Effect of prickly pear cactus (Opuntina ficus-indica) intake on blood lipids, platelet aggregation, antioxidant and liver parameters in volunteer diving woman (손바닥 선인장 열매 섭취가 한림지역 해녀의 혈중지질, 혈소판 응집성 및 항산화 효과에 미치는 영향)

  • We investigated dietary effects of prickly pear cactus (Opuntina ficus-indica) on plasma lipids, platelet aggregation (PA), hemolysis, plasma TBARS and liver enzymes. Twenty eight volunteer diving women in Jeju island had daily 20 g cactus tea containing 27% prickly pear cactus (PPC) powder for 4 weeks, and data for the study subjects were analyzed, on the basis of diagnostic criteria for blood pressure (BP)(${\geq}$ 140/90 mmHg), plasma cholesterol (${\geq}$ 200 mg/dL) and triglyceride (${\geq}$ 150 mg/dL). The subjects with higher BP had higher plasma total cholesterol (TC) and triglyceride (TG) concentrations than those with normal BP. Those with higher TC also had higher TG. Subjects with normal BP or normal TC had higher initial slope of PA than their higher counterpart in BP and TC. PPC intake decreased plasma TG in those with higher BP. PPC intake significantly decreased the elevated initial slope in groups with normal BP, TC, and TG. Hemolysis after PPC intake decreased significantly in all the subjects and plasma TBARS decreased in the subjects with higher plasma TC and higher TG. Glutamic-oxaloacetic transaminase (GOT) significantly increased and total bilirubin significantly decreased in all the subjects after PPC intake. The present study with diving women showed that beneficial effects of short term intake of prickly pear cactus might differ depending on the subject conditions in term of blood pressure, and plasma lipids. However, long term usage of prickly pear cactus may provide preventive effects of cardiovascular diseases to all the population, presumably by hypolipidemic, antithrombotic, and antioxidant actions of its bioactive flavonoids and soluble fiber.

Effects of Conjugated Linoleic Acid-Triglyceride Additives on Quality Characteristics of Pressed Ham (Conjugated Linoleic Acid-Triglyceride 첨가가 프레스햄의 품질특성에 미치는 영향)

  • Conjugated linoleic acid (CLA) was chemically synthesized using the alkaline isomerization method of com oil. CLA-TG was synthesized by reaction with sodium methoxide. Five different treatments were devised based on differences in the amount of CLA-TG added into the pressed han. for controls, 10% of pork back fat among the total component was only added without any CLA-TG. For the first treatment, 5% of CLA-TG among the lard component added into the press ham was replaced. For the 2nd, 3rd and 4th treatments, 10%, 15% and 20% of CLA-TG was respectively replaced. Pressed ham manufactured using CLA-TG was vacuum packaged and then stored for 1, 7, 14, 21 and 28 days at $4^{\circ}C$. Samples were analyzed for chemical composition, physico-chemical properties (pH, color), and texture characteristics. Typical chemical composition characteristics (moisture, crude protein, crude fat) were not significantly different between controls and CLA-TG treatment groups. Crude fat content of CLA-TG treatment groups was significantly lower than that of controls (p<0.05). pH values of controls was higher than that of CLA-TG treatment groups. The pH of control and CLA-TG treatment groups increased significantly as the storage period increased (p<0.05). Meat color (CIE $L^*,\;b^*$) of CLA-TG treatment groups was higher than that of controls. $a^*$ values were decreased by replacement of CLA-TG, but appeared to be unaffected by storage length. There was no significant difference in texture between controls and CLA-TG treatment groups. Based on these findings, we conclude that the physico-chemical properties and texture characteristics of manufactured pressed ham were not affected by CLA-TG addition. Also, our results indicate that high-quality pressed ham can be manufactured with CLA accumulation.

The opposite correlation between calcium ion and cyclic-AMP regarding the activation of microsomal triglyceride transfer protein in rat liver

  • In this study, the effects of $Ca^{2+}$ and cyclic adenosine monophosphate (cAMP) on microsomal triglyceride (TG) transfer protein (MTP) activity were investigated in rat liver. MTP activity was high when liver contained low levels of cAMP, which was induced by administration of glucose, or high levels of total $Ca^{2+}$ and TG. However, MTP activity increased by high levels of $Ca^{2+}$ and TG was reduced in a dose-dependent manner by treatment with dibutyryl-cAMP (db-cAMP), a cAMP analogue. Conversely, when homogenates of liver from normal rats, with low levels of total $Ca^{2+}$ and high levels of cAMP, were incubated with thapsigargin, a $Ca^{2+}$-inducer, MTP activity was increased in a dose-dependent manner compared to control. Therefore, our results suggest that high levels of $Ca^{2+}$ cause hypertriglyceridemia through the elevation of MTP activity, as opposed to high levels of cAMP, which suppress MTP activity and inhibit hypertriglyceridemia.

