Enzymatic Hydrolysis Performance of Biomass by the Addition of a Lignin Based Biosurfactant

  • Hydrolysis of biomass for the production of fermentable sugar can be improved by the addition of surfactants. In pulp and paper mills, lignin, which is a by-product of the pulping process, can be utilized as a fine chemical. In the hydrolysis process, lignin is one of the major inhibitors of the enzymatic breakdown cellulose into sugar monomer. Therefore, the conversion of lignin into a biosurfactant offers the opportunity to solve the waste problem and improve hydrolysis efficiency. In this study, lignin derivatives, a biosurfactant, was applied to enzymatic hydrolysis of various lignocellulosic biomass. This Biosurfactant can be prepared by reacting lignin with a hydrophilic polymer such as polyethylene glycol diglycidylethers (PEDGE). In this study, the effect of biosurfactants on the enzymatic hydrolysis of pretreated sweet sorghum bagasse (SSB), oil palm empty fruit bunch, and sugarcane trash with different lignin contents was investigated. The results show that lignin derivatives improve the enzymatic hydrolysis of the pretreated biomass with low lignin content, however, it has less influence on the enzymatic hydrolysis of other pretreated biomass with lignin content higher than 10% (w/w). The use of biosurfactant on SSB kraft pulp can increase the sugar yield from 45.57% to 81.49%.

  • 1978년도부터 2007년도까지 30년간 해양경찰청 소속 해양환경감시원이 선박에 직전 승선하여 해양오염방지설비의 정상가동 여부 및 각종 오염물질 기록부의 비치 기록 관리여부와 폐유 및 폐기물 처리 실적을 확인하고 선외 배출여부 등을 점검하여 오던 해양환경관리법 제 115조에 좌한 사후적 해양오염 점검 성격의 선박출입검사 제도를 선박이 스스로 해양오염을 관리 점검하고 선박 종사자의 불편과 경제적 활동에 대한 부담을 덜어 주기 위해 해양오염을 사전에 예방할 수 있는 선박자율점검 제도에 대하여 제시하였다.

Purification, Characterization and Immobilization of Lipase from Proteus vulgaris OR34 for Synthesis of Methyl Oleate

  • A newly isolated strain, Proteus vulgaris OR34, from olive mill waste was found to secrete an alkaline extracellular lipase at 11 U·ml-1 when cultivated on an optimized liquid medium. This lipase was purified 94.64-fold with a total yield of 9.11% and its maximal specific activity was shown to be 3232.58 and 1777.92 U·mg-1 when evaluated using the pH-stat technique at 55℃ and pH 9 and Tributyrin TC4 or olive oil as the substrate. The molecular mass of the pure OR34 lipase was estimated to be around 31 kDa, as revealed by SDS-PAGE and its substrate specificity was investigated using a variety of triglycerides. This assay revealed that OR34 lipase preferred short and medium chain fatty acids. In addition, this lipase was stable in the presence of high concentrations of bile salt (NaDC) and calcium ions appear not to be necessary for its activity. This lipase was inhibited by THL (Orlistat) which confirmed its identity as a serine enzyme. In addition, the immobilization of OR34 lipase by adsorption onto calcium carbonate increased its stability at higher temperatures and within a larger pH range. The immobilized lipase exhibited a high tolerance to organic solvents and retained 60% of its activity after 10 months of storage at 4℃. Finally, the OR34 lipase was applied in biodiesel synthesis via oleic acid mediated esterification of methanol when using hexane as solvent. The best conversion yield (67%) was obtained at 12 h and 40℃ using the immobilized enzyme and this enzyme could be reused for six cycles with the same efficiency.

