Biochemical characteristics and serotypes of Salmonella spp isolated from domestic animal in western Gyeongbuk province (경북서부지방 가축에서 분리된 Salmonella 속균의 생물화학적 특성 및 혈청형)

  • The present study was conducted to investigate the prevalence, biochemical properties and serotypes of Salmonella organisms in the domestic animals in Western Gyeongbuk province during the period from January to December 2000. Salmonella spp were isolated from 51(1.62%) of 3,141 cases of domestic animals. Serotypes of isolates were S enteritidis 17(29.6% of isolates), S agona 11(20.3%), S rissen 8(14.8%), S gallinarum 4(7.4%), S derby 3(5.5%), S typhimurium 2(3.7%), S travis 1(1.8%), S montevideo 1(1.8%) and untypable 4(12.9%), in order. The majority of isolates were highly susceptible to amikacin, ampicillin, ciprofloxacin, cephalothin, chloramphenicol, trimethoprim-sulfamethoxazole, norfloxacin and gentamicin, whereas all isolates were resistant to bacitracin, erythromycin, tiamulin and tylosin, and the majority of them were highly resistant to penicillin, streptomycin and tetracycline.

  • Methicillin-resistant Staphylococcus aureus (MRSA), a leading cause of nosocomial infections, has been increasingly recognized in communities of the United States. This article will review the clinical spectrum and treatment of MRSA infections in children in the context of recent epidemiological changes of MRSA infections. In general, community-associated (CA) MRSA most frequently causes skin and soft tissue infections and has an increased association with invasive infections, particularly pneumonia and musculoskeletal infections. Hospital-associated (HA) MRSA strains tend to be associated with bloodstream infections, pneumonia, and surgical site infections. Different from the United States, CA-MRSA infections are not common in Korea (only 5.9%); however, there are some CA-MRSA clones that are different from HA-MRSA clones in Korea and from CA-MRSA clones in other countries. The treatment of MRSA infections should be guided by antimicrobial susceptibility testing, the site of infection, and the infection severity. Vancomycin is the treatment of choice for invasive MRSA infections. Other agents such as trimethoprim-sulfamethoxazole, clindamycin, linezolid, quinupristin-dalfopristin, and daptomycin have been used for some conditions.

Maxillary Sinusitis by Staphylococcus aureus Infection in a Thoroughbred Gelding: Case Report

  • A 4-year-old gelding Thoroughbred racehorse, which had been undergoing antibiotic therapy at a local veterinary clinic, was referred to the KRA veterinary center with a 20-day history of continuous right nasal discharge. Patient's history, endoscopic examination, and radiographic examination revealed primary maxillary sinusitis. Under sedation, surgical intervention was performed to collect samples and remove the accumulated mucopurulent exudate in the sinus. Swab samples were collected from the sinus during surgery for cytology and antimicrobial susceptibility testing. Only one type of bacteria was cultured, and molecular analyses of 16S ribosomal RNA gene sequences identified it as Staphylococcus aureus (S. aureus). The isolate was resistant to multiple antibiotics, which are frequently used in equine practice. Trimethoprim-sulfamethoxazole was chosen based on antibiotic susceptibility test, trephination, and sinus lavage using saline were applied to treat bacterial sinusitis. The clinical signs improved after 1 month and the patient resumed training. This report describes S. aureus isolated from bacterial maxillary sinusitis in a horse and its antibiotic susceptibility.

Complete Genome of Methicillin-Resistant Staphylococcus pseudintermedius Z0118SP0130 Isolated from a Companion Dog

  • Methicillin-resistant Staphylococcus pseudintermedius Z0118SP0130 was isolated from eye specimen of a companion dog in South Korea. The complete genome of Z0118SP0130 consisted of a 2,663,277 bp chromosome and there was no plasmid. The strain was identified as the sequence type 45 and contained a mecA gene which comprised of staphylococcal cassette chromosome mec type Vb (5C2&5). Antimicrobial resistance to erythromycin, clindamycin, quinupristin-dalfopristin, trimethoprim-sulfamethoxazole, mupirocin, oxacillin, streptomycin, and gentamicin was observed in the strain.

