Rumen 미생물을 이용한 주방폐기물 혐기성소화의 효율증진 방안 (Enhanced Anaerobic Degradation of Food Waste by Employing Rumen Microorganisms)

  • 우리나라에서 년간 3,367만톤(1991년) 배출되는 생활쓰레기의 28% 이상이 주방폐기물로 통칭되는 음식찌꺼기, 채소류등으로 구성되는데 수분함량이 75~85%로 높고 발열량 낮아 혐기성 소화에 의한 처리법은 폐기물의 안정화 및 메탄가스 회수를 통한 효율적인 자원화가 가능하여 매우 실용적인 것으로 고려된다. 그러나, 주방폐기물은 셀룰로즈, 단백질등의 복잡한 고분자 고형물질들로 구성되어 있어 혐기성 소화시 가수분해반응이 율속단계로 인식되고 있으며, 이로 인한 유기물 부하율이 작고 반응조 용적이 커져 경제적이지 못한 것으로 알려져 있다. 따라서, 본 연구에서는 셀룰로즈 등의 고분자물질의 가수분해 반응에 활성이 뛰어난 rumen 미생물을 이용하여 주방폐기물의 혐기성 소화의 효율을 증진시키기 위한 연구가 600 mL의 회분식반응조에서 수행되었다. 주방폐기물의 가수분해 및 산생성 반응에 대한 중온산생성균과 rumen 미생물의 산생성반응 효율을 비교하였으며, rumen 미생물을 이용한 산형성 반응에서 pH, 온도, 고형물 농도등 환경인자의 영향을 평가하였다. Rumen 미생물을 이용한 주방폐기물의 산형성 반응에서 최종휘발산수율은 이론치의 약 95%에 해당하는 8.4meq/g VS를 보여 중온산형성균을 이용하였을 때와 비슷하였으나, 산생성반응의 속도는 중온산형성균을 이용했을 때 보다 약 2배 이상 빨랐다. Rumen 미생물을 이용한 주방폐기물의 산생성 반응에 대한 기질 농도의 영향에서는 TS 15%일 때 급격한 pH저하 및 비이온화된 VFA 등의 저해작용으로 휘발산수율은 TS 1%인 반응조에서의 절반이하의 값을 보여 유출수의 연속적인 희석이 중요한 인자임을 알 수 있었다. Rumen 미생물을 이용한 산생성 반응의 최적 pH 및 온도 범위는 각각 6.0~7.5, $35{\sim}45^{\circ}C$였다. 이상과 같은 결과로부터 rumen 미생물을 주방폐기물의 혐기성소화 공정에 이용할 경우 혐기성 반응의 율속단계로 고려되는 가수분해 및 산형성 반응의 속도를 증가 시킬 수 있어 반응조 용적 감소 및 유기물 부하 증가 등의 소화 효율향상이 가능함을 알 수 있었다.

반추가축전용 기후변화대응 비식량자원 거대억새의 생육부위 및 시기에 따른 체내 이용가치 비교 연구 (Effect of Different Parts and Growing Stages of Miscanthus sacchariflorus as a non-Food Resource that does not Contribute towards Climate Change on Metabolic Availability in Ruminants)

  • 거대억새 1호는 한국에서 새로이 개발된 품종이다. 본 연구의 목적은 거대억새 1호의 사료적 가치를 평가하기 위해 in vitro 반추위 발효 시험 및 in situ 건물소화율을 조사하였다. in vitro 발효에 따른 pH는 전반적으로 거대억새 1호 시험구들이 RS에 비해 높은 수치를 보였다. 1y4m에서 다른 시험구에 비해 가장 높은 acetate 생성량을 나타내었다. propionate 및 전체 휘발성 지방산 생성량에서 시험구들간 유의성이 배양 9, 24 및 48시간에서 나타났다. in situ 건물소실율 시험 결과 RS(60.51%) > 1y4m(57.65%) > 2y4m(57.63%) > 2y8m(46.28%)순으로 나타났다. 본 연구의 결과는 이른 예취시기가 거대억새 1호의 영양적 가치를 향상시키는 것을 반영한다. 그러나 거대억새 1호가 반추가축을 위한 양질의 조사료로 이용되기 위해서는 실제 사양시험이 우선되어야 한다.

