Pasteurella anatipestifer infection in ducklings in Korea (오리의 Pasteurella anatipestifer 감염증 발생)

  • This study was conducted to investigate the cause of a new duck disease occured in southern part of Korea. A meat type duck farm located in Kangjin, Chonnam Province had experienced outbreaks of septicemic disease at around 20 days of age in nearly every batch of ducklings from early spring to early summer in 1989. Main symptoms of the birds were eye and nasal discharge, depression, inappetence, diarrhea and nervous signs such as tremor and ataxia. Some birds died suddenly without any signs. Mortality reached from 20% to 80% in severe cases. The autopsy findings of the affected ducklings revealed consistantly severe airsacculitis, fibropericarditis, perihepatitis and occasionaly enteritis and distended ureter with urate deposit. A rod shaped gram-negative bacterium was isolated purely from brain and liver of the diseased ducks by culturing the specimens on blood agar for 48 hours in candle jar. The isolate neither produced hemolysis nor grew on MacConKey Agar. It formed colony relatively slowly being recognizable at least 36 hours after culturing, reaching colony size of about 1mm in diameter at 48 hours culture. The colony looked iridescent under oblique light and had muddy odor. The isolate did not ferment carbohydrates tested but produced gelatinase, hippuricase and oxidase which were considered as characteristics of P anatipestifer. The isolate induced similar signs and lesions when infected experimentally into ducks of 3 to 38 days age via intraperitoneum or intratrachea. However it did not produce any clinical signs wen inoculated via intranasal route. It produced only mild signs in chicken just injected with a very large dose. The bacteria did not produce any signas or lesions in mice. It was concluded through biochemical and physiological tests and animal inoculation tests that the new disease was caused by P anatipestifer.

  • Pigmented villonodular synovitis, synovial chondromatosis, long-standing rheumatoid arthritis, hemophilic arthropathy, chronic tophaceous gout, amyloid arthropathy, tuberculous arthritis, and hemangioma are the synovial diseases showing low signal intensity on T2-weighted image. Synovial deposition of hemosiderin, urate, and amyloid and fibrosis or caseous necrosis of hypertrophied synovium are known as the pathologic causes of T2 signal intensity. Because of the low incidence of the synovial lesions showing T2 low signal intensity, recognition of these diseases would be helpful for the exact diagnosis.

Structure and Isolation of Xanthine Oxidase Inhibitor from Oolong Tea (우롱차로부터 Xanthine Oxidase 저해물질 분리 및 구조)

  • Xanthine oxidase involved in pruine metabolism oxidizes hypoxanthine to xanthine and xanthine to uric acid. The derangement of pruine metabolism results in gout that associates painful deposit of monosodium urate in the cartilage of joints. In the continuous study for natural compound, six flavan-3-ols have been isolated from the leaves of Oolong tea. The structures of procyanidin B-1, B-3, procyanidin B-3-3-O-rhamnose, procyanidin B-1-3-O-gallate, (-)-epicatechin, (-)-epicatechin-3-O-gallate were established by NMR and their inhibitory effect on xanthine oxidase activity was investigated. Flavan-3-ols containing the gallate had a high inhibitory capacity. Procyanidin B-1-3-O-gallate showed complete inhibition at $50\;{\mu}M$ and inhibited on the xanthine oxidase competitively.

Hypouricemic and xanthine oxidase inhibitory activities of the fractions of Coccinia grandis L. Voigt

  • The present study was aimed at investigating the hypouricemic and xanthine oxidase inhibitory activities of the various fractions of the hydromethanolic extract of the leaves of Coccinia grandis L. Voigt (Cucurbitaceae). The leaves of this species was used in traditional medicinal system for the treatment of gout, rheumatism, jaundice, bronchitis, fever, skin eruptions, wounds, etc. The degree of xanthine oxidase inhibition was determined in vitro by measuring the increase in absorbance at 295 nm associated with uric acid formation. Among the fractions tested, the chloroform fraction exhibited highest potency ($IC_{50}$ $17.8\;{\mu}g/ml$). This was followed by the pet-ether ($IC_{50}$ $29.7\;{\mu}g/ml$), ethyl acetate ($IC_{50}$ $41.2\;{\mu}g/ml$) and residual ($IC_{50}$ $47\;{\mu}g/ml$) fractions. The $IC_{50}$ value of allopurinol was $6.1\;{\mu}g/ml$. In addition, the hypouricemic and hepatic xanthine oxidase (XO)/xanthine dehydrogenase (XDH) inhibitory activities of the fractions were examined in vivo using oxonate (280 mg/kg, i.p.) induced hyperuricemic mice. At a dose of 200 mg/kg orally for 7 days, the pet-ether, chloroform and ethyl acetate fractions produced a significant (P < 0.01) reduction in serum urate level and also inhibited hepatic XO/XDH activities when compared to hyperuricemic mice. These inhibitory effects were weaker than that observed for the standard drug, allopurinol (10 mg/kg, p.o.). Lineweaver-Burk analysis of the enzyme kinetics indicated that the mode of inhibition was of a mixed type. These results suggest that the use of Coccinia grandis leaves for the treatment of gout could be attributed to its XO inhibitory activity.

