효소 가수분해 방법을 이용한 스피루리나 추출물의 제조 (Production of Spirulina Extract by Enzymatic Hydrolysis)

  • 세포벽 분해 효소와 단백질 분해 효소를 이용하여 스피루리나 추출물을 효율적으로 생산할 수 있는 방법을 조사하였다. 세포벽 분해 효소인 Tunicase의 사용 농도는 2%가 적당하였다. 고형분 회수율과 핵산 관련 성분의 함량을 나타내는 spiruina extraction(SE) index를 기준으로 상업용 단백질 분해 효소를 선별하였다. 일곱 종류의 효소를 조사한 결과, Esperase가 가장 우수하였으며, 최적 사용량은 2%이었다. Tunicase와 Esperase를 순차전으로 반응시키거나 동시에 반응시켜도 고형분 회수율과 SE index는 매우 유사하였으며 동시에 사용하는 것이 반응 시간을 단축시킬 수 있었다. 두 효소를 동시에 반응시키면 단순 열수 추출보다 고형분 회수율은 약 45%$(45.2%{\rightarrow}65.3%)$, SE index는 약 75%$(11.4{\rightarrow}20.0)$ 증가하였다.

Hepatotoxicity in Rats Treated with Dimethylformamide or Toluene or Both

  • The effects of toluene in dimethylformamide (DMF)-induced hepatotoxicity were investigated with respect to the induction of cytochrome P-450 (CYP) and the activities of related enzymes. The rats were treated intraperitoneally with the organic solvents in olive oil (Single treatment groups: 450 [D1], 900 [D2], 1,800 [D3] mg DMF, and 346 mg toluene [T] per kg of body weight; Combined treatment groups: D1+T, D2+T, and D3+T) once a day for three days, while the control group received just the olive oil. Each group consisted of 4 rats. The activities of the xenobiotic metabolic enzymes and the hepatic morphology were assessed. The immunoblots indicated that the expression of CYP2E1 was considerably enhanced depending on the dosage of DMF and the CYP2E1 blot densities were significantly increased after treatment with both DMF and toluene, compared to treatment with DMF alone. The activities of glutathione-S-transferase and glutathione peroxidase were either decreased or remained unaltered after treatment with DMF and toluene, whereas the lipid peroxide levels were increased with increasing dosage of DMF and toluene. The liver tissue in the D3 group (1,800 mg/kg of DMF) showed signs of microvacuolation in the central vein region and a large necrotic zone around the central vein, in rats treated with both DMF (1,800 mg/kg) and toluene (D3T). These results suggest that the expression of CYP2E1 is induced by DMF and enhanced by toluene. These changes may have facilitated the accelerated formation of N-methylformamide (NMF) from toluene, and the generated NMF may directly induce liver damage.

초음파치료가 종양세포에 미치는 효과 (Effects of Continuous Therapeutic Ultrasound on Subcutaneous Murine Melanoma)

  • The use of therapeutic ultrasound(US) in humans with malignant neoplasms has been contraindicated in physical therapy practice. Some studies have shown that results after application of US differ according to tumor type and penetration depth. The purposes of this study were to determine the effects of US on melanoma in mice and to determine treatment dosage. Twenty-four female C57BL/6 mice, age 8 weeks. The right flank of all mice was shaved, and a 0.1 ml suspension of cells was injected subcutaneously into the animals' right flank. In this study, 24 subjects were randomly divided into three groups: experimental group 1(n=8), experimental group 2(n=8), control group(n=8). In the experimental group 1, animals received continuous 3 MHZ US treatment, administered at $2.0W/cm^2$ for five minutes. In experimental group 2, animals received continuous 3 MHz US treatment, administered at $1.0W/cm^2$ for 5 minutes. The control group received the same handling as other experimental groups, including rodent chow, water, US gel application but US head pressure without the power turned on. After 10 days treatment, all mice were killed with a potassium solution. Tumors were excised and weighed on an electrical balance and fixed in a 10% neutral buffered formalin solution. Tumor weights were smaller in experimental group 2(0.3838 g) than in the control group(0.6275 g). Tumor weights of the experimental group 1(0.015 g) were smaller than those of experimental group 2. Continuous therapeutic US decreased the weight of subcutaneous melanoma tumors in mice. The treatment dosage($2.0W/cm^2$) we suggest was more effective than earlier studies on decreasing tumor size with ultrasound.

