Amphidinium stirisquamtum sp. nov. (Dinophyceae), a new marine sand-dwelling dinoflagellate with a novel type of body scale

  • Amphidinium species are amongst the most abundant benthic dinoflagellates in marine intertidal sandy ecosystems. Some of them produce a variety of bioactive compounds that have both harmful effects and pharmaceutical potential. In this study, Amphidinium cells were isolated from intertidal sand collected from the East China Sea. The two strains established were subjected to detailed examination by light, and scanning and transmission electron microscopy. The vegetative cells had a minute, irregular, and triangular-shaped epicone deflected to the left, thus fitting the description of Amphidinium sensu stricto. These strains are distinguished from other Amphidinium species by combination characteristics: (1) longitudinal flagellum inserted in the lower third of the cell; (2) icicle-shaped scales, 276 ± 17 nm in length, on the cell body surface; (3) asymmetrical hypocone with the left side longer than the right; and (4) presence of immotile cells. Therefore, they are described here as Amphidinium stirisquamtum sp. nov. The molecular tree inferred from small subunit rRNA, large subunit rRNA, and internal transcribed spacer-5.8S sequences revealed that A. stirisquamtum is grouped together with the type species of Amphidinium, A. operculatum, in a fully supported clade, but is distantly related to other Amphidinium species bearing body scale. Live A.stirisquamtum cells greatly affected the survival of rotifers and brine shrimp, their primary grazers, making them more susceptible to predation by the higher tropic level consumers in the food web. This will increase the risk of introducing toxicity, and consequently, the bioaccumulation of toxins through marine food webs.

  • − A rotordynamic system consists of components that undergo rotational motion. These components include shafts, impellers, thrust collars, and components that support rotation, such as bearings and seals. The motion of this type of rotating system can be modeled as two-dimensional motion and, accordingly, the equa- tion of motion for the rotordynamic system can be represented using complex coordinates. The directional fre- quency response function (dFRF) can be derived from this complex coordinate system and used as an effective analytical tool for rotating machinery. However, the dFRF is not widely used in the field because most pre- vious studies and commercial software are based on real coordinate systems. The objective of the current study is to introduce the dFRF and show that it can be an effective tool in rotordynamic analysis. In this study, the normal frequency response function (nFRF) and dFRF are compared under rotordynamic analysis for isotropic and unisotropic rotors. Results show that in the nFRF, the magnitude of the response is the same for both pos- itive and negative frequencies, and the response is similar under all modes. Consequently, the severity of the mode cannot be identified. However, in the dFRF, the forward and backward modes are clearly distinguishable in the frequency domain of the isotropic rotor, and the severity of the mode can be identified for the uniso- tropic rotor.

The Infectivity of Recombinant Porcine Endogenous Retrovirus (PERV-A/C) Is Modulated by Membrane-Proximal Cytoplasmic Domain of PERV-C Envelope Tail (C형 돼지 내인성 레트로바이러스(PERV)의 C-말단 외막당단백질에 의한 재조합 PERV-A/C의 감염력 조절)

  • Xenotransplantation of pig organs is complicated by the existence of polytropic replication-competent porcine endogenous retroviruses (PERV) capable of infecting human cells. Two classes of infectious human-tropic replication-competent PERVs (PERV-A and PERV-B) and one class of ecotropic PERV-C are known. The potential for recombination between ecotropic PERV-C and human-tropic PERVs adds another level of infectious risk. A recombinant PERV-A/C (PERV-A14/220) virus is 500-fold more infectious than PERV-A. Two determinants of this high infectivity was identified; one was isoleucine-to-valine substitution at position 140 in RBD (receptor binding domain), and the other lies within the PRR (proline rich region) of the envelope protein. To examine whether the effects of the cytoplasmic tail of the PERV-C Env on fusogenesity also influences infectivity, we constructed a pseudotype retroviral vectors containing MoMLV core protein and PERV envelopes. Pseudotyping experiments with the PERV envelope glycoproteins indicated that recombinant PERV-A/C virus is 10-fold more infectious than PERV-A by lacZ staining. This result supports the suggestion that viral transduction of PERV-A/C is enhanced by a membrane-proximal cytoplasmic amphiphilic ${\alpha}$-helix in PERV-C Env tail.

