닭 도계 및 가공공정 중 유해미생물의 분포와 항생제 감수성 (The distribution and antimicrobial susceptibility of pathogenic microorganisms isolated from chicken slaughtering and processing procedure)

  • This study was performed to analyze the distribution and antimicrobial resistance of pathogenic microorganisms isolated from the carcass and environments of chicken processing plant located in Gyeonggi province from October to November in 2010. Chicken slaughterhouse was visited 3 times and totally 40 samples were collected from chicken carcass before and after washing (n=14), chicken cuts (n=7), cooling water (n=8), brine (n=2), cutting knives (n=7) and working plate (n=2). Whole-chicken rinsing technique (for chicken carcasses) and swab technique (for working plate and knives) were used to analyze the distribution of pathogenic microorganisms. In addition, brine and chilling water from storage tanks were gathered using sterilized tubes and used as samples. The matrix-assisted laser desorption/ionization mass spectrometry (MALDI MS) for whole cell fingerprinting in combination with a dedicated bioinformatic software tool was used to identify the isolated microorganisms. The pathogenic microorganisms, such as Bacillus cereus (n=8) and Staphylococcus aureus (n=9), were isolated form the chicken processing process (chicken carcasses of before and after chilling, chicken cuts, and working plate). The antimicrobial susceptibility of those isolated microorganisms was analyzed using 21 antimicrobial agents. In the case of B. cereus, it showed 100% of resistance to subclasses of penicillins and peptides, and it also resistant to cephalothin, a member of critically important antimicrobials (CIA), however there was no resistance (100% susceptible) to vancomycin and chloramphenicol. S. aureus showed 100% resistance to subclasses of peptides and some of penicillins (penicillin and oxacillin), however, it showed 100% susceptibility to cephalosporins (cefazolin and cephalothin). All of the tested pathogens showed multi drug resistance (MDR) more than 4 subclasses and one of B. cereus and S. aureus showed resistance to 9 subclasses. After the ban on using the antimicrobials in animal feed in July 2011, there would be some change in microbial distribution and antimicrobial resistance, and it still has a need to be analyzed.

광주지역내 바닥분수 재이용수에서 분리한 장구균의 항생제 내성 연구 (A Study of Antibiotic Resistance of Enterococci Isolated from Recycling Water of Floor Fountains in Gwangju Area)

  • Objectives: This study aims to evaluate water quality in terms of microorganisms and identify the antibiotic resistance of Enterococci isolated from the recycling water in floor fountains at three parks and one reservoir in the Gwangju area. Methods: Water samples were analyzed for Enterococci using membrane Enterococcus indoxyl ${\beta}$ d glucoside agar (mEI) as described in USEPA Method 1600. The vancomycin-resistant Enterococci with VanA and VanB were identified by PCR. An examination of the antibiotic resistance of isolates against 14 antibiotics was performed by the disk diffusion method. Results: The drinking water quality criterion was exceeded for total colony counts in 68% of all recycling water samples. The average concentration of total califorms and fecal coliforms was 139,325 and 413 CFU/100 mL, respectively. VanA and VanB were not detected from the isolates. We found the antibiotic resistant Enterococci strains to be E. faecalis, E. faecium, E. durans, E. mundtii, E. hirae, and E. thailandicus. The isolates were resistant to Rifampin (50%), Erythromycin (25.8%), Tetracycline (10.2%), Nitrofurantoin (8.1%), Minocycline (3.1%), Erythromycin (1.2%), Penicillin (0.7%), Norfloxacin (0.5%), and Teicoplanin (0.5%) among the 14 antibiotics tested. Antibiotic resistance tests for Enterococci from the recycling water of floor fountains resulted in 30.2% showing resistance to two or more antibiotics. Conclusions: These results showed that the multi-antibiotic resistance of Enterococci, E. coli, and others should be investigated continuously in each environment field.