Effects site concentrations of propofol using target-controlled infusion in dental treatment under deep sedation among different intellectual disability types

  • Background: We aimed to assess the dose needed to achieve the propofol effect-site concentration using target-controlled infusion in intellectually disabled patients and to detail the most effective method for achieving a safe level of consciousness without hemodynamic changes as well as detail any resulting adverse effects. Methods: We performed a retrospective review of sedation service records of 138 intellectually disabled patients (51, mental retardation; 36, autism; 30, brain lesion, 12 genetic diseases, 9 dementia) aged over 15 years and weighing over 30 kg. These patients had received propofol via target-controlled infusion in the special care dental clinic of Seoul National University Dental Hospital from May 2008 to September 2018 for restorative treatment (112), minor surgery (13), prosthodontics (7), periodontics treatment (5), and implant (1). Results: For all groups, the duration of dental treatments was $43{\pm}18$ minutes, total sedation time was $73{\pm}23$ minutes, and total BIS values was $57{\pm}12$. The propofol maintenance dosage values for each group were: mental retardation, $3{\pm}0.5(2-4){\mu}g/ml$; autism, $3.1{\pm}0.7(2-5){\mu}g/ml;$; brain lesion, $2.8{\pm}0.7(1.5-5){\mu}g/ml;$; genetic disease, $2.9{\pm}0.9(1-4){\mu}g/ml;$ and dementia $2.3{\pm}0.7(1-3.4){\mu}g/ml;$. Conclusions: The dementia group needed a lower dosage to reach a safe, effective propofol effect-site concentration than the other groups. Since there were no complications, deep sedation is a great alternative to general anesthesia for dental treatment of intellectually disabled patients.

  • A 14-year-old, spayed female Labrador Retriever breed dog with housesoiling problem was presented to the Animal Behavior Clinic, College of Veterinary Medicine, Gyeongsang National University. Housesoiling problem was diagnosed as geriatric incontinence based on the questionnaire and interview on the dog. Pharmacological treatment was recommended. Phenylpropanolamine was prescribed at a dosage of 1.19 mg/kg PO q48h for 3 weeks. Housesoiling problem caused by geriatric incontinence disappeared after the treatment.

Clinical application of gonadotropin-releasing hormone analogs in children and adolescents

  • Although the increasing incidence of central precocious puberty (CPP) in Korea has recently raised public concerns about health and growth problems, there are many areas of uncertainty regarding the pathogenesis, diagnosis, and management of CPP. In this paper, we review the definition of precocity, the assessment of CPP, and the hormonal abnormalities that support the diagnosis. In addition, we review the practical guidelines regarding the clinical use of gonadotropin-releasing hormone analogs in children with CPP. Indications for treatment, determination of dosage, monitoring during treatment, and discontinuation of therapy are discussed.

연속식 오존처리공정을 이용한 낙동강 상수원수의 정수처리 효과 (Effect of Water Treatment for Nakdong River Raw Water by Continuous Ozone Process)

  • 낙동강 상수원수를 대상으로 연속적 오존처리에서 오존주입농도에 따른 유기물 분해 특성을 조사하기 위하여 오존처리에 의한 DOE성분변화, UV254/DOC의 변화와 소비오존에 따른 DOC 제거효율을 조사한 결과는 다음과 같다. 유입원수의 수질변화에 따라 다소 차이는 있었으나 전반적으로 오존주입농도가 증가할수록 DOC 처리효율도 증가되었다. 오존처리에 따른 DOC 성분변화는 전오존을 3 mg/L 농도로 처리시 TDOC, ADOC, A &BDOC 및 NRDOC는 각각 0.6, 0.2, 0.1 및 1.0 mg/L 정도 오존처리에 의하여 감소되었으나 BDOC는 0.8 mg/L 증가하였다. UV254/DOC의 분율은 오존주입농도를 1, 3 및 5 mg/L처리하였을 때 전오존의 경우는 각각 0.048, 0.044 및 0.037 이였고, 후오존의 경우는 각각 0.018, 0.015 및 0.012로 나타나 오존주입농도가 증가할수록 감소하는 경향을 보였다. 초기 DOC mg당 소비되는 오존량에 대한 DOC 제거율은 전오존과 후오존처리의 초기 DOC에 대한 소비오존량이 각각 약 1.5와 2.3일 때 각각 25%와 32%로 서 최대의 DOC 처리효율을 나타내었다. 낙동강 상수원수에 전오존을 처리할 경우 오존주입량은 초기 DOC mg당 소비된 오존이 1.5 mg 이내의 농도가 되도록 주입하구 후오존은 유입수인 모래여과수의 초기 DOC mg당 소비된 오존이 2.3 mg 이내의 농도가 되도록 조절하여 주입하면 적당할 것으로 생각되었다.