  • Sex and its tropic hormones influence the lacrimal system, corneal anatomy and disease, aqueous humor dynamics and glaucoma, crystalline lens and cataract, and retinal disease. Dry eye occurs especially frequently during pregnancy, oral contraceptive use, and after menopause, during which androgen levels decrease. Androgen control development, differentiation, and lipid production of sebaceous glands throughout the body, and androgen deficiency also leads to meibomian gland dysfunction and evaporative dry eye. On the other hand, estrogen causes a reduction in size, activity, and lipid production of sebaceous glands. Sex and its tropic hormones also influence the corneal anatomy and disease, and corneal thickening occurred on the second day of the menstrual cycle and around the time of ovulation and appeared to be related to estrogen levels. Fuchs' dystrophy is more commonly seen in postmenopausal women than men and may be linked to hormonal changes that occur with aging. In addition, overexpression of estrogen and progesterone receptors in the conjunctiva of vernal keratoconjunctivitis patients. Serum progesterone levels also may be associated with intraocular pressure especially in pregnant women, and for the women. For women with cataracts, hormone levels were typical of menopause, and there was a significant negative correlation between estradiol and follicular stimulating hormone levels. In addition, serum testosterone levels are associated with the development of diabetic retinopathy. Although the role of sex hormones on the eye is largely unknown, and the results should be interpreted with caution until replicated, the functions of sex hormones in ocular disease remains to be investigated, because they may be involved in structure and function of the ocular components, which are important in the pathogenesis of ocular disease.

우리나라 인공호의 부영양화 평가 및 예측에 관한 연구

  • The purpose of this study is to evaluate and to predict of eutrophication in lakes by using VollenweiderGECD model and total phosphorus concentration and inflow rate which were measurded in 1993-1996. The results of study was as follows. The annual total phosphorus loading from the watershed was calculated to be 181-195tP /yr at lake Soyang, 591-680tP/yr at lake Chungju, 420-466tP/yr at lake Taechong, 229-278tP/yr at lake Andong, 103-106tP/yr at lake Hapchon, 57-59tP/yr at lake Imha, 194-244tP/yr at lake Namgang, 8386tP /yr at lake Chuam, 99-109tP /yr at lake Somjin. These are discharged, for the most parts, from population and ftshfarm facility. TP loading on the surface area at lake Soyang was 3.0lgP/$m^2$/yr, 2.82gP/$m^2$/yr, 2.84gP/$m^2$/yr, 3. 03gP/$m^2$/yr, at lake Chungju 7.91gP/$m^2$/yr, 6.87gP/$m^2$/yr, 7.38gP/$m^2$/yr, 7.l8gP/$m^2$/yr, at lake Taechong 6.7lgP/$m^2$/yr, 7.25gP/$m^2$/yr, 7.24gP/$m^2$/yr, 6.53gP/$m^2$/yr and TP loading on the surface area of Nakdong river basin, that is, lake Andong, Imha, Hapchon and Namgang were 5.39gP/$m^2$/yr, 4.47gP/$m^2$/yr, 4. 56gP/$m^2$/yr, 4.45gP/$m^2$/yr and 2.20gP/$m^2$/yr, 2.23gP/$m^2$/yr, 2.24gP/$m^2$/yr, 2.l7gP/$m^2$/yr and 4.50gP/$m^2$/ yr, 4.50gP/$m^2$/yr, 4.54gP/$m^2$/yr, 4.43gP/$m^2$/yr and 8.25gP/$m^2$/yr, 8.48gP/$m^2$/yr, 8.48gP/$m^2$/yr, 10. 39gP/$m^2$/yr respectively. Also those of lake Chuam was 2.51gP/$m^2$/yr, 2.61gP/$m^2$/yr, 2.52gP/$m^2$/yr, 2. 54gP/$m^2$/yr and TP loading on the surface area at lake Somjin was analysed 4.09gP/$m^2$/yr, 4.l0gP/$m^2$/yr, 3.98gP/$m^2$/yr,3.73gP/$m^2$/yr. The tropic states of nine lakes can be assessed as eutrophy because phosphorus loading exceeds the critical phosphorus loading by Vollenwelder-GECD model.

