Antimicrobial Activity of Korea Traditional Prescription(Bangyak-Happyon) and Herb Simplexes Extracts to Vibrio parahemolyticus (Vibrio parahemolyticus에 대한 한방처방 "방약합편(方藥合編)${\rfloor}$ 및 그 단미제의 항균활성에 관한 연구)

  • Vibrio are become prevailing if superficial temperature of ocean is raised and their activities of area are expanded and most of ocean creatures(fishes, oysters etc) are polluted with vibrio. The one who has taken these poluted fishes and aysters uncooked caused foodpoisoning and diarrhea from Vibrio. Frequencies of these diseases breakout is disposed in westsea shore of Korea. According to ancient and traditional Korean medical book -${\ulcorner}$Bangyak Happyeon${\lrcorner}$(Collection of Local Medicines, 1884) - and their single prescribes, we carried out experiment check the activities of natural medicinal effects on Vibrio parahemolyticus. The prescriptions of trial materials are processed from extraction boiling water and 80% methanol and followed freeze dried and adsorbed to every discs in dosage of 10mg. Gentamycin of 10mg were used for control. The result of compound prescription displayed special diseases in antimicrobial activities of boiling water and MeOH extraction compared with control. In compound prescription, extraction MeOH of Sashinhwan(clear zone : 17mm) presented extraordinaire antimicrobial activity. In single prescription, extraction of boiling water(clear zone : 16mm) and MeOH(clear zone : 18mm) of Fructus Chebulae presented extraordinaire antimicrobial activity. The MBC of Fructus Chebulae extracts was expressed in boiling water(1.28mg/ml) and MeOH(0.64mg/ml).

  • It has been suggested that bisphenol A (BPA), a known endocrine disruptor, interferes with the endocrine system, causing reproductive dysfunction. Recently, BPA has been found in waste water due to incomplete sewage purification, possibly threatening health through its ingestion via tap water. In this study, young male mice (6 - 7 weeks old) were administered water containing BPA (50 mg·kg-1) for four weeks, while control mice consumed water without BPA. Serum, epididymal spermatozoa and testicular sections were assessed after sacrificing the mice on day 28. No significant differences were obtained between the groups in the body, testis and seminal vesicle weights. However, the epididymal sperm motility and count levels were significantly reduced in BPA-fed mice. Significantly higher hepatotoxicity levels were also observed in mice ingesting BPA as compared to the control mice. The level of serum testosterone was reduced, and testicular sections revealed incomplete and irregular spermatogenesis in BPA-ingested mice. The sperm proteasomal-proteolytic activity level has been implicated in sperm function and is measured in motile spermatozoa using fluorometric substrates. High ubiquitin C-terminal hydrolase activity levels were observed in the control mice without BPA. During a mating trial, a low pregnancy rate (71.4%) was observed in females mated with males who had consumed BPA (100% in the control mice). Overall, BPA adversely affected spermatogenesis and quality, as indicated by decreased sperm motility, concentration and serum testosterone levels, resulting in reduced fertility competence.

Antioxidant Activity of Ethanol-Extracts of Defatted Soybean, Sesame, and Perilla Flours in a Soybean Oil-Water Emulsion System (탈지(脫脂) 콩, 참깨 및 들깨박(粕)의 에탄올 추출물(抽出物)의 콩기름-물 기질(基質)에서의 산화억제효과(酸化抑制效果))

  • The antioxidant activity of ethanol-extracts of defatted soybean, sesame, and perilla flours was compared with that of 0.02% BHT in a soybean oil-water emulsion system. The emulsion substrates and control were stored at $46.0{\pm}0.5^{\circ}C$ for 25 days. The peroxide and TBA values of the substrates and control were determined regularly. The activity of the oilseed flour extracts and BHT was estimated by comparing the POV development of the substrates with that of the control. The POVs of the substrates containing the soybean, sesame, and perilla flour extracts and BHT and that of the control after 25 day storage were respectively $43.3{\pm}0.1,\;22.6{\pm}0.7,\;21.5{\pm}0.2,\;38.6{\pm}0.4,\;and\;80.1{\pm}0.8$. The TBA values after 20 day storage were $0.91{\pm}0.05,\;0.67{\pm}0.02,\;0.68{\pm}0.01,\;0.38{\pm}0.01,\;and\;0.62{\pm}0.01$ The soybean, sesame, and perilla flour extracts exhibited considerable antioxidant activity in the oil-water emulsion system. The activity of the sesame and perilla flour extracts was far stronger than that of 0.02% BHT in the emulsion system. The abnormally high TBA values of the oilseed flour extracts in the present study might be attributed to the interference of some carbonyl compounds in the extracts in the TBA value determination.

