Effect of Dietary Fibers Isolated from Tangerine Peels on Lipid and Cadmium Metabolism in the Rat (감귤과피로부터 분리한 식이섬유가 흰쥐의 지방 및 Cadmium 대사에 미치는 영향)

  • This study was performed to investigate the effects of dietary fibers from tangerine peels on lipid and cadmium metabolism. And effects were compared with those of commercial dietary fibers($\alpha$-cellulose, citrus pectin). Sixty male rats of Sprague-Dawley strain weighing 186.7$\pm$2.6g were blocked into 12 groups according to body weight, and were raised for 2 weeks. Cadmium chloride was given at the levels of 0 of 400 ppm in diet. Various dietary fibers were given at the level of 0 or 4%(w/w) of diet. The results are summarized as follow. In lipid metabolism, insoluble fibers[insoluble dietary fibers from tangerine peels(IDE), $\alpha$-celluolse] increased fecal excretion of lipids by inhcreasing feces weight, and decreased the concentrations of serum triglyceride and liver lipids. Soluble dietary fibers from tangerine peels(SDF) decreased the concentrations of serum cholesterol and liver lipids by increasing fecal lipids, too. In cadmium metabolism, soluble fivers(SDF, pectin) inhibited Cd absorption by increasing fecal Cd excretion and decreased Cd concentrations of intestion, liver and kidney. In conclusion, among the extracted fibers, SDF were more effective on lipid and Cd lowering activity and IDF had effect of increasing fecal lipid excretion. This result is useful to reduce food waste and utilize waste products.

Effects of Coated Liposome from Discorea rhizoma Extract (DRE) -on Hypoglycemic, Serum Insulin, and Lipid Levels in Streptozotocin-Induced (산약 추출물의 리포좀 처리가 Streptozotocin으로 유발된 당뇨 마우스의 혈당 강하 효과와 혈장 인슐린 및 지질에 미치는 영향)

  • To investigate the effects of coated liposome from Discorea rhizoma extract (DRE) in streptozotocin (STZ)-induced, we evaluated changes in body weight, fasting blood glucose, blood insulin and blood lipid concentrations in mice. Mice were divided into four groups: (DC), diabetic DRE at 10 mg/kg (DRE-1), diabetic DRE at 50 mg/kg (DRE-2), and diabetic DRE at 250 mg/kg (DRE-3). Mice had free access to water and diet (10 weeks). The DC group showed higher blood cholesterol than the DRE-1, DRE-2, DRE-3 groups. In glucose tolerance test, the DRE-1, DRE-2, and DRE-3 groups increased after 30 minutes in decremental glycemic response area under the curve. Fasting blood glucose levels in the DRE groups significantly decreased through 4 weeks. Plasma total cholesterol, triglyceride and LDL-cholesterol concentrations were also lower in the DRE groups. On the other hand, the DRE-1, DRE-2 and DRE-3 groups showed higher HDL-cholesterol and insulin levels than the DC group. Moreover, blood glucose and lipid levels significantly decreased in streptozotocin (STZ)-induced diabetic mice treated with DRE. These results indicate that DRE may reduce elevated blood glucose levels and serum lipid concentrations in hypoglycemic and diabetic mice, suggesting its usefulness as a functional food.

Effects of the Zizyphus jujuba Seed Extract on the Lipid Components in Hyperlipidemic Rats

  • The purpose of this study was designed to observe the effects of Zizyphus jujuba seed extract on the concentrations of the lipids and blood glucose in the S.D. rats fed the experimental diets for 4 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, triglyceride (TG), phospholipid (PL) and blood glucose in serum were significantly higher in the cholesterol administration groups (group 2 (cholesterol+water), group 3 (cholesterol+Zizyphus jujuba seed extract)) than those in the control group (group 1, basal diet+water). But the concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum ware remarkably lower in the group 3 than those in the group 2. In the ratio of HIDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Zizyphus jujuba seed extract administration group was higher percentage than in the group 2. The activities of aspartate aminotransferase (AST), alanine amino-transferase (ALT), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum were rather lower in the Zizyphus jujuba seed extract administration group (group 3) than in the cholesterol diet group (group 2). From the above research, Zizyphus jujuba seeds were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic rats. And particularly, Zizyphus jujuba seeds were more effective as a therapeutic regimen for the control of metabolic derangements in adult disease.