t10,c12 Conjugated Linoleic Acid Upregulates Hepatic De Novo Lipogenesis and Triglyceride Synthesis via mTOR Pathway Activation

  • In mice, supplementation of t10,c12 conjugated linoleic acid (CLA) increases liver mass and hepatic steatosis via increasing uptake of fatty acids released from adipose tissues. However, the effects of t10,c12 CLA on hepatic lipid synthesis and the associated mechanisms are largely unknown. Thus, we tested the hypothesis that gut microbiota-producing t10,c12 CLA would induce de novo lipogenesis and triglyceride (TG) synthesis in HepG2 cells, promoting lipid accumulation. It was found that treatment with t10,c12 CLA ($100{\mu}M$) for 72 h increased neutral lipid accumulation via enhanced incorporation of acetate, palmitate, oleate, and 2-deoxyglucose into TG. Furthermore, treatment with t10,c12 CLA led to increased mRNA expression and protein levels of lipogenic genes including SREBP1, ACC1, FASN, ELOVL6, GPAT1, and DGAT1, presenting potential mechanisms by which CLA may increase lipid deposition. Most strikingly, t10,c12 CLA treatment for 3 h increased phosphorylation of mTOR, S6K, and S6. Taken together, gut microbiota-producing t10,c12 CLA activates hepatic de novo lipogenesis and TG synthesis through activation of the mTOR/SREBP1 pathway, with consequent lipid accumulation in HepG2 cells.

Antidiabetic Effect of Ethanol Extract on Astragali Radix (황기 에탄올 추출물의 항 당뇨 효과)

  • This study was carried to investigate the antidiabetic effect of ethanol extract of Astragali Radix(A.R) in Streptozotocin(STZ) induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg dissolved in citrate buffer. The ethanol extract of A. R was orally administrated once a day for 7 days at a dose of 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in A.R treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucokinase(GK) and glucose-6-phosphate dehydrogenase(G-6-PDH) were significantly increased, and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in A.R treated group compared to the those of STZ-control group, These results indicated that ethanol extract of A.R would have antidiabetic effect in STZ-induced diabetic rats.

Anti-hyperlipidemic Effects of Bacillus strain-fermented Cheonggukjang Products in Mice

  • To evaluate the hypolipidemic effects of Cheonggukjang (CGJ), which is frequently used in Korea similar to Natto in Japan and Douchi in China like a dairy product, boiled soybeans were fermented with two Bacillus strains, B. subtilis and B. licheniforms, isolated from rice straw and their antihyperlipidemic effects of their products were investigated. Treatment with the CGJs significantly reduced blood triglyceride (TG) and total cholesterol (TC) levels and increased HDL cholesterol levels in Triton WR-1339-induced hyperlipidemic mice. The treatment of non-fermented soybeans alone also reduced blood TG and TC levels, but not significantly. Feeding the CGJs significantly lowered high blood TG and TC levels as well as body and epididymal mass weights in hyperlipidemic mice induced by the long-term feeding of a high-fat diet that increased blood HDL cholesterol levels. The B. subtilis-fermented CGJ products more potently reduced TG and TC levels, although the differences between the starters were not significant. These finding suggest that CGJ products may be effective as hypolipidemic foods by the synergistic interaction of soy and Bacillus strains.

Effect of Ginseng Saponin on Adrenaline-Induced Hyperglycemia and Hyperlipidemia (인삼사포닌이 Adrenaline에 의한 고혈당 및 고지방혈증에 미치는 영향)

  • To elucidate the effect of ginseng saponin on adrenaline-induced hyperglycemia and hyperlipidemia, ginseng saponin was administered before and after administration of adrenaline, and concentration of glucose, triglyceride(TG), and free fatty acid (FFA) as well as lipase activity in plasma was determined. 1) Glucose concentration was slightly increased by administration of ginseng saponin at 1 hour before and 10 minutes after adrenaline administration, but decreased by administration of ginseng saponin at 7 and 4 hours before adrenaline administration. TG and FFA concentration was also greatly inhibited by administration of ginseng saponin in advance. 2) When gineseng saponin 3, 10, 30mg/kg were administered 7 hours before adrenaline administration, glucose and TG concentration as well as lipase activity were inhibited in proportion to the dose of ginseng saponin, but FFA concentration was slightly inhibited. 3) It was suggested that protopanaxatriol group have potentiating effect on adrenaline induced hyperglycemia but protopanaxadiol group have inhibitory effect. And ginseng saponin seems to have strong inhibitory action on TG mobilization into blood and stimulatory effect on esterification of FFA in liver and adipose tissue.

