Biotransformation of Reactive Red 141 by Paenibacillus terrigena KKW2-005 and Examination of Product Toxicity

  • A total of 37 bacterial isolates were obtained from dye-contaminated soil samples at a textile processing factory in Nakhon Ratchasima Province, Thailand, and the potential of the isolates to decolorize and biotransform azo dye Reactive Red 141 (RR141) was investigated. The most potent bacterium was identified as Paenibacillus terrigena KKW2-005, which showed the ability to decolorize 96.45% of RR141 (50 mg/l) within 20 h under static conditions at pH 8.0 and a broad temperature range of 30-40℃. The biotransformation products were analyzed by using UV-Vis spectrophotometry and Fourier-transform infrared spectroscopy. Gas chromatography-mass spectroscopy analysis revealed four metabolites generated from the reductive biodegradation, namely sodium 3-diazenylnaphthalene-1,5-disulfonate (I), sodium naphthalene-2-sufonate (II), 4-chloro-1,3,5-triazin-2-amine (III) and N1-(1,3,5-triazin-2-yl) benzene-1,4-diamine (IV). Decolorization intermediates reduced phytotoxicity as compared with the untreated dye. However, they had phytotoxicity when compared with control, probably due to naphthalene and triazine derivatives. Moreover, genotoxicity testing by high annealing temperature-random amplified polymorphic DNA technique exhibited different DNA polymorphism bands in seedlings exposed to the metabolites. They compared to the bands found in seedlings subjected to the untreated dye or distilled water. The data from this study provide evidence that the biodegradation of Reactive Red 141 by P. terrigena KKW2-005 was genotoxic to the DNA seedlings.

Physicochemical Properties and Antioxidant Activity of Extract from Astragalus membranaceus Bunge Leaf Fermented with Lactic Acid Bacteria (유산균으로 발효한 황기 잎 추출물의 이화학적 특성 및 항산화 활성)

  • Background: This study aimed to investigate the quality characteristics of Astragalus membranaceus Bunge leaf (AMBL) fermented with lactic acid bacteria and the applicability of its biologically active compounds. Methods and Results: An assessment of physicochemical properties such as pH, total acidity, free sugars, and isoflavonoid (calycosin-7-o-β-d-glucoside, ononin, calycosin, and formononetin) was conducted. Furthermore, the levels of antioxidant compounds, including polyphenols and flavonoids, and radical scavenging activities of the extracts using 2,2-Diphenyl-1-picryl-hydrazyl-hydrate and 2,2-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) were investigated. The calycosin content in the water extract of AMBL fermented with Leuconostoc mesenteroides increased by approximately twice as much as the control. Conclusions: These results indicate that L. mesenteroides can be used to improve biological activity through fermentation, and that AMBL can be used as a functional materials and edible resource in industrial areas.

Evaluation of the acute toxicity of theoredoxin (TRX) transgenic soybean to Daphnia magna

  • Theoredoxin (TRX) transgenic soybeans were developed using the human Theoredoxin gene under the control of the ��-conglycinin promoter with a selection marker, the phosphinothricin acetyltransferase (PAT) gene. This study was done to assess the acute toxicity of a genetically modified (GM) soybean using the fresh water planktonic crustacean Daphnia magna. The acute toxicity effect of the TRX soybean and non-GM soybean (Gwangan) on D. magna was investigated at different concentrations (0, 156, 313, 625, 1,250, 2,500, and 5,000 mg·L-1). The TRX soybean used for the test was confirmed to express the TRX/PAT genes by PCR and enzyme-linked immunosorbent assay (ELISA). D. magna feeding tests showed no significant differences in the cumulative immobility or an abnormal response with either the TRX soybean or non-GM soybean. The feeding study showed a similar abnormal response and cumulative immobility of the D. magna between the TRX soybean and Gwangan treatments. Additionally, the 48 h-EC50 values for the TRX and Gwangan soybeans were 755.6 and 778 mg·L-1, respectively. The soybean NOEC (no observed effect concentration) value for D. magna was suggested to be 156 mg·L-1. These results suggest that there is no significant difference in toxicity to Daphnia magna between the TRX soybean and its non-GM counterpart.

