder consisted of (all w/w) 12.7-13.2% water, 15.7-17.8% crude protein, 1.3% crude fat, 7.5-7.7% free sugar, 14.7-18.6% crude fiber, 3.4-4.9% pollen, and 49.7-55.9% soluble nitrogen-free extract (NFE). Analysis of the physical properties of dough mixed with pollen powder showed that as more pollen powder was added, the absorption rate increased, but dough stability decreased. With increasing levels of cattail pollen powder, the falling number decreased, and amylase activity increased. Fermentability was highest in dough made with 3% by weight of pollen powder, and the bread product made from such dough had the greatest volume. As more cattail pollen powder was added, the moisture activity in dough tended to decrease to a greater extent than seen in control dough, and this tendency increased with time. We found that longer storage periods were associated with greater hardness and springiness, which indicated degradation in product quality. Therefore, it is suggested that bread products containing cattail pollen powder should be consumed within 3 days of preparation. In a taste survey, bread baked with 3% (w/w) cattail pollen powder scored highest in all questionnaire items.

Antimicrobial Activity Screening of Sixty-four Evergreen Woody Species According to Extraction Conditions against Trichophyton mentagrophytes (상록성 목본 64종의 추출조건에 따른 무좀원인균의 항균활성 스크리닝)

  • This study was performed to investigate and measure the antimicrobial activity of evergreen woody species extracts on Trichophyton mentagrophytes. To do this, leaves and stems were collected from Wando and Jeju islands, and were used for the extraction with different solvents (i.e., distilled water, 80% ethanol, and 100% methanol), and at different ultrasonic extracting times (i.e., 15, 30, and 45 minutes). The experiment was conducted by using the agar diffusion method. The clear zone was measured after incubating the paper disc containing the plant extract in a bacterial culture medium. The controls were synthetic antimicrobials, methylparaben and phenoxyethanol, at concentrations of 0.4, 1, 2, and 4 mg/disc. Altogether, extracts of 56 out of 64 species used in this study had inhibitory activity, which confirmed their antimicrobial activity against Athlete's foot. Among them, the crude ethanolic extract of Elaeocarpus sylvestris in 45 min showed a zone of inhibition < 20.2 mm, while the clear zone of Actinodaphne lancifolia ethanolic extraction for 30 min was 23.5 mm. Also, Quercus acuta, Dendropanax morbiferus and Daphne odora showed clear zones of 28.0 mm (45 minutes ethanolic extraction), 20.5 mm (45 minutes crude methanolic extraction) and 19.7 mm (45 minutes methanolic extraction), respectively. Thus, these results confirm that the extracts of evergreen woody species have therapeutic potential against Athlete's foot, and suggest that in order to extract adequate amounts of antimicrobial substance from the plant sources, ideal extraction condition has to be considered.

  • Recently, spillways are need to control stable water level for supporting main dams because of floods by unusual change of weather such as Typhoon Rusa. This study has been focused on the amount of leakage through the rock mass distributed fractures and joints under the opened emergency spillway. It is very important to evaluate the amount of leakage as these affect stability of spillway by interaction between effective stress and pore pressure. The commercial program MAFIC has been used for analyzing groundwater flow in fractured rock mass. The results showed that the values of range, average and deviation of leakage were 2.85∼ 3.79×10-1, 3.32×10-1 and 1.70×10-2 m3/day/m2 respectively. Secondary, we have estimated the effect of grouting after the transmissivity(Tf) of joint 1 as main pathway of leakage known from above results was changed from 1.78×10-7 to 1.59×10-9 m2/s. The results showed that the values of range, average and deviation of leakage were 7.80×10-4∼1.53×10-3, 1.18×10-3 and 1.32×10-4 m3/day/m2 respectively. As the result, the amount of leakage after grouting has been decreased by a ratio of 1 to 277.

Evaluation of Natural Attenuation by Addition of Fumarate as Carbon Source and Gene Analysis in Groundwater Sample (지하수 중 탄소원으로 fumarate 주입과 유전자분석을 통한 질산성질소 자연저감도 평가)

  • In the results of monitoring nitrate concentration in more than 8,000 groundwater wells around agro-livestock, the average and maximum nitrate concentration was 9.4 mg/L and 101.2 mg/L, respectively. Since about 31% of the monitoring wells was exceed the quality standard for drinking water, nitrate control such as remediation or source regulation is required to conserve safe-groundwater in South Korea. Typical nitrate-treatment technologies include ion exchange, reverse osmosis, and biological denitrification. Among the treatment methods, biological denitrification by indigenous microorganism has environmental and economic advantages for the complete elimination of nitrate because of lower operating costs compared to other methods. Major mechanism of the process is microbial reduction of nitrate to nitrite and nitrogen gas. Three functional genes (nosZ, nirK, nirS) that encode for the enzyme involved in the pathway. In this work, we tried to develop simple process to determine possibility of natural denitrification reaction by monitoring the functional gene. For the work, the functional genes in nitrate-contaminated groundwater were monitored by using PCR with specific target primers. In the result, functional genes (nosZ and nirK) encoding denitrification enzymes were detected in the groundwater samples. This method can help to determine the possibility of natural-nitrate degradation in target groundwater wells without multiplex experimental process. In addition, for field-remediation application we selected nitrate-contaminated site where 200~600 mg/L of nitrate is continuously detected. To determine the possibility of nitrate-degradation by stimulated-natural attenuation, groundwater was sampled in two different wells of the site and nitrate concentration of the samples was 300 mg/L and 616 mg/L, respectively. Fumarate for different C/N ratio was added into microcosm bottles containing the groundwater to examine denitrification rate depending on carbon concentration. In the result, once 1.5 times more than amount of fumarate stoichiometry required was added, the 616 mg/L of nitrate and 300 mg/L of nitrate were completely degraded in 8 days and 30 days. The nitrite, byproduct of denitrification process, was also completely degraded during the experimental period.

