Cavitation Condition Monitoring of Butterfly Valve Using Support Vector Machine (SVM을 이용한 버터플라이 밸브의 캐비테이션 상태감시)

  • Butterfly valves are popularly used in service in the industrial and water works pipeline systems with large diameter because of its lightweight, simple structure and the rapidity of its manipulation. Sometimes cavitation can occur. resulting in noise, vibration and rapid deterioration of the valve trim, and do not allow further operation. Thus, the monitoring of cavitation is of economic interest and is very importance in industry. This paper proposes a condition monitoring scheme using statistical feature evaluation and support vector machine (SVM) to detect the cavitation conditions of butterfly valve which used as a flow control valve at the pumping stations. The stationary features of vibration signals are extracted from statistical moments. The SVMs are trained, and then classify normal and cavitation conditions of control valves. The SVMs with the reorganized feature vectors can distinguish the class of the untrained and untested data. The classification validity of this method is examined by various signals that are acquired from butterfly valves in the pumping stations and compared the classification success rate with those of self-organizing feature map neural network.

  • A Web-based design automation system for concrete slab bridge has been developed to manage the resultant documents as well as to speed up the repetitive design process. Manipulation of engineering drawings in the Web page is one of the critical functions needed for Web-based design automation. Active Server Pages (ASP) is used to that collects design parameters in the database, and eXtensible Markup Language (XML) and XML-based vector graphics are expected to facilitate the representation of engineering drawings in the Web page. In this paper, we present how we used XML and Scalable Vector Graphics (SVG) to compose engineering drawings and represent them in the Web page. XML Data Island we designed to define drawing components turned out effective in manipulating the engineering drawings in the Web page.

Protoplast-Mediated Transformation of the Filamentous Fungus Cladosporium phlei: Evidence of Tandem Repeats of the Integrative Transforming Vector

  • To facilitate the genetic manipulation of Cladosporium phlei, a causal agent of leaf spot disease in timothy (Phleum pretense), protoplast-mediated transformation of C. phlei has been developed and the resulting transformants were characterized in this study. Hygromycin B resistance was applied as a dominant selection marker due to the sensitivity of C. phlei to this antibiotic. The transformation efficiency ranged from approximately 20-100 transformants per experiment. Southern blot analysis of stable transformants revealed that transformation occurred by way of stable integration of the vector DNA into the fungal chromosome. PCR analysis and plasmid rescuing of randomly selected transformants suggested that integration of tandem repeat copies of vector DNA was common. In addition, multiple integrations of the transforming vector at different chromosomal sites were also observed. The establishment of a transformation method for C. phlei facilitates strain improvement of this fungus and can be applied as an initial step in the molecular analysis of pigment production in this fungus.

Development of Selectable Vector Plasmid in Bacteriophage P2-P4 System and Its Stability (박테리오파지 P2-P4 시스템을 위한 벡터 플라스미드 개발과 안정성)

  • While bacteriophage P2-P4 system is very useful experimental tool for the study of viral capsid assembly, there is no useful plasmid vector for the DNA manipulation in bacteriophage P2-P4 system. In this study, a new vector plasmid, P4 ash8 (sid71) kmr, was constructed by swapping the non-essential region of P4 DNA for kanamycin resistance(kmr) gene cassette of plasmid pUC4-K. P4 ash8 sid71 was starting material for the construction, since it tends to be maintained as a plasmid in the absence of the helper phage. The total size of this chimera was designed to be packaged into P4 or P2 size heads with induction by P2 infection. The conversion of plasmid P4 ash8 (sid71) kmr to bacteriophage was proved by burst size determination experiment and CsCl buoyant equilibrium density gradient experiment. Integrase destructed P4 derivative, P4 ash8 sid71 kmr intS, was able to be constructed easily by in vitro DNA manipulation of P4 ash8 sid71 kmr. The plasmid stability experiment with P4 ash8 sid71 kmr if/tS showed that the integrase of P4 affects the stable maintenance of plasmid P4 state.

  • In this paper, we propose a new channel identification technique for long code DS/CDMA DMB down link system which estimate the channel response based on the signal space vector only, unlike the most conventional subspace method relying on the orthogonal property of noise space vectors to the signal space vector. Because of this property of the proposed method, it is optimum and practical in manipulation of the covariance matrix to be analyzed. In the paper, we derive the mathematical expression necessary to clarify the proposed method and show the relevant simulation and numerical results to verify the validity of the proposed algorithm.