A Field Study on the Application of Pilot-scale Vertical Flow Reactor System into the Removal of Fe, As and Mn in Mine Drainage (현장 파일럿 실험을 통한 광산배수 내 Fe, As, Mn 자연정화처리 효율평가)

  • This study aimed to monitor a pilot-scale vertical flow reactor (VFR) system being operated in long-term for water quality control of pH-neutral mine drainage containing Fe, Mn and As, discharged in D mine site. The treatment systems of VFR and zero manganese reactor (ZMR) consisted of sand/limestone, and steel slag/limestone, respectively. The systems were operated during about six months in order to evaluate their long-term treatment efficiency It was observed that both pH and alkalinity of mine drainage were remarkably increased and more than 98% of Fe, As and Mn ions was continuously removed during the tested period of time. In conclusion, the field results of this work demonstrated that the vertical flow reactor system can effectively treat mine drainage contaminated by Fe, As and Mn.

Effect of Cryoprotectants on the Cryopreservation of Manila Clam, Ruditapes philippinarum Embryo (바지락 발생배의 냉동보존에 관한 보존액의 효과)

  • The possibility and effectiveness of cryopreservation was determined to assess survival rates and improve stock management of thawed embryos of Manila clam, Ruditapes philippinarum. The ideal freezing rates were designed and tested to allow cryoprotectants to equilibrate across the membrane during freezing. Survival rates ranging from 0 to 64.3% were obtained using a stepwise freezing protocol compared with 82.3% control rates. Embryos of Ruditapes philippinarum were equilibrated in 2 CPAs plus sea water for 10 min at 25℃ and then cooled at -1℃/min from 20℃ to -12℃. Straws containing more than 100 embryos were held at 12℃ for 5 min allowing equilibration after seeding and slowly cooled at 2℃/min. to -35℃ for 30 min for equilibration before quenching in liquid nitrogen. Dimethyl sulfoxide (DMSO) is the best cryoprotectant indicated for embryos of R. philippinarum with a survival rate of 64.3±3.28% in the presence of 2.0 M DMSO.

  • This paper studies the problems and improvements of government real estate policies. Moon Jae-in government shifted toward regulation and pledge to curb the tax imposed by speculators. It strengthened regulations on reconstruction and bank loans rather than supply, and raised capital gains taxes. As the government implemented measures, emphasizing political logic rather than the economy, the market is unstable and the economy is in a recession. Land has increased the vicious cycle of problems due to population growth, industrialization, urbanization, and wealth growth. Mis-established land policies not only accelerate land prices, but also accelerate the use of disordered land and lead to disruptions in the trading order. In addition, real estate is so difficult to recover from the land problem that it is difficult to contain water that has been spilled once. This is called the irreversible nature of land. Once the land price rises, it is difficult to regain control and reckless development leads to the destruction of the ecosystem, making it difficult to return. This is why such a complex real estate issue should not be implemented as if it were a punishment in a short period of time with government policies. This paper aims to examine the problems of real estate policies and to examine ways to improve them.

  • In this study, pollution characteristics were investigated for the Andong Dam sediments in the Nakdong River. Sediment samples were collected from the middle, left, and right points of the lake up to 40km upstream of the Andong Dam at intervals of about 5km by stratification and turnover period. In the case of nutrients and total organics such as loss of ignition, total nitrogen, and phosphorus, the degree of contamination is low, but heavy metals of arsenic and cadmium is very high. The contamination level of sediments in the dam area is higher than that of the branch of river and the control group, and concentration of chromium, copper, and lead are higher in the stratification period, and that of zinc is higher in turnover period. Arsenic, cadmium, manganese, and zinc showed contamination variation between upstream and downstream but chromium, copper, mercury, iron and lead didn't show the variation.