Enhancing Butyrate Production, Ruminal Fermentation and Microbial Population through Supplementation with Clostridium saccharobutylicum

  • Butyrate is known to play a significant role in energy metabolism and regulating genomic activities that influence rumen nutrition utilization and function. Thus, this study investigated the effects of an isolated butyrate-producing bacteria, Clostridium saccharobutylicum, in rumen butyrate production, fermentation parameters and microbial population in Holstein-Friesian cow. An isolated butyrate-producing bacterium from the ruminal fluid of a Holstein-Friesian cow was identified and characterized as Clostridium saccharobutylicum RNAL841125 using 16S rRNA gene sequencing and phylogenetic analyses. The bacterium was evaluated on its effects as supplement on in vitro rumen fermentation and microbial population. Supplementation with $10^6CFU/ml$ Clostridium saccharobutylicum increased (p < 0.05) microbial crude protein, butyrate and total volatile fatty acids concentration but had no significant effect on $NH_3-N$ at 24 h incubation. Butyrate and total VFA concentrations were higher (p < 0.05) in supplementation with $10^6CFU/ml$ Clostridium saccharobutylicum compared with control, with no differences observed for total gas production, $NH_3-N$ and propionate concentration. However, as the inclusion rate (CFU/ml) of C. saccharobutylicum was increased, reduction of rumen fermentation values was observed. Furthermore, butyrate-producing bacteria and Fibrobacter succinogenes population in the rumen increased in response with supplementation of C. saccharobutylicum, while no differences in the population in total bacteria, protozoa and fungi were observed among treatments. Overall, our study suggests that supplementation with $10^6CFU/ml$ C. saccharobutylicum has the potential to improve ruminal fermentation through increased concentrations of butyrate and total volatile fatty acid, and enhanced population of butyrate-producing bacteria and cellulolytic bacteria F. succinogenes.

호맥 사일리지의 급여가 비육돈의 생산성, 혈액성상 및 도체특성에 미치는 효과 (Effects of Feeding Rye Silage on Growth Performance, Blood, and Carcass Characteristics in Finishing Pigs)

  • 본 연구는 조사료인 호맥 사일리지를 배합사료와 혼합급여 시 비육돈의 성장, 혈액성상 및 도체특성에 미치는 영향을 알아보기 위해 시험을 실시하였다. 3원 교잡종($Landrace{\times}Yorkshire{\times}Duroc$) 비육돈 18두를 공시하였으며 시험개시시 평균체중은 $74.22{\pm}0.71kg$이었고 28일간 개체별 사양시험을 실시하였다. 시험설계는 호맥 사일리지 첨가에 따라 1) Con(basal diet), 2) S1(basal diet+1.66% rye silage), 3) S2(basal diet+3.32% rye silage)로 3처리로 하여 처리당 6반복, 반복당 1마리씩 $1.8{\times}1.8m$ 크기의 돈방에 개별 수용하였다. 생산성에서 $4{\sim}7$주 동안 일당사료 섭취량에서는 CON구가 S1구와 S2구와 비교하여 유의적으로 높게 나타났다(p<0.05). 전체 사양시험기간 동안($0{\sim}7$주) 일당사료섭취량에서는 CON구와 S2구가 S1구와 비교하여 유의적으로 높게 나타났다(p<0.05). 혈중 스트레스 관련 호르몬인 코티졸 함량의 변화는 대조구와 비교하여 호맥 사일리지를 급여한 S1구와 S2구에서 유의적으로 감소하였다(p<0.05). 육색에서 명도를 나타내는 $L^*$값은 S2구가 CON구와 비교하여 유의적으로 높게 나타났고(p<0.05) 황색도를 나타내는 $b^*$값은 S1구와 S2구가 CON구와 비교하여 높게 나타났다(p<0.05). 도체 등지방 두께는 CON구가 S1구와 S2구와 비교하여 유의적으로 높게 증가하였다(p<0.05). 살코기 내 지방산 조성에서는 포화지방산 중 palmitic acid와 stearic acid의 함량은 CON구가 S1구와 S2구와 비교하여 유의적으로 높은 함량을 나타내었다(p<0.05). 불포화 지방산에서는 eicosenoic acid와 linolenic acid의 함량이 S2구가 CON구와 S1구와 비교하여 유의적으로 높았다(p<0.05). 총 포화지방산 함량은 CON구가 다른 처리구와 비교하여 가장 높았으며(p<0.05) 포화지방산과 불포화지방산 비율에서는 S1구와 S2구가 CON구와 비교하여 유의적으로 높은 결과를 보였다(p<0.05). 지방 내 지방산 조성에서는 불포화지방산 중 linolenic acid의 함량은 S2구가 S1구와 CON구와 비교하여 유의적으로 높았다(p<0.05). 건물 소화율에서는 S1구와 S2구가 CON구와 비교하여 유의적으로 높게 나타났고(p<0.05) 질소 소화율에서는 S1구가 처리구중에서 가장 높은 결과를 보였다(p<0.05). 분변 내 휘발성 지방산 발생량에서는 분을 1일간 발효 후 측정한 결과 S1구와 S2구가 CON구와 비교하여 유의적으로 높게 나타났다(p<0.05). 결론적으로, 비육돈 사료 내 3.32%의 호맥 사일리지의 혼합급여는 혈액 내 코티졸 함량, 도체육의 명도와 황색도, 지방산 조성 및 영양소 소화율에 영향을 미치는 것으로 사료되나 이에 대한 보다 많은 연구가 필요할 것으로 판단된다.