Vibrio parahaemolyticus의 환경인자(環境因子)에 대(對)한 저항성(抵抗性) (Resistance of Vibrio parahaemolyticus Against the Environmental Factors)

  • 본 연구는 식중독 환자와 1985년부터 1986년까지 2년간 경상북도 동해안에서 어획한 어패류로부터 분리 동정된 장염 비브리오균의 저항성에 대해서 관찰한 것으로, 대조균은 VP ATCC 17802와 NAG V. ATCC 6538이었다. 어패류와 해초 둥에서는 온도가 상승할수록 생존기간이 짧았다. 해수온도가 상승할수록 생존기간이 길었으며 특히 $37^{\circ}C$$25^{\circ}C$에서 6개월이 지난 현재도(1987. 6. 10) 생존을 계속하고 있다. 상수에서 150분내 사멸하였고 지하수에서 11.5일간 생존하였다. 숙성된 김치(pH 3.9)가 냉장고에서 보관된 김치(pH 4.5)보다 균의 사멸시간이 빨랐으며, 우유에서 57.1일, 요구르트에서 49.2분간 생존하였다. $-70^{\circ}C$의 냉동상태에서는 6개월이 경과한 현재까지도 생존하고 있으나$60^{\circ}C$ 항온조에서는 20분내에 전 균주가 사멸하였다. 내건성 실험에서 10-11% 함수율을 기점으로 생존수가 급격히 떨어지는 양상을 보였다. 자외선 $2538{\AA}$의 조사에서 20분내에 전 균주가 사멸하였다. 알콜에 대한 시험결과 25%미만의 발효주에서는 0.5내지 4분간 생존하였고 25% 이상의 증류주 경우 0.1분내 사멸하였다. 소독액의 유효 살균농도는 석탄산 1%, 크레졸 3%이었다. 0.1M 빙초산과 유산 함유배지에서 60분간 배양후, 생균수가 급격히 저하하였으나 구연산의 경우는 그렇지 않았으며 염도가 증가할수록 생균수 감소폭이 줄어 들었다. Vancomycin, Chloramphenicol, Gentamicin, Cephalothin 및 Nitrofurantoin 등에 감수성을 보였으며 Carbenicillin, Ampicillin 및 Kanamycin 등에 저항성을 나타내었다. 또 1일 배양균을 25일간 증균시킬 때 Tetracycline, Cephalothin 등에 대한 저항성이 감수성으로 변화하였다.

소아의 지역사회 획득 장구균 요로감염의 임상 양상 (Clinical Characteristics of Community Acquired Enterococcal Urinary Tract Infections In Children)

  • 목적: 장구균은 원내 감염의 중요한 세균으로 최근 그 빈도가 많아지며, 항생제 내성을 갖는 경우도 증가하면서 활발한 연구가 이루어지고 있다. 하지만 소아의 지역사회 획득 요로감염의 원인으로서 장구균에 대한 연구는 소수에 불과하여 이에 저자들은 부산 경남 지역에서 발생하는 장구균에 의한 지역사회 획득 요로감염의 임상적 특징과 항생제 감수성 등을 조사하고자 하였다. 방법: 20010년 1월부터 2013년 1월까지 첫 번째 요로감염으로 부산대학교 어린이병원에 입원 하였던 18세 미만의 환자 중 장구균과 대장균이 배양된 환자를 대상으로 하여 의무 기록을 후향적으로 분석하였다. 의무 기록을 통해 환자의 성별, 진단 시 연령, 기저 신 질환, 최근 항생제 복용력, 영상 검사의 결과, 세균의 항생제 감수성, 처음 사용한 항생제에 대한 반응 유무 등을 조사하였다. 결과: 2010년 1월부터 2013년 1월까지 첫 번째 요로감염이 확인되어 치료한 환자는 201명으로 이 중 대장균은 154명(76.6%)에서 분리되었고, 장구균은 11명(5.5%)에서 분리되었으며 배양된 장구균은 모두 Enterococcus feacalis였다. 장구균이 배양된 집단에서 배뇨방광요도조영술 검사를 시행한 7명 중 4명(57.1%)에서 방광요관역류가 발견되었고, 이들 모두 DMSA 신스캔에서 신 반흔을 보였으며 현재까지 3명의 환자가 방광요관역류에 대한 비뇨기과적 교정 시술을 시행 받았다. 대장균이 배양된 집단에서는 121명의 환자에서 배뇨방광요도조영술 검사가 시행되어 23명이 방광요관역류로 진단되었고, 현재까지 11명이 비뇨기과적 교정술을 시행 받았다. 대장균에 의한 요로감염 환자들과 비교하였을 때 장구균 요로감염 환자에서 방광요관역류와 수술적 교정술의 빈도가 통계학적으로 의미 있게 높았다. 분리된 장구균의 항생제 감수성 검사에서 ampicillin, vancomycin, linezolid에 대해서는 100% 감수성을 보였으며 tetracycline에 대해서는 100% 내성을 보였고 Trimethoprim/sulfamethoxazole에는 71.4%, quinolone계열인 ciprofloxacin에는 20% 내성을 나타냈다. 결론: 부산 경남 지역의 지역사회 획득 장구균 요로감염은 흔하지 않지만, 근본적인 요로계의 이상을 동반할 가능성이 높으므로 환자에 대한 영상학적 평가가 이루어져야 할 것으로 생각된다.

