Ligusticum chuanxiong (Umbelliferae) is a perennial herb that has been used for invigoration of blood in Korean traditional medicine. It is especially important in gynecological therapy of amenorrhea and dysmenorrhea. In this study, the essential oil of L. chuanxiong was obtained by steam distillation and its main components of L. chuanxiong, Z-ligustilide and butylidene phthalide, were isolated by silica gel column chromatography. We investigated the cytotoxic effects of the essential oil fraction of L. chuanxiong and its main components on MCF-7, HeLa and SK-Hep-1 cell lines by measuring the number of surviving cancer cells after treatment through direct cell counting and MTT analysis, and by examining the morphological changes under the microscope. The essential oil from the rhizomes of L. chuanxiong and its main components showed significant cytotoxic activities for all three tested cell lines. We also observed morphological changes of shrinking and blebbing in the membranes of the three cell lines, depending on the concentration of L. chaunxiong oil or its main components.

The application of antique medical instructions, practices, skills and knowledge has been considered as the most affordable treatment in many developing countries. The use of these preparations and prescriptions over generations has made a useful and valuable guide for drug discovery in modern medicine. Medical herbs have been of a high importance for this purpose. The genus Dorema, of Apiaceae family (Umbelliferae) has a wide use in ethnobotany and traditional medicine around the world. It has been used as a treatment for CNS disease, convulsion, upper respiratory tract problems, gastrointestinal disorder and high blood sugar. Furthermore, phytochemical investigations have reported Dorema species to contain a wide range of constituents including terpenes, coumarins and phenolic compounds. The current review summarizes comprehensive information regarding botany, phytochemistry and pharmacological aspects of Dorema spp.
The root of Angelica tenuissima Nakai (Umbelliferae) has been used in traditional medicines of Korea as a headache, common cold and a fever remedy. A. tenuissima contains ferulic acid and various compounds of essential oil group such as limonene, 3-butylidenephthalide, ${\gamma}$-terpinene, neocnidilide, ligustilide, senkyunolide and neocnidilide. This study carried out to compare the contents of ferulic acid, z-ligustilide and n-butylidenephthalide between native and cultivated species of A. tenuissima by HPLC. The average contents of ferulic acid, z-ligustilide and n-butylidenephthalide indicated that native species (9 samples) were 0.060%, 0.616%, 0.025% and cultivated species (15 samples) were 0.037%, 0.141%, 0.029%, respectively. All samples were collected from different places in Korea.

A mouse whole animal bioassay was employed to screen for potential mutagenicity of ethanol/water extracts of 16 Chinese herbal drugs that are commonly prescribed in Korea. Specific cytogenetic toxicity was measrured by recording evidence of clastogenesis toxicity was measured by recording evidence of clastogenesis via the mouse bone marrow micronucleus test. Male ICR mice administered ethanol extract of Pinelliae tuber (Pinellia eternata Breitenbach, ARACEAE, 양복) and ddY female mice administered extract of Angelica Koreanae radix(Angelica Koreana Maximowicz, UMBELLIFERAE, ) (both by oral administration, at a dose of 600 mg/kg), in a short-term dosing schedule, demonstrated significant increase in micronucleated polychromatophilic erythrocytes, indicating the increase of clastogenicity.

