Experimental studies about the inhibitory effect on tyrosinase and elastase activities by various herb medicines (수종(數種)의 한약재의 Tyrosinase와 Elastase 활성 억제 효과에 대한 실험적 연구)

  • Objectives : The purpose of this study is to investigate whitening effects and anti-wrinkle effects of a few 80% ethanol extracted herbal medicines. Methods : In the first study, a few 80% ethanol extracted herbal medicines were screened for their inhibitory activities against the tyrosinase. In the second study, a few 80% ethanol extracted herbal medicines were screened for their inhibitory activities against elastase. Results : 1. We showed 28%, 27% and 19% inhibitions of mushroom tyrosinase at 500 $\mu$g/ml concentration of ASR, AIF and ABR extracts and they were showed higher anti-tyrosinase activity than arbutin's. We also could observe that the decreased mushroom tyrosinase activities in RR, CML, LR, AGR and TH extracts. 2. RR, AF and ABR (final concentrstion 1 mg/ml) were appeared 60%, 98%, 83% of inhibitions of elastase activity, and they were showed higher anti-elastase activity than that of ursolic acid. We also could observe that the decreased elastase activities in AIF, AR, LR and CML extracts. Conclusions : These results suggest that ASR, AIF and ABR extracts contribute to the anti-melanin activities and represent potential sources of whitening agent, and RR, AF and ABR extracts contribute to the anti-elastase activities and represent potential sources of anti-wrinkle agent. These results suggest that some herbal medicines could be strong potential sources of inhibition about anti-aging and whitening effects for the skin.

Antioxidative Activities and Inhibitory Effects on Tyrosinase and Elastase by Water Extracts of Allium sp. (Allium 속 식물 물 추출물의 항산화 활성과 Tyrosinase 및 Elastase 저해 효과)

  • Three different Allium plants, including wild garlic (Allium monanthum Max; AMM), onion (Allium cepa L.; ACL) and green onion (Allium fistulosum L; AFL), have been widely used as spicy food in daily life. This study was attempted to investigate the properties of antioxidant, whitening and anti-wrinkle activities in water extracts in Allium plants. The antioxidant, whitening and anti-Wrinkle activities were evaluated by performing DPPH and ABTS radical scavenging activities, inhibitory effects on tyrosinase, and elastase, respectively. The water extract (10 mg/mL) of onion, wild garlic and green onion exhibited DPPH radical scavenging activities of 28.46%, 28.45%, and 15.91%, respectively. The inhibitory effects of tyrosinase activities by wild garlic, green onion, and onion were increased by heating the extracts at $90^{\circ}C$. Additionally, the wild garlic extract showed higher elastase inhibitory activity than those of other plant extracts. These results suggest that water extracts of Allium plants may be useful as potential sources of beauty foods due to higher antioxidant, anti-melanin and anti-wrinkle activities.

Effects of Paeoniae radix alba(PRA) on Skin whitening and Elasticity using Melanoma cells (백작약(白芍藥)이 피부 미백 및 주름에 미치는 영향)

  • Objective : Paeoniae radix alba(PRA) can enrich the blood and regulate menstruation, astringe yin and arrest sweating, calm the liver and arrest pain. This study was designed to investigate effects of PRA on skin whitening and elasticity using melanoma cells. Methods : In this experiment, effect of PRA on cell viability, inhibition of melanin synthesis and inhibitory effect on tyrosinase and elastase. Results : 1. More than $1,000\;{\mu}g/ml$ of PRA treated group showed lowered proliferation rates significantly compared to non-treated control group. 2. All of treated groups were lower levels of melanin synthesis respectively. 3. PRA did not show inhibitory effect on tyrosinase activities in vitro. But, PRA suppressed tyrosinase activities in B16F10 cells significantly. 4. PRA suppressed elastse type 1 activities in dose-dependent manner in vitro. But, PRA slightly suppressed elastase type 4 activities in vitro, and PRA also slightly suppressed elastase activities in vivo. Conclusion : These results suggest that PRA can inhibit melanin synthesis through ihhibitory action on tyrosinase activity and inhibt elastase activity, and also suggest that these results can be used for the study on maintaining skin whitening or elasticity.

Physiological Effects of Formulations Added with Black Garlic Extract on Skin Care: Oxidative Stress, Tyrosinase and Elastase Activities (흑마늘 추출물을 첨가한 화장품의 기능성 평가: Tyrosinase와 Elastase 저해 활성 및 산화 스트레스에 미치는 영향 중심으로)

  • We evaluated the physiological effects of formulations added with black garlic extract on skin care. Black garlic was made by spontaneous fermentation for 40 days at $60{\sim}70^{\circ}C$ and 85~95% RH without any additives. When black garlic extract was added to formulation, the tyrosinase activity was decreased significantly (p<0.05, p<0.01, p<0.001). Base formulation inhibited slightly elastase activity (<5%), while black garlic formulation had about 8-fold higher elastase inhibitory activity (p<0.01). It was observed that formulations, with or without black garlic extract, decreased thiobarbituric acid reactive substances (TBARS) and hydrogen peroxide which were increased by UVB irradiation, although there were no significant differences of these contents between black garlic formulation and base formulation. In conclusion, black garlic formulation had high inhibitory activities for tyrosinase and elastase, suggesting that black garlic may have beneficial properties as a material for cosmeceuticals.

Physiological Activities of Hot Water Extracts from Ecklonia cava Kjellman (감태 열수 추출물의 생리활성)

  • The biological activity of hot water extract from Ecklonia cava Kjellman (ECE) was investigated to assess antioxidative, anti-skin aging, and nitrite scavenging abilities, as well as alcohol metabolizing activities. Antioxidant activity of ECE was measured by using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD)-like activity. DPPH radical scavenging activity and SOD-like activity of ECE increased in a remarkably dose-dependent manner, and were about 91.4% and 75% at 1 mg/ml, respectively. The xanthine oxidase inhibitory activity was indicated to be about 70% at 1 mg/ml of ECE. Nitrite scavenging ability of ECE showed to be 93.6% at 1 mg/ml and pH 1.2. The influence of ECE on alcohol metabolism was demonstrated through the generating activity of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). The facilitating rate of ADH and ALDH activity by ECE was 167.2% and 334% at 10 mg/ml, respectively. In addition, tyrosinase and elastase inhibitory activities of ECE were 58% and 72% at 10 mg/ml, respectively. These results indicated that ECE has valuable biological attributes owing to its antioxidant, nitrite scavenging, alcohol metabolizing, and elastase and tyrosinase inhibitory activities.

Inhibitory Effect of Elastase and Tyrosinase of Ginsenoside $F_1$ Isolated from Panax ginseng Leaves (인삼잎으로부터 분리된 진세노사이드 $F_1$의 Elastase 및 Tyrosinase 억제 효과)

  • This study was carried to establish a simple isolation and purification method of ginsenoside $F_1$ from leaves of Panax ginseng and was to evaluate the inhibitory effect of purified ginsenoside $F_1$ on the activities of elastase and tyrosinase. The content of ginsenoside $F_1$ was 90-fold higher in leaves than in root of ginseng. Ginsenoside $F_1$ was isolated from EtOAc fraction between EtOAc and alkalized water of 80% EtOH extract after remove of hydrophobic components. The 50% inhibitory concentration ($IC_{50}$) of ginsenoside $F_1$ on elastase activity and tyrosinase activity was 1.07 mM and 1.81 mM, respectively. Especially, inhibitory effect of ginsenoside $F_1$ on tyrosinase activity was higher than that of arbutin ($IC_{50}$; 2.20 mM). These results indicate that ginsenoside $F_1$ have a potential for industrial cosmetic materials.

The effect of Elastase inhibition and Tyrosinase by Rheum undulatum Stokes extracts (대황(大黃)의 Elastase 활성 억제와 Tyrosinase 억제연구)

  • Objective : Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search new materials from the traditional herbal medicines which inhibit the aging process of skin in vitro. Methods : herbal medicines(80% ethanol or water extracts) were screened for their inhibitory activities against elastase. Results : Rheum undulatum (final concentrstion 1 mg/ml) appeared over 30% of inhibition of elastase activity. so we are investigated anti wrinkle effects of Rheum undulatum look through MMP-1 inhibition activity. also Extracts of Rheum undulatum showed higher anti tyrosinase activity than arbutin (final concentration 1 mg/ml). Conclusion : These results suggest that herbal medicines could be strong potential sources of inhibition of anti-aging and whitening effects for the skin.

Antioxidant and Antibacterial Activities, and Tyrosinase and Elastase Inhibitory Effect of Fermented Omija (Schizandra chinensis Baillon.) Beverage (오미자 발효액의 항산화 및 항균 활성과 미용효과)

  • This study was aimed to investigate the contents of chemical components and the biological activity of fermented Omija (Schizandra chinensis Baillon.) beverage (FOB), which have not been well known up to now. General compositions and the contents of mineral and vitamin of FOB were lower than Omija before fermentation (OBF). In addition, most sucrose was digested into glucose and fructose in FOB. This suggests that the beneficial contents may be made from general compositions by fermentation, though more evidences are needed. Antioxidant activity of FOB was measured by using 2,2-diphenyl-l-picryl-hydrazyl (DPPH) free radical scavenging activity and superoxide dismutase-like activity (SODA). DPPH radical scavenging activity and SODA were increased in a dose-dependent manner, and were about 79.7% and 60.8% at four times diluted FOB (25% of FOB), respectively. The FOB also showed strong antibacterial activity on Escherichia coli. Beauty effect of FOB was demonstrated by the analysis of tyrosinase and elastase inhibitory activities, and they were remarkably increased in a dose-dependent manner. Tyrosinase inhibitory activity was about 82% at two times diluted FOB (50% of FOB) and elastase inhibitory activity was 78.2% at five times diluted FOB (20% of FOB). These results suggest that FOB has a strong antioxidant activity, and tyrosinase and elastase inhibitory effects.

α-Glucosidase, Tyrosinase, and Elastase Inhibitory Effects of Enzymatic Extracts from Ecklonia cava and its Alcohol Metabolizing Activity (감태(E. cava Kjellman) 효소분해산물의 항당뇨 및 알코올 분해능과 미용효과)

  • Microbulbifer sp. was used to acquire the degrading products from Ecklonia cava (DPEC) and the products were investigated to determine the physiological activities. Firstly, 2,2-diphenyl-1-picrylhydrazyl (DPPH) activity and superoxide dismutase (SOD) assay were about 84.1% and 89.6% at 2.5 mg/ml, respectively. In addition, nitrite scavenging ability was shown to be 56.3% at 0.5 mg/ml on pH 1.2. ${\alpha}$-Glucosidase inhibitory activity was increased in a dose-dependent manner and was about 58.7% at 2.5 mg/ml. To determine the influence of DPEC on alcohol metabolism, the generating activity of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) were measured. Facilitating rates of ADH and ALDH activities by DPEC were 123.3% and 215.2% at 2.5 mg/ml, respectively. For analyses of anti-wrinkling and whitening effects, its elastase and tyrosinase inhibitory activities were measured and were about 73.1% and 42.2% at 2.5 mg/ml, respectively. These results indicated that DPEC has valuable biological attributes owing to its antioxidant, nitrite scavenging, and alcohol metabolizing activities and ${\alpha}$-glucosidase, elastase, and tyrosinase inhibitory activities.