Effects of Dried Leaf Powders, Water and Ethanol Extracts of Persimmon and Green Tea Leaves on Lipid Metabolism and Antioxidative Capacity in 12-Month-Old Rats (감잎, 녹차의 건분 및 물, 에탄올추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향)

  • The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level. Physiological Studies on Acute Water-temperature Stress of Juvenile Abalone, Haliotis discus hannai (급격한 수온 스트레스에 따른 전복, Haliotis discus hannai 치패의 생리학적 연구)

  • This study was conducted to investigate antioxidant enzyme activity (catalase and superoxide dismutase) and Heat Shock Protein 70 (HSP70) mRNA variation in hepatopancreas of abalone (Haliotis discus hannai) cultured under several acute water temperatures. Abalones were cultured at 10, 15, 20, 25 and $30^{\circ}C$, for 0, 6, 12, 24 and 48 hours, respectively. The HSP70 mRNA expression in hepatopancreas was more increased at $30^{\circ}C$ compared to those at 10. 15, 20 (control) and $25^{\circ}C$. The superoxide dismutase (SOD) activity was increased in hepato-pancreas at all water temperature conditions compared to the control ($20^{\circ}C$). The SOD activity at high water temperature (25 and $30^{\circ}C$) tended to be increased after 12 hours, and was increased immediately after exposure to low water temperature (10 and $15^{\circ}C$). and then was recovered to starting level after the increase. Also, catalase (CAT) activity in hepatopancreas was increased in all the groups except for at $10^{\circ}C$ than the control ($20^{\circ}C$). Survival rate of abalone was $100\%$ at 10, 15, 20 and $25^{\circ}C$, but $92\%$ at $30^{\circ}C$. Thus, according to our study, when abalone is appeared at $20^{\circ}C$, defense mechanism against stress at low water temperature can be accelerated to be stabilized at about $5^{\circ}C$. In the case of exposure of abalone to high water temperature, antioxidant enzyme and HSP70 expression were increased due to elevated physiological stimulation factor, such as temperature.

  • Objective : This study was conducted in order to find the effects of Gammaekdaejo-tang (Ganmaidazao-tang, GDT) by subjecting rats to immobilization stress, thereby inducing depression, anxiety and acquisition-retention defects. Method : Rats treated with normal saline, GDT 200mg/kg and GDT 400mg/kg were subjected to stress by immobilization. Afterwards, behavior changes were observed by elevated plus maze test, acquisition test and retention test in the Morris water maze. The results were obtained by immunohistochemically measuring stress hormone (corticosteroid) levels in the blood. Results and Conclusions : 1. The open arm test in the elevated plus maze showed that compared with the normal group, the time spent decreased in the control group and increased in the GDT 400mg/kg group. 2. The locomotor activity test in the elevated plus maze revealed that the control group showed significant activity decrease compared with the normal group but significant increase in the GDT 400mg/kg group. 3. The acquisition test in the Morris water maze showed that the acquisitive ability of the control group significantly deteriorated on the 3rd and 4th day compared with the normal group, but improved significantly in the GDT 200mg/kg and GDT 400mg/kg groups. 4. The retention test on the 7th day in the Morris water maze revealed that the retentive ability of the control group significantly deteriorated compared with the normal group, but the retentive ability of the GDT 400mg/kg group significantly improved. 5. The blood levels of corticosteroid in the control group increased significantly compared with the normal group but the levels of corticosterone in the blood of the GDT 400mg/kg group significantly decreased.

