발효 미역부산물이 반추위 발효특성에 미치는 영향 (Effect of Fermented Brown Seaweed Waste (FBSW) on in vitro Rumen Microbial Fermentation)

  • DS-01 균주 접종에 의한 미역부산물 발효산물이 분해 정도, 영양소 변화 및 미생물오염 정도에 관한 관찰과 반추위 발효성상에 미치는 영향을 관찰하여 미역부산물의 저장성 확보와 반추동물의 사료로서의 이용가능성을 발효단계에 따라 조사하였다. DS-01 균주와 함께 배양된 미역부산물은 발효 1개월부터 현저한 분해율을 보이기 시작했다. 미역부산물중의 영양성분 함량은 발효와 함께 커다란 변화를 나타내지 않았으며 모든 처리에서 반추동물에 병원성을 가지는 미생물은 검출되지 않았다. In vitro 실험에서는 발효시간 및 첨가농도에 따른 pH 변화, 암모니아태 질소와 휘발성지방산의 생성에 대한 영향을 조사하였다. pH의 경우 6% FBSW는 1개월 및 2개월 발효조건에서 대조구, 2% 및 4% FBSW 보다 pH 값이 증가한 것을 나타내었다. 암모니아태 질소의 생성에 미치는 영향을 보면 1개월 FBSW 경우 실험 3시간 경과 후가 대조구에 비해 높은 농도를 나타내었지만 시간이 경과에 따라 대조구보다 감소한 경향을 보인다. 2개월 FBSW의 경우 4% 첨가량만 24시간 경과 후 타 처리구에 비해 높은 농도를 나타내었다. 특히, 전 발효기간 동안 6% FBSW 처리구가 발효 3시간과 24시간 경과 후 타 처리구에 비하여 낮게 나타나는 경향을 보였다. 휘발성지방산의 경우 발효 1개월 미역부산물의 4, 6% FBSW 첨가구가 6시간 경과 후 총 VFA의 농도가 현저하게 증가하는 양상을 나타내었고(p<0.05), 발효 2개월 미역부산물의 2, 4% FBSW 첨가구가 9시간 경과 후 총 VFA 농도가 6% FBSW 첨가구에 비해 현저하게 증가하는 양상을 나타냈다(p<0.05).

허브부식토의 사료내 첨가에 따른 In Vitro 발효특성과 젖소의 유생산성에 미치는 영향 (Effects of Dietary Herbaceous Peat on In Vitro Fermentation and Milk Production in Dairy Cows)

  • 본 연구는 허브 부식토를 이용하여 첨가 수준별 in vitro 반추위 발효특성 평가와 젖소를 이용하여 급여시 유생산성에 미치는 영향을 조사하기 위하여 본 연구를 수행하였다. 시험 1에서는 티머시 건초를 기질로 하여 허브부식토(herbaceous peat)를 0,1 및 5%를 3반복으로 각각 첨가하여 in vitro 반추위내 pH, 가스발생량, VFA (volatile fatty acid), ammonia-N 및 건물분해율을 조사하여 반추위내 발효성상의 변화를 평가하였다. pH 변화는 0, 3, 12 및 24시간 배양에 있어서 모든 처리구에서 유의한 차이를 보이지 않았지만 6시간. 대조구에서 1 및 5% 첨가구와 비교하여 유의하게 낮은 pH를 나타냈다(p<0.05).가스 발생량은 배양 12시간까지 처리구에서 대조구와 비교하여 유의하게 증가 하였으며(p<0.05), 반추위액내 암모니아 농도는 모든 처리구에서 24시간까지 증가하는 경향을 나타냈으며, 처리간 유의한 차이는 없었다. 건물분해율은 모든 시간에서 5%구가 대조구 및 1%구와 비교하여 건물분해율이 높았다(p< 0.05) 따라서 허브부식토를 0, 1 및 5% 수준으로 각각 첨가하여 반추위내 in vitro 배양시간 별 반추위 발효특성은 대부분 조사항목에서 처리구가 대조구에 비해 개선효과를 나타냈으며, 시험 2에서는 젖소를 이용하여 사양시험을 통해 유생산성을 평가하였으며, 우유생산량 변화와 유성분 및 체세포 수 변화를 조사하기 위해 홀스타인 착유우 16두를 공시하여 4주간 사양 시험을 실시하였다. 산유량은 T3구가 25.0 kg으로 대조구(23.2 kg), 부식토 처리구(23.1 kg), 비타민 C 처리구(23.4 kg)와 비슷하여 수치적으로 증가하였다. 대조구와 처리구별 시험 시작 전 유성분(유지방, 유단백, 유당, MUN 및 SNF) 및 체세포 수의 변화는 일부 유의성이 인정되었다(p<0.05). 4주간의 시험 기간 중 전반기(1~2주)와 후반기(3~4주)의 유성분 변화를 보면 부식토 첨가구에 있어서 유단백질과 SNF가 유의적으로 증가하는 경향을 나타내었다. 대조구와 처리구의 시험 시작 전 의 혈액내 영양성분 (총단백질, 콜레스테롤, NEFA, BUN), 간기능 성분(AST, GGT) 및 무기물(Ca, P, Mg)에서 뚜렷한 변화를 나타내지 않았다. 이상의 결과를 종합해 볼 때 착유우에 부식토를 급여하는 것이 유성분, 체세포 수 및 생리적으로 긍정적인 영향을 줄 것으로 판단된다.