Molecular Typing and Antimicrobial Susceptibility of Staphylococcus aureus Strains Isolated from Raw Milk, Cheese, Minced Meat, and Chicken Meat Samples

  • The objectives of this study were: i) to detect the presence of Staphylococcus aureus and methicillin-resistant S. aureus (MRSA) in raw milk, cheese, beef minced meat, and chicken meat samples; ii) to evaluate the antimicrobial susceptibility of the isolates; and iii) to determine clonal relation among the isolates by using pulsed-field gel electrophoresis (PFGE) method. Therefore, a total of 160 food samples were randomly collected between August 2014 and May 2015 in Hatay province, located in the southern Turkey. Twenty (12.5%) of the samples were found to be contaminated with S. aureus. A total of 40 isolates from the 20 positive samples were confirmed to be S. aureus by multiplex PCR based on 16S rRNA and nuc gene. The mec A gene was not detected in any of the S. aureus strains. In the present study, 39 out of 40 (97.5%) isolates were found to be resistant to one or more antibiotics. All of isolates were susceptible to gentamicin, oxacillin, and vancomycin. The highest resistance rate was detected in penicillin (95%) and ampicillin (92.5%), followed by tetracycline (30%), erythromycin (20%), ciprofloxacin (12.5%). Nine major patterns were determined by PFGE. In 6 of these patterns, thirty-six strains (90%) had identical PFGE profiles.

Purification and Characterization of Antistaphylococcal Substance from Pseudomonas sp. KUH-001

  • A bacterium producing unique antistaphylococcal substance (ASS) was isolated from soil samples. The isolated strain KUH-001 was identified to belong to Pseudomonas species from the characteristic properties of its fluorescence and cellular 3-hydroxy fatty acid composition, etc. The ASS component was purified by procedures employing activated carbon adsorption, column chromatography with silica gel, preparative TLC and HPLC. This compound could also be purified mainly by repeating of trituration and precipitation with chilled ether. Purified ASS with a m.p. value of $140~142^{\circ}C$ showed marked stability at high temperature (at $121^{\circ}C$ for 10 min) and extreme pHs (in 1N HC1 and 1N NaOH for 1 day) without significant loss of antibiotic activity. From spectral data of UV, IR, NMR, and FAB-MS, the compound was elucidated as 2-heptyl-4-hydroxyquinoline N-oxide (HHQO). Under the conditions employed, HHQO exhibited a narrow antimicrobial spectrum. active particularly against Staphylococcus aureus including the methicillin resistant strain. Moreover, it did not induce resistance, and besides, interacted synergistically with certain antibiotics such as vancomycin or erythromycin.