헴프 부산물의 열분해 특성 연구 (Pyrolysis Characteristics of Hemp By-products (Stem, Root and Bast))

  • 헴프(Hemp)는 빠른 성장과 재배가 용이한 특성을 가지고 있으며 전통적으로 인피(Bast)는 섬유산업에 이용되고 있다. 그러나 인피를 제외한 줄기(Stem)와 뿌리(Root)는 활용분야가 없어 대부분 부산물로서 폐기되고 있다. 이러한 헴프 부산물은 바이오-오일과 같은 바이오연료나 활성탄의 원료물질로 활용될 수 있다. 본 연구에서는 헴프 줄기, 뿌리 및 인피를 대상으로 열화학적 특성을 파악하였다. TGA를 이용해 열중량분석을 수행한 결과 헴프 부산물들의 분해영역은 대부분 $270{\sim}370^{\circ}C$ 라는 것을 확인하였다. TGA 분석으로부터 얻은 실험데이터는 미분법을 적용하여 전화율 변화에 따라 활성화에너지와 전지수인자를 계산하였다. 열분해반응에서 활성화에너지는 전화율 증가에 따라 증가하였다.

여수산업단지의 생태효율성지표 개발에 관한 연구 (Development of Eco-Efficiency Indicators for Yeosu Industrial Park)

  • 본 논문은 생태효율성 지표 작성을 통하여 여수산업단지(여수산단) 지표를 개발하였다. 여수산단 매출의 경우 원화의 가치와 원부원료의 수출입 원가 변동 폭이 큰 관계로 제품의 가치(기능)에서 제외하고 대신 총 생산액, 에틸렌 생산량, 경질유 생산량으로 제품의 기능 부문을 구성하고, 환경부하로서는 현재 공식적으로 입수 가능한 자료인 전력 소모량, 용수 사용량 및 폐수 배출량을 사용하였다. 2004년을 기준으로 할 때 2006년의 환경경제효율 지수를 계산한 결과 0.954로 지수가 나빠졌으나, 여수산단의 생태산업단지사업이 성공할 경우 2015년에는 지수가 1.153으로 약 15% 정도 개선될 것으로 예측되었다.

