  • A macromolecular fraction from fresh ginseng root containing mainly polysaccharide showed mild mitogenic activity in human cord blood lymphocytes. 2) When lymphocytes were transformed by Con A or PHA, this fraction could greatly enhance the activity of these lectin mitogens, thus showing a potentiation effect. 3) Although this macromolerular fraction contains saponin and is susceptible to trypsin digestion, it is probably a peptido-glycan in nature on account of its important carbohydrate content and thermal stability. 4) This fraction could not support cancer cell (EAT, K562) growth : its inhibitory effect on these cells remains to be explored.

  • Hypocholesterolemic peptide isolated from glycimin (11S protein) hydrolyzate by trypsin was purified and identified as LPYP and IAVPGEVA. To investigate the effects of phyiscal properties of side chains of the hypocholesterolemic activity, some of mutant peptides were designed and synthesized chemically. The structure related structures of each peptide were simulated and constructed and their conformations were observed by using spectropolarimeter. The hypocholesterolemic activities were monitored by assaying the inhibition of 3-hydroxy-3-methylglutaryl CoA reductase (HMG-CoA reductase) in vitro and by the determination of cholesterol content in mice serum. For LPYP derivatives, Hypocholesterolemic activity was lost when hydrophobic leucine residue at N-terminus was not so critical for maintaining hypocholesterolemic activity. For idealogical design of hypocholesterolemic peptides, the structure of HMG-CoA reductase are shown and inhibition mechanism of some peptides or inhibitors will be presented. For IAVPGEVA derivative inhibition of HMG-CoA reductase has been studied. For detail study of hypocholesterolemic activity, kinetic study of inhibition of peptides on HMG-CoA reductase and structural view of ligand binding should be investigated.

Levamisole이 양식뱀장어의 면역조절작용 및 항균효과에 미치는 영향 (In vivo, Immunomodulatory and Antibacterial Reaction of Levamisole in Cultured Eel, Anguilla japonica)

  • This experiment was carried out in order to evaluate the immunomodulatory activity of levamisole (LMS) in 5. fgrjn challenged eels with different treatment regimens: 7-day LMS treatment before the challenge, 7-day LMS treatment started simultaneously with the challenge, 14-day treatment before and after the challenge. The antibacterial effect was activated in all treated groups, with the best being obtained in the simultaneously treated group. LMS stimulated the defense mechanisms of the eel as demonstrated by increase in the level of total protein, albumin, trypsin inhibitor capacity, lysozyme activity, antibody titers antibacterial activity and survival rate. These results suggest that antibacterial effects of LMS was achieved by not only non-specific immune response but also specific one in eel.

Effects of Supplementing Different Levels of a Commercial Enzyme Complex on Performance, Nutrient Availability, Enzyme Activity and Gut Morphology of Broilers

  • A trial was conducted to study the influence of different levels of a commercial enzyme complex on performance, nutrient availability, blood parameters, digestive tract measurements, amylase and trypsin activity of the digestive tract and gut morphology in broilers fed the typical diets in north China. There were four treatments: the control diet and the other three enzyme complex supplemented diets which were 180 mg/kg, 360 mg/kg and 720 mg/kg enzyme complex supplemented to the control diet, respectively. The birds fed the diets supplemented with 180 mg/kg and 360 mg/kg enzyme complex had better performance and nutrient availability, the activities of amylase and trypsin in the digestive tract in the two treatments were improved, the villus height and surface area of villus in the small intestine increased and the crypt depth and epithelial thickness of small intestine decreased. Relative weights of pancreas and relative weights and lengths of small intestine decreased. However, the addition of 720 mg/kg enzyme complex had no effects on these parameters and increased crypt depth and epithelial thickness of the small intestine. The data suggested that suitable supplementation of enzyme complex was beneficial for the birds, while excess enzyme complex inhibited secretion of endogenous enzyme and destroyed the structure of the small intestine.

  • We compared the nutritional requirements of whole larvae of the black seabream Acanthopagrus schlegeli, sweet fish Plecoglossus altivelis, olive flounder Paralichthys olivaeus and rock fish Sebastes schlegeli. The larvae were 20, 30, 14 and 5 DAH (or spawning) of black seabream, sweet fish, olive flounder and rock fish, respectively. Specific ${\alpha}$-amylase activity (mU/mg protein) was highest (8,324.9 mU/mg protein) in rock fish larvae (P<0.05). Specific trypsin-like protease activity was highest (11,330.1 mU/mg protein) in black seabream larvae (P<0.05), which also exhibited the highest activity, 685.5 mU/mg dry weight (P<0.05). The specific activities per mg protein and mg dry weight of black seabream were the highest (187.4 mU/mg protein and 11.3 mU/mg dry weight, respectively) (P<0.05). A/P, P/L and A/L ratios of rock fish were 1.47, 90.3 and 133.1, respectively (P<0.05). We present here basic larval digestive enzymatic nutritional requirement data.

