우엉뿌리추출물이 ICAM-1과 NO조절에 미치는 항염증효과 (Anti-inflammatory Effects by Arctium lappa L. Root Extracts through the Regulation of ICAM-1 and Nitric Oxide)

  • 본 연구는 우엉추출물의 항염증효과를 측정하였다. 인간 유래 폐상피세포 A549를 이용하여 염증반응을 유발하고 유지시키는데 중요한 역할을 나타낸다고 알려진 ICAM-1의 억제적 조절 효과를 평가하였으며, 염증매개인자로 알려진 NO와 이의 생성에 영향을 미치는 iNOS의 발현조절에서도 억제효과가 있음을 확인하였다. 또한, 우엉추출물이 인간각질형성세포주의 세포증식에 효과가 있음을 나타내었다. 본 연구에서 사용되는 폐상피세포는 호흡기질환의 천식 등의 알러지 및 만성염증연구에 주로 활용되고 있으며, 각질형성세포는 아토피 피부염의 만성 염증반응에서 주요하게 확인하는 요소이다. 따라서 본 연구결과에서 나타난 우엉추출물의 항염증 효과는 다양한 염증반응에서도 특히, 알러지 및 아토피 피부염 질환을 더욱 악화시키는 만성 염증에 있어서 우엉추출물이 유용한 소재로 사용될 수 있음을 시사한다고 할 수 있다.

  • 본 논문은 한 중 FTA로 농수산업의 피해가 가장 클 것으로 예상되는 전남의 농수산업에 대해 무역경쟁력을 분석하고 경쟁력 강화 방안을 제시하였다. 무역경쟁력 분석 결과 채소와 과일류에서는 수출특화, 식품가공품에서는 수입특화가 이루어지고 있었다. 중국 수입시장에서 전남의 해조류, 과실 견과류, 효모 등이 경쟁력 있는 것으로 나타났다. 또한 전남의 대부분의 농수산업 품목에서 고품질 상품을 수출하고 저품질 상품을 수입하는 고품질 산업내 무역이 이루어지고 있는 것으로 분석되었다. 분석결과를 바탕으로 농업부문과 수산업부문의 생산체제의 혁신, 유통현대화, 부가가치 극대화, 한 중 간 협력 체계 구축 측면에서 경쟁력 강화 방안을 제시하였다.

보리순의 영양성분과 항산화 효과 (Nutrient Composition and Antioxidative Effects of Young Barley Leaf)

  • This study was conducted to investigate the proximate composition and antioxidative activities of young barley leaf(YBL). YBL powder(all w/w) was 2.98% moisture, 17.13% crude protein, 4.00% crude fat, 10.72% crude ash, and 65.17% carbohydrate. The contents of total, insoluble, and soluble dietary fiber were $36.62{\pm}2.33$, $19.05{\pm}1.04$, and $17.57{\pm}1.01g/100g$, respectively. The essential and non-essential amino acids contained in the YBL powder accounted for 46.56% and 53.44% of the total amino acids, respectively. The major unsaturated fatty acid was linolenic acid. The ratio of polyunsaturated fatty acids to saturated fatty acids was 4.84. Only tartaric acid was detected. The contents of vitamins A, C, and E were 0.761, 398.05, and 0.936 mg%, respectively. The mineral contents of YBL powder were in the order of Na$IC_{50}$ value for the DPPH radical scavenging of the YBL ethanol extract was $365.74{\pm}6.98mg/mL$. The antioxidative index was high and was similar to that of t-butylated hydroxytouene. These results suggest that YBL can be recommended as a baby vegetable of high nutritional quality and antioxidative properties.

총 디젤분진의 DNA 손상작용과 야채 및 과일추출물의 보호효과 (Chemopreventive Effect of Vegetable or Fruit Extract Against Total Diesel Exhaust Particle Extract in NIH/3T3 Cells Using Alkaline Single Cell Gel Electrophoresis)

  • In urban areas, diesel exhaust particles (DEP) are probably a major component of particulate matters, especially in Korea where drive many diesel vehicles. The aim of this study was to investigate genotoxic effects of DEP using single ceil gel electrophoresis. In order to evaluate the mechanisms of DEP genotoxicity, the rat microsome mediated and DNA repair enzyme treated comet assays together with conventional comet assay were performed. Total diesel particles (DEPT) was collected without site fractionation from diesel engine bus and dichloromethane extract was obtained. The organic extract of DEPT revealed DNA damage itself in NIH/3T3 cells. The level of DNA breaks plus oxidative DNA lesions and microsome mediated DNA damage was assessed by modified single cell gel eletrophoresis. DEPT was able to induce oxidative DNA damage as well as microsome mediated DNA damage. Vitamin C as an model antioxidant reduced DNA damage in endonuclase III treated comet assay. One of flavonoid, galangin as a CYP1A1 inhibitor. reduced DNA damage in the presence of S-9 mixture. $DEP_T$ is the sources of oxidative stress, but antioxidants can significantly reduce oxidative DNA dmage. And $DEP_T$ may contain indirect mutagens which can be inhibited by CYP1A1 inhibitors. The ethanol extracts of the mixed vegetables (BV) or the mixed fruits (BF) were evaluated for their in vitro antigenotoxic effects. BV and BF showed potent Inhibitory effects against DEPT induced DNA damage with oxidative DNA lesions and in the prescence of S-9 mixture. These results indicate that BV and BF could prevent cellular DNA damage by inhibiting oxidative stress and suppressing cytochrome P4501A1 in cell culture.