조사료와 농후사료의 급여 순서가 In vitro 반추위 발효성상, 총 가스 발생량과 메탄 발생량에 미치는 영향 (The Effects of Feeding Order of Roughages and Concentrates on in vitro Rumen Fermentation, Total Gas and Methane Production in Hanwoo)

  • 본 연구는 국내 고유종인 한우에서 1) 농후사료 급여 후 조사료 급여, 2) 조사료 급여 후 농후사료 급여, 3) TMR 급여의 총 세 가지 형태의 사료급여가 반추위 발효성상, 총 가스 및 메탄 발생량에 미치는 영향을 in vitro로 평가하기 위해 수행되었다. 사료 급여 형태에 따라 반추위 환경이 변화할 것으로 가설을 세우고, 옥수수 (M)와 티모시 (T)를 기질로 하여 반추위액의 24시간 in vitro 배양을 실시하였다. 따라서 시험 처리구는 총 6개로 구성되었다: M1, M2, M3, T1, T2, T3. 반추위 pH는 사료 급여방식 2에서 다른 처리구보다 높았고 (P < 0.001), 티모시 기질에서 옥수수 기질보다 높았다 (P < 0.001). 휘발성 지방산 (VFA; volatile fatty acid) 조사항목들 (Total VFA, acetate, propionate, butyrate, valerate)은 사료 급여방식 1에서 가장 높았고 급여방식 2에서 가장 낮았으며 (P < 0.001), 옥수수 기질에서 더 많이 생산되었다 (P < 0.001). 반추위액 내 acetate:propionate 비율은 사료 급여방식에 따른 효과는 없었으나 (P = 0.116), 티모시 기질이 옥수수 기질보다 더 높은 값을 나타냈다 (P < 0.001). 암모니아태 질소 (NH3-N)는 사료 급여방식 1에서 다른 처리구보다 높았으며(P < 0.001), 티모시 기질에서 높은 수치를 보였다 (P < 0.001). 총 가스 및 메탄가스 발생량은 사료 급여방식 1에서 가장 높았고(P < 0.001), 옥수수 기질에서 더 많이 생산되었다 (P < 0.001). 메탄은 이산화탄소의 20배 이상의 지구온난화효과를 갖는 것으로 알려져 있고, 반추가축이 섭취한 에너지의 손실로 작용하고 있어 장내발효 메탄 발생량을 저감하는 것은 매우 중요한 과제라 할 수 있다. 반추가축에서 사료급여 방식에 따른 메탄 발생량 차이에 대한 결론을 얻기 위해서는 보다 다각적인 형태의 연구가 필요할 것으로 사료된다.

Comparison of Rabbit Caecal Content and Rabbit Hard Faeces as Source of Inoculum for the In vitro Gas Production Technique