Dephosphorylation of Phytate from Rice Bran and Soybean Meal Using Phytases from Aspergillus sp. 5990

  • Total phosphorus contents in rice bran and soybean meal were determined to be 5.81 and $2.77\%$, respectively, and $97.2\%$ of phosphorus in rice bran and $66.4\%$ in soybean meal were presented as phytate phosphorus. Optimum pH condition for hydrolysis of phytate in rice bran and soybean was determined to be in the pH range of 3.7 and 5.3. The highest activity of phytase for hydrolysis of phytate in both samples was determined to be at $55^{\circ}C$ for rice bran and $55-60^{\circ}C$ for soybean. Hydrolysis of phytate in soybean meal at pH 5.0 increased with the co-reaction or consecutive reaction with protease; however, in rice bran hydrolysis decreased with co-reaction with protease. Phytate degradation of soybean meal in the presence of pepsin at pH 2.5 showed higher than that of rice bran. Phytate degradation of rice bran in the presence of trypsin or pancreatin at pH 7.0 increased the activity around 2-times compared with the activity in the absence of trypsin or pancreatin. The results of this study suggest that hydrolysis of phytate in rice bran or soybean meal with phytase and protease may provide an alternative process for the preparation of aquacultural feed with a low level of organic phosphorus.

Effects of Different Levels of Supplementary Alpha-amylase on Digestive Enzyme Activities and Pancreatic Amylase mRNA Expression of Young Broilers

  • Four hundred and forty 1-day-old Arbor Acre broilers were fed commercial starter diets with 0, 250, 750 and 2,250 mg/kg of an alpha-amylase preparation from 1 to 21 days of age to investigate the effects of an exogenous enzyme on growth performance, activities of digestive enzymes in the pancreas and anterior intestinal contents and pancreatic amylase mRNA expression. Body weight gain (BWG) and average daily gain (ADG) increased linearly (p<0.01) with increasing levels of supplementary amylase but feed conversion ratio (FCR) was not affected. There was a negative quadratic change of protease and amylase in the small intestinal contents with the increase of supplementary amylase level. The activity of intestinal trypsin was also increased (p<0.05). Lipase was unaffected by amylase supplementation (p>0.05). The pancreatic protease, trypsin, and lipase were not affected by exogenous amylase levels. Consistent with the tendency for a linear depression of amylase activity, pancreatic ${\alpha}$-amylase mRNA was down-regulated by dietary amylase supplementation. The present study suggested that oral administration of exogenous amylase affected activities of intestinal enzymes and the production of pancreatic digestive enzymes in a dose-dependent manner.

Bacillus subtilis cx1이 생산하는 박테리오신의 특성 (Characterization of Bacteriocin from Bacillus subtilis cx 1)

  • B. subtilis cx1이 생산하는 박테리오신(BSCX1)을 부분 정제하고 특성을 규명하였다. BSCX1은 pH 안정성 실험에서 pH 2.5~9.5 구간에서도 안정되게 항균활성을 유지하였다. 각종 효소에 대한 안정성 실험에서는 protease, trypsin, proteinase K, 그리고 carboxypeptidase로 처리하였을 때는 완전히 항균활성이 사라졌고, $\alpha$-amylase, lipase, aminopeptidase는 항균활성에 영향을 미치지 못하였다. 이상의 결과로 본 항균물질이 단백질계열의 물질이며 당이나 지질 결합은 항균활성에 큰 영향을 미치지 않는 것으로 추정되었다. 그러나 열에는 불안정하여 $60^{\circ}C$이상에서는 15분 안에 완전히 그 항균활성을 상실하였으며 $50^{\circ}C$ 에서는 15분만에 역가의 50%가 실활 되었다. 이에 반하여 저온에서는 무척 안정하여 $-20^{\circ}C$$-70^{\circ}C$에서 수개월간 보관하여도 여전히 항균활성을 그대로 유지하였다. Tricin-SDS-PAGE를 통하여 BSCX1의 분자량은 약 9,500 dalton으로 확인되었ekl. BSCX1은 pH2.5에서 9.5에 이르는 넓은 pH 영역에서 그 활성을 유지하므로, 기존에 개발된 nisin이나 유산균 유래의 항균물질이 pH 안정성이 떨어진다는 단점을 보완할 수 있는 생물학적 식품보존제로서의 개발 가능성을 높이 시사하였다.

