정향(Clove) 추출물이 티로시나아제 유전자의 발현에 미치는 효과 (Effect of Clove Extracts on Tyrosinase Gene Expression)

  • Clove extract by methanol increased expression of the tyrosinase gene on B16 mouse melanoma cells containing tyrosinase promoter. $10{\mu}g/mL$ and $100{\mu}g/mL$ of the extract showed expression rate of the tyrosinase gene about 138% and 245%, respectively, compared with control. At $500{\mu}g/mL$, expression rate of the extract was impossible to measurement by high cytotoxicity. The solvent fraction of methylene chloride also exhibited highly expression rate as methanol extract. However, the solvent fractions of butyl alcohol and water showed repressive effect on expression of tyrosinase gene at $500{\mu}g/mL$. In MTT assay, cell survival rate of the extract exhibited similar to expression rate of tyrosinase gene. That is, $10{\mu}g/mL$ and $100{\mu}g/mL$ of the extract showed the cell survival rate about 128% and 187%, respectively.

  • Chestnut bark extract by methanol repressed the expression of tyrosinase gene of B16 mouse melanoma cell containing tyrosinase promoter. $10{\mu}g/ml{\ell}\;100{\mu}g/m{\ell}$, $1mg/m{\ell}$ of the extract repressed expression of tyrosinase gene about $38\%,\;47\%,\;and\;78\%$, respectively, compared with control. In the MTT assay, the same extract exhibited very low cytotoxicity at $1{\mu}g/m{\ell},\;10{\mu}g/m{\ell},\;100{\mu}g/m{\ell},\;and\;1mg/m{\ell}$, respectively. The fractions of Methylene chloride and ethyl acetate did not showed the repressive effect on the expression of tyrosinase gene, but the fraction of butyl alcohol repressed highly at $10{\mu}g/m{\ell}\;and\;100{\mu}g/m{\ell}$.

  • In this study, we found that the methanolic extract of Areca catechu repressed expression of the tyrosinase gene in B16 mouse melanoma cells containing a tyrosinase promoter. Extract concentrations of 100 ${\mu}$g/ml and 500 ${\mu}$g/ml exhibited tyrosinase gene expression rates of aproximately 62% and 48%, respectively, compared to the control. The fraction layers consisting of ethyl acetate, butyl alcohol, and water showed repressive effects on the tyrosinase gene. In particular, the butyl alcohol fraction highly repressed at 100 ${\mu}$g/ml and 500 ${\mu}$g/ml. In the MTT assay, the methanolic extracts exhibited very low cytotoxicities at 1 ${\mu}$g/ml, 10 ${\mu}$g/ml, and 100 ${\mu}$g/ml.

어성초(Houttuynia cordata) 추출물이 Tyrosinase유전자 발현에 미치는 효과 -연구노트- (Effect of Houttuynia cordata Extracts on Tyrosinase Gene Expression)

  • 멜라닌 색소 생성에 관여하는 tyrosinase 유전자의 발현을 억제하는 물질을 탐색하고자 tyrosinase 프로모터를 지닌 Bl6 mouse melanoma cell에 어성초 메탄올 추출물을 처리한 결과 메탄올 추출물은 10$\mu$g/mL와 100$\mu$g/mL의 농도에서 대조군에 비해서 약 13$\%$와 45$\%$의 억제효과를 나타냈으며, 세포생존율은 1$\mu$g/mL, 10$\mu$g/mL, 100$\mu$g/mL의 농도에서 약 104$\%$, 101$\%$, 74$\%$로서 세포독성이 낮게 나타났다. Butyl alcohol과 물 분획물은 tyrosinase 유전자의 발현을 억제하는 효과가 없 었지만 methylene chloride 분획물은 tyrosinase 유전자의 발현을 억제하는 효과를 나타내었으며, 특히 methylene chloride 분획물은 10$\mu$g/mL와 100$\mu$g/mL의 농도에서 각각 약 82$\%$와 48$\%$의 발현율을 나타냄으로써 대조군에 비해 tyrosinase유전자의 발현을 크게 억제하였다.