Removal of Cyanogenic Compounds in Apricot Kernel during Heating Process (가열조리방법을 통한 행인 내 시안화합물의 저감화)

  • Apricot Kernel, consumed as herbal medicine, contains amygdalin which generate HCN upon hydrolysis. Dyspnea was reported by ingesting large amount of apricot kernel, and neurological disorders such as tropic ataxic neuropathy or konzo were known as chronic toxicity of amygdalin. Other cyanogen containing plants, including flaxseed and almond, are consumed as food around the world. Moreover, some of them are promoted as functional food, leading to higher consumption, and posing health risk by cyanogenic components. The objective of this study was to find a method for the reduction of the cyanogenic compound, using apricot kernel as a model food. The most effective reduction was obtained by boiling the slices of the kernel for one hour in pH 1 HCl solution, showing 90% removal. However, the common process known to reduce the cyanogen contents, i.e., long incubation at the low temperature, did not show significant change in cyan concentration. Our data contribute to the safety of the plants containing cyanogenic compounds if they were to be developed as foodstuff.

Kinetic Analyses for Enzymatic Properties of Trypsins Purified from Dark-Fleshed Fish (혈합육어 Trypsin의 효소적 성질에 대한 반응속도론적 해석)

  • Kinetic properties of typsins purified from dark-fleshed fish (anchovy, mackerel, yellowfin tuna, and albacore) were examined and analyzed on $benzoyl-_{D,L}-arginine-p-nitroanilide\;(BAPNA)$. The values of Km' and $k_{cat}$ of the purified trypsins from the four dark-fleshed fish were found to be $49.3{\mu}M$ and $90.9\;min^{-1}$ for anchovy, $53.7{\mu}M$ and $61.2min-^{-1}$ for mackerel A, $96.5{\mu}M$ and $76.6min^{-1}$ for mackerel B, $62.8{\mu}M$ and $46.6min^{-1}$ for yellowfin tuna, and $98.3{\mu}M$ and $47.7min^{-1}$ for albacore, respectively. The values of $K_i$ on $tosyl-_L-lysine$ chloromethyl ketone (TLCK) were determined to be $20.90{\mu}M$ for anchovy trypsin, $2.86{\mu}M$ for mackerel trypsin A, $3.90{\mu}M$ for mackerel trypsin B, $0.96{\mu}M$ for yellowfin tuna trypsin, and $1.82{\mu}M$ for albacore trypsin. Thus yellowfin tuna trypsin was the most sensitive to TLCK among all trypsins. The activities and catalytic efficiency of the trypsins purified from the temperate zone fish, anchovy and mackerel, were higher than those of the trypsins purified from yellowfin tuna and albacore which migrate widely from the tropic zone to the temperate zone.

Characteristics of ecological structure and spatial distribution of micro-plankton in relation to water masses in the northern East China Sea(nECS) in summer 2019 (2019년 여름 동중국해 북부해역의 수괴 분포에 따른 미소플랑크톤의 공간분포 및 생태구조 특성)