Lipoxygenase 결핍 콩의 이화학적 특성 (Physicochemical Characteristics of Lipoxygenase - Deficient Soybeans)

  • 본 실험은 lipoxygenase 결핍 콩인 진품콩(수원169호; lipoxygenase-2, 3 결핍)과 진품콩 2호(수원177호; lipoxygenase-1, 2, 3 결핍) 및 lipoxygenase 활성이 큰 황금콩에 대한 일반적인 종자 특성과 종자의 저장성을 살펴봄으로서 lipoxygenase 결핍 콩의 식품학적 이용과 이들의 품종 개발을 위한 기초 자료를 제공하고자 시도하였다. 실험에 사용된 콩은 모두 노란 색 종피로 중콩에 속하는 것이었으며 일반 성분분석시 진품콩 2호의 조단백질과 조지방함량이 황금콩과 진품콩에 비하여 낮은 것으로 나타났다. Trypsin inhibitor의 활성은 진품콩이 가장 컸고, 수분 흡수 속도도 진품콩이 가장 빨랐으며 황금콩이 가장 느렸는데 익힘 속도는 황금콩이 가장 컸다. 종실의 노화촉진($40^{\circ}C$, 90% RH, 96 hours)시 황금콩의 lipoxygenase 활성이 낮아졌으나 발아율은 황금콩이 가장 높았고 저장에 따른 발아율의 저하 현상도 거의 나타나지 않았다. 그러나 진품콩과 진품콩 2호는 노화촉진시 발아율과 발아 속도가 급격히 감소하였으며 전기 전도도는 노화촉진시 급속히 증가하였다. 이상으로 미루어 보아 lipoxygenase 결핍콩의 더 폭 넓은 활용을 위해 종자활력개선에 대한 연구가 더 필요한 것으로 생각된다.

메주로부터 분리한 항진균 및 항세균 활성의 Bacillus polyfermenticus CJ9 (Bacillus polyfermenticus CJ9, Isolated from Meju, Showing Antifungal and Antibacterial Activities)

  • 메주로부터 항진균 및 항세균 활성을 나타내는 균주를 분리하고 동정하여 B. polyfermenticus CJ9로 명명하였다. B. polyfermenticus CJ9의 생육에 따른 항균 활성을 측정한 결과 항세균 활성은 배양 12시간에 최대 활성을 나타내며 72시간까지 90% 이상 활성을 유지하다 120시간에 활성을 완전히 상실하였다. 항진균 활성은 배양 24시간 이후부터 최대 활성을 나타내었고, 사멸기 이후 활성이 다소 감소되었으나 배양 120시간까지 활성이 유지되었다. B. polyfermenticus CJ9은 식품과 인체에 유해한 곰팡이, 효모, 그람 양성 및 음성 세균에 대한 항진균 활성과 항세균 활성을 동시에 나타내었다. B. polyfermenticus CJ9의 항세균 활성은 $37^{\circ}C$ 에서 24시간 열처리 후에 활성을 상실하였으며, pH 5.0~9.0 구간에서는 안정한 활성을 나타내었으나 pH 3.0~4.0 구간에서 활성이 감소하였다. 항진균 물질은 $121^{\circ}C$에서 15분간 열처리시 활성이 감소되었으나 역가가 완전히 소실되지 않았으며, pH 3.0~9.0 구간에서 안정한 활성을 나타내었다. 항세균 물질과 항진균 물질은 proteinase K, protease, trypsin, $\alpha$-chymotrypsin 등의 단백분해효소 처리로 역가를 상실하거나 일부 감소되어 단백질성 물질임을 추정하였다. B. polyfermenticus CJ9의 항세균 물질과 항진균 물질을 $C_{18}$ Sep-Pak column에 흡착된 분획으로부터 역가를 확인하여 소수성 물질임을 알 수 있었으며, Tricine-SDS-PAGE 및 direct detection 실험을 통하여 분자량을 확인한 결과 항진균 물질은 약 1.4 kDa의 물질임을 확인하였다. 그러나 항세균 활성 물질은 열 불안정성 때문에 동 실험법상에서 그 분자량을 확인할 수 없었다. B. polyfermenticus CJ9이 생산하는 항균 물질은 항세균 및 항진균 활성을 동시에 가지는 단백질성 물질로서 천연 식품보존제 및 정장제재로 활용이 기대되며, 이를 위하여 항세균 물질과 항진균 물질의 정제 및 구조분석 등의 연구가 필요하다.