A case report : Iguana iguana with cystic calculus (녹색 이구아나의 방광결석증)

  • A female Green iguana (Iguana iguana) was brought to the small animal clinic in Hannover Veterinary school because of anorexia and convulsion. Two months ago, the owner bought two iguanas, one female and one male and had to take a female to the one of local small animal clinic because she, born in 19931 didn't eat well and showed the tremor of the legs. The veterinarian suspected it to have a calcium deficiency and gave an injection of calcium and vitamin D3. But still during the last two months, the female iguana did only eat some lettuce while the other male ate lettuce, fruits and commercial cat-food ($Whiskas^{\circledR}$, Effem, Verden). It was not possible to evaluate the rations of these different "food-components". They are kept in the terrarium with water container. By palpation of the abdomen, hard thing like stone was palpated in the mid-region. The blood test wasn't carried out because of the difficulties of getting blood and because of her bad condition. It was firmly diagnosed as cystic calculus through radiography. Since the animal is in shock state already, the prognosis after surgery was bad. It was assumed to be too late to expect its surviving with the surgery carried out or without it. Cystotomy was performed in the cranial ventral paramedian incision in order to preserve the large ventral abdominal vein in the linen alba region. After two stay sutures ("holding suture" ) with 5-0 polyglactin 910 ($Vicryl^{\circledR}, Ethicon$) on the bladder, cystotomy was carried out and the stone inside was removed. Then, the bladder was closed in one-layer sutures. The abdomen was closed in simple interrupted suture with same material after the instillation of 10 ml saline solution into the abdominal cavity. The cystic calculus was in a size of $5.8{\times} 5.O{\times} 3.5 cm$. The analysis of the stone was carried out in two ways which are the infrared spectroscopy of the stone and quick analysis by $Merckognost^{\circledR}$(Diagnostica Merck, Merck). In the result of the infrared spectroscopy, the stone wee composed of 10% ammonium urate and 90% dihydrated uric acid. This iguana wasn't able to survive.n't able to survive.

Pathological evaluation of renal changes induced by multiple nephropathogenic factors in SPF chickens II. Clinicopathological observation (신부전 요인에 의해 유발된 닭 신장변화의 병리학적 관찰 II. 임상병리학적 관찰)

  • Renal failure is one of the main causes of economic impacts in the poultry industry and complex syndrome with different severity of clinical signs caused by multiple nephropathogenic factors such as infectious bronchitis viral infection and excess salt and calcium in diet. To evaluate the correlation between severity of renal failure and the causative nephropathogenic factors, one-day-old specific pathogen free chickens were treated with either single causative factor or multiple causative factors described as above. Each group was designed as control for non-treated control, IB for infectious bronchitis virus (IB virus) infection, IBHNa for IB virus infection with high diet salt, IBHCa for IB virus infection with high diet calcium, IBHNC for IB virus infection with high diet salt and calcium, HNa for high diet salt, HCa for high diet calcium and HNC for high diet salt and calcium. Chickens were inoculated with IB virus at 1-day-old and remained on their respective diets until 21 day of age. Plasma $Na^+$, $Cl^-$, BUN, creatinine, calcium and uric acid values were examined. The results obtained were as follows ; IB virus and high dietary calcium combined treatment showed elevated plasma uric acid. BUN and creatinine values were not characteristic on chicken renal failure. But plasma uric acid values were increased according to renal lesion. Hypercalcemia and hyperuricemia did not induce urate deposition and mineralization in the kidney.

Computed tomographic assessment of retrograde urohydropropulsion in male dogs and prediction of stone composition using Hounsfield unit in dogs and cats

  • Background: Persistent uroliths after a cystotomy in dogs are a common cause of surgical failure. Objectives: This study examined the following: the success rate of retrograde urohydropropulsion in male dogs using non-enhanced computed tomography (CT), whether the CT mean beam attenuation values in Hounsfield Units (mHU) measured in vivo could predict the urolithiasis composition and whether the selected reconstruction kernel may influence the measured mHU. Methods: All dogs and cats that presented with lower urinary tract uroliths and had a non-enhanced CT preceding surgery were included. In male dogs, CT was performed after retrograde urohydropropulsion to detect the remaining urethral calculi. The percentage and location of persistent calculi were recorded. The images were reconstructed using three kernels, from smooth to ultrasharp, and the calculi mHU were measured. Results: Sixty-five patients were included in the study. The success rate of retrograde urohydropropulsion in the 45 male dogs was 55.6% and 86.7% at the first and second attempts, respectively. The predominant components of the calculi were cystine (20), struvite (15), calcium oxalate (8), and urate (7). The convolution kernel influenced the mHU values (p < 0.05). The difference in mHU regarding the calculus composition was better assessed using the smoother kernel. A mHU greater than 1,000 HU was predictive of calcium oxalate calculi. Conclusions: Non-enhanced CT is useful for controlling the success of retrograde urohydropropulsion. The mHU could allow a prediction of the calculus composition, particularly for calcium oxalate, which may help determine the therapeutic strategy.