  • In order to find an alternative source of inoculum to caecal content for studying the fermentation activity of rabbit hindgut, caecal content and faeces of 25 hybrid Hyla rabbits were used as inocula for an in vitro gas production trial. About 1 g of three substrates (dehydrated alfalfa meal, dehydrated beet pulp, barley) was weighed, in quadruplicate per inoculum, in 120 ml bottles; 75 ml of anaerobic medium and 4 ml of reducing solution were added and bottles were placed at $39^{\circ}C$. Caecal content and faeces were diluted respectively 1:2 (CI) and 1:8 (FI) with anaerobic medium and were introduced in the respective bottles (10 ml). Gas production was recorded 20 times at 2-24 h intervals throughout fermentation (96 h). The fermentation characteristics (i.e. degraded organic matter, OMd; potential gas production, A; fermentation rate, Rmax; time at which it is reached, Tmax; pH, volatile fatty acid, VFA) were studied by inoculum and feedstuffs. The feedstuffs, according to their chemical composition, showed very different fermentation characteristics. In particular, OMd, A and Rmax allowed feedstuff classification as follows: barley>beet pulp>alfalfa. The inocula differ (p<0.05) in Tmax, were higher for CI (15.53 vs. 11.96 h) and in VFA production. In particular, CI produced higher levels of acetate (38.9 vs. 33.4 mM/g OM incubated, p<0.01) and isobutyrate (0.72 vs. 0.42, p<0.01) but less propionate (7.1 vs. 10.3, p<0.01) and butyrate (11.3 vs. 14.0, p<0.01). However, the trend of gas production, similar for the inocula according to the fermented substrate, and the good regression equation to estimate some caecal fermentation parameters from faeces suggest that, after standardisation, the faeces could be used as an alternative inoculum for gas tests in rabbit.

Effects of Medicinal Herb Extracts on In vitro Ruminal Methanogenesis, Microbe Diversity and Fermentation System

  • This study was aimed to evaluate the in vitro effects of medicinal herb extracts (MHEs) on ruminal fermentation characteristics and the inhibition of protozoa to reduce methane production in the rumen. A fistulated Hanwoo was used as a donor of rumen fluid. The MHEs (T1, Veratrum patulum; T2, Iris ensata var. spontanea; T3, Arisaema ringens; T4, Carduus crispus; T5, Pueraria thunbergiana) were added to the in vitro fermentation bottles containing the rumen fluid and medium. Total volatile fatty acid (tVFA), total gas production, gas profiles, and the ruminal microbe communities were measured. The tVFA concentration was increased or decreased as compared to the control, and there was a significant (p<0.05) difference after 24 h incubation. pH and ruminal disappearance of dry matter did not show significant difference. As the in vitro ruminal fermentation progressed, total gas production in added MHEs was increased, while the methane production was decreased compared to the control. In particular, Arisaema ringens extract led to decrease methane production by more than 43%. In addition, the result of real-time polymerase chain reaction indicted that the protozoa population in all added MHEs decreased more than that of the control. In conclusion, the results of this study indicated that MHEs could have properties that decrease ruminal methanogenesis by inhibiting protozoa species and might be promising feed additives for ruminants.

Effects of Essential Oils Supplementation on Growth Performance, IgG Concentration and Fecal Noxious Gas Concentration of Weaned Pigs

  • Ninety six crossbred pigs (Landrace${\times}$Yorkshire${\times}$Duroc) were used to determine the effects of essential oils (Fresta F $Conc^{(R)}$) supplementation on growth performance, immune response and fecal noxious gas of weaned pigs. Treatments were 1) NC (negative control; basal diet without antibiotics), 2) PC [positive control; basal diet+CSP (CTC+Sulfathiazole+Penicillin) 0.1%], 3) NCF (basal diet+Fresta F $Conc^{(R)}$ 0.03%) and 4) PCF [basal diet+CSP (CTC+Sulfathiazole+Penicillin) 0.1%+Fresta F $Conc^{(R)}$ 0.02%]. From d 0 to 14, ADFI was increased in pigs fed PCF diet (p<0.05). From d 14 to 28, pigs fed PCF diet had greater ADG and ADFI than pigs fed NC diet (p<0.05). From d 28 to 49, ADG and ADFI in pigs fed PCF diet were higher than in pigs fed NC diet (p<0.05). Through the entire experimental period, ADG and ADFI in pigs fed PCF diet were the highest compared to pigs fed NC and PC diets (p<0.05). There was no significant difference in fecal consistency score among the treatments (p>0.05). No statistical differences (p>0.05) were found in red blood cells (RBC) counts, white blood cells (WBC) counts, lymphocyte counts, total protein and albumin. Serum IgG concentration of PCF treatment was greater than that of other treatments (p<0.05). From d 0 to 14, there was no significant difference in digestibility of dry matter and nitrogen among the treatments (p>0.05). From d 14 to 28, digestibility of dry matter in pigs fed PC, NCF and PCF diets was higher than that of pigs fed NC diet (p<0.05) and treatments with added essential oils were higher than other diets on digestibility of nitrogen (p<0.05). Also, from d 28 to 49, digestibility of nitrogen in pigs fed PCF diet was the highest among others (p<0.05). On d 14 and 28, no statistical differences (p>0.05) were found in volatile fatty acid (VFA), ammonia nitrogen ($NH_3$-N) and hydrogen sulfide ($H_2S$) concentrations among treatments. On d 49, there was no significant difference in VFA concentration among the treatments (p>0.05). $NH_3$-N concentration in pigs fed PCF diet was lower than in pigs fed other diets (p<0.05). $H_2S$ concentration in pigs fed diets with added essential oils was lower than others. In conclusion, the results suggest that the dietary addition of essential oils and antibiotics into diets for weanling pigs improved growth performance, IgG concentration and nitrogen digestibility and decreased noxious gas concentration. Essential oils can be used to partly replace antibiotics in diets for weaned pigs without negative affects on growth performance.