토양 및 지하수 Investigation 과 Remediation에 대한 현장적용

  • Situated close to Heathrow Airport, and adjacent to the M4 and M25 Motorways, the site at Axis Park is considered a prime location for business in the UK. In consequnce two of the UK's major property development companies, MEPC and Redrew Homes sought the expertise of Intergeo to remediate the contaminated former industrial site prior to its development. Industrial use of the twenty-six hectare site, started in 1936, when Hawker Aircraft commence aircraft manufacture. In 1963 the Firestone Tyre and Rubber Company purchased part of the site. Ford commenced vehicle production at the site in the mid-1970's and production was continued by Iveco Ford from 1986 to the plant's decommissioning in 1997. Geologically the site is underlain by sand and gravel, deposited in prehistory by the River Thames, with London Clay at around 6m depth. The level of groundwater fluctuates seasonally at around 2.5m depth, moving slowly southwest towards local streams and watercourses. A phased investigation of the site was undertaken, which culminated in the extensive site investigation undertaken by Intergeo in 1998. In total 50 boreholes, 90 probeholes and 60 trial pits were used to investigate the site and around 4000 solid and 1300 liquid samples were tested in the laboratory for chemical substances. The investigations identified total petroleum hydrocarbons in the soil up to 25, 000mg/kg. Diesel oil, with some lubricating oil were the main components. Volatile organic compounds were identified in the groundwater in excess of 10mg/l. Specific substances included trichloromethane, trichloromethane and tetrachloroethene. Both the oil and volatile compounds were widely spread across the site, The specific substances identified could be traced back to industrial processes used at one or other dates in the sites history Slightly elevated levels of toxic metals and polycyclic aromatic hydrocarbons were also identified locally. Prior to remediation of the site and throughout its progress, extensive liaison with the regulatory authorities and the client's professional representatives was required. In addition to meetings, numerous technical documents detailing methods and health and safety issues were required in order to comply with UK environmental and safety legislation. After initially considering a range of options to undertake remediation, the following three main techniques were selected: ex-situ bioremediation of hydrocarbon contaminated soils, skimming of free floating hydrocarbon product from the water surface at wells and excavations and air stripping of volatile organic compounds from groundwater recovered from wells. The achievements were as follows: 1) 350, 000m3 of soil was excavated and 112, 000m3 of sand and gravel was processed to remove gravel and cobble sized particles; 2) 53, 000m3 of hydrocarbon contaminated soil was bioremediated in windrows ; 3) 7000m3 of groundwater was processed by skimming to remove free floating Product; 4) 196, 000m3 of groundwater was Processed by air stripping to remove volatile organic compounds. Only 1000m3 of soil left the site for disposal in licensed waste facilities Given the costs of disposal in the UK, the selected methods represented a considerable cost saving to the Clients. All other soil was engineered back into the ground to a precise geotechnical specification. The following objective levels were achieved across the site 1) By a Risk Based Corrective Action (RBCA) methodology it was demonstrated that soil with less that 1000mg/kg total petroleum hydrocarbons did not pose a hazard to health or water resources and therefore, could remain insitu; 2) Soils destined for the residential areas of the site were remediated to 250mg/kg total petroleum hydrocarbons; in the industrial areas 500mg/kg was proven acceptable. 3) Hydrocarbons in groundwater were remediated to below the Dutch Intervegtion Level of 0.6mg/1; 4) Volatile organic compounds/BTEX group substances were reduced to below the Dutch Intervention Levels; 5) Polycyclic aromatic hydrocarbons and metals were below Inter-departmental Committee for the Redevelopment of Contaminated Land guideline levels for intended enduse. In order to verify the qualify of the work 1500 chemical test results were submitted for the purpose of validation. Quality assurance checks were undertaken by independent consultants and at an independent laboratory selected by Intergeo. Long term monitoring of water quality was undertaken for a period of one year after remediation work had been completed. Both the regulatory authorities and Clients representatives endorsed the quality of remediation now completed at the site. Subsequent to completion of the remediation work Redrew Homes constructed a prestige housing development. The properties at "Belvedere Place" retailed at premium prices. On the MEPC site the Post Office, amongst others, has located a major sorting office for the London area. 만경강 유역의 토양과 수도체중 Cd 및 Zn 함량의 변화 (Variation of Cadmium and Zinc Content in Rice and Soil of the Mangyeong River Area)

  • 수질오염에 따른 Cd 및 Zn의 함량변화 차이를 구명하기 위하여 도시하수 및 공단배출수의 영향을 받는 만경강 유역의 논토양을 중심으로 1982년도와 1990년도에 표토와 심토로 구분하여 채취한 토양시료를 그리고 1990년도의 토양시료 채취지역에서 재배된 수도체 시료중 Cd 및 Zn 함량을 분석하여 그 변화를 분석하였다. 만경강 유역의 토양 중 이들 중금속 분포는 Cd이 0.38-1.17, Zn이 33.8-464.6mg $kg^-$이었다. 년도별 함량변화에서 Cd 함량은 1990년도 토양이 1982년도 보다 낮았고, Zn 함량은 반대로 1982년도 토양이 1990년도 토양 보다 낮았다. Cd의 경우 도시하수 및 공장폐수 배출지로부터 거리별 변화가 분명하지 않았으나, 토양의 유기물함량, 유효규산, CEC, 치환성 $Ca^{++}$ 함량과 유의성 있는 상관관계를 나타내었다. 또한 이 지역의 토양 중 Cd 함량은 1982년도 심토 중 Cd 함량은 토양 중 Zn, Cu 및 Pb 함량과 유의한 상관관계를 나타내었다. Zn의 경우는 거리가 멀 수록 그 함량이 낮아지는 경항이 있었으며, 1982년도 토양 중에서 유기물 함량과 치환성 $Ca^{++}$ 함량과, 그리고 년도에 관계없이 토양 중 Cu와 Pb 함량과 유의성있는 상관관계를 나타내었다. 한편 이 지역의 수도체 부위별 Cd 함량은 현미중 농도에 비해 엽신 중의 농도가 최고 7배 이상이 되었고, Zn의 경우는 엽신과 화서축 중에서 흡수축적량이 높았으며, 토양 중 Zn, Cu, Pb의 존재는 수도체의 화서축 중의 Cd 함량과 또 수도체의 모든 부위별 Zn 함량과 유의성 있는 상관관계를 나타내었다. 만경강유역의 현미 중 Cd의 함량은 0.10-0.90mg $kg^{-1}$이었고, Zn의 함량은 4.2-95.9mg $kg^{-1}$ 범위였다.

