It is generally known that mutations in any of several genes encoding photoreceptor-specific proteins have resulted in retinitis pigmentosa (RP), a disease characterized by losing photoreceptor function with progressive degeneration of photoreceptor cells and eventually leading to blindness. To study the procure and cure of photoreceptor degeneration, we produced transgenic mice. Transgene consisted of a 12.5kb genomic DNA fragment that contains mutated pig rhodopsin gene (Pro-347-Ser) including both the 5'-franking (4.0 kb) and the 3'-franking (2.9 kb) sequences. This gene was used for the production of transgenic mouse. The mutated rhodopsin DNA was microinjected into male pronuclei of fertilized mouse (C57BL /6]) embryos. We detected transgenic animals harboring mutated rhodopsin gene by PCR and Southern blot analysis. These transgenic mice showed stable transmission of microinjected rhodopsin gene into their offspring. Therefore these animals will provide a novel approach to study the mechanism of the photoreceptor degeneration and be provided as a disease model for the treatment of the blind in human.
The objectives of this study are to investigate soil contamination in the vicinity of abandoned Au-Ag mine and to apply a remediation technique of liming to tailings. In the study area of the Imcheon Au-Ag mine, soils were sampled in and around the mine the analyzed by Atomic Absorption Spectrometry extracted by both 0.1N HCl and aqua regia. Elevated levels of Cd, Cu, Pb and Zn concentrations extracted by 0.1N HCl were found in soils taken from tailings site. These high contents directly influenced metal concentrations in soils taken in the vicinity of the site. This is mainly due to clastic movement by wind and effluent of mine waste water. In addition, relatively enriched concentrations of the metals were found in soils extrated by aqua regia due to strong decomposition of the samples compared with 0.1N HCl extration. According to the statistical approach, metal concentrations in soils by 0.1N HCl had a positive correlation with those by aqua regia extraction. Mine waste waters and stream waters were also sampled around the mine in spring and summer and analyzed by AAS for Cd, Cu, Pb and Zn, and by Ion Chromatography for anions. Like soils developed over tailings, significant levels of metals and sulphates were found in the mine waste waters ranging of 0.2~0.3, 0.5~2.0, 0.2~2.8, 30~50 and 1,240~4,700 mg/l of Cd, Cu, Pb, Zn and $SO_4^{2-}$, respectively. These elevated levels influenced in the stream waters in the vicinity of the tailings site. In seasonal variation of metal and anion contents, relatively high levels were found in waters sampled on summer due to leaching the metals and anions from tailings by rain. This study also examined the possibility of lime treatment for remediation of acid mine tailings and assumed to be 46 tones of pulverized lime for neutralization of the tailings.
The purpose of this study was to classify the courses of the dental-hygiene curricula into several categories by field, to incorporate the subjects in the same category into an integrated course, and to suggest how to ensure the successful phase-in application of integrative education according to Ronald M. Harden's 11-stage integrative ladder model. The findings of the study were as follows: 1. When the existing curricula were analyzed, it's found that many credits were provided to the courses in the area of basic dentistry that offered both theory and practice. In particular, the subjects tested by the national examination were offered by every college. In the field of public oral health, the largest number of credits was allocated to theory of oral prophylaxis and practice courses. In clinical area, clinical practice, in the area of dental office management, dental insurance course was given the largest credit. There were 31 to 61 major subjects in the colleges, which indicated that the major subjects were segmented in detail. 2. It seemed necessary to incorporate the subjects in the field of basic dentistry into oral biology, and theory of oral prophylaxis/practice, preventive dentistry/practice, preclinical stage, emergency treatment and introduction to dental hygiene should be integrated in to clinical dental hygiene. The courses in clinical area should be combined into clinical dentistry, and in the field of medical management, dental insurance, hospital management and medical relation law should be incorporated into management of dental clinic. 3. In the 11-stage integrative ladder, the subjects in the same field could perfectly be incorporated as the stages advanced. Each of the subjects was less emphasized, and communication and joint plans among teachers who were respectively in charge of the courses were increasingly considered important. Specifically, there should be a consensus among the teachers in regard to the outline of teaching programs, order of education. objects and objectives of programs and what and how to evaluate.

The publication of books on modern agriculture started under the influence of Silhak Thought developed from the late Chosun Dynasty. The common intention found in these publications is the objective to enrich the country through increased agricultural productivity and thus to secure national independence from the surrounding powers. The study reviews three different categories in this area; five general books on agriculture, three on agriculture related legal regulations, and ten books introducing techniques of commercial agriculture. The first category is comprehensive treatment of general agricultural content and the second is legal regulations which affected the contemporary agriculture. The third category is introductory books on commercial techniques aimed at accumulating wealth through agriculture. Silkworm cultivation occupies an important place in these publications owing to the active encouragement given by the Section of Silkworm Cultivation in the Ministry of Agriculture, Commerce and Industry. We can recognise the extensive involvement of Suh Pyung Sook, who served as the Section Chief of Silkworm Cultivation in 1904, by examining introductions and prefaces of many books in this category. The examples of general books on agriculture are 'New Agricultural Administration' (농정신편), 'General Introduction on Agiculture' (농업대요), 'New Textbook on Agricuture' (신찬농업교과서), 'Pragmatic Agriculture' (실리농방신편), Of these, 'New Agricultural Administration' (농정신편) is evaluated as the first publication on modern agriculture. It was written in 1881, and the first edition was published was published in 1901 with the second edition following in 1905. Examples of the second category are; 'The regulation on utilizing uncultivated state owned land' (국유미간지리용법) legislated and declared with the intention of expanding cultivated area, 'Mining and Forestry Regulation' (광임법규), 'Forest Land Regulation' (임야법령) to administer mining and forestry. Books on commercial agriculture take up the highest proportion in the agricultural publication of this period. These cover silkworm cultivation, chicken farming, vegetables and fruits. The books introduce Western techniques with more scientific and rational approach especially on Silkworm cultivation and chichen farming which had become increasingly significant as commercial agriculture from the late Chosun Dynasty.
Dehydroxylation and mullitization of primary and secondary kaolins were investigated in order to compare and understand the differences in thermal behaviours by DTA and TG up to $1,100^{\circ}C$. Chemical analyses and EPMA of the samples revealed nearly ideal unit-cell formulae of kaolins. The weight losses of dickite and halloysite are 14% and 12.5% on the average, respectively. The activation energies of dehydroxylation of kaolin minerals were calculated according to Kissinger's approach which uses various heating rates in DTA to estimate the activation energy of thermal reactions. The activation energies of dehydroxylation of halloysites from Daemoung and Buksam mines are about $163kJmor^{-1}$ (white), $168kJmor^{-1}$ (pink), and $176kJmor^{-1}$ respectively. The activation energies of dickites collected from Sungsan and Ogmae mines are about $166kJmor^{-1}$ and $387kJmor^{-1}$. The asymmetric shape of endothermic peak in DTA, the relative intensities of OH-stretching bands in FTIR spectroscopy and the existence of residual XRD peaks of the samples which were heated at $550^{\circ}C$ for 2 hours indicate that Sungsan dickite may be more disordered than Ogmae dickite. The new phase formed in thermally treated samples in the range of $900^{\circ}C$ to $1,100^{\circ}C$ was identified as mullite by XRD on the basis of disappearing of the characteristic peaks of kaolins and increasