사료자원으로서 Chufa (Gyperus esculentus L.)의 생장특성과 사료가치 (Growth Characteristics and Nutritive Value of Chufa (Cyperus esculentus L.) for Forage Resource)

  • Chufa(Cyperus esculentus L.)는 고온성 사초과 식물의 일종으로 여름기간에 잘 자란다. 지상부는 주로 잎으로 구성되어 있으며 지하부는 수염뿌리가 뭉쳐 나있고 끝에 괴경이 달린다. 파종 당년에는 출수가 되지 않고 괴경을 많이 생산한다. 번식은 괴경에서 분얼이 많이 나오는 영양번식을 하며 이 분얼이 다발모양(bunch type)의 포기를 형성한다. 성숙기의 초고는 73~75cm 전후이며 7월 중순에 거의 성장이 완료된다. 포기당 분얼의 수는 7월말까지 급격히 증가하고 8월 이후는 서서히 증가하지만 생장이 빈약하였다. 지상부의 최종 생초수량은 40.3ton/ha, 건물수량은 12.2ton/ha이다. 재생력은 생장초기에는 강하지만 2회 예취후는 재생력이 급격히 떨어진다. 무예취구에서 최종 수확기의 포기당 괴경의 수는 722개, 1$m^2$ 당 생체량은 4.2kg, 건물량은 1.9kg이었다. 예취구의 경우 괴경의 생산량은 예취시기가 늦어질수록 감소하였으나 8월 이후에는 이미 괴경생산이 상당히 진행된 것으로 보였다. chufa의 영양생장기 지상부 사료가치는 우수하였으나 생장이 진행될수록 낮아졌다. 최종 수확한 지상부의 조단백질, NDF, ADF, DMD, TDN 함량은 각각 6.1%, 81.5%, 39.8%, 33.2%39.40%, 괴경의 영양성분은 각각 6.0%, 68.3%, 15.5%, 50%, 51.8%였으며, 특히 지방 함량은 16.2%로 높았다.

  • 단마의 종근부위에 따른 수량성을 밝혀 적정 종근부위를 확인하고 새로운 지주설치 방법이 단마와 장마의 수량 및 생육에 미치는 영향을 구명하여 다수확 재배기술을 확립코자 수행한 실험의 결과를 요약하면 다음과 같다. 1. 출현기는 동부과 미부가 노두나 두부에 비하여 15일 정도 늦고, 출현율은 동부에서 다소 낮은 경향이였다. 2. 종근부위별 지상부의 생육은 두부가 가장 왕성하였고 지하부의 생육은 두부와 미부가 가장 왕성하였다. 3. 10a당 수량은 노두에 비하여 두부와 미부가 각각 16, 15% 증수를 보였으나 동부는 8%의 증수를 보였다. 4. 개화기와 영여자착생기는 모든지주설치 방법에서 비슷하였으나 지상부 생체중은 관행지주법에서 보다 네트를 이용한 ${\wedge}$, I형 지주에서 높았다. 5. 단마는 지주형에 따라 큰 수량의 차이를 보이지 않았으나 장마는 ${\wedge}$, I형 지주에서 각각 11.8%의 증수를 보였다. 6. 경영비를 고려할 때 네트지주형은 I형이 적합하였다.

환상 및 선상감자 걀쪽바이로이드 RNA분자의 전기영동적 분리를 위한 효과적인 조건에 관한 연구 (Determining an Effective Electrophoretic Gel System for Separation of the Circular and Linear Potato Spindle Tuber Viroid RNA Molecules)

  • 감자 걀쪽바이로이드(PSTV) RNA분자를 포함한 저분자량의 식물성 RNA분자에 대해서 0M부터 8M까지의 요소농도구배를 가진 폴리아크릴아마이드겔을 이용하여 1/40, 1/10로 희석한 완충액조건과 $17^{\circ}C,\;37^{\circ}C,\;57^{\circ}C$의 온도조건에서 전기영동을 실시하였다. 바이로이드의 RNA분자가 환상과 선상의 두가지 분자로 나뉘어지는 변성조건에서는 1/40로 희석한 TBE완충액을 포함하는 요소농도구배겔을 $57^{\circ}$에서 전기영동을 실시하는 것이 효과적이었다. 변성된 환상의 바이로이드분자에 대한 전기 영동적 이동성은 주로 요소의 농도에 따라 영향을 받는 것으로 보이는데, 이와 더불어 저 농도의 TBE완충액이 요소농도구배겔에서 환상과 선상바이로이드분자 사이에서 전기영동적 분리를 증가시켜 주었다.

Effects of Vermicompost on Growth of Fall-Cropping Potato in Volcanic Ash Soil

  • Recently, with increasing concern for sustainable agriculture and safe agricultural products, organic farming has become widely adapted as an alternative to conventional farming. This study was conducted to investigate the effects of earthworm casts (EWC) with $100\%$ organic compost on the growth and yield characteristics of fall-cropping potato (Solanum tuberosum L. cv. Dejima) in Jeju. The treatments consisted of seven plots: 2-, 4-, 6-, 8-, and 10-ton $ha^{-1}$ EWC-treated plots, 1.2-ton $ha^{-1}$ complex fertilizer $(N-P_2O_5-K_2O,\;10-10-14)-for-potato$ (CEP)-treated plot as conventional practices, and a control plot. The plant heights were greater in the plots where EWC and CFP were applied than in the control plot. Tuber diameter, number of stems per plant, and chlorophyll level tended to increase in the plots where 8-10 tons $ha^{-1}$ of EWC were applied. The application of CFP and EWC showed an increment in the average tuber weight per plant, but there was minimal significant difference. The application of 8-10 tons $ha^{-1}$ of EWC resulted in an increase in the total tuber yield (21.61-21.87 tons $ha^{-1}$) as compared to the other plots. The highest yield of market able tubers was $69.8\%$ of the total yield from the 10-ton $ha^{-1}$ EWC-treated plot. Consequently, with regard to the growth and yield characteristics of fall-cropping potato, the effects of EWC application were more favorable than the effects of the application of a chemical fertilizer.

기내증식 대반하의 상토 조성별 괴경 비대 조건 비교를 통한 최적 배양묘 생산 조건 확립 (Optimized Production through Enlargement Comparison Grown in Various Mixed Soils using Tubers of In vitro Pinellia triparita(Blume) Schott)

  • 대반하[大半夏: Pinellia tripartita(Blume) Schott]는 천남성과에 속하는 다년생 초본식물로써 중국, 일본, 우리나라를 포함한 아시아 전역에 분포하며 꽃과 전초가 아름다워 최근 원예용으로도 많이 이용되고 있으나 체계적인 대량번식 체계가 구축되어 있지 않다. 본 연구에서는 조직배양을 통해 증식된 대반하 기내 식물의 토양조성별 괴경 생육을 조사하여 비교하였다. 기내 증식된 대반하 괴경의 크기에 따라 직경이 1cm 이상인 경우 Type I, 1cm 이하를 Type II로 나누어 6종의 조합상토에 파종하여 괴경의 비대와 생육을 조사하였다. 괴경의 비대와 전초의 생육변화를 확인하기 위하여 초장, 잎 수, 마른 잎 수, 괴경 수, 괴경 크기, 생체중 및 건물중을 8주간 변화를 측정한 결과, Type I와 II 모두 코이어 68.0%, 피트모스 14.7%, 펄라이트 3.0%, 버미큘라이트 7.0% 및 제오라이트 7.0%로 구성된 조합상토 B에서 가장 우수한 생육을 보였다. 특히 Type I은 조합상토 B에서 괴경 크기와 생체중이 각각 45%와 101%를 보였으며, 이는 생육이 가장 좋지 못했던 처리구인 조합상토 E(코이어 14.3%, 피트모스 14.3%, 펄라이트 42.9%, 버미큘라이트 14.3% 및 제오라이트 14.3%)와 약 2배의 차이를 보였다. 이러한 연구결과는 종자번식이 어려운 대반하의 기내 대량증식을 통한 육묘에 있어 안정적인 토양 순화와 대규모 배양묘 생산을 위한 중요한 기초자료로 활용할 수 있을 것으로 사료된다.

Relationship between Planting Material Conditions and Major Agronomic Characters in Cassava

  • This study was carried out to investigate the effect of stem-cutting (SC) length and portion on the major agronomic characters related with dry matter production and accumulation, and to examine their relation­ships in cassava. When cassava was planted with longer SCs or with older portion SCs, major canopy or source characters like leaf number and leaf area index developed excessively, while tuber yields could be reduced due to the decrease of root/shoot ratio, relative growth rate, root dry weight, and harvest index, particularly in the bitter varieties (high cyanide-level varieties). It was considered that the sweet varieties (low cyanide-level varieties) be early­bulked with higher tuber yield when they are planted with the 25-30 cm long SCs or with the SCs from young type to semi-mature portions of mother stems, while the bitter varieties with the 15-20 cm long SCs or with the SCs from semi-mature to hardwood portions of mother stems, respectively. However, a significant interaction between length and portion of SC was not observed in all agronomic characters.

Endophytic Bacteria Improve Root Traits, Biomass and Yield of Helianthus tuberosus L. under Normal and Deficit Water Conditions

  • Drought is more concerned to be a huge problem for agriculture as it affects plant growth and yield. Endophytic bacteria act as plant growth promoting bacteria that have roles for improving plant growth under stress conditions. The properties of four strains of endophytic bacteria were determined under water deficit medium with 20% polyethylene glycol. Bacillus aquimaris strain 3.13 showed high 1-aminocyclopropane-1-carboxylate (ACC) deaminase production; Micrococcus luteus strain 4.43 produced indole acetic acid (IAA). Exopolysaccharide production was high in Bacillus methylotrophicus strain 5.18 while Bacillus sp. strain 5.2 did not show major properties for drought response. Inoculation of endophytic bacteria into plants, strain 3.13 and 4.43 increased height, shoot and root weight, root length, root diameter, root volume, root area and root surface of Jerusalem artichoke grown under water limitation, clearly shown in water supply at 1/3 of available water. These increases were caused by bacteria ACC deaminase and IAA production; moreover, strain 4.43 boosted leaf area and chlorophyll levels, leading to increased photosynthesis under drought at 60 days of planting. The harvest index was high in the treatment with strain 4.43 and 3.13 under 1/3 of available water, promoting tuber numbers and tuber weight. Inulin content was unchanged in the control between well-watered and drought conditions. In comparison, inulin levels were higher in the endophytic bacteria treatment under both conditions, although yields dipped under drought. Thus, the endophytic bacteria promoted in plant growth and yield under drought; they had outstanding function in the enhancement of inulin content under well-watered condition.

Curing period affect the occurrence of black dot, weight loss and sprouting of potato tubers during storage

  • A potato cultivar, Haryeong, was released for table stock in 2005 and has been supplied to farmers since 2010. Black dot has been known as a disease causing tuber blemishes in the cultivar, which is primarily an issue in storage. To investigate the effect of curing periods on disease occurrence, four periods (1, 3, 6 and 9 weeks) of curing were applied on Haryeong tubers harvested from the highlands (800 m a.s.l) in September. Tubers were stored at $2-3^{\circ}C$ and 80-90% RH for 6-7 months and were visually checked for disease occurrence. Tuber infection was characterized by barely visible small black spots on tubers and dark sooty lesions in stolon end of tubers. Another symptoms were observed on the upper surface of tubers which showed irregular, small to large, and depressed areas of brown to blackish skin with necrotic lesions inside. Results showed decrease in black dot depending on the curing period that ranges 18-48, 3-35, 0-14 and 0-3% at 1, 3, 6 and 9 weeks curing, respectively. During the storage, however, percentage weight loss of tubers and percentage of tubers with sprouts increased slightly with increasing the curing period from 1-3 to 6-9 weeks.

Chemopreventive Effect of Amorphophallus campanulatus (Roxb.) blume tuber against aberrant crypt foci and cell proliferation in 1, 2-dimethylhydrazine induced colon carcinogenesis

  • Colorectal cancer is one of the leading causes of cancer death, both in men and women. This study investigated the effects of Amorphophallus campanulatus tuber methanolic extract (ACME) on aberrant crypt foci (ACF) formation, colonic cell proliferation, lipid peroxidative damage and the antioxidant status in a long term preclinical model of 1, 2-dimethylhydrazine (DMH) induced colon carcinogenesis in rats. Male Wistar rats were divided into six groups, viz., group I rats served as controls; group II rats treated as drug controls receiving 250 mg/kg body weight of ACME orally; group III rats received DMH (20 mg/kg body weight) subcutaneously once a week for the first 15 weeks; groups IV, V and VI rats received ACME along with DMH during the initiation, post-initiation stages and the entire period of the study, respectively. All the rats were sacrificed at the end of 30 weeks and the intestinal and colonic tissues from different groups were subjected to biochemical and histological studies. Administration of DMH resulted in significant ($p{\leq}0.05$) intestinal and colonic lipid peroxidation (MDA) and reduction of antioxidants such as catalase, glutathione peroxidase, glutathione reductase, glutathione-Stransferase and reduced glutathione. Whereas the supplementation of ACME significantly ($p{\leq}0.05$) improved the intestinal and colonic MDA and reduced glutathione levels and the activities of antioxidant enzymes in DMH intoxicated rats. ACME administration also significantly suppressed the formation and multiplicity of ACF. In addition, the DMH administered rats showed amplified expression of PCNA in the colon and decreased expression of this proliferative marker was clearly noted with initiation, post-initiation and entire period of ACME treatment regimens. These results indicate that ACME could exert a significant chemopreventive effect on colon carcinogenesis induced by DMH.