이비인후, 안, 치아 및 피부증상의 민간요법에 관한 고찰 (Study on the Herb Remedies of ENT, Eyes, Teeth and Skin Problems)

  • The intention of this study is to apprehend the con. tents and methods of herb remedies that are commonly conducted when there are health-problem cases of ENT, eyes, teeth and skin. Methods of this study are divided into two stages : 1) For a period of six months from December 1994 to June 1995, some 40 persons who are believed to be well versed with herb remedies have been randomly chosen, and we made a survey on herb remedies by symptoms: and 2) we have endeavored to make their grounds evident through the studies on literatures with the focus on the basic data collected. Their results are as follows: 1) When one feels a pain in ears, such herb remedies are employed as pouring the vapor into ears, which is made by steaming Alaska pollack, or as applying or wiping with the juices of radish or the ginkgo, or' alum. Applying the radish juice is effective for sterilization and fever removal: and applying the ginkgo juice is effective for cleanliness. But, plastering alum, sesame oil or castor oil, or pouring the vapor of Alaska pollack into ears are perhaps effective but do not have any pharmacologic grounds. 2) When one bleeds at the nose, such kinds of herb remedies are applied as stimulating nose or head with cold water, pressing nose or ridge of nose, or filling up nares with mashed mugworts. In addition, they have utilized garlic or leeks. Such methods as stimulating with cold water or just pressing nose and ridge of nose is based on reasonable grounds, i.e. vasoconstriction and vascular compression ; and applying mashed garlic on the sole of foot is good for the circulation of Qui ; and the use of mugworts and leeks is based upon the pharmacological function of hemostasis. 3) When one feels a sore throat such kinds of herb remedies are employed as gargling or rinsing throat with brine, drinking hot gruel or water, or drinking the juice of mugwort, radish, ginger or Chinese quince. Gargling with brine or drinking the juice of mugwort, radish or ginger is based upon the pharmacological function of pain alleviation, fever removal, and detoxication. 4) When a boil is formed in mouth, such herb remedies are applied as spreading honey, brine or alum water, and taking gall nut, Chinese matrimony vine, lotus root, etc, for drugs. Spreading honey, brine or water that is made by infusing gallnut, Chinese matrimony vine, lotus root is based upon such functions as hematosis, astriction, antibacterial, and antiphlogistic, Alum, eggplant and licorice are said to be effective, but their pharmacological effects have no grounds. 5) When one has conjunctivitis such herb remedies are commonly applied as irrigation with brine and dropping breast milk in eyes. Moreover, such other drugs are used as plantain. shepherd's purse, and purslane, etc. The use of brine, breast milk, plantain, shepherd's purse and purslane is based upon such functions as sterilization, antiphlogistic, disinfection and pain relieving. Eriocaulon sieboldianum, bean stem, bean pod and narcissus leaves are said to be effective, but their pharmacological action have no basis. When one has a stye, such herb remedies are applied as extracting eyelashes, stimulating by a massage of middle finger, third finger or big toe, as well as sear ing with a heated bamboo comb that is fine-toothed. Other than these, plantain and nightshade's nuts are used as drugs for it. Extracting eyelashes corresponds with exclusing suppurative node and draining the stye of pus ; and the use of plantain is based upon disinfection: and nightshade's nuts are said to be effective, however, their pharmacological action has no grounds. 6) For a treatment of toothache, such herb remedies are commonly employed as rinsing mouth with brine and holding cold water or gasoline in the mouth ; and as the drugs that are believed to be effective have been Welsh onion, ginger and castor-oil, plant, etc. The use of Welsh onion is based upon pain killing, antiinflammatory actions, and the use of ginger is based upon detoxication and disinfection ; and seeds of castor-oil plants are said to be effective, but they have no pharmacological basis. 