Clozapine과 Risperidone에서 Olanzapine으로 교체 연구 : 12개월 추적연구 (Switch to Olanzapine from Clozapine or Risperidone and 12-months Follow Up)

  • In clinical setting, treatment-refractoriness, medication induced tardive dyskinesia and amenorrhea in chronic schizophrenia are frequently problematic. However, there are few guideline solving these problem available to clinicians. The goal of this study was collecting clinical data on clinical effectiveness and predictors of response of switching to olanzapine. We attempted to switch to olanzapine from risperidone and clozapine in chronic 31(risperidone 17, clozapine 14) schizophrenia and schizoaffective disorder patients suffering from sustained symptoms, weekly blood monitoring, medication induced tardive dyskinesia and amenorrhea. Previous antipsychotics dosage was gradually decreased for 2 or 3weeks, at the same time olanzapine dosage was gradually increased. At baseline, after 1 week, after 2 weeks and after 4 weeks we checked Brief Psychiatric Rating Scale, Clinical Global Impression Scale, Sympson-Angus Rating Scale, Barnes Akathisia Rating Scale and followed up after 12 months. Successful switch after 4 weeks was achieved in 25 patients(clozapine 9(64.2%), risperidone 16(94.1%)). Overall, mean BPRS and CGI scores increased significantly. Successful maintenance after 12 months was achieved in 17 patients(clozapine 5(35.7%), risperidone 12(70.5%)). Overall, mean BPRS and CGI scores increased significantly too. Switching to olanzapine from other atypical antipsychotics is recommendable in chronic schizophrenia with treatment refractoriness and drug induced side effect.

5-HT 수용체 길항제를 이용한 파킨슨 환자의 정신 증상의 치료 (Psychosis in Parkinson's Disease-Treatment with 5-HT Receptor Antagonist)

  • Current treatment strategies for levodopa-induced psychosis in advanced Parkinson's disease have had limited success. Reduction or discontinuation of levodopa and coadministration with dopamine-blocking neuroleptics may attenuate the psychotic symptoms, but these strategies are associated with worsening of parkinsonian symptoms. Administration of 5-HT3 receptor antagonist ; ondansetron, a newer strategy to attenuate psychosis of Parkinson'disease without motor deterioration was introduced. A 41-year-old young-onset male, who was diagnosed as Parkinson's disease 7 years ago, was treated with levodopa therapy, and had levodopa-induced psychosis(delusion, hallucination, paranoid, insomnia). After trial of ondansetron, he showed improvement in the Brief Psychiatric Rating Scale(from 21 points to 9 points) in spite of increasing the dosage of levodopa. With ondansetron, we could increase the dosage of levodopa without psychotic complications(esp, hallucination), and he showed improvement in the motor fluctuation.

근로자의 건강보호를 위한 알릴 염화물의 포유류 배양세포 염색체이상시험 (In Vitro Mammalian Chromosomal Aberration Test of Allyl Chloride for Workers' Health)

  • Objectives: Chemical hazard evaluations are important for workers' health and working environments. Allyl chloride (CAS No. 107-05-1) is used in many industries, leading to concerns about the possibility of threats to the health of workers. Since only insufficient or controversial information is available about potential related hazards, an in vitro mammalian chromosomal aberration (CA) assay was conducted in order to gain additional information concerning any such hazards. Moreover, toxicological information from this study could be applied for workers' rights to know, and to prepare or update the Materials Safety Data Sheet (MSDS) for a number of industries. Methods and Results: The assay was performed using the Chinese hamster lung fibroblast cell (ATCC, CRL-1935), by the direct method (-S9) and by the metabolic activated method (+S9 mix). Using the direct method, the seven dosages in the 48-hour treatment group did not show that the frequency of CA is proportionate to the dosage. The frequency of CA is not proportionate to the dosage addition for a six-hour treatment using the metabolic activated method. Conclusions: From these findings, it was decided that this chemical does not induce chromosomal aberrations under the tested conditions.