Comparative Study of Antioxidant Activity of Apple and Pear Peel

  • Apple and pear are popular fruits consumed in Korea and are common fruit in daily diet. In order to compare the antioxidant activity of the apple and pear peels, total polyphenol contents, total flavonoid contents, $ABTS^+$ free radical scavenging activity, and DPPH free radical scavenging activity were measured from hot water, ethanol, and methanol extracts of the two fruit peels. The total polyphenol and flavonoid contents were highest in 95% methanol extracts of the apple peelsand 70% ethanol extract of the pear peels, respectively. Total polyphenol contents of the pear peels were higher than that of apple peels, and total flavonoid contents of the apple peels were higher than that of pear peels. The apple and pear peels had the highest $ABTS^{+{\cdot}}$ and DPPH free radical scavenging activity in 95% methanol extracts and 70% ethanol extracts, respectively. $ABTS^{+{\cdot}}$ and DPPH free radical scavenging activity of pear peels was higher than that of apple peels, and the DPPH free radical scavenging activity of apple and pear peels were detected in hot water, 95% methanol, and 70% ethanol extracts, respectively. Ascorbic acid, a synthetic antioxidant used as positive control, had significantly higher scavenging activity than the apple and pear peels. In conclusion, the apple and pear peelshave great potential as natural antioxidants. Therefore, above results should be considered to provide the possibility for the development of high functional antioxidants.

Effect of the Extruded Ginseng on Antioxidant Activity (압출 성형 인삼의 항산화 활성 증강 효과)

  • This study was conducted in order to evaluate the antioxidant activity of extruded ginseng in different extracted fractions. Each of the fractions obtained from extruded ginseng and ginseng (control) were extracted with 80% ethanol, and then the lipophilic components were removed with ether while the hydrophilic components were separated with water-saturated butanol. Each of the 80% ethanol/butanol/water layers were collected and evaporated to acquire samples for tests of saponin content and antioxidant activity. The antioxidant activity of extruded ginseng fractions and ginseng fractions were determined via the oxygen radical absorbance capacity (ORAC) assay. Overall, the extruded ginseng samples harbored saponin contents of 2.2 (Rg1), 2.3 (Re), 1.2 (Rc), 1.3 (Rb2), and 2.2 (Rd) times that measured in the ginseng prior to extrusion. Antioxidant capacity was also higher, not only in the 80% ethanol/butanol which harbor a significant quantity of saponin, but also in the water fractions, which harbor relatively low quantities of saponin as compared to the control samples. Effects of Water Extracts Green Tea Scented with Lotus Nelumbo nucifera Gaertner Flower on Serum Lipid Concentrations in Rats Fed High Fat (백련향차의 열수 추출물이 고지방식이 급여에 의한 흰쥐의 혈청 지질 함량에 미치는 영향)

  • The effects of water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower on serum lipid concentrations were evaluated in rats. Forty-eight male Sprague-Dawley rats weighing l00±l0 g were divided into six groups and fed high fat diets for six weeks. Experimental groups were administered with following diets; Control diet, animal, plant high fat diet and control and high fat diets with 2% water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower. Tissue weights of liver, lung, stomach, heart, kidney and spleen of high fat diet exposed rats were reduced by water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower groups. The concentrations of serum triglyceride in rats fed the water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower were lower than those in other groups. The concentrations of total cholesterol in water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower group were lower than those in high fat diet groups. The concentrations of HDL-cholesterol in serum of the water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower groups were significantly higher than those of other groups. The levels of LDL-cholesterol in serum of the water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower groups were tended to be lower than those of other groups. GPT and GOT activities were decreased in water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower groups and than in the high fat group. LDH activity was lower in the water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower groups than in the high fat group. These results suggest that water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower groups may reduce elevated levels of serum lipid concentrations in rats fed high fat diets.

  • This study was performed to estimate the clinical signs and biochemical para meters of indicator on acute hepatic injury induced by the administration of CCl$_{4}$ in rats. Minor behavioral changer brittleness of skin hair and decreased volume of water and food intake were observed in rats after 2 hours of $CCl_{4}$ administration compared to control group. Concentration of serum albumin has shown lower than that of control group. However concentration of total bilirubin has shown higher than that of control group. As times go onto serum LDH activity was significantly increased compared to control Broup. Serum CPK activity hasn't shown change compared to control group. Passive hemagglutination that of afetoprotein was shown negative reaction in all the treatment groups and control group.