  • Staphylococcus epidermidis is a normal flora of human skin and is occasionally associated with pathogenic infections. We report the complete genome sequence of methicillin-resistant Staphylococcus epidermidis strain Z0118SE0272 isolated from the residential environment sharing by a companion dog and dwellers. Resistance to cefoxitin was observed in the strain, whereas it was susceptible to erythromycin, clindamycin, quinupristin-dalfopristin, trimethoprim-sulfamethoxazole, mupirocin, vancomycin, teicoplanin, linezolid, and tigecycline. The strain Z0118SE0272 identified as sequence type 130 possessed the mecA gene responsible for methicillin resistance, which composed the new type of staphylococcal cassette chromosome mec elements lacking mecRI.

The Causative Organisms of Otitis Media Accompanying Otorrhea in Children and Their Antimicrobial Susceptibility (소아에서 이루를 동반한 중이염의 원인 및 항균제 감수성)

  • Purpose : A great deal of youngsters suffer from otitis media, for which antimicrobials are frequently prescribed. Increased antimicrobial resistance forces physicians to judiciously use antimicrobial agents in treating patients with acute otitis media. There have however been few references with regard to otitis media in Korean children, and authors proceeded investigation to look for the causative agents of otitis media in Korean children and their antimicrobial susceptibility. Methods : The study included 65 patients younger than 15 years old who had been cared at the department of pediatrics and otolaryngology in Hanyang University Hospital from July 1994 to June 1999, and diagnosed of otitis media with otorrhea which contained microorganisms isolated in otorrhea culture. The medical records were reviewed for demographic data, isolated organisms and their antimicrobial susceptibility. Results : Among 65 patients, 37(57%) were boys and 28(43%) girls. Distribution of the patients was reciprocal to the age of the patients; 27 patients(41.5%) were younger than 1 year old, 24(36.9%) were 1 to 3 years old with the average of 2.9 years of age. Staphylococus aureus was isolated in 32 patients(49.2%), Streptococcus pneumoniae in 19 patients(29.2%) Haemophilus influenzae in 9 patients(13.8%), Streptococcus oralis in 3 patients(4.6%), Moraxella catarrhalis in 1 patient(1.5%). The isolated microorganisms were not different whether patients had cleft lip/palate or not. The antibiotic resistance rates of S. aureus were ${\geq}90%$ to erythromycin, imipenem, cephalothin, and clindamycin, 86.2% to oxacillin, 25% to chloramphenicol, 12.5% to trimethoprim/sulfamethoxazole(TMP/SMX), and 0% to vancomycin and teicoplanin. The antibiotic resistance rates of S. pneumoniae were 71.4% to penicillin and greater than 60% to erythromycin, tetracycline, TMP/SMX, 7.1% to chloramphenicol, and 0% to vancomycin and teicoplanin. The antibiotic resistance rates of H. influenzae were 55% to ampicillin and TMP/SMX, and 0% to chloramphenicol, ceftriaxone, aztreonam, imipenem and ciprofloxacin. Conclusion : With otorrhea culture, the causative organisms of otitis media appear to be S. aureus, S. pneumoniae and H. influenzae. The high antibiotic resistance rates of the isolated organisms should affect the choice of antibiotics in treating patients with otitis media. Prospective investigations utilizing tympanocentesis in microbiologic studies are needed.

Experimental activation of cryptosporidiosis in mice by immunosuppression (면역억제에 의한 마우스의 Cryptosporidium 발현 실험)

  • Cryptosporidium, a coccidian parasite first described by Tyzzer (1907) from a laboratory mouse, has become an important human enteric pathogen causing overwhelming diarrhea especially in immunocompromised patients such as AIDS. This parasite has been reported from over 20 countries and is recognized as a cosmopolitan species. In Korea, however, thEre has been no report on human as well as animal cryptosporidiosis. This study was performed so as to verify the presence of Cryptosporidium in Korea by activating the parasite from laboratory mice by immunosuppression. Total 65 conventionally.bred ICR mice including a control (5 mice) and 3 experimental groups (20 each) were used for this study. Group I was immunosuppressed with Prednisolone injection (1 mg IM, every other day) for 7 weeks. Group II (prednisolone injection and tetracycline administration) and Group III. (prednisolone injection and trimethoprim-sulfamethoxazole administration) were prepared to observe the effect of antibacterial agents on the activation of cryptosporidiosis. In fecal examinations of mice Cryptosporidium oocysts($4-6{\mu\textrm{m}}$ in size) were detected from 1 week after the start of immunosuppression and the mice began to die. In H-E stained tissue sections of the lower jejunum, numerous very small ($2~4{\;}{\mu\textrm{m}}$), dense, ovoid or spherical, slightly basophilic bodies were seen attached on the free border of mucosal epithelial cells. In scanning and transmission electron microscopic observations, these organisms were identified as various developmental stages of Cryptosperidium. The species is considered to be C. parvum. Cryptosporidiosis was activated not only in Group I but also in Group II and III, indicating no protective effects of the antibacterial agents used, although the mice in Group II and III lived longer than those in Group I. The present study confirmed that Cryptosporidium exists in laboratory mice bred in Korea, and predicts possible occurrence of human cryptosporidiosis in Korea.