Study on the Lowering of NaCl Content by Co-composting Food Wastes (저농도 식염을 함유하는 음식물쓰레기 퇴비제조)

  • High salt content in food waste limit use as the potential source of compost. Since sodium chloride content should be lower than one percent to meet requirement for agricultural use of compost, we attempted use of three kinds of co-composting material for food wastes compost, which were fermented, dried, and fresh types of amendments which included pig manure, sawdust and puffed rice hull. Food wastes were composted well and stabilized after around the 40th days of composting. Little difference were found between composts amended sawdust and puffed rice hull. The result indicated that puffed rice hull could be used as a good alternative of sawdust. Because sawdust or puffed rice hull were mixed as amendments to control water contents and to dilute high NaCl content of compost material, the C/N ratios of the final products were significantly high over 40. However, NaCl contents of final products were dropped to less than percent on the fresh weight basis by mixing food wastes and the above amendments with the optimum rates.

A Study on the Tannin Weighting of Silk (견의 탄닌가공에 관한 연구)

  • The studies were performed to investigate the optimum conditions for tannic processing of silk by use Chinese Gallotannin and synthesized tannic acid, which are aimed at weighting, dyeing and physical properties of tannin treated silk. 1. It was reasonable that the concentration of tannin solution is 30 grams per liter of Chinese Gallotannin, 15 grams per liter of tannic acid for the efficient weighting of processed silk. The temperature and time for tannin treatment was optimum at 80$^{\circ}C$, 60 minutes and the acidity of tannin solution at pH 2 to 3. 2. In dyeing the tannintreated silk by Acid dye Orange II, the temperature and time was reasonable at 60$^{\circ}C$, 90 minutes to control the desorption of tannin components weighted onto silk. 3. The colour differences ($\Delta$E) of dyed silk fabric by soaping could be remarkably narrowed by tannin treatment, resulting in improving the washing fastness of tannin treated silk by two grades more than that of untreated one. 4. The light fastness of tannin treated silk could be drastically improved by reducing the dye-loss of dyed silk fabric which was coused from the Ultra-violet ray irrdiation. 5. The rubbing fastness and water repellency of tannin treated silk was at the same level with that of untreated one. However, the Drape coefficient of tannin treated silk was decreased more than that of untreated one, which is closely related with fabric softness and dressing appearence.

Prioritizing Environmental Risks of Veterinary Antibiotics Based on the Use and the Potential to Reach Environment (사용량과 잠재적 환경 유출 가능성을 기준으로 한 축산용 항생제의 우선순위 결정)

  • Veterinary antibiotics have been widely used to increase feed efficiency, to prevent disease, and to promote growth as well as to control disease. The antibiotics administered can be excreted through the urine and feces. One of the major routes of veterinary antibiotics entering soil and water environment is via the application of animal manure to agricultural land as an organic fertilizer source. Since little is known about impacts of antibiotics on the environment, this study was conducted to prioritize the veterinary antibiotics based on the consumption and potential to reach the environment. Among 83 veterinary antibiotics consumed in Korea, ten antibiotics were used at the greater dose than 25 Mg in 2004. Potential to reach the environment was determined according to excretion rate after administered to animals and sorption affinity to soil solids after applied to agricultural land. Seventeen antibiotic active ingredients (Ais) were classified as 'High' priority in terms of the potential to reach the environment. An overall priority score was determined by combining priority score based on consumption with the degree of potential environment exposure. Twenty veterinary antibiotic AIs were classified as 'Very high' or 'High' priority requiring detailed assessment. The antibiotic AIs were identified four aminoglycosides, two macrolides, two penicillins, five sulfonamides, three tetracyclines, two quinolones, and two miscellaneous. Eight veterinary antibiotic AIs including amoxicillin, carbadox, chlortetracycline, neomycin, oxytetracycline, sulfamethazine, sulfathiazole, and tylosin were identified to have a greater priority of environmental risk in Korea.

Factors Controlling the Losses of Urea through Ammonia Volatilization (암모니아 휘산에 의한 요소비료의 손실에 미치는 요인)

  • Volatilization of ammonia from N fertilizer is the major mechanism of N losses that occur naturally in all soils and is influenced by numerous soils, environmental and N fertilizer management factors. Vegetables are often damaged by $NH_3$ gas volatilized from the high rates of N fertilizer. We determined the rate of $NH_3$ volatilization from urea applied to surface of the alluvial soil (coarse silty, mixed, mesic family of Dystric Fluventic Eutrochrepts, Ihyeon series) as affected by fertilizer management factors such as rate of urea application, irrigation schedule and temperature. The $NH_3$ volatilization was triggered about 3 d after urea application and reached at maximum level in general within 15 days. Cumulative amounts of 3.0, 4.4, and 8.0 kg of $NH_3$ N after 17 d were volatilized at application rates of 200, 400, and $600kg\;N\;ha^{-1}$, respectively, which were equivalent to the N losses of 15.0, 10.9, and 13.0% of N applied. Masses of N volatilization were 5, 21, 75 and $87kg\;NH_3\;N\;ha^{-1}$ at 5, 8, 22, and 28, respectively. Total amounts of 21.3, 21.2, and $16.6kg\;N\;ha^{-1}$ were volatilized at control, 5 and 10 mm water irrigation before fertilization, respectively. However, those at 5 mm irrigation after fertilization were only $10.44kg\;N\;ha^{-1}$. Results showed that urea loss can be avoided by incorporating with the recommended level, applying when temperatures are low or irrigating immediately to carry the urea into soil.