Assessment of Potential Probiotic and Starter Properties of Pediococcus spp. Isolated from Turkish-Type Fermented Sausages (Sucuk)

  • In this study, the metabolic activities of five strains of Pediococcus spp., in terms of the quantities they produced of lactic acid, hydrogen peroxide, exopolysaccharides, and proteolytic activity, were determined. Lactic acid levels produced by these strains were found to be in the range of 2.5-5.6 mg/ml. All strains produced hydrogen peroxide. The P. pentosaceus Z13P strain produced the maximum amount (0.25 mg/ml) of proteolytic activity. Exopolysaccharide (EPS) production by the Pediococcus strains during growth in MRS (de Man, Rogosa, and Sharpe) medium was in the range 25-64 mg/l. The susceptibility of 10 different antibiotics against these strains was also tested. All strains were found to be resistant to amoxicillin, gentamicin, and vancomycin. Antimicrobial effects of the Pediococcus spp. on pathogens were also determined by an agar diffusion method. All of the strains were able to inhibit L. monocytogenes. The tolerance of the strains to low pH, their resistance to bile salts of strains, and their abilities to autoaggregate and coaggregate with L. monocytogenes were also evaluated.

경북지역 젖소 유방염 우유 및 각종 동물로부터 분리한 포도구균의 동정 및 항생제 감수성 (Species identification and antibiotics susceptibility of Staphylococci isolated from bovine mastitic milk and several animals in Kyungbuk province)

  • This study was carried out species identification and antibiotics susceptibility of Staphylococci Isolated from bovine mastitic milk, chicks, Korean indigenous goats, pigs and mice in northern area of Kyungbuk. The result were summarized as follows ; A total of 71 Staphylococci were Isolated from bovine mastitic milk, chicken, pig, Korean indigenous goat and mouse. The results of identification of 71 Staphylococci revealed that S. aureus was most important pathogen in animals tested. Of 39 Staphylococci from bovine mastitic milk, 16 of 39 isolates (41%) were S. aureus and 9 of 39 isolates (23% ) were S. hyicus subsp chromogens. The results of susceptibility test to 16 antibiotics revealed that 91.5% of all isolates were resistant to more than 1 antibiotic and resistance to penicillin was most high (76.1%), All Isolates were susceptible to vancomycin.

소와 돼지 도체표면에서 황색포도상구균의 분리 및 장독소 검출 (Isolation of Staphylococcus aureus and detection of enterotoxin from pigs and cattle carcass by PCR)

  • At the present study, it was aimed to explore the states of antimicrobial resistant Staphylococcus aureus isolates from 320 pigs and cattle carcass (160 pigs and 160 cattle) slaughtered in Busan province from March 2008 to November 2009. Among 320 samples, 26 of Staphylococcus aureus were isolated from pigs (10.6%) and cattle (5.6%). In antimicrobial susceptibility test, all of the isolates were demonstrated susceptibility to oxacillin, cefoxitin, cephalothin, vancomycin, rifampin and linezolid. But the isolates were showed resistance other antibiotics in order of penicillin (92.3%), gentamicin (76.9%), tetracycline (69.2%), erythromycin (65.4%), and clindamycin (61.5%). In case of enterotoxin production, 7.7% of 2 strains produced enterotoxin A.

  • The antimicrobial susceptibility of 73 strains of Staphylococcus aureus isolated from dogs and cats was determined against 16 antimicrobial agents. All isolates were susceptible to cephalothin (CF), vancomycin (VA), rifampin (RA), quinupristin-dalfopristin (SYN) and linezolid (LZD), whereas more than 86.3% of the isolates were resistant to penicillin (P). In the case of enterotoxin production, 8.2% of 73 strains produced enterotoxin A and 1 strain produced enterotoxin C. Appropriate protocols for antimicrobial agents and strategies to reduce antimicrobial resistance will need to be investigated in the future.