Triglycerides increase mRNA Expression of Pro-inflammatory Cytokines Via the iNOS in Jurkat T lymphocyte and U937 Monocyte Cell Lines (Jurkat T 림프구와 U937 단핵구에서 중성지방 처리 시 iNOS를 통한 염증성 사이토카인의 mRNA 발현 증가)

  • Triglycerides (TG) are one of the triggers of chronic inflammatory lesions in the blood vessels. In the key factors in the development of inflammatory diseases, Pro-inflammatory cytokines such as tumor necrosis factor-alpha $(TNF-){\alpha}$ and interleukin-1 beta ($IL-1{\beta}$) contribute to the development of inflammatory lesions by recruiting other immune cells in the inflamed area or causing cell necrotic death. In this study, I investigated the effect of Jurkat T lymphocytes and U937 monocytes involved in vascular inflammation development on the expression of $TNF-{\alpha}$ and $IL-1{\beta}$ on exposure to TGs. In Jurkat cells, mRNA expression of $TNF-{\alpha}$ is increased by exposure to TGs. However, the expression levels of $TNF-{\alpha}$ and $IL-1{\beta}$ were increased by TGs in U937 cells. To investigate whether inducible nitric oxide synthase (iNOS) is involved in the increase of expression of $TNF-{\alpha}$ and $IL-1{\beta}$ by TGs, treatment of W1400 (an iNOS inhibitor) resulted in recovery of expression level both $TNF-{\alpha}$ and $IL-1{\beta}$. Based on the present study, it was confirmed that the expression of $TNF-{\alpha}$ and $IL-1{\beta}$ in monocytes and T lymphocytes. This increased cytokines contribute to development of vascular inflammatory lesions. In addition, iNOS is involved in the increase of $TNF-{\alpha}$ and $IL-1{\beta}$ expression by TGs.

Gender Difference in Associations between Serum Cholesterol Levels and Depression Symptoms in Healthy General Population (건강한 성인에서 성별에 따른 혈중 콜레스테롤 농도와 우울증상의 연관성 : 후향적 의무기록 조사)

  • Objectives : The purpose of this study was to determine the association between serum lipid profiles and depression according to gender difference. Methods : This retrospective cohort study included 27,452 subjects(15044 men and 12408 women) who underwent health examination. The duration was from January 2013 to December 2013. We estimate the correlation between serum lipid profile and Beck Depression Inventory(BDI) scores. We compare the effect size using beta coefficient. Results : In men, serum Triglyceride level was correlated positively with BDI scores(r=0.020, p<0.01). Serum LDL-C and HDL-C were negatively correlated with BDI scores(r=-0.015, p<0.01 ; r=-0.016, p<0.05). In women, Triglyceride level was also correlated positively with BDI scores(r=0.020, p<0.01), Serum HDL-C were negatively correlated with BDI scores(r=-0.019, p<0.01). There was no statistical significance between Serum LDL-C and Beck Depression Inventory(BDI) score. Conclusions : Both men and women had more depressive symptoms when they had low serum HDL-C level or high serum Triglyceride level. The depression symptoms were more severe when serum LDL-C level was low only in men.

Change of Fatty Acid of Non-Polar Lipid in Flint Corn During the Growth Process (경립종 옥수수의 성장과정중 비극성 지질의 지방산 변화)

  • In this study the quantity and the ratio of saturated and unsaturated fatty acids were investigated with using gas chromatography after non-polar lipid of the flint corn in the growing period was extracted with the solution of chloroform : methanol (2 : 1) and then separated by the SACC method. Neutral lipid was separated into monoglyceride(MG), 1, 3-diglyceride (1, 3-DG) , free fatty acid (FFA) , triglyceride (TG), cholesteryl ester (CE). Fatty acid of each spot was mainly composed of linoleic , oleic and palmitic acids. Lonolieic acid decreased in MG(54.5-51.4%), CE(31.3-28.9%) but increased ini TG(57.2-63.8%) during growth process. Olec acid increased in MG(25.7-29.3%), 1, 3-DG(24.7-28.9%), CE(16.7-19.9%) but decreased in TG28.6-23.1%). Palmitic acid decreased in MG(12.8-11.5%), FFA(25.7-24.1%), TG(10.4-9.3%) but increased in CE(26.4-31.5%) during a growth process. The ratio of unsaturated to saturated fatty acid in TG(7.01-7.84%) was higher in five spots of neutral lipid and in 1, 3-DG(4.61-4.16%) decreased growth process but increased in MG(5.06-5.60%), TG(7.01-7.84%).