B16 Melanoma Cell에서 티로시나아제 유전자 발현에 황금(Scutellaria baicalensis) 추출물이 미치는 효과 (Effects of Scutellaria baicalensis Extracts on Tyrosinase Gene Expression in B16 Melanoma Cells)

  • 유전자 발현 조절 수준에서 멜라닌 색소의 생합성을 조절하는 천연물질을 탐색하고자 황금으로부터 유용성 물질을 추출 및 분획하여 티로시나아제 프로모터 부위를 도입하여 형질전환된 B16 melanoma cell에 처리한 결과, 그 메탄올 추출물의 티로시나아제 유전자 발현율은 $10\;{\mu}g/m{\ell}$$100\;{\mu}g/m{\ell}$의 농도에서 대조군 세포에 비해 각각 약 231과 256%로 매우 크게 증진하는 효과를 보여 주었다. 극성도가 서로 다른 용매 분획물들을 형질전환된 세포에 처리하였을 때 methylene chloride와 ethyl acetate 용매 분획물들은 $10\;{\mu}g/m{\ell}$의 농도에서, butyl alcohol 용매 분획물은 $10\;{\mu}g/m{\ell}$$100\;{\mu}g/m{\ell}$의 농도에서, 그리고 물 용매 분획물은 $500\;{\mu}g/m{\ell}$의 고농도에서 티로시나아제 유전자의 발현을 증진시키는 효과를 보여 주었다. 그러나 그들의 티로시나아제 발현 증진 효과는 메탄올 추출물보다 낮았다. 황금 메탄올 추출물을 B16 melanoma cell에 처리한 다음 MTT assay를 실시한 결과, $10\;{\mu}g/m{\ell}$$100\;{\mu}g/m{\ell}$의 농도에서는 대조군 세포와 세포활성능이 유사할 정도로 세포독성이 매우 낮게 나타났다. 그러나 그 이상의 농도에서는 세포 활성능이 현저하게 낮았으며, 특히 $1,000\;{\mu}g/m{\ell}$의 농도에서는 세포 독성이 심하여 세포의 활성능을 측정하는 것이 불가능하였다. 본 연구결과, 황금 메탄올 추출물 내에는 티로시나아제 유전자의 발현을 증진시키는 데에 관여하는 성분들이 함유되어 있는 것으로 추측된다. 따라서 황금 추출물은 멜라닌 색소의 생합성에 관여하는 유전자의 발현하는 증진시키는 목적으로 이용함이 바람직하며, 이를 산업적으로 응용하기 위해서는 앞으로도 보다 많은 연구가 필요하다.

Effect of Cordyceps militaris mycelia containing improved cordycepin on expression gene in the melanin biosynthesis pathway

  • This study aimed to verify the whitening effect of Cordyceps militaris, which is distributed in several countries worldwide, including Korea, Japan, and China, and has various medical effects. To screen the efficacy of C. militaris, the inhibitory activity of tyrosinase, which was 66% at a concentration of 1 mg/mL, was measured. Thereafter, the survival rate of melanoma cells was measured, and cell experiments were conducted at a concentration of 90% or more in which C. militaris was not toxic to cells. After measuring the inhibitory effect of TRP-1, TRP-2, tyrosinase protein, and mRNA expression, which are factors influencing melanin synthesis, C. militaris was found to decrease in all factors, with an expression level that was significantly lower compared to quercetin. This confirmed that C. militaris stimulated with LED has excellent whitening activity and can be used as a functional whitening cosmetics material.