  • We conducted a field survey to analyze the ecological structure and spatial distribution of microplankton (phytoplankton and ciliates) in relation to water masses at 21 stations on the surface and chlorophyll-a maximum layers (CML) in the Northern East China Sea (nECS; 32°-33°N; 124°00'-127°30'E) from August 3 to August 6, 2019. The results showed that the water masses were divided into Chinese Coastal Waters (CCW) and the Tsushima Warm Current (TWC). The CCW showed the environmental characteristics of high temperature and low salinity, and the TWC showed high temperature and high salinity. The characteristics of the phytoplankton community in the CCW showed various community structures related to the nutrients supplied from the large rivers of the Chinese continent. However, the TWC had simple community structures because it originated near the equator and moved northward. The standing crops of phytoplankton and ciliates were very high in the CCW but showed low at the TWC. In particular, from the higher standing crops of protozoa than plant plankton at the TWC, the energy flow at the lower tropic levels caused by the microbial loop that fed on heterotrophic bacteria played an important role in the production of resource organisms. In other words, the marine ecological structure of the nECS in summer could be estimated as a bottom-up system at the CCW and a top-down system at the TWC.

Applications and Assessments of a Multimetric Model to Namyang Reservoir (남양호에서 다변수 메트릭 모델 적용 및 평가)

  • The purpose of this study was to evaluate fish metric attributes using a model of Lentic Ecosystem Health Assessment (LEHA) and apply the model to the dataset sampled from six sites of Namyang Reservoir during October 2005$\sim$May 2006. The model was composed of 11 metries and the metric attributes were made of physical, chemical and biological parameters. Trophic composition's metrics showed that tolerant species ($M_3$, 80%) and omnivore species ($M_4$, 92%) dominated the fish fauna, indicating a biological degradation in the aquatic ecosystem. The metric of $M_7$, relative proportions of exotic species, also showed greater than 8% of the total, indicating a ecological disturbance. The average value of LEHA model was 24.3 (n= 12) in the reservoir, indicating a "poor condition" by the criteria of An and Han (2007). Spatial variation based on the model values was low (range: $21{\sim}26$), and temporal variation occurred due to a monsoon rainfall. Electrical conductivity (EC) and tropic state index of chlorophyll-$\alpha$ [TSI(CHL)] was greater in the premonsoon than the postmonsoon.

A Study on Spatial Characteristics of Post-Disaster Interim Housing - Focusing on Asian Precedents of Natural Disasters - (재난 이후 임시주거의 공간특성 연구 - 아시아지역에서 발생한 자연재난을 중심으로 -)

  • This study intends to research the spatial characteristics of Asian interim housing that accommodates sufferers pro tempore after disasters. The scope of this research covers the interim spaces used for housing people after natural disasters that occurred in Asia for the past fifteen years. Within this scope, literature review was conducted as the basis to derive the characteristics and environmental elements of interim housing, which provided the criteria to compare and evaluate cases of interim housing along with characteristic elements required of interim housing found in previous studies. According to literature review, interim housing can be classified by life-span, region, economy, climate, type, number of household, square measure, residential cost, structure/material, and service life. Within the scope of the present research, literature review showed a total of twenty-eight cases of interim housing in fifteen countries revealing a high rate of disaster occurrence in the subtropic and tropic climate of Southeast Asia. A great percentage of interim housing was used for long-term stay of over a year. The structure of interim housing varied from lightweight steel, wooden, masonry, membrane, to traditional structure and the type were divided into temporary shelter, transitional housing, temporary housing, and permanent housing. Followed by literature review, the characteristics required of post-disaster interim housing were analyzed based on previous research and case studies. The characteristics of interim housing can be divided into environmental, technological, and socio-cultural ones. Sub-characterical items according to such division include amenity, health, surroundings, structure, convenience, eco-friendliness, safety, communication, and locality. As a result of evaluation, most items met the required characteristics of interim housing, while technological characteristics such as structure and convenience varied with the types of interim housing and appeared even unnecessary in some cases. According to analysis, amenity is maintained through the structural and material characteristics of interim housing and is also facilitated by increasing number of infrastructure such as educational, sanitary, and convenience facilities provided by the governmental and organizational bodies. It is expected that this study will be utilized as preliminary data for follow-up studies that improve the environment of post-disaster interim housing suitable for domestic circumstances in environmental, technological, and socio-cultural respects.