Evaluation of the nutritional value of locally produced forage in Korea using chemical analysis and in vitro ruminal fermentation

  • Objective: The use of locally produced forage (LPF) in cattle production has economic and environmental advantages over imported forage. The objective of this study was to characterize the nutritional value of LPF commonly used in Korea. Differences in ruminal fermentation characteristics were also examined for the LPF species commonly produced from two major production regions: Chungcheong and Jeolla. Methods: Ten LPF (five from each of the two regions) and six of the most widely used imported forages originating from North America were obtained at least three times throughout a year. Each forage species was pooled and analyzed for nutrient content using detailed chemical analysis. Ruminal fermentation characteristics were also determined by in vitro anaerobic incubations using strained rumen fluid for 0, 3, 6, 12, 24, and 48 h. At each incubation time, total gas, pH, ammonia, volatile fatty acid (VFA) concentrations, and neutral detergent fiber digestibility were measured. By fitting an exponential model, gas production kinetics were obtained. Results: Significant differences were found in the non-fiber carbohydrate (NFC) content among the forage species and the regions (p<0.01). No nutrient, other than NFC, showed significant differences among the regions. Crude protein, NFC, and acid detergent lignin significantly differed by forage species. The amount of acid detergent insoluble protein tended to differ among the forages. The forages produced in Chungcheong had a higher amount of NFC than that in Jeolla (p<0.05). There were differences in ruminal fermentation of LPF between the two regions and interactions between regions and forage species were also significant (p<0.05). The pH following a 48-h ruminal fermentation was lower in the forages from Chungcheong than from Jeolla (p<0.01), and total VFA concentration was higher in Chungcheong than in Jeolla (p = 0.05). This implies that fermentation was more active with the forages from Chungcheong than from Jeolla. Analysis of gas production profiles showed the rate of fermentation differed among forage species (p<0.05). Conclusion: The results of the present study showed that the nutritional values of some LPF (i.e., corn silage and Italian ryegrass) are comparable to those of imported forages widely used in Korea. This study also indicated that the nutritional value of LPF differs by origin, as well as by forage species. Detailed analyses of nutrient composition and digestion kinetics of LPF should be routinely employed to evaluate the correct nutritional value of LPF and to increase their use in the field.

The Influence of Feed Energy Density and a Formulated Additive on Rumen and Rectal Temperature in Hanwoo Steers

