초임계유체 추출을 이용한 포도씨 tocotrienol 추출조건 최적화 (Optimization of Supercritical Fluid Extraction of Tocotrienol from Grape Seed)

  • 포도가공 시 발생되는 부산물인 포도씨로부터 유용성분인 vitamin E 및 tocotrienol을 초임계 유체추출방법으로 최적화 하였다. 추출 온도(30-$50^{\circ}C$, $X_1$), 추출압력(17-25MPa, $X_2$) 및 추출시간(1-5 hr, $X_3$)을 변수로 반응표면 분석법으로 vitamin E 및 tocotrienol 함량을 측정하였다. 초임계 이산화탄소상에서 vitamin E 및 tocotrienol 함량은 추출온도, 압력 및 시간이 증가함에 따라 증가하였다 (plt;0.001). 각각의 추출조건 가운데 $40^{\circ}C$, 20 MPa 및 5 hr에서 vitamin E는 8.65 mg/100 g 그리고 tocotrienol은 7.88 mg/100 g으로 가장 높은 함량을 나타내었다. 반응표면분석에 의한 최적추출조건은 $40^{\circ}C$, 20.7MPa 및 3.8 hr으로 나타났으며, 이 때의 vitamin E 및 tocotrienol 함량은 각각 8.20 mg/100 g 및 7.42 mg/100 g으로 나타났다. Vitamin E 및 tocotrienol 함량에 대한 실측값은 통계적 예측값의 각각 99.27-106.10% 및 96.09-106.60%로 나타나 추출조건의 유효성이 인정되었다. 또한 유기용매 추출시 vitamin E 및 tocotrienol 함량은 각각 8.18 mg/100 g 및 7.24 mg/100 g이었다. 따라서 용매추출법과 초임계 이산화탄소 추출법을 비교했을 때 초임계 이산화탄소 추출법을 이용하는 것이 더 효과적으로 vitamin E 및 tocotrienol을 추출할 수 있을 것으로 판단되었다.

조리 방법이 상용채소의 비타민 및 무기질 함량에 미치는 영향 -시금치, 양배추, 콩나물, 당근을 중심으로 - (Effects of Cooking Method on the Vitamin and Mineral Contents in Frequently Used Vegetables)

  • Purpose: We have investigated for the purpose of studying change of vitamins and minerals in frequently used vegetables by low moisture cooking method. Methods: Vitamin B complex, vitamin C and mineral (Mg, Ca, K, Zn, Fe) contents are analyzed in vegetables such as spinach, cabbage, bean sprouts and carrot which are heat-treated with degrees of water contents. Low moisture cooking method represents heat-treated vegetable (LM experiment group) with 25 wt.% water content for the vegetable weight while general blanching method includes heat-treated vegetable (GB experiment group) with 500-1,000 wt.% water content for the vegetable weight. Results: Retention rate of vitamin B1 (thiamin) in the LM experiment group is relatively high (87.50-95.68%) and dosen't show considerable differences from raw vegetables. On the other hand, the retention rate of vitamin $B_1$ in GB group's cabbage and spinach dramatically decreased to 19.46-25.00%. Retention rate of vitamin B2 (riboflavin) is sustained stably in LM experiment group (75.00-87.50% in bean sprouts, cabbage and spinach), represents that low moisture cooking method has relatively higher contents in vitamin $B_2$. The LM experiment group has 71.43-85.71% on retention rate of niacin while the GB group shows relatively lower niacin (57.14-64.58%), represents conflicting results from the general idea that vitamin $B_3$ is relatively stable in heat treatment and blanching. Retention rate of pantothenic acid is considerably higher in both LM and GB experiment group, indicates vitamin pantothenic acid is relatively stable in the various cooking condition such as amount of water and heat treatment. In the experiment under various water contents, vitamin C is not detected in both bean sprouts and carrots, while the contents in cabbage and spinach are 19.87 mg/100 g and 26.65 mg/100 g respectively. In the same experiment, the retention rate of Vitamin C in LM experiment group (91.65%, 92.23%) is considerably higher than GB group (58.08%, 61.61%). Retention rate of Mg, K, Fe in the LM experiment group is relatively higher than GB group. Conclusion: Resultingly, the observations suggests that minimum water quantity and minimum heat treatment processes should be established in cooking vegetables which have soluble vitamins and minerals.