7) When one has hives, such herb remedies are commonly applied as rubbing burned straw in affected parts, exposing to its smoke, rubbing with salt, sweeping down with a broom, and spreading and drinking boiled water of trifoliate orange. The use of cassia tora seeds, walnut, aloe and radish is said to be effective. The use of cassia tor a seeds has the functions of intestinal order, anti-paralysis, etc. The use of walnut has resulted in an increase of blood by invigorating spirits ; and the use of aloe is based upon disinfection, antibiotic, anti-salt, antihistamine and detoxication action. But, the effects of radish juice and straw's smoke have no pharmacological grounds. 8) When one gets a boil, such herb remedies are commonly used as applying a plaster, paste of flour mixed with yolk, soy sauce or honey, as well as spreading pounded elm tree. Other remedies that have been said to be effective are ; heating with mugwort, brine, wild rocambole, aloe, onion, squid's bone, etc. The use of mugwort is based upon pain killing, astringent antiinflammatory and tranquility. Wild rocambole is based upon the generation and maintenance functions of cell-joining textures ; elm tree upon antiphlogistic ; aloe upon fever removal and antiphlogistic ; onion on pain killing, fever removal, antiphlogistic and tranquility ; squid's bone on astriction: and brine or vinegar on sterilization. Pine resin and gardenia seed are said to be effective, but they have no pharmacological basis. 9) When one cuts his skin, such herb remedies are commonly employed as spreading mugwort's juice or squid's bone powder, or pressing the wounds. In addition, kalopanax, onion and fine soil are employed. The use of mugwort, kalopanax and squid's bone is based upon such functions as hemostasis, sedation, pain killing, antibacterial ; and fine soil is said to be effective, but it has no pharmacological basis. 10) When one suffers from whitlow, such herb remedies are commonly utilized as heating with boiled soy sauce, spreading soybean paste, or dipping into eggs, etc. Other drugs that have been employed are onion root, brine, eggplant, potato, loach, etc. The use of onion is based upon pain killing and antiphlogistic functions ; and that of brine upon antiphlogistic function. The use of soy sauce or soybean paste, fomentation, eggplant, potato and loach is said to be effective, but it has no pharmacblogic ground. 11) For the treatment of frostbite, such herb remedies are commonly used as dipping the affected part into frozen soybean sack, using boiled water of eggplant stem, garlic caulis, onion, hot pepper, caulis. Onion is based upon antiphlogistic and tranquility actions garlic upon disintection, metabolic exacerbation, tonic and aphrodisiac actions and the use of eggplant and hot pepper is based upon help blood circulation, dissolution and excretion of waste matters in vein. 12) For the treatment of burn, such herb remedies or drugs are commonly used as cleansing with Korean gin, spreading eggs, cleansing with cold water and soap water ; and as brine, cactus, moss, soybean paste, oil, etc. The cleansing with Korean gin, cold water, soap water, brine, vinegar is based upon cleaning and sterilizing functions ; and the use of cucumber is based upon nu. trition provision, and strengthening of resisting power by adjustment of metabolism. The use of potato, cactus, moss, oil and eggs is said to be effective, but their phamacological functions are not clarified. In view of the above results, we can realize that the drugs that have been employed in herb remedies are quite diverse. However, in regard to majority of herb remedies that have been employed by symptoms, the pharmacological functions of their drugs have not been clarified, and they are merely known as effective. Furthermore, they have not been recorded in the literature as yet ; and we confirm that there have been many herb remedies that were executed without the proper knowlege of their effects. It is now our view that the results of this survey may be utilized for consulting data in regard to the use of herb remedies.

