Effects of dietary glutamine and arginine supplementation on performance, intestinal morphology and ascites mortality in broiler chickens reared under cold environment

  • Objective: An experiment was conducted to evaluate the effects of dietary glutamine (Gln) and arginine (Arg) supplementation on performance, intestinal morphology and ascites mortality in broilers. Methods: A total of 675 day old chicks were randomly allocated to 9 experimental groups in a $3{\times}3$ factorial arrangement based on a completely randomized design with 5 replicates of 15 chicks. Three levels of dietary Gln (0%, 0.5%, and 1%) and Arg (100%, 130%, and 160% of Ross recommendation) supplementation were used in ascites inducing condition ($15^{\circ}C{\pm}1^{\circ}C$) from 7 to 42 days of age. Results: Dietary supplementation of Gln increased body weight gain during grower, finisher and total periods (p<0.05) and increased feed intake during total period. Ascites mortality was decreased by Gln supplementation (p<0.05). Gln supplementation increased the villus height (VH) and crypt depth (CD) in duodenum and jejunum (p<0.05). Arg supplementation decreased CD in duodenum and jejunum, and increased ileum villus width (VW) and also VH/CD ratio in duodenum and jejunum (p<0.05). Both Gln and Arg increased the goblet cell number (GCN) in duodenum whereas Gln supplementation decreased GCN in jejunum and ileum (p<0.05). The $Gln{\times}Arg$ interaction were observed for right ventricle (RV)/total ventricular (TV) ratio, VH, VW, CD, VH/CD. Conclusion: It was concluded that dietary 0.5% Gln alone or along with 130% Arg of Ross requirement, improve the intestinal morphology and performance and hence decrease the ascites mortality in broiler chickens with cold induced ascites.

Effects of soft pellet creep feed on pre-weaning and post-weaning performance and intestinal development in piglets

  • Objective: This study aimed to determine the effects of soft pellet creep feed (SPCF) on growth performance and intestinal development in piglets. Methods: A total of 18 sows and their litters of crossbred piglets (14±2 days, 3.73±0.72 kg) were assigned to one of three dietary groups receiving i) powder creep feed (PCF), ii) hard pellet creep feed (HPCF) or iii) SPCF during the pre-weaning period. After weaning, piglets were selected for continuous evaluation of the three diets on growth performance and intestinal health. Results: In the pre-weaning period, the average daily feed intake and average daily dry matter intake were significantly higher in the SPCF group than the HPCF group (p<0.05). In the post-weaning and entire experimental period, the different diets had no significant effect on growth performance. At 10 d after weaning, the serum glucose concentration was lower in the SPCF group (p<0.05) than the other groups; a higher (p<0.05) villus height and lower (p<0.05) crypt depth in the jejunum were also observed in the SPCF group than the other groups; Meanwhile, in the duodenum and jejunum, the SPCF group had a higher (p<0.05) villus height to crypt depth ratio than the other groups; Furthermore, the higher (p<0.05) threshold cycle values of lactic acid bacteria and lower (p<0.05) threshold cycle values of Clostridium, Enterobacter and Escherichia coli were also observed in the SPCF group, and the sucrase and maltase activity was higher (p<0.05) in the SPCF group than the other groups in duodenum and ileum. Conclusion: The SPCF improved pre-weaning feed intake and decreased the negative effects of weaning stress in the intestine in piglets.

Microencapsulation of Lactobacillus plantarum MB001 and its probiotic effect on growth performance, cecal microbiome and gut integrity of broiler chickens in a tropical climate

  • Objective: Microencapsulation technologies have been developed and successfully applied to protect the probiotic bacterial cells damaged by environmental exposure. This study aimed to investigate the effects of microencapsulation of Lactobacillus plantarum MB001 on the growth performance, ileal nutrient digestibility, jejunal histomorphology and cecal microbiome of broiler chickens in a tropical climate. Methods: A total of 288 one-day-old female broilers (Ross 308) were randomly allocated into 4 groups (6 replicates of 12 birds). Treatments included, i) a basal diet (NC), ii) NC + avilamycin (10 mg/kg) (PC), iii) NC + non-encapsulated L. plantarum MB001 (1×108 colony-forming unit [CFU]/kg of diet) (N-LP), iv) NC + microencapsulated L. plantarum MB001 (1×108 CFU/kg of diet) (ME-LP). Results: Dietary supplementation of ME-LP improved average daily gain, and feed conversion ratio of broilers throughout the 42-d trial period (p<0.05), whereas ME-LP did not affect average daily feed intake compared with NC group. Both N-LP and ME-LP improved apparent ileal digestibility of crude protein and ether extract compared with NC group (p<0.05). The broilers fed ME-LP supplemented diet exhibited a beneficial effect on jejunal histomorphology of villus height (VH), crypt depth (CD) and villus height to crypt depth ratio (VH:CD) of broilers compared to NC group (p<0.05). At the phylum level, Firmicutes was enriched (p<0.05) and Proteobacteria was decreased (p<0.05) only in the ME-LP group. At the genus level, the ME-LP diets increased (p<0.05) the number of both Lactobacillus and Enterococcus compared to NC, PC, and N-LP groups (p<0.05). Conclusion: Microencapsulation assists the efficient functioning of probiotics. ME-LP could be potentially used as a feed additive for improvement of cecal microbiota, gut integrity and nutrient utilization, leading to better performance of broilers.