Effects of Liquid Culture of Coriolus versicolor on Lipid Metabolism and Enzyme Activities in Rats fed Cholesterol Diet. (운지버섯 배양액이 콜레스테롤 식이를 섭취한 흰쥐의 지질 대사에 미치는 영향)

  • The effects of liquid culture of Coriolus (C) versicolor on weight gain, food intakes, food efficiency ratios, serum and hepatic lipid concentrations were investigated in male rats fed the high cholesterol diets. Eight weeks old Sprague-Dawley rats were given three different types of diet for six weeks, respectively: a control diet ($ 20\% fat +0.5 \% $ cholesterol), two kinds of C versicolor diet (control diet + $ 30\% or 40\%$C. versicozor in water) according to the levels of C. versicolor supplementation. The body weight gains of the rats fed $ 30\% or 40\% $ C. versicolor diets were lower than those in the rats fed the control diet. The food intake, food efficiency ratios, and liver, kidney, epididymal fat pad weights of the rats fed $ 30\% or 40\%$ C. versicoEer diets were similar to those of the rats fed the control diet. The concentrations of hepatic cholesterol and triglyceride in the rats fed $ 30\% or 40\% $ C. versicolor diets were significantly lower than those in the rats fed the control diet. The concentrations in serum total cholesterol, LDL-cholesterol, and atherogenic index ratios were significantly lower in the rats fed $ 30\% or 40\%$ C. versicolor diets compared to those fed the control diet. The HDL-cholesterol/total-cholesterol ratios was significantly higher in the rats fed $ 30\% or 40\% $ C. versicolor diets compared to those fed the control diet. The fecal excretion of total lipid and triglyceride in the rats fed $ 40\% $ C. versicolor diet was significantly higher than that of the rats fed the control diet. There were no significant difference found in the serum trigly-ceride, phospholipid and HDL-cholesterol concentrations among the experimental groups. These results showed that the C. versicolor feeding decreased the hepatic cholesterol, triglyceride, and the serum total cholesterol, LDL-cholesterol and atherogenic index, and increased the serum HDL-cholesterol/ total-cholesterol ratio of the rats.

Effects of Tea Fungus/Kombucha Beverage on Lipid Metabolism and Protein Level in Diabetic Female Rats (발효홍차 음료가 당뇨 암쥐의 지질대사 및 단백질농도에 미치는 영향)

  • We investigated the effect of tea fungus/kombucha beverage(TF) on the body weights, the blood glucose levels, lipid and protein concentrations, and enzyme activities in diabetic female rats. Sprague-Dawley rats were fed drinking water supplemented with 20% or 40% TF groups, respectively for 7 weeks. The female rats (mean weight 155.5$\pm$9.3 g) were assigned to one control and three diabetic groups. Diabetic groups were divided into diabetic control (TF free water), 20% or 40% TFD groups (20% or 40% TF in water) according to the levels of TF supplementation. Diabetes was experimentally induced by intraperitonially administration of streptozotocin in citrate buffer(pH 4.3) after 2 weeks feeding of four experimental water. Animals were sacrificed at the 5 weeks of diabetic state. The diabetic groups showed significantly decrease of body weight(6.8-7.5 g) compared with the control group(48.3 g). The hepatic, kidney and pancreatic weights of 20% or 40% TFD groups were not significantly different with D-control group. The fasting serum glucose level were higher in all diabetic groups than that of the control group. The concentrations of serum triglyceride in 40% TFD group and serum LDL-cholesterol in 20% TFD group were significantly decreased compared with the D-control group. The concentrations of serum total cholesterol and HDL-cholesterol, HDL-cholesterol/total cholesterol ratio, and atherogenic index in 20% or 40% TFD groups were similar to those in D-control group. The concentrations of hepatic triglyceride in 20% or 40% TFD groups were significantly decreased compared with the D-control group, but the concentrations of hepatic cholesterol and phospholipid were similar to all diabetic groups. The concentrations of serum and hepatic total protein, serum albumin, and the activities of GOT, GPT and LDH in the serum were the same levels of all diabetic groups.

Isolation of salt-tolerant bacteria from rhizosphere and rhizoplane of halophyte plant Suaeda japonica in Gochang·Buan tidal flat (고창·부안 갯벌에 자생하는 염생식물 칠면초 근권 및 근면으로부터 내 염성 세균 분리와 그 특성화)

  • Nine strains of high concentrations of salt-tolerant bacteria were isolated from the rhizosphere and rhizoplane of the halophyte plant Suaeda japonica grown in Gochang Buan tidal flat. The isolated bacteria were classified as genera Vibrio (strains JRS-1, -2, -3, -4, and -5, and JRL-1 and -4) and Bacillus (strains JRL-2 and -3) based on the 16S rRNA gene sequence similarity. The optical growth condition for salt concentration was examined on the selected, representative strains. Strain JRS-1 with the closest relative of Vibrio neocaledonicus showed the highest growth rate at the total salt concentration of 6% among the incubation conditions of 3-8% salt concentrations. Strain JRL-2 with the closest relative of Bacillus thuringiensis showed the tendency that growth rate increased with increasing salt concentrations and the maximum growth rate at 7% of the total salt concentration. The isolated bacteria showed salt-resistances to higher salt concentrations than their habitat soils with 3%. In addition, we identified evidences of potentially plant interaction-relevant enzymatic activities, from utilization of some substrates rich in plants, such as triglyceride, ${\rho}$-nitrophenyl-${\alpha}$,$\text\tiny{D}$-glucoside, and ${\rho}$-nitrophenyl-${\beta}$,$\text\tiny{D}$-glucoside.

Effects of the Feeding Mixed with Various Levels of n-3 and n-6 Polyunsaturated Fatty Acid on the Lipid Components and Fatty Acid Metabolism of Serum Lipoprotein in Hyperlipidemic Rats (n-3 및 n-6계 다불포화 지방산의 함유비율이 다른 유지가 고지혈증 흰쥐의 혈청 지단백 지질성분 및 지방산 대사에 미치는 영향)

  • This study was designed to observe the effects of the feeding mixed with the sardine oil containing n-3 EPA, DHA and the safflower oil in which rich n-6 linoleic acid on the improvement of the lipids and on the fatty acid matabolism of serum lipoprotein in the dietary hyperlipidemic rats. Experimental oils mixed with 16% butter(control group) and 8% butter+8% olive oil, 8% butter and various level of sardine and safflower oils were administered to the male rats of the Sprague Dawley for 4 weeks. Concentrations of total cholesterol in serum lipoproteins were highest in the LDL, followed by VLDL, HDL and chylomicron components. in the triglyceride concentrations, chylomicron were highest, and much VLDL, LDL, HDL components. There were highest in the HDL, followed by LDL, VLDL and chylomicron in phospholipid concentrations of serum lipoproteins. Concentrations of total cholesterol, triglyceride and phospholipid in serum lipoprotein were lower in the other groups than in the control groups, and particularly, lowest in the group 5(4% sardine oil+4% safflower oil). From this results, the feeding equal quantity mixed oil with n-3 PUFA rich sardine oil and n-6 PUFA rich safflower oil were effective on the improvement of the lipid composition in the serum lipoprotein. It might be due to the effects of appropriate rations of P/S, 0.85 and n-6/n-3P, 2.85 in the test lipids. In the fatty acid composition of serum lipoprotein, ratio of P/S and n-3/n-6P were influenced from the fatty acid compositions of the mixed test lipids. And EPA contents were higher in the chylomicron components.

Effects of Agaricus blazei Murill on Lipid Metabolism in Rats Fed High Fat Diet (신령버섯이 고지방 식이를 급여한 흰쥐의 지질대사에 미치는 영향)

  • The effects of the fruiting body of Agaricus blazei Murill on the weight gains, food intakes, food efficiency ratios, serum and hepatic lipids concentrations were investigated in male rats. Sprague-Dawley rats, 21 weeks old, were given four different types of diets for a succeeding period of 10 weeks: either a normal diet (5% corn oil), a high fat diet (high fat; 20% lard), a 3% or 5% Agaricus diet (high fat diet+3% or 5% Agaricus powder). The body weight gains and food efficiency ratios of the rats fed 5% Agaricus diet were significantly lower than those of the rats fed high fat diet. The hepatic and kidney weights of the rats fed Agaricus diets were similar to those of the rats fed high fat diet. The epididymal fat pad weights of the rats fed 3% or 5% Agaricus diets were significantly lower than those of the rats fed high fat diet. The concentrations of hepatic and serum triglyceride in the rats fed 5% Agaricus diet were significantly lower than those in the rats fed high fat diet. But the hepatic total cholesterol of rats fed the 3% or 5% Agaricus diets were similar to those of rats fed the high fat diet. The concentrations of serum total cholesterol, LDL-cholesterol and atherogenic index in rats fed the 3% or 5% Agaricus diets were significantly decreased compared with those of rats fed the high fat diet. The HDL-cholesterol/total- cholesterol ratios of the rat fed 3% or 5% Agaricus diet were significantly increased compared with those of rats fed the high fat diet. There were no differences in serum concentrations of HDL-cholesterol and phospholipid among the experimental groups. These results showed that the 5% Agaricus diet feeding decreased the total cholesterol, the triglyceride, the LDL-cholesterol and the atherogenic index, and increased the HDL-cholesterol/total-cholesterol ratio in serum of rats.

Short-term Administration of Conjugated Linoleic Acid Reduces Liver Triglyceride Concentration and Phosphatidate Phosphohydrolase Activity in OLETF Rats

  • The present study explored the short-term effects of dietary conjugated-linoleic acid (CLA) on liver lipid metabolism in starved/refed Otsuka Long Evans Tokushima Fatty (OLETF) rats. Male OLETF rats (12 weeks old) were starved for 24 hours, then refed for 48 hours with either a CLA diet [7.5% CLA and 7.5% Safflower oil (SAF)] or a SAF control diet (15% SAF). The results demonstrated a 30% reduction of hepatic triglyceride (TG) concentration in the CLA group when compared to the control group. Liver cholesterol concentration was also 26% lower in the CLA fed rats. The activity of mitochondrial carnitine palmitoyltransferase, the rate-limiting enzyme of fatty acid oxidation, was moderately elevated by 1.2-fold in the livers of the CLA group when compared to the control. In contrast, phosphatidate phosphohydrolase, the rate-limiting enzyme for TG synthesis, was found to be 20% lower in the livers of the CLA-fed rats. Therefore, dietary CLA evidently lowers liver lipid concentrations through a reduced TG synthesis and enhanced fatty acid oxidation in starved/refed OLETF rats.

The Effect of Jeseubsungi-tang on Dietary Hyperlipidemia-induced Mice (제습순기탕(除濕順氣湯)이 고지혈증(高脂血症) 병태(病態) 모델에 미치는 영향(影響))

  • Objectives : This experiment was performed to determine the effects of the Jeseubsungi-tang(JSSGT) on hyperlipidemia-induced by hypercholesterolemic diet in mice. Methods : C57BL/6 mice were sensitized with a hypercholesterolemic diet for 6 weeks. Experimental group was treated with $150mg/d{\ell}$ concentrations of JSSGT for 4 weeks (once a day). Results: JSSGT-treated mice showed a body weight decrease compared to normal group. JSSGT-treated mice showed a significant decrease in cholesterol, total cholesterol, triglyceride, ALT and LDL-cholesterol levels compared to hyperlipidemia-induced control group. However, HDL-cholesterol levels in JSSGT-treated group significantly increased compared to the hyperlipidemia-induced control group. Conclusion : These results suggest that JSSGT is effective in treating hyperlipidemia. Because only hyperlipidemic mice were studied, further research is needed.