Intrameniscal Gouty Tophus Originating from Medial and Lateral Meniscus (내측 및 외측 반월연골판에서 기시한 반월연골판 내 통풍 결절)

  • In advanced gout, monosodium urate crystals are deposited in the joints and surrounding tissues, resulting in gouty tophi. A 45-year-old man was admitted to hospital due to pain and a limited range of motion in his right knee. Magnetic resonance imaging revealed a mass, and arthroscopic surgery and biopsy were then performed. Gouty tophi invading the anterior horns of the medial and lateral menisci were identified and surgically removed. To the best of the author's knowledge, there is no case report of intrameniscal gouty tophi invading both the medial and lateral menisci. This paper presents this case along with a review of the relevant literature.

A case report of runting-stunting syndrome in a broiler farm (육계에서 발생한 흡수불량증후군 증례)

  • A broiler farm in Jeonbuk province experienced high mortality due to decreased feed intake and poor growth, and pathological evaluation of 11- and 17-day-old broilers was performed, which led to the diagnosis of runting-stunting syndrome. From the start of the rearing period to shipment, 18.4% of the chickens in the three barns experiencing continuous culling and mortality were affected, compared to 7.7% in the other five barns. Gross findings on the 11-day-old broiler chicken revealed proventricular dilatation and hemorrhage, intestinal hemorrhage, urate deposition in the pericardium and renal tubule, nephropathy, and mild hepatic capsulitis. Similar proventricular dilatation and hemorrhage were observed in the 17-day-old broiler chickens. In addition, hepatitis and pericarditis were observed with the progression of secondary bacterial infection, and pathogenic Escherichia coli was isolated from these lesions. As a result of PCR, Newcastle disease virus, fowl adenovirus, chicken anemia virus and Marek's disease virus were not detected in the all tissue samples. In contrast, infectious bronchitis virus was detected in the proventriculus, kidney and cecal tonsil. chicken astrovirus was detected in the intestine, cecal tonsil and bursa of Fabricius, and chicken parvovirus was detected in proventriculus, intestine, cecal tonsil and bursa of Fabricius. By sharing the diagnostic process of a case of malabsorption syndrome through this case report, we hope that it can be widely utilized in the diagnostic process of livestock disease pathognomonic institutions.

Modulation of Inflammation by Plant Resources (식물 자원을 활용한 염증반응 조절)

  • Chrysanthemum zawadskii (C. zawadskii) is used in traditional East Asian medicine for the treatment of various diseases, including inflammatory disease. However, it has remained unclear whether extracts of C. zawadskii inhibit inflammasome activation in macrophages. The present study assessed the inhibitory effect of an ethanol extract of C. zawadskii (CZE) on the activation of the inflammasome in macrophages and the underlying mechanism. Bone marrow[-derived macrophages (BMDMs) were obtained from wild-type C57BL/6 mice. The release of IL-1β and lactate dehydrogenase in response to nucleotide-binding oligomerization domain-like receptor (NLR) family pyrin domain containing 3 (NLRP3) inflammasome activators, such as ATP, nigericin and monosodium urate (MSU) crystals, was significantly decreased by CZE in lipopolysaccharide(LPS)-primed BMDMs. Western blotting revealed that CZE inhibited ATP-induced caspase-1 cleavage and IL-1β maturation. To investigate whether CZE inhibits the priming step of the NLRP3 inflammasome, we confirmed the role of CZE at the gene level using RT-qPCR. CZE also downregulated the gene expression of NLRP3 and pro-IL-1β as well as NF-κB activation in BMDMs in response to LPS. Apoptosis associated speck-like protein containing a caspase-recruitment domain (CARD) oligomerization and speck formation by NLRP3 inflammasome activators were suppressed by CZE. By contrast, CZE did not affect NLR family CARD domain containing protein 4 (NLRC4) or absent in melanoma 2 (AIM2) inflammasome activation in response to Salmonella typhimurium and poly(dA:dT) in LPS-primed BMDMs, respectively. The results revealed that three key components of CZE, namely linarin, 3,5-dicaffeoylquinic acid and chlorogenic acid, decreased IL-1β secretion in response to ATP, nigericin and MSU. These findings suggest that CZE effectively inhibited activation of the NLRP3 inflammasome.