Effects of dietary methyl sulfonyl methane and selenium on laying performance, egg quality, gut health indicators, and antioxidant capacity of laying hens

  • Objective: This study investigated the effects of dietary methyl sulfonyl methane (MSM) and selenium (Se) on the laying performance, egg quality, gut health indicators, egg yolk Se content, and antioxidant markers in laying hens. Methods: One hundred ninety-two 73-wk-old laying hens were randomly divided into four groups with eight replicates of six hens each. Four diets were prepared in a 2×2 factorial arrangement with or without MSM and Se. The trial lasted for 12 wk. Results: There were no interaction effects or main effects (p>0.05) on laying performance and egg quality. However, feed intake increased in Se-fed hens (p = 0.051) and decreased in MSM-fed hens (p = 0.067) compared with that of hens in the control group. Dietary MSM increased (p<0.05) the ileal villus height and villus height:crypt ratio in hens compared with those receiving the non-supplemented control diet. Dietary MSM and Se did not affect the percentage of short-chain fatty acids in the ileal contents. Dietary Se enriched the Se content in egg yolk compared with that of the non-supplemented control diet (p<0.05). Dietary Se increased (p<0.05) glutathione peroxidase levels in the liver and serum samples compared to the control diet. The total antioxidant capacity in the liver increased (p<0.05) in laying hens that were fed MSM-supplemented diets than in hens fed the control diet. Dietary MSM significantly increased the relative superoxide dismutase levels in serum samples (p<0.05). Conclusion: Supplementation with either MSM or Se independently improved the antioxidant capacity of laying hens. Furthermore, dietary Se produced Se-enriched eggs, but this effect was neither additive nor synergistic with dietary MSM.

Morphological differences according to uterine tissue remodeling during estrus between sika deer and water deer

  • Background: Water deer and sika deer, which breed in the wild environment, are known to have similar reproductive physiology mechanisms. Therefore, this study aimed to analyze the differences in uterine development between water deer and sika deer during estrus. Methods: MMPs and uterine development-related factors were analyzed and morphological differences were compared in the uterus of sika deer captured near Russia near Korea and water deer captured in the wild in Korea. Results: In terms of morphological differences in the uterus, the glands that form villus within the endometrium of the water deer were newly developed, and the formation of small glands was high, but the villus and glands of the sika deer were expanded, and the stroma zone in the myometrium was higher than that of the water deer. Development has increased. Additionally, the expression of PAPP-A and VEGF factors was increased in the endometrium of water deer than in sika deer, but the actions of MMPs were increased in sika deer. Conclusions: As a result of this study, there is a significant difference in the development of glands in the endometrium of water deer and sika deer during estrus, and it is believed that there is a significant difference in the development of the uterus due to the physiological effects of estrus between water deer and sika deer. Additionally, it is believed that there will be differences in the timing at which pregnancy can be decided.

Effects of Feeding and Processing Methods of Diets on Performance, Morphological Changes in the Small Intestine and Nutrient Digestibility in Growing-Finishing Pigs

  • These experiments were conducted to investigate the effects of different feeding and processing methods of diets on performance, morphological changes in the small intestine and nutrient digestibility of growing-finishing pigs. One-hundred fifty growing pigs ($Yorkshire{\times}Landrace{\times}Duroc$; initial body weight of $23.33{\pm}0.75kg$) and one-hundred twenty finishing pigs ($Yorkshire{\times}Landrace{\times}Duroc$; initial body weight of $59.22{\pm}0.56kg$) were used in Exp. 1 and Exp. 2, respectively. Pigs were grouped on the basis of body weight and gender, and randomly allotted into 6 different treatments with 5 replications in each treatment in a $2{\times}3$ factorial arrangement. Treatments were 1) dry feeding with a mash diet (DM), 2) dry feeding with a pelleted diet (DP), 3) dry feeding with an expanded crumble diet (DEC), 4) dry/wet feeding with a mash diet (WM), 5) dry/wet feeding with a pelleted diet (WP), and 6) dry/wet feeding with an expanded crumble diet (WEC). In Exp. 1 (growing phase), there was no significant difference in average daily gain (ADG) and average daily feed intake (ADFI) among treatments during the entire experimental period, but feed conversion ratio (FCR) was significantly (p<0.05) improved in pigs fed pelleted diets regardless of feeding method. FCR was best in pigs fed a DP diet and worst in pigs fed a WM diet. Pigs fed a pelleted diet showed a 6.2% or 4.0% improvement in FCR compared with those fed a mash diet or an expanded crumble diet. Water disappearance was not significantly affected by dry/wet feeding or feed processing. Significant differences in villus height were not found among treatments, but villus height tended to be improved by dry/wet feeding. Dry/wet feeding or feed processing did not affect crypt depth. Digestibilities of calcium and phosphorus were significantly (p<0.05) improved in pigs fed an expanded crumble diet compared with pigs fed mash diets. Especially, pigs fed a WEC diet digested 8.1% more P than those fed a DM diet. Feed cost per kg weight gain (FCG) tended to be increased by dry/wet feeding rather than dry feeding. In Exp. 2 (finishing phase), ADG and ADFI were not significantly different among treatments, but a significant difference in FCR was found among feed processing forms. The best FCR was obtained in pigs fed a pelleted diet. Pigs fed a DP diet showed a 11.3% improvement compared with those fed a DEC diet. Water disappearance was significantly (p=0.0408) decreased by feeding the mash diet. However, water disappearance was not affected by dry/wet feeding during the finishing period. The villus height and crypt depth were not significantly different among treatments. However, crypt depth tended to be decreased by dry/wet feeding at the mid part of the small intestine. Fat digestibility was improved by dry feeding rather than dry/wet feeding, and was improved by 4.8% by feeding pellet diets compared with expanded crumble diets. Except for carcass grade, carcass characteristics were not significantly (p<0.05) different among treatments. Carcass grade was the best in pigs fed a WP diet. Feed cost per kg weight gain (FCG) was significantly decreased in pigs fed a pelleted diet compared with those fed an expanded crumble diet, and tended to be decreased by dry/wet feeding. In conclusion, these studies suggest that feeding the pelleted diet to growing-finishing pigs can be beneficial in terms of FCR and production cost. Dry/wet feeding can be helpful for the maintenance of villus height, but may not be reflected in improved growth performance or reduction of production costs.