  • Background: Vitamin C is an essential nutrient, taken as a daily supplement by many people. Recently, high-dose vitamin C is considered as a therapeutic regimen in some clinical situations. Until now, few studies have been done with the effects of high-dose vitamin C on the immune response. Methods: In this experiment, the effects of high-dose vitamin C on cell-mediated immune response in immunologically competent mice were evaluated. After intraperitoneal injection of 2.5, 5, or 10 mg/day of vitamin C for 10 days, delayed type hypersensitivity (DTH) was provoked against DNFB in the pinnae as a model for cell-mediated immune response. Severity of DTH reaction was evaluated as the thickness of pinnae, and the vitamin C levels were measured in the serum, liver, kidney, lung, pinnae, and splenocytes. Results: After challenge, the thickness increased at its peak on the $2^{nd}$ day in all groups. On the first day, the pinnae were thicker in the injected groups than in the control. On the contrary, the increment of the pinnae thickness was attenuated and the number of cells infiltrated in the site of DTH decreased proportionately to the amount of vitamin C administered from the second day on. With vitamin C exogenously given, the serum level peaked at 30 min after injection, and returned abruptly to its basal level without accumulation. However, it accumulated in the liver, kidney, and especially in the pinnae inflamed and splenopcytes, proportionately to the amount administered. Conclusion: Based on these results, it is suggested that, in one hand, exogenously administered high-dose vitamin C accumulated in the splenocytes and presumably changed the function of them resulting in the augmented cell-mediated immune response, as was revealed in the first day of DTH reaction. On the other hand, it seems likely that the vitamin C also showed anti-inflammatory effects.

Association of Vitamin D Level with Clinicopathological Features in Breast Cancer

  • A population-based relationship between low vitamin D status and increased cancer risk is now generally accepted. However there were only few studies reported on prognostic impact. To determine the effect of low vitamin D on progression of breast cancer, we conducted a cross-sectional analysis of vitamin D levels and clinico-pathological characteristics in 200 cases of breast cancer diagnosed during 2011-2012 at the National Cancer Institute of Thailand. Vitamin D levels were measured by high-performance liquid chromatography (HPLC). Clinical and pathological data were accessed to examine prognostic effects of vitamin D. We found that the mean vitamin D level was $23.0{\pm}6.61ng/ml$. High vitamin D levels (${\geq}32ng/ml$) were detected in 7% of patients, low levels (<32 ng/ml) in 93% Mean vitamin D levels for stages 1-4 were $26.1{\pm}6.35$, $22.3{\pm}6.34$, $22.2{\pm}6.46$ and $21.3{\pm}5.42ng/ml$ respectively (P=0.016) and 24.1 and 21.3 ng/ml for lymph node negative and positive cases (P=0.006). Low vitamin D level (<32 ng/ml) was significantly found in majority of cases with advanced stage of the disease (P=0.036), positive node involvement (P=0.030) and large tumors (P=0.038). Our findings suggest that low and decreased level of vitamin D might correlate with progression and metastasis of breast cancer.

DFT/B3LYP Study to Investigate the Possible Ways for the Synthesize of Antioxidants with High Efficiency Based on Vitamin E

  • The possible ways for increasing the antioxidant properties of vitamin E have been investigated with density function theory. The effect of replacing three methyl groups of vitamin E with various substituents such as electron donating and electron withdrawing groups on the antioxidant properties of vitamin E were investigated. Also the effects of the reducing the number of atoms in the heterocyclic ring and replacing the oxygen heteroatom with other heteroatoms on the antioxidant properties of vitamin E were investigated. The novel structures that obtained from replacing methyl groups with substituents such as $NH_2$, OH, COOH and NHMe have greater antioxidant activity than vitamin E. Obtained results reveal that novel structure that obtained with replacing O with NH hetroatom would be a better antioxidant than vitamin E. The results reveal that reducing the number of atoms in the heterocyclic ring is a better way to synthesize novel antioxidants.