  • The present study investigated the optimum blending condition of protected fat, choline and yeast culture for lowering of rumen temperature. The Box Benken experimental design, a fractional factorial arrangement, and response surface methodology were employed. The optimum blending condition was determined using the rumen simulated in vitro fermentation. An additive formulated on the optimum condition contained 50% of protected fat, 25% of yeast culture, 5% of choline, 7% of organic zinc, 6.5% of cinnamon, and 6.5% of stevioside. The feed additive was supplemented at a rate of 0.1% of diet (orchard grass:concentrate, 3:7) and compared with a control which had no additive. The treatment resulted in lower volatile fatty acid (VFA) concentration and biogas than the control. To investigate the effect of the optimized additive and feed energy levels on rumen and rectal temperatures, four rumen cannulated Hanwoo (Korean native beef breed) steers were in a $4{\times}4$ Latin square design. Energy levels were varied to low and high by altering the ratio of forage to concentrate in diet: low energy (6:4) and high energy (4:6). The additive was added at a rate of 0.1% of the diet. The following parameters were measured; feed intake, rumen and rectal temperatures, ruminal pH and VFA concentration. This study was conducted in an environmentally controlled house with temperature set at $30^{\circ}C$ and relative humidity levels of 70%. Steers were housed individually in raised crates to facilitate collection of urine and feces. The adaptation period was for 14 days, 2 days for sampling and 7 days for resting the animals. The additive significantly reduced both rumen (p<0.01) and rectal temperatures (p<0.001) without depressed feed intake. There were interactions (p<0.01) between energy level and additive on ruminal temperature. Neither additive nor energy level had an effect on total VFA concentration. The additive however, significantly increased (p<0.01) propionate and subsequently had lower acetate:propionate (A/P) ratios than non-additive supplementation. High concentrate diets had significantly lower pH. Interactions between energy and additive were observed (p<0.01) in ammonia nitrogen production. Supplementation of diets with the additive resulted in lower rumen and rectal temperatures, hence the additive showed promise in alleviating undesirable effects of heat stress in cattle.

Effects of Non-ionic Surfactant Supplementation on Ruminal Fermentation, Nutrient Digestibility and Performance of Beef Steers Fed High-roughage Diets

  • Three experiments were conducted to determine the effects of non-ionic surfactant (NIS) supplementation on ruminal fermentation, nutrient digestibility and performance of beef steers fed high-roughage diets. The objective of experiment 1 was to investigate the effects of NIS supplementation on in vitro ruminal fermentation of cultures administered with corn and barley as grain substrate and rice straw and timothy hay as roughage substrate. The in vivo ruminal fermentation, nitrogen balance and digestibility of nutrients were also examined with steers fed a high-roughage diet in experiment 2. The aim of experiment 3 was to determine the responses to NIS of growing steers fed a high-roughage diet. In experiment 1, ammonia nitrogen concentration for NIS supplementation was higher (p<0.05) than for the control with all substrates. However, concentrations of total volatile fatty acid (VFA), acetate, butyrate and valerate of the incubated roughage substrates, rice straw and timothy hay, were higher (p<0.05) for NIS supplementation than for the control whereas VFA concentrations in the cultures of corn and barley were unaffected. These results indicated that effects of NIS on ruminal fermentation are diet dependent, specifically on roughage sources. In experiment 2, ruminal pH of steers supplemented with NIS was lower (p<0.05) than the control. Ruminal concentrations of ammonia nitrogen, acetate, total VFA and urinary concentrations of purine derivatives were increased (p<0.05) by NIS supplementation. In experiment 3, supplementation of NIS increased (p<0.05) intakes of total feed and corn silage, average daily gain, and feed efficiency of growing steers although they varied depending on supplementation level. Due to the roughage-specific feature of NIS effects, NIS appears to enhance ruminal fermentation of fibrous parts of feeds and, consequently, performance of steers fed a high-roughage diet.

생육 후기 거대억새의 In vitro 반추위 발효특성 및 건물 소화율 (Effect of Mature Miscanthus sacchariflorus var. No. 1 on In Vitro Rumen Fermentation Characteristics and Its Dry Matter Digestibility)

  • 본 연구는 반추동물의 조사료 자원으로서 거대억새를 개발하기 위한 목적으로 수행되었다. 우리나라에서 새롭게 개발된 품종인 거대억새 1호를 완숙기 이후에 채취하여 in vitro 반추위 발효를 이용해 반추위내 pH, 암모니아태 질소, 가스발생량, 휘발성 지방산 생성량 및 건물소화율을 조사하였으며, 볏짚과 비교하여 평가하였다. 거대억새는 볏짚에 비하여 유의적으로 높은 반추위내 pH를 나타내었다(p<0.01). 암모니아태 질소의 경우 배양 12시간 이후에는 두 처리구간의 유의적인 차이를 나타내지 않았다 (p>0.05). 배양 6시간 이후 부터는 거대억새의 가스발생량이 볏짚에 비하여 유의적으로 낮게 나타났다 (p<0.05). 휘발성 지방산 생성량에 있어 acetate, propionate, butyrate, valerate 및 총생상량에서 볏짚이 거대억새보다 높게 나타났다. 그러나 iso-butyrate와 iso-valerate에서는 두 조사료원별 차이는 발견되지 않았다. 건물소화율에 있어 배양 12~24시간 사이의 거대억새 소화율이 볏짚에 비하여 유의적으로 나타났다. 결론적으로 거대억새의 이용성은 볏짚의 약 80% 수준인 것으로 나타났다.