바다사자에서 Clostridium perfringens 감염에 의한 괴사성 장염증례 보고 (Necrotic Enteritis Caused by Clostridium perfringens in California sea lion (Zalophus californianus))

  • A case of enetritis caused by Clostridium perfringens was observed in 4years old, male California sea lion(Zalophus californianus). Clinical signs were included in anorexia, depression, diarrhea and vomitting, and the animal died on 4 days after showing clinical signs. Grossly, bloody contents were presented in the intestine and the intestinal mucosa were congested and dark redcolored, and also there were serous fluid in the peritoneal cavity. Histopathologically coagulative necrosis destroyed two third of the villus. Clostridium perfringens were isolated from the intestinal contents. Based on the gross and histopathological findings, this case was diagnosed as necrotic enteritis caused by Clostridium perfringens infection.

방사선에 의한 미토콘드리아 손상의 형태학적 고찰 (Morphological Review on Mitochondria Damage by Irradiation)

  • Mitochondria was observed much around the nuclear membrane of liver tissue where the energy metabolism process is active. Testis tissue had a large number of undifferentiated cells, and cristae in Inner membrane of Mitochondria was not observed clearly. Morphological damage occurred first in Inner membrane rather than the outer membrane. The kidney tissue was clearly observed in the form of cristae. Radiation-induced damage occurred at the edges of both ends, and the membrane was observed bursting with the thickness of the outer membrane. Small intestine cells were observed in many mitochondria in the tissues around the villus, where bowel movements were active. Morphological damage occurred with the outer and inner membranes getting tangled. Mitochondria sensitivity to radiation was sensitized in testis and small intestine tissues, and kidney, ovary and liver tissues were found to be resistant.

Studies on the Fine Structure of Caeca in Domestic Geese

  • The objective of this study was to investigate the villus distribution in the caeca of domestic geese based on the fine structure. The caeca of White Roman geese, 14-week old, were sampled and specimens were detected under photomicroscope and scanning electron microscope (SEM). The results indicated that the villi existed at the proximal caecum. The morphologies of these villi showed finger-like, peak-like or tongue-like shapes. The heights of the villi decreased far from the proximal caecum. No villi were found in the middle and distal caecum. It was obvious that the proximal segment was the main portion for absorbing food nutrients in the caeca. The caecal content particles were small and possessed a viscid character. The large particles filtered out at the proximal caecum just like a mesh. The surface of the middle caecum exhibited parallel ridges with no villi. There were band plicae circular shapes found in the middle caecum under scanning electron microsopy.

태반성성선자극(胎盤性性腺刺戟)홀몬에 관(關)한 검토(檢討) (Studies on Placental Chorionic Gonadotropin)

  • Human chorionic gonadotropin hormone(H.C.G.) is secreted from the villus tissues of the placenta and excreted in lange amount into the urine. Its isolation is chiefly made from the urine of a pregnant woman. Recently, Matsushima attempted isolation of H.C.G. directly from the placenta itself. In order to prepare H.C.G. from human placenta, general method of extractiag and purifying proteins was applied. Its way was as follow: Crude H.C.G. was extracted from placenta with pH 9.0 and pH 5.0 aqua ammonia, and purified with pH 8.0 ammonia and 50% ethanol at pH 4.8. The purified H.C.G. showed two moving bands on the anode by paper electrophoresis. On the other hand, the H.C.G. from pregnancy urine (Standard. Pharm. Co.) showed same two bands but their moving ratio were different. The purified H.C.G. showed gonadotropin effect when it was injected young fomale rats 40r/cc per day for 5 days and weighted the increased ovary weight.