국내산 여주의 지역별, 수확시기별, 숙기별 Vitamin C 함량 (Vitamin C Quantification of Korean Momordica charantia by Cultivar, Harvest Time, and Maturity)

  • 본 연구에서는 vitamin C 분석법을 검증하고, 국내산 지역별 여주와 재배 시기별, 숙기별 여주의 vitamin C 함량을 분석하였다. Vitamin C 분석법을 검증하기 위해 직선성, 검출한계, 정량한계, 정밀성 및 정확성을 확인하였고, 그 결과 직선성의 상관계수 값은 0.9994, 검출한계는 $0.05{\mu}g/mL$, 정량한계는 $0.16{\mu}g/mL$, 정밀성의 상대표준편차는 intra-day test에서는 2.34 %, inter-day test에서는 1.34%이었고, 정확성인 회수율은 98.66~101.74%로 95% 이상의 높은 회수율을 보였다. 여주는 지역별로 vitamin C를 분석한 결과 20.75~107.31 mg/100 g, FW로 함량 차이가 높았으며, 평균 함량은 $68.85{\pm}25.57mg/100g$, FW로 나타났다. 평균 함량을 기준으로 구분하였을 때, 전라북도 지역에서 재배한 여주는 평균보다 낮은 함량을 보였고, 전라남도 지역에서 재배한 여주는 영암을 제외하고 모두 평균보다 높은 함량을 보였다. 수확시기별 여주의 vitamin C의 함량은 재배 초기 6월까지 vitamin C의 함량이 $113.20{\pm}1.89mg/100g$, FW로 증가하였다가 점차 감소하는 경향을 보였다. 또한 숙기별 vitamin C의 함량은 성장 초기에는 vitamin C의 함량이 0.53~0.68 mg/100 g, FW로 낮은 수준이었지만 숙기 15일차에서는 vitamin C의 함량이 $48.59{\pm}0.87mg/100g$, FW까지 증가하였다가 점차 감소하는 것을 확인하였다. 이처럼 여주의 vitamin C 함량은 지역별 생육환경에 따라, 재배 시기별, 숙기별에 따라 함량 차이가 나타났으며, 기능성 식품원료의 개발과 추후 연구의 기초자료로 활용이 가능할 것으로 기대된다.

  • 본 연구는 맥주박과 옥수수 위주의 발효사료에 대한 coated vitamin C+E 및 면실의 첨가 급여가 거세한우의 육조성분, 육색 및 도체등급에 미치는 영향을 구명하고자 평균체중 550.4±42.8kg의 거세한우 24두를 공시하여 실시하였다. 시험구 배치는 발효사료급여구(T1)와 coated vitamin C+E 및 면실 첨가 발효사료구(T2)의 2처리로 하였다. 배최장근의 조지방 함량은 T2구에서 T1구에 비해 높았다(p< 0.05). 배최장근의 표면 육색 중 황색도와 색상색은 T2구에서 T1구에 비해 낮았고(p<0.05) 명도는 T1구에 비해 T2구에서 높았다(p<0.05). 육량 A 등급의 출현은 T2구에서 T1구에 비해 높았다. 근내지방도는 T1구에 비해 T2구에서 높았다. 이상의 결과에서 맥주박과 옥수수 위주의 발효사료에 대한 CVCE 및 면실의 첨가는 거세한우 배최장근의 조지방 함량, 육색 및 근내지방도에 효과적인 것으로 판단된다.