Use of condensed molasses fermentation solubles as an alternative source of concentrates in dairy cows

  • Objective: The purpose of present study was to investigate the effects of condensed molasses fermentation solubles (CMS) on lactation performance, rumen fermentation, nutrient digestibility, and serum parameters of dairy cows. Methods: A total of 75 healthy Holstein cows with the same parity (milk production = 35±2.5 kg, body weight = 570±28 kg) were randomly selected and divided into 5 groups. One group served as control group (CON; no CMS), whereas the other 4 groups were CMS1 (accounted for 1% of the diet), CMS2 (2%), CMS3 (3%), and CMS4 (4%). All cows were fed regularly three times each day at 0800, 1600, and 2400 h. Cows received diet and water ad libitum. The experiment lasted for 60 days. Results: Results showed that the dry matter intake, milk yield, and protein of CMS2 were maximum and higher (p<0.05) than CMS4. The ruminal pH was observed less than 6 in CMS3 and CMS4 groups. No noticeable difference of microbial protein was found between CON and CMS2 groups, while the microbial protein in these groups was higher (p<0.05) than CMS3 and CMS4 groups. The apparent digestibility of dry matter, organic matter, and crude protein in CMS2 group was higher (p<0.05) than CMS3 and CMS4 groups. Compared to CMS3 and CMS4 groups, the CMS2 group increased (p<0.05) the serum concentrations of immunoglobulin G and immunoglobulin M on d 60. Conclusion: Therefore, it is practicable that CMS substitutes for a part of concentrates in lactating cows' diets, but higher addition of CMS (more than 3% of the diet) could decrease production performance of dairy cows as seen in the present study.

Effects of Faecal Sludge and Food Waste Composts on Seed Germination and Initial Growth Performance of Acacia auriculiformis (A. Cunn. ex Benth.) and Swietenia mahagoni (L.) Jacq.

  • The study was conducted to observe the effects of compost of faecal sludge (FSC) and food waste (FWC) on seed germination and initial growth performance of Acacia auriculiformis and Swietenia mahagoni seedlings at the nursery of Chittagong University, Bangladesh. Before sowing the seeds, different combinations of FSC and FWC were incorporated with nutrient-deficient natural forest soils. Seed germination and growth parameters of seedlings were recorded after two, three and four month of seed sowing. We observed that seed germination (%) and seedlings growth parameters varied significantly (p<0.05) in the soil added with FSC and FWC in comparison to control. The highest germination was observed 88.89% for A. auriculiformis in combination of soil and FSC at 3:1 and 92.59% for S. mahagoni where soil combined with FWC at 4:1. The seedling biomass and other growth parameters of A. auriculiformis and S. mahagoni was also observed with combinations of soil with FSC 3:1 and soil with FWC 4:1. Therefore, the study revealed that the compost of both FS and FW can be a good soil conditioner for the initial growth of forest seedlings and the proven combinations can help to grow quality seedlings in the nursery.

Effect of Deer Antler Extract on Muscle Differentiation and 5-Aminoimidazole-4-Carboxamide Ribonucleoside (AICAR)-Induced Muscle Atrophy in C2C12 Cells

  • The effect of deer antler extract on muscle differentiation and muscle atrophy were evaluated to minimize muscle loss following aging. Various deer antler extracts (HWE, hot water extract of deer antler; FE, HWE of fermented deer antler; ET, enzyme-assisted extract of deer antler; UE, extract prepared by ultrasonication of deer antler) were evaluated for their effect on muscle differentiation and inhibition of 5-aminoimidazole-4-carboxamide ribonucleoside (AICAR)-induced muscle atrophy in C2C12 cells. Morphological changes according to the effect of antler extracts on muscle differentiation were confirmed by Jenner-Giemsa staining. In addition, the expression levels of genes related to muscle differentiation and atrophy were confirmed through qRT-PCR. In the presence of antler extracts, the length and thickness of myotubes and myogenin differentiation 1 (MyoD1) and myogenic factor 5 (Myf5) gene expression were increased compared to those in the control group (CON). Gene expression of AMP-activated protein kinase (AMPK), MyoD1, and myogenin, along with the muscle atrophy factors muscle RING finger-1 (MuRF-1) and forkhead box O3a (FoxO3a) upon addition of deer antler extracts to muscle-atrophied C2C12 cells was determined by qRT-PCR after treatment with AICAR. The expression of MuRF-1 and FoxO3a decreased in the groups treated with antler extracts compared to that in the group treated with AICAR alone. In addition, gene expression of MyoD1 and myogenin in the muscle atrophy cell model was significantly increased compared that into the CON. Therefore, our findings indicate that antler extract can increase the expression of MyoD1, Myf5 and myogenin, inhibit muscle atrophy, and promote muscle differentiation.