미백산(美白散)이 멜라닌 생성 및 유전자 발현에 미치는 영향 (The Effect of Mibaeksan(MB) on Melanin Synthesis and Gene Expression)

  • Purpose: This study was performed to elucidate the inhibitory effect of Mibaeksan (MB) on melanin synthesis in B16F10 mouse melanoma cell. Methods: To demonstrate the inhibitory effects of MB on melanin synthesis, we measured the amount of released and produced melanin in B16F10 melanoma cell. Also, we evaluated tyrosinase-activity in vitro as well as in B16F10 melanoma cell. And to investigate the action mechanism, we assessed the gene expression of tyrosinase, TRP-1, TRP-2, MMP-2, PKA, $PKC{\beta}$, ERK-1 ERK-2, AKT-1 and MITF in B16F10 melanoma cells. Results: 1. MB decreased the release and production of melanin in B16F10 melanoma cells. 2. MB decreased tyrosinase activity in vitro and in B16F10 melanoma cells. 3. MB decreased the expression of tyrosinase, TRP-1, TRP-2, PKA, $PKC{\beta}$ and MMP-2 in B16F10 melanoma cells. 4. MB increased the expression of ERK-1, ERK-2 and AKT-1 in B16F10 melanoma cells. 5. MB decreased the expression of MITF in B16F10 melanoma cells. Conclusion: From these results, it may be concluded that MB has the antimelanogenetic effects.

육미지황탕가감방(六味地黃湯加減方)이 멜라닌 생성 및 유전자발현에 미치는 영향 (The Effect of Yukmijihwangtang -gagambang (YMG) on Melanin Synthesis and Gene Expression)

Inhibitory Effect of Prunus persica Flesh Extract (PPFE) on Melanogenesis through the Microphthalmia-associated Transcription Factor (MITF)-mediated Pathway

  • Novel tyrosinase inhibitors are important for pigmentation in the skin. Following extraction of tyrosinase inhibitors from edible vegetables or fruits, we found that the Prunus persica flesh extract (PPFE) exhibited potential inhibitory activity for melanogenesis. PPFE showed tyrosinase inhibitory activity in an enzymatic assay and PPFE also significantly inhibited the melanin formation in cultured mouse melan-a cells. Moreover, real-time RT-PCR analysis revealed that the inhibition of melanin production by PPFE was closely related to marked suppression of mRNA expression of tyrosinase and tyrosinase-related protein-1 and -2 (TRP-1 and TRP-2) in melan-a cells. Further investigation found that the modulation of tyrosinase expression by PPFE was associated with the transcriptional regulation of the microphthalmia-associated transcription factor (MITF). PPFE inhibited the promoter activity of MITF and suppressed MITF mRNA expression in melan-a cells. These results indicate that PPFE down-regulates melanogenesis-associated gene expression through MITF-mediated transcriptional regulation and these events might be related to the hypopigmentary effects of PPFE.

독활기생탕(獨活寄生湯)이 멜라닌 생성억제 및 유전자 발현에 미치는 영향 (Effects of Dokhwalkisaeng-tang on Melanin Synthesis Inhibition and Gene Expression in B16F10 Melanoma Cells)

  • The aim of this study was to elucidate the antimelanogenic effect of Dokhwalkisaeng-tang(Duohujisheng-tang) in B16F10 melanoma cells. Dokhwalkisaeng-tang(DKT) was used to develop the effective prescription of inhibition of melanin production. We determined inhibitory effects of DKT on melanin-release, melanin production, and tyrosinase activity in B16F10 melanoma cells. And to explicate the action-mechanism of DKT, melanin-related gene expressions were determined using RT-PCR and real time RT PCR technique in B16F10 melanoma cells. DKT inhibited melanin-release, melanin production in B16F10 melanoma cells considerably. DKT inhibited tyrosinase activity in vitro and in B16F10 melanoma cells. DKT inhibited the expression of tyrosinase, TRP-1, TRP-2 in B16F10 melanoma cells. DKT inhibited the expression of PKA, PKC, MMP-2 and MITF in B16F10 melanoma cells. On the other hand, DKT increased the expression of ERK-1, ERK-2, AKT-1 in B16F10 melanoma cells. From these results, we propose that DKT may have effect on the antimelanogenesis.