An Efficient Markerless Deletion System Suitable for the Industrial Strains of Streptomyces

  • The genus Streptomyces is intensively studied due to its excellent ability to produce secondary metabolites with diverse bioactivities. In particular, adequate precursors of secondary metabolites as well as sophisticated post modification systems make some high-yield industrial strains of Streptomyces the promising chassis for the heterologous production of natural products. However, lack of efficient genetic tools for the manipulation of industrial strains, especially the episomal vector independent tools suitable for large DNA fragment deletion, makes it difficult to remold the metabolic pathways and streamline the genomes in these strains. In this respect, we developed an efficient deletion system independent of the episomal vector for large DNA fragment deletion. Based on this system, four large segments of DNA, ranging in length from 10 kb to 200 kb, were knocked out successfully from three industrial Streptomyces strains without any marker left. Notably, compared to the classical deletion system used in Streptomyces, this deletion system takes about 25% less time in our cases. This work provides a very effective tool for further genetic engineering of the industrial Streptomyces.

Modern Methods of Text Analysis as an Effective Way to Combat Plagiarism

  • The article presents the analysis of modern methods of automatic comparison of original and unoriginal text to detect textual plagiarism. The study covers two types of plagiarism - literal, when plagiarists directly make exact copying of the text without changing anything, and intelligent, using more sophisticated techniques, which are harder to detect due to the text manipulation, like words and signs replacement. Standard techniques related to extrinsic detection are string-based, vector space and semantic-based. The first, most common and most successful target models for detecting literal plagiarism - N-gram and Vector Space are analyzed, and their advantages and disadvantages are evaluated. The most effective target models that allow detecting intelligent plagiarism, particularly identifying paraphrases by measuring the semantic similarity of short components of the text, are investigated. Models using neural network architecture and based on natural language sentence matching approaches such as Densely Interactive Inference Network (DIIN), Bilateral Multi-Perspective Matching (BiMPM) and Bidirectional Encoder Representations from Transformers (BERT) and its family of models are considered. The progress in improving plagiarism detection systems, techniques and related models is summarized. Relevant and urgent problems that remain unresolved in detecting intelligent plagiarism - effective recognition of unoriginal ideas and qualitatively paraphrased text - are outlined.

Distinctive Characteristics of an Autonomous Replication Sequence of Cephalosporium acremoniurn in Yeast (Cephalosporium acremonium의 자율복제 기점의 특성)

  • An autonomous replication sequence (ARS) derived from Cephalosporium acremonium ATCC 20339 was cloned in Sarchuromyces cerevisiae SHY 3 using YIp5 as a cloning vector. A new recombinant plasmid, designated pCY-2, which contained a 3.7 kb BamHI fragment of C. acrenzonium DNA showed the highest stability among the 40 recombinant plasmids composed of the YIp5 2nd ARS of C. ucremoniztm. Also, Southern hybridization and transformation of E, cull with DNA purified from yeast transformants verified that pCY-2 autonomously replicates in yeasts. Transformation efficiency and plasmid stability of pCY-2 in yeast were higher than those ol YRp 7 containing ARS which originated from yeast. Detailed studies by subcloning revealed that two ARSs existed within 2.6 kb of the insert, which is a novel discovery. However, it was concluded that these two ARSs were ligated during the gene manipulation in vitro.

The Road Alignment Optimization Modelling of Intersection Based on GIS (GIS를 이용하여 교차로를 고려한 도로선형 최적화 모델링)

  • This study develops modeling processes for alignment optimization considering characteristics of intersections using genetic algorithms and GIS for road alignment optimization. Since existing highway alignment optimization models have neglected the characteristics of intersections, they have shown serious weaknesses for real applications. In this paper, intersection costs include earthwork, right-of-way, pavement, accident, delay and fuel consumption costs that are sensitive and dominating to alignments. Also, local optimization of intersections for saving good alignment alternatives is developed and embedded. A highway alignment is described by parametric representation in space and vector manipulation is used to find the coordinates of intersections and other interesting points. The developed intersection cost estimation model is sufficiently precise for estimating intersection costs and eventually enhancing the performance of highway alignment optimization models. Also, local optimization of intersections can be used for improving search flexibility, thus allowing more effective intersections. It also provides a basis for extending the alignment optimization from single highways to networks. The presented two artificial examples show that the total intersection costs are substantial and sensitive to highway alignments.