-Aspergillus Parasiticus R-716의 생육 및 aflatoxin 생성에 미치는 채소추출물의 영향- (Effects of Vegetable Extracts on the Growth and the Aflatoxin Production by Aspergillus Parasiticus R-716)

  • 15종의 채소를 Chloroform으로 추출물을 조제하여 공시균(Aspergillus parasiticus R-716)의 생육 및 aflatoxin 생성에 미치는 영향을 조사하였다. 그 결과 마늘, 생강, 무우 및 양배추의 순으로 공시균의생육을 저해하였으나, 가지, 상치 등은 오히려 생육을 촉진하였다. 또한 균체생성시 공시균의 aftlatoxin기 생성은 무우, 생강, 쑥갓, 양배추의 순으로 저해하였으나, 우엉, 고추는 오히려 증가시켰다. 특히 무우는 균체 g당 aflatoxin 함량을 감소시켜 가장 효과적인 채소로 나타났으며, 고체배지에서도 무우 30g에 해당되는 추출물의 첨가로 약 $80\%$의 aflatoxin생성이 저해되어 쌀, 보리배지에서 각각 484 및 $191{\mu}g$의 aflatoxin만이 생성되었다.

한국 산채류 8종 추출물의 폴리페놀 함량분석 및 Peroxynitrite 소거효과 (Polyphenol Analysis and Peroxynitrite Scavenging Effect of the Extracts from Eight Korean Mountainous Vegetable)

  • Mountainous vegetables are usually used in the early stage of plant growth. We attempted to identify the quantity of polyphenols (caffoeoylquinic acids (CQs) and flavonoids) and peroxynitrite scavenging effect ($ONOO^-$) of eight Korean mountainous vegetables. The prominent characteristics were as follows: Only the roots of two plants Taraxacum platycarpum and Ixeris dentata contained chicoric acid. Five CQs were identified in the leaves of Cacalia firma whereas only 3-p-coumaroylquinic acid in the petioles of the plant was shown. The quantities of polyphenols such as quinic acid or tartaric acid derivatives in the mountainous vegetables were generally high, though those of flavonoids were very low. The $IC_{50}$ of chlorogenic acid, chicoric acid and kaempferol were 0.31, 0.12 and $0.25\;{\mu}g/ml$, respectively.

The antihypertensive effect of ethyl acetate extract of radish leaves in spontaneously hypertensive rats

  • Radish (Raphanus sativus L.) is a cruciferous vegetable, and its leaves have antioxidant and anticancer properties. This study was conducted to evaluate the effects of ethyl acetate extracts from radish leaves on hypertension in 11-week-old spontaneously hypertensive rats (SHRs). The SHRs were randomly divided into 3 groups of 6 rats each on the basis of initial systolic blood pressure (SBP) and were treated with oral administration of radish leaf extract (0, 30, or 90 mg/kg body weight [bw], respectively) for 5 weeks. Six Wistar rats were used as normotensive controls. The amount of the radish leaf extract had no effect on body weight. The SBP of the SHRs showed a decreasing trend with the consumption of the radish leaf extract. In the third week, the SBP of the group fed 90 mg extract/kg bw reduced from 214 mmHg to 166 mmHg and was significantly lower than that of the normotensive and hypertensive controls. The extract did not show a significant effect on the angiotensin-converting enzyme (ACE) activity in the serum, kidney, and lung. The extract increased the concentration of NO in serum and the activities of antioxidant enzymes such as glutathione peroxidase and catalase in red blood cells (RBCs). The serum concentrations of $Na^+$ and $K^+$ were not significantly different between all groups. However, the fecal concentrations of $Na^+$ and $K^+$ increased; the fecal concentrations of $Na^+$ and$K^+$for the normotensive and hypertensive controls were not different. Urinary excretion of $Na^+$ was higher in the normotensive Wistar rats than in the SHRs, while that of $K^+$ was not significantly different. These findings indicate that consumption of radish leaves might have had antihypertensive effects in SHRs by increasing the serum concentration of NO and fecal concentration of $Na^+$ and enhancing antioxidant activities.