Consumption and Digestion of Forages by Male Rusa (Cervus timorensis russa) Deer-the Effects of Castration and Season

  • In a split-unit design, 12 Javan rusa (Cervus timorensis russa) stags (6 castrated and 6 entire) were used to investigate seasonal (winter, spring and summer) effects on intake, digestibility of food constituents, volatile fatty acid profile, and nitrogen retention, when given lucerne (Medicago sativa) or rhodes grass (Chloris guyana) hays. Entire stags ate 9% more dry matter (DM) than castrates (p<0.05). Both castrated and entire stags exhibited seasonal changes in DM intake, these being higher (p<0.05) in winter (62.3 g/kg $W^{0.75}$) than spring (56.9 g/kg $W^{0.75}$) or summer (55.3 g/kg $W^{0.75}$). Intakes of lucerne hay (211 g protein/kg DM) were significantly higher than of rhodes grass hay (49 g protein/kg DM) in all seasons (p<0.05). Digestibilities of DM (0.58), neutral detergent fibre (0.59) and acid detergent fibre (0.47) were similar between castrates and entires. DM digestibility was higher (p<0.0001) for lucerne (0.66) than rhodes grass (0.55), and was higher (p<0.05) in winter (0.60) than spring (0.58) or summer (0.57). The ruminal acetate concentrations were higher (p<0.001) in spring than summer (78 and 73 molar % respectively). Apparent digestibility of protein was significantly less (p<0.0001) for rhodes grass (0.37) than lucerne (0.75). N retention was positive for lucerne (15.2 g/d) but negative for rhodes grass (-2.8 g/d) (p<0.0001), and was higher (p<0.001) in summer (12.0 g/d) than spring (4.3 g/d) or winter (2.4 g/d). The tropical rusa deer exhibits seasonal variations in feed intake, food constituent digestibilities, VFA profile and N retention. Castration did not alter these traits. The results do not support the view that rusa deer can thrive on low-quality pastures. The productivity and commercial exploitation of rusa deer could be optimised if they are given high-protein feed during spring and summer.

Effect of Monensin and Live Yeast Supplementation on Growth Performance, Nutrient Digestibility, Carcass Characteristics and Ruminal Fermentation Parameters in Lambs Fed Steam-flaked Corn-based Diets

  • In performance, digestibility and slaughter trials, a total of forty five male weaned lambs were used to examine the effects of monensin and live yeast supplementations on growth performance, nutrient digestibility, carcass characteristics and ruminal fermentation parameters when the lambs were fed steam-flaked corn-based diets. Animals were allotted to one of three treatment diets in a completely randomized design. The three treatment diets were: (1) basal diet (CON) with steam-flaked corn as a sole grain source, (2) basal diet supplemented with monensin (MO), and (3) basal diet supplemented with live yeast (LY). Total average daily intake (ADI) was unaffected by MO and LY supplementations. LY supplementation increased (p<0.05) average daily gain (ADG) by 13.1% compared with the CON diet. Both MO and LY supplementations resulted in a significant improvement (p<0.05) of feed efficiency over the CON diet (4.47, 4.68 vs. 5.05). Hemicellulose digestibility was higher (p<0.05) for lambs in the LY supplementation group (62.4%) as compared with the CON group (55.7%), but no differences were observed in digestibilities of dry matter (DM), organic matter (OM), crude protein (CP), neutral detergent fiber (NDF) and acid detergent fiber (ADF). All carcass traits were not influenced by dietary supplementations. Ruminal pH in lambs fed the LY supplemental diet was more stable than that with the CON diet (6.57 vs. 6.17). Neither MO nor LY supplementation influenced the concentration of ruminal ammonia-N and total volatile fatty acid (VFA), and molar percentages of individual VFA. Plasma urea-N concentration was decreased (p<0.05) by MO and LY supplementations, while plasma ${\beta}$-hydroxybutyrate (BHBA), glucose and other blood parameters were unaffected. In conclusion, while both MO and LY supplementations had a positive impact on feed efficiency and LY supplementation stabilized ruminal pH and improved fiber utilization, none of the supplements had the capacity to significantly enhance the carcass characteristics.