Vitamin C Stimulates Epidermal Ceramide Production by Regulating Its Metabolic Enzymes

  • Ceramide is the most abundant lipid in the epidermis and plays a critical role in maintaining epidermal barrier function. Overall ceramide content in keratinocyte increases in parallel with differentiation, which is initiated by supplementation of calcium and/or vitamin C. However, the role of metabolic enzymes responsible for ceramide generation in response to vitamin C is still unclear. Here, we investigated whether vitamin C alters epidermal ceramide content by regulating the expression and/or activity of its metabolic enzymes. When human keratinocytes were grown in 1.2 mM calcium with vitamin C ($50{\mu}g/ml$) for 11 days, bulk ceramide content significantly increased in conjunction with terminal differentiation of keratinocytes as compared to vehicle controls (1.2 mM calcium alone). Synthesis of the ceramide fractions was enhanced by increased de novo ceramide synthesis pathway via serine palmitoyltransferase and ceramide synthase activations. Moreover, sphingosine-1-phosphate (S1P) hydrolysis pathway by action of S1P phosphatase was also stimulated by vitamin C supplementation, contributing, in part, to enhanced ceramide production. However, activity of sphingomyelinase, a hydrolase enzyme that converts sphingomyelin to ceramide, remained unaltered. Taken together, we demonstrate that vitamin C stimulates ceramide production in keratinocytes by modulating ceramide metabolicrelated enzymes, and as a result, could improve overall epidermal barrier function.

매생이소스 개발에 관한 선호도 조사와 개발된 매생이소스별 선호도 비교분석 (Preference Investigation for Maesengee Sauce Development and Comparative Analysis of Preference for Developed Maesengee Sauce)

  • 매생이 소스 개발을 위한 선호도를 조사하였고, 세 종류의 매생이소스를 개발하였으며, 개발된 매생이소스법 선호도를 비교분석함으로써 건강기능성 식품으로서의 매생이소스 개발과 향후 매생이 가공 식품을 통한 지역 경제 활성화에 목적이 있다. 매생이 성분분석은 2009년 7월에 전남 강진군에서 구입한 매생이틀 재료로 하였고, 매생이 소스 개발을 위한 선호도 조사 결과를 바탕으로 세 종류의 매생이 소스 즉 매생이 플레인 요거트 소스, 매생이 홍초 소스, 매생이 밀쌈 소스를 개발하였다. 개발된 매생이소스별 선호도를 비교분석함으로써 완성도를 높이고자 하였다 개발된 시제품은 광주 소재 조리과학과 학생 42명을 대상으로 2009년 11윌 9일부터 11윌 16일 까지 설문조사를 실시하였다. 통계기법은 빈도분석을 이용하였다. 매생이 성분분석 결과 무기질로는 K 227.98mg/100g, Ca 58.32mg/100g, Fe 7.8mg/100g 로 나타났고, 바타민은 Vitamin A(${\mu}gRE$/100g) No Detection Vitamin $B_1$ 0..0837mg/100g Vitamin $B_2$ 0.0641 mg/100g Vitamin B6 0.0261 mg/100g Vitamin C 19.9877 mg/100g Vitamin E 0.3607 mgaTE/100g Vitamin K 7.2079 ${\mu}g$/100g Niacin 1.5057 mgaNE/100g으로 나타났다. 설문조사 결과를 통한 매생이 플레인 요거트 소스, 매생이 홍초 소스 매생이 밀쌍 소스 시제품 제작 및 조리법을 개발하였다.

  • 비타민 K 결핍은 드물지만, 소량의 출혈이 지속적일 때 의심할 수 있으며, 담즙정체증과 성장 부진이 있을 때 동반될 수 있다. 저자들은 신생아기와 영아기의 비정상적인 출혈을 주소로 내원하여 담즙 정체증에 의한 비타민 K 결핍으로 진단되었던 2예를 경험하였기에 문헌 고찰과 함께 보고한다.