Quantitative Determination of Five Phenolic Peroxynitrite-scavengers in Nine Korean Native Compositae herbs

  • Peroxynitrite (ONOO)-scavenging activities of nine Compositae herbs consisting of three Ixeris, two Youngia, two Cirsium and one of each Lactuca and Taraxacum species were evaluated. The contents of their ONOO scavengers in the extracts were also determined on a HPLC using seven standard compounds, chlorogenic acid (CGA), chicoric acid (CA), luteolin 7-glucoside (luteolin-7-glc), luteolin 7-glucuronide (luteolin-7-glcU), luteolin, linarin and pectolinarin. Five of those compounds exhibited potent ONOO-scavenging activities: IC50, CA (0.76 μM), CGA (1.34 μM), luteolin (0.81 μM), luteolin-7-glc (0.86 μM) and luteolin-7-glcU (3.13 μM). Both CA and luteolin-7-glc were highly contained in I. dentata (19.71 mg/g and 13.58 mg/g, respectively), I. dentata var. albiflora (17.58 mg/g and 23.83 mg/g, respectively) and I. sonchifolia (65.71 mg/g and 6.99 mg/g, respectively). Among the nine herbs, those three Ixeris species had very low IC50 values over the range of 0.48 - 1.74 μg/mL, suggesting that they could be potential therapeutic vegetables, particularly for preventing diabetic complications or obesity, which can be caused by an excess production of ONOO.

아산 지역특산물과 설화를 활용한 Food Tourism 제품 개발 (Study on the Development of Food Tourism Products Based on the Local Food and Folktale)

  • This study aimed to develop unique, local "food tourism" products by finding specialized items that combine tourist attractions, such as folklore or hot springs. Traditional ingredients were analyzed with ancient texts for the methods of research. A brand image was made possible utilizing hot spring lore and other regional stories. The tofu products were produced using local specialty beans. Products, such as tofu residue cake and willow bean tea, were made with the tofu residue. After the products were completed, the sensory test began at the local tourist attraction. Asan City's food tourism product willow tofu was made with beans that were given as compensation for building the Onyang temporary palace according to the Annals of the Joseon Dynasty and the willow tree that appears in Sunshin Lee's anecdotes. After the preference test between normal tofu and willow-extract tofu was conducted to measure the product potential of willow tofu, among the sample extracts, 0.04% of the willow extract showed a significant preference. The hot spring tofu-residue cake was baked using tofu residue and vegetable olive oil to substitute for animal oil, such as butter, or margarine. After the sensory test targeting the adults was conducted, both products displayed significant product potential with average scores above 5.0. Willow tree bark, which has antioxidation and anti-inflammatory effects without a bitter taste or strong smell, was proven to bean appropriate ingredient for leached tea. The nutty flavor of leached tea was enhanced by roasted green kernel black beans and willow tree bark. The sensory test showed that the leached tea and tofu received a high preference rating on both color and flavor.

적갓김치의 발효특성 및 항산화성 (Fermentation Characteristics and Antioxidative Effect of Red Mustard Leaf Kimchi)

  • 본 연구에서는 적갓김치의 발효과정에 따른 발효특성과 항산화성을 규명하기 위해 갓김치와 가열 쇠고기를 이용한 model system을 만들어 쇠고기 지방질에 대한 항산화성을 살펴보았고 용매별로 추출한 김치성분의 항산화성을 살펴보았다. 적 갓김치는 15$^{\circ}C$에서 발효가 진행되면서 6일째 김치의 pH가 4.5에 도달하였다. 발효진행에 따라서 산도는 증가하였고 환원당은 서서히 감소하였으며 이때 젖산균수 역시 최대치에 도달하였다. 또한 a-scorbic acid 함량은 발효가 진행되면서 감소하였다가 발효 6일째에 다시 다소 증가하였다. 총 페놀함량은 생 시료에서보다는 갓김치에서 높게 나타났으며 발효가 진행됨에 따라 총 페놀함량은 증가하였다. 적 갓김치를 가열쇠고기와의 model system을 조제하여 TBA가를 통해 항산화성을 살펴본 결과 첨가한 김치의 양이 많을수록 항산화성이 높게 나타났으며 생시료일 때보다는 김치의 항산화성이 우수하였으며 발효가 진행된 숙성상태일 때 더 항산화성이 우수하였다. 동결건조시킨 적 갓김치를 물, 메탄올, 핵산 세가지 용매로 추출하여 항산화성을 살펴보았을 때 메탄을 추출물의 항산화성이 가장 높게 나타났다. 또한 담금직후보다는 숙성된 상태의 갓김치가 항산화성이 높았다.