Replacing Concentrate with Wheat Straw Treated with Urea Molasses and Ensiled with Manure: Effects on Ruminal Characteristics, In situ Digestion Kinetics and Nitrogen Metabolism of Nili-Ravi Buffalo Bulls

  • To evaluate the effects of replacing concentrate with urea molasses treated fermented wheat straw (FWS) ensiled with cattle manure (CM) on ruminal characteristics, in situ digestion kinetics and nitrogen (N) metabolism was studied in Nili Ravi cannulated buffalo bulls in a $4{\times}4$ Latin Square Design. Wheat straw treated with urea (4%) and molasses (6%) was ensiled with cattle manure (CM) (70:30) and fermented for 40 days. Four iso-nitrogenous and iso-caloric diets were formulated. In the FWS0, FWS10, FWS20 and FWS30 diets 0, 10, 20 and 30% of the concentrate was replaced with FWS, respectively. Daily intake by bulls was restricted to 1.5% dry matter (DM) of body weight. Ruminal ammonia nitrogen concentration was greater (p<0.05) in bulls fed FWS diet than for those fed FWS0 diet at 3, 6, 9 and 12 h post-parandial. Bulls fed FWS 20 and FWS 30 diets had higher ruminal pH at 3 and 6 h post-parandial than bulls fed FWS10 and FWS0. Ruminal total volatile fatty acid (VFA) concentrations 3 h post-parandial were greater (p<0.05) in bulls fed FWS0 than those fed FWS diets. However ruminal VFA tended to increase at 6, 9 and 12 h post-parandial as the level of FWS increased. In situ ruminal DM and neutral detergent fiber (NDF) degradation, rates of disappearance and extent of digestion were higher (p<0.05) for bulls fed FWS30 diet than those fed FWS0. Ruminal DM and NDF lag time tended to decrease (p<0.05) as FWS concentration in the diet increased. Feed intake, nitrogen intake, N-balance and blood urea-N did not differ (p>0.05) in buffalo bulls fed different diets. Wheat straw treated with urea and molasses and ensiled with CM enhanced the nutritive value of wheat straw and improved nutrient utilization in buffalo bulls when up to 30% of the concentrate was replaced with FWS; no adverse effects on ruminal characteristics and nutrients digestibilities were detected.

Effects of Gelidium amansii extracts on in vitro ruminal fermentation characteristics, methanogenesis, and microbial populations

  • Objective: Gelidium amansii (Lamouroux) is a red alga belonging to the family Gelidaceae and is commonly found in the shallow coasts of many East Asian countries, including Korea, China, and Japan. G. amansii has traditionally been utilized as an edible alga, and has various biological activities. The objective of this study was to determine whether dietary supplementation of G. amansii could be useful for improving ruminal fermentation. Methods: As assessed by in vitro fermentation parameters such as pH, total gas, volatile fatty acid (VFA) production, gas profile (methane, carbon dioxide, hydrogen, and ammonia), and microbial growth rate was compared to a basal diet with timothy hay. Cannulated Holstein cows were used as rumen fluid donors and 15 mL rumen fluid: buffer (1:2) was incubated for up to 72 h with four treatments with three replicates. The treatments were: control (timothy only), basal diet with 1% G. amansii extract, basal diet with 3% G. amansii extract, and basal diet with 5% G. amansii extract. Results: Overall, the results of our study indicate that G. amansii supplementation is potentially useful for improving ruminant growth performance, via increased total gas and VFA production, but does come with some undesirable effects, such as increasing pH, ammonia concentration, and methane production. In particular, real-time polymerase chain reaction indicated that the methanogenic archaea and Fibrobacter succinogenes populations were significantly reduced, while the Ruminococcus flavefaciens populations were significantly increased at 24 h, when supplemented with G. amansii extracts as compared with controls. Conclusion: More research is required to elucidate what G. amansii supplementation can do to improve growth performance, and its effect on methane production in ruminants.