Studies on characteristics and pathogenicity of Yersinia sp. Isolated from cultured olive flounder, Paralichthys olivaceus (양식넙치에서 분리한 Yersinia sp.의 일부 특성과 병원성에 관한 연구)

  • Characteristics and pathogenicity of Yersinia sp. strain isolated from diseased cultured olive flounder, Paralichthys olivaceous were studied. The causative organisms were identified as Yersinia sp. by biochemical and biophysical characteristics. The strain was named KF-1, and it showed the optimal growth rate at pH 9.0 and 1% NaCl. Total cell protein peptide bands of the Yersinia sp. KF-1 were between 14.4-100.6 Kd in molecular weight by the electrophoretic analysis, and a total of 28 bands appeared. The band found at 32.8 Kd in molecular weight was the major one of electrophoretic phase. In the pathogenicity test of the isolate to the flounder injecting with $1.0{\times}10^7$ cfu/fish 9 died out of 10 within 60 hrs, and in the group with $1.0{\times}10^7$ cfu/fish 5 died within 60 hrs. Thus the $LD_{50}$ was presumed to be $1.0{\times}10^7$ cfu/fish. In the drug sensitivity test KF-1 strain was sensitive to chloramphenicol, gentamycin, kanamycin, streptomycin and pefloxacin, and intermediate to erythromycin, and resistant to ampicillin, cephalothin, sulfamethoxazole/trimethoprim, tetracycline and vancomycin.

Characterization of Verotoxin-producing Escherichia coli Isolated from Domestic Foods (국내 유통식품에서 분리된 Verotoxin 생성 Escherichia coli의 특성)

  • The incidence of verotoxin-producing Escherichia coli(VTEC) was surveyed in domestic foods including hamburger, raw meats and vegetables from 1997 to 1999. The molecular biological characteristics of the isolates were analyzed. Three VTEC strain were isolated from 1,700 samples. Serotypes of those isolates were 0157 : H7, 026 H4, and 056 : Hl2, respectively. Serotype O26 : H4 produced VT I and VT II, and 055 Hl2 isolate produced VT I, however the 60 MDa plasmid DNA and eae gene were not found from both strains. One 0157 : H7 isolate produced VT II and harbour 60 MDa plasmid DNA, however eae gene was not found in the strain. Although they produced VT, it seemed that the virulence of two strains were relatively weak because of the lack of the eae gene. In addition, the serotype O157 : H7 isolate resistant to ampicillin and streptomycin, while isolates of serotype O26 : H4 and O55 : Hl2 were multi-resistant to antibiotics including ampicillin, carbenicillin , cephalothin, trimethoprim/sulfamethoxazole and tetracycline. Characteristics of Yersinia enterocolitica Isolated from Foods (식품에서 분리한 Yersinia enterocolitica의 특성조사)

  • The incidence of Yersinia enterocolitica in raw meat was determined over 10 month period. Y. enterocolitica was isolated from 8.5% of beef, 17.0% of pork and 25.6% of chicken. Overall prevalence of Y. enterocolitica in raw meat was 17.5%. Seasonal difference was observed in isolation rate in which pork and chicken samples collected in the second half of the year twice was more than that of the first half of the year. Serotypes of Y. enterocolitica isolates were O:5 (17.3%), O:8 (8.6%), O:3 (6.2%), O:1,2 (1.2%), and others. The antibiotics susceptibility tests showed Y. enterocolitica isolates were resistant to carbenicillin, ampicillin, erythromycin, penicillin and bacitracin. In contrast it showed sensitivity to polymyxin B, kanamycin, ciprofloxacin, gentamicin, trimethoprim/sulfamethoxazole, nalidixic acid, and tetracycline. PCR with specific primers derived from ail gene of Y. enterocolitica was applied to detect and confirm pathogenic Y. enterocolitica. About 10% of the isolated Y. enterocolitica proved to be pathogenic and most of them were found in pork. However, proper cooking and meat process can kill and remove all Y. enterocolitica in meat, because the organism is very sensitive to heat.