Obesity increases the risk of mortality and morbidity because it results in hypertension, heart disease, and type 2 diabetes. Therefore, there is an urgent need for pharmacotherapeutic drugs to treat obesity. We performed a screening assay using natural products with anti-adipogenic properties in 3T3-L1 cells and determined that tschimganidine, a terpenoid from the Umbelliferae family, inhibited adipogenesis. To evaluate the anti-obesity effects of tschimganidine in vivo. Mice were fed either a normal chow diet (NFD) or a high-fat chow diet (HFD) with or without tschimganidine for 12 weeks. Treatment with tschimganidine decreased lipid accumulation and adipogenesis, accompanied by reduced expression of adipogenesis and lipid accumulation-related factors. Tschimganidine significantly increased the phosphorylation of AMP-activated protein kinase (AMPK) and decreased that of AKT. Depletion of AMPK relieved the reduction in lipid accumulation resulting from tschimganidine treatment. Moreover, tschimganidine administration drastically reduced the weight and size of both gonadal white adipose tissue (WAT) and blood glucose levels in high-fat diet-induced obese mice. We suggest that tschimganidine is a potent anti-obesity agent, which impedes adipogenesis and improves glucose homeostasis. Tschimganidine can then be evaluated for clinical application as a therapeutic agent.
Pine (Pinus densiflora) forests of 58 sites on the ridge of the Baekdudaegan, South Korea were classified by applying the phytosociological method and TWINSPAN. The floristic composition and life-form characteristics of the vegetation types analyzed and documented. The vegetation types were classified into five groups including four groups of Carex humilis var nana type and one group of Abies koreana-Sasa borealis type. Compositae was the most diverse family (26 species), Liliaceae (21 species), Rosaceae (18 species) and Betulaceae (11 species) whereas 27 families were represented by only one species. Taxonomically, Compositae was the most diverse (17 genera), Liliaceae and Rosaceae(12 genera) whereas 41 families were represented by only one genus. The Fagaceae was the most important family in total coverage (19.0) and followed Pinaceae (16.7) and Ericaceae (15.1). Five families such as Compositae, Liliaceae, Rosaceae, Betulaceae, and Umbelliferae contributed little to total coverage, although represented by numerous species. In the correlation between species richness and life forms, the species richness was highly correlated with growth and disseminule forms. In general, the correlation coefficients between life forms represented lower values than those between species richness and life-forms.
Discovery and isolation of compounds capable of blocking the interactions between VCAM-1 and VLA-4, a major pair of adhesion molecules contributing to the different steps of leukocyte migration across the endothelium in inflammatory responses, has been a major goal of this lab. Through bioactivity-guided fractionation, five saikosaponins were subsequently isolated from the methanol extracts of the roots of Bupleurum falcatum L. Their structures were elucidated by spectroscopic analysis ($^1H-$, $^{13}C$-NMR and 2D-NMR), as follows, saikosaponins: A (1); D (2); C (3); B3 (4); B4 (5). Compounds 1 and 2 inhibited interaction of sVCAM-1 and VLA-4 of THP-1 cells with respective $IC_{50}$ values of 7.8 and 1.7 ${\mu}M$. The aglycone structure of 2 also showed cell adhesion inhibitory activity with an $IC_{50}$ value of 21.1 ${\mu}M$. With these results, we suspect these two saikosaponins from the Bupleurum falcatum L. roots to be prime candidates for therapeutic strategies towards inflammation.
This review deals briefly with the various medicinal components(mainly saikosaponins), their biological activities and the variation of their contents by different cultivation environment and plant parts in Bupleurum species. Bupleuri radix, a crude drug, is the root of Bupleurum falcatum L. (Korea, Japan), B. chinense(China), and their related species (Umbelliferae). There are over 120 species in Bupleurum genus throughout world, mainly Asian area, and over 5 species in Korea, investigated up to now. These plants contain many physiological active compounds and the principal components are saikosaponins. Major activities of this crude drug and saikosaponins are the anti-inflammatory and antihepatotoxic activities. Saikosaponins and their derivatives in Bupleurum spp. have been chemically studied, isolated and identified over 70 compounds in over 50 species. Other components, physiologically active ones, also have been investigated, which are the groups of lignan, flavonoid, essential oil, polyacetylene, polysaccharide, etc. Saikosaponins belong to the group of triterpenoid saponin chemotaxonomically and occur the accumulation and turnover in plant tissues through secondary metabolism, mevalonic acid pathway. The contents and kinds of saikosaponins and other components in Bupleurum spp. plants are various due to different species and growing environments, as the plant growth characters and yield are various. Most of medicinal plants as well as Bupleurum species are very useful as agricultural products and traditional medicines, and also are very valuable as genetic resources and natural products. So we need to collect, evaluate, preserve, and utilize various medicinal plants, and also to under-stand secondary metabolism and improve the breeding and cultivation techniques for the safe production of crude drugs with high quality and yielding.

Imperatorin, a biologically active furanocoumarin from the roots of Angelica dahurica (Umbelliferae), was mutagenic and induced transformation of mouse fibroblast cell lines, whereas it provided inhibiting effects on mutagenesis and carcinogenesis induced by various carcinogens. Furthermore, it has been suggested that imperatorin may have potential anticarcinogenic effects when administered orally in the diet. In addition to its anticarcinogenic properties, imperatorin has been shown to possess anticancer activities. We investigated the macro scale gene expression analysis on the HL-60 cells treated with imperatorin. Imperatorin (10μM) were used to treat the cells for 6h, 12h, 24h, 48h, and 72h. In a human cDNAchip study of 10,000 genes evaluated 6, 12, 24, 48, 72 hours after treated with imperatorin in HL-60 cells. Hierarchical cluster against the genes which showed expression changes by more than 2 fold. Three hundred eighty six genes were grouped into 6 clusters by a hierarchical clustering algorithm. Bupleuri radix (Umbelliferae), the dried root of Bupleurum Chinense DC, has been clinically used to mitigate pain sensation. The descending pain control system consists of three major components, and modulation of pain in the periaqueductal gray is the most extensively studied descending pain control system. However, the relation of Bupleuri radix on the descending pain control system has not been clarified. In the present study, modulation of the aqueous extract of Bupleuri radix on glycine-induced ion current in the acutely dissociated periaqueductal gray neurons was investigated by using nystatin-perforated patch-clamp technique under voltage-clamp condition. In the present results, the glycine-induced ion current was significantly suppressed by 0.1 mg/ml Bupleuri radix, while treatment with $10^{-5}\;M$ naltrexone, opioid receptor antagonist, alleviated Bupleuri radix-induced inhibition on glycine-induced ion current. The present study showed that the aqueous extract of Bupleuri radix may activate descending pain control system through inhibition on glycine-induced ion current in the periaqueductal gray neurons and this effect is mediated by opioid receptors.