천연 약용식물의 미생물 발효를 통한 장내 메탄 생성 억제 효과 비교 연구 (A Comparative Study between Microbial Fermentation and Non-Fermentation on Biological Activities of Medicinal Plants, with Emphasis on Enteric Methane Reduction)

  • 본 연구는 반추위 메탄저감용 약용식물 첨가제 개발을 위해 유산균을 이용하여 발효한 두충 및 감초 추출물의 항균활성, 항산화활성 및 in vitro 반추위 발효시험을 실시하였다. 발효 약용식물 추출물 제조를 위해 접종된 종균의 성장효율을 조사하기 위해 실시한 생균수 측정 결과 L. curvatus NJ40, L. brevis NJ42 및 L. plantarum NJ45가 두충과 감초 모두에서 유의적으로 높은 균주 성장을 나타냈다(p<0.05). 항균활성측정 결과는 두충과 감초 추출물에서 공통적으로 L. curvatus NJ40 및 L. plantarum NJ45 로 발효한 추출물이 일부 병원균에 대한 항균효과를 나타내는 것으로 조사되었다. In vitro 반추위 발효시험에서 두충 및 감초발효 추출물을 적용한 결과, 감초 추출물에서 메탄 저감효과가 나타났다. 특히 반추위내 미생물 발효 특성을 대표할 수 있는 휘발성지방산 생성효율을 향상 시키면서, 전반적 반추위 발효에 부정적 영향이 없는 것으로 나타났다.

Effects of Addition Level and Chemical Type of Propionate Precursors in Dicarboxylic Acid Pathway on Fermentation Characteristics and Methane Production by Rumen Microbes In vitro

  • Two in vitro experiments were conducted to examine the effects of propionate precursors in the dicarboxylic acid pathway on ruminal fermentatation characteristics, $CH_4$ production and degradation of feed by rumen microbes. Fumarate or malate as sodium salts (Exp. 1) or acid type (Exp. 2) were added to the culture solution (150 ml, 50% strained rumen fluid and 50% artificial saliva) to achieve final concentrations of 0, 8, 16 and 24 mM, and incubated anaerobically for 0, 1, 3, 6, 9 and 12 h at $39^{\circ}C$. For both experiments, two grams of feed consisting of 70% concentrate and 30% ground alfalfa (DM basis) were prepared in a nylon bag, and were placed in a bottle containing the culture solution. Addition of fumarate or malate in both sodium salt and acid form increased (p<0.0001) pH of culture solution at 3, 6, 9 and 12 h incubations. The pH (p<0.0001) and total volatile fatty acids (VFA, p<0.05) were enhanced by these precursors as sodium salt at 3, 6 and 9 h incubations, and pH (p<0.001) and total VFA (p<0.01) from fumarate or malate in acid form were enhanced at a late stage of fermentation (9 h and 12 h) as the addition level increased. pH was higher (p<0.001) for fumarate than for malate as sodium salt at 3 h and 6 h incubations. Propionate ($C_3$) proportion was increased (p<0.0001) but those of $C_2$ (p<0.05) and $C_4$ (p<0.01 - p<0.001) were reduced by the addition of sodium salt precursors from 3 h to 12 incubation times while both precursors in acid form enhanced (p<0.011 - p<0.0001) proportion of $C_3$ from 6h but reduced (p<0.018 - p<0.0005) $C_4$ proportion at incubation times of 1, 3, 9 and 12 h. Proportion of $C_3$ was increased (p<0.05 - p<0.0001) at all incubation times by both precursors as sodium salt while that of $C_3$ was increased (p<0.001) from 6h but $C_4$ proportion was decreased by both precursors in acid form as the addition level increased. Proportion of $C_3$ was higher (p<0.01 - p<0.001) for fumarate than malate as sodium salt from 6 h incubation but was higher for malate than fumarate in acid form at 9 h (p<0.05) and 12 h (p<0.01) incubation times. Increased levels (16 and 24 mM) of fumarate or malate as sodium salt (p<0.017) and both precursors in acid form (p<0.028) increased the total gas production, but no differences were found between precursors in both chemical types. Propionate precursors in both chemical types clearly reduced (p<0.0001 - p<0.0002) $CH_4$ production, and the reduction (p<0.001 - p<0.0001) was dose dependent as the addition level of precursors increased. The $CH_4$ generated was smaller (p<0.01 - p<0.0001) for fumarate than for malate in both chemical types. Addition of fumarate or malate as sodium type reduced (p<0.004) dry matter degradation while both precursors in both chemical types slightly increased neutral detergent fiber degradability of feed in the nylon bag.