호기순환 호흡계를 이용한 토양처리 석탄바닥재의 유기물 분해에 미치는 영향 (Effects of Soil-Amended Bottom Ash on Decomposition Rates of Organic Matter as Investigated by an Enforced-Aeration Respirometer)

  • 석탄 연소 부산물인 비산회 (fly ash)는 이산화탄소 발생을 저감하고 토양 탄소를 격리하는 효과가 있음이 보고된 바 있다. 같은 연소 부산물인 석탄바닥재 (bottom ash)는 비산회와 유사한 화학적 성질을 가지고 있고, 아울러 다공성으로 토양미생물들이 정착하는 적절한 담체로서의 기능을 가지고 있다. 이에 본 연구는 성질이 다른 유기물들 (헤어리베치, 청보리, 유박비료)이 처리된 토양에서 석탄바닥재가 토양 미생물들에 의한 유기물 분해 및 호흡량에 미치는 영향을 조사하였다. 미생물에 의한 유기물의 분해속도는 유박비료, 헤어리베치, 청보리의 순으로 높게 나타났고, 유기물과 석탄바닥재를 함께 처리하였을 때, 유의하게 감소하였다. 이산화탄소 발생량의 경시적인 변화는 유기물의 이분해성에 의존하였으나, 석탄바닥재를 처리하였을 때 유의하게 발생량이 감소하였다. 총 누적 이산화탄소 발생량 또한 이와 유사한 결과를 보였다. 석탄바닥재가 함유하고 있는 중금속 등과 같은 유해물질의 농도가 높지 않다면, 본 연구의 결과들은 석탄바닥재가 토양 처리 유기물의 이산화탄소 발생 저감 및 토양 탄소격리에 긍정적으로 기여 할 수 있는 소재로서 가능성이 있음을 보여준다.

  • 태안반도 원유 유출사고, 구미 불산누출사고 등 2000년 전후로 하여 환경관련 사건과 사고가 빈번히 발생하고 있다. 이러한 환경오염사고를 계기로 선제적 대응 방안이 필요하다는 사회적 공감대가 형성되어 2014년 환경오염피해 배상책임 및 구제에 관한 법률이 제정되고, 2016년 1월부터 시행되고 있다. 이에 따라 도입된 환경책임보험제도는 국내 보험산업계에서 환경위험을 관리하는 새로운 보험모델의 표준화 정립을 통해 관리될 필요가 있다. 지금까지 보험산업에서 표준화에 대한 노력은 다양한 위험 유형의 보장성 보험모델들의 출현에 따라 진행되어 왔다. 이에 따라 본 연구에서는 지식 기반 서비스 중의 하나인 보험 영역에서 기업에 의해 발생 가능한 대기, 수질, 화학, 폐기물, 해양, 토양 등의 환경오염 발생에 대해 6가지 보장 매체에 관한 검증 자료를 의미적 상호운용이 가능한 온톨로지를 통해 표현하였으며, 사업장의 인허가를 토대로 보험 가입 매체 간의 관계를 추론하여 보험모델을 설계하고 제시하였다. 각 사업장 담당자에 의한 물질량 정산 과정과 검증자를 통한 정산결과의 편차를 줄이기 위해, 추상적인 개념을 흐름도로 객관화, 구체화 하였으며, 해마다 많은 비용과 소모되는 자원을 줄이기 위해 온톨로지 기반의 의사결정지원 시스템의 향후 구축 방안을 제시하고 일부 구현 하였다. 이를 통해 물질량 검증 기준을 표준화함으로써 오류를 최소화하고, 검증에 소요되는 시간과 자원을 감소시킬 수 있을 것으로 기대된다.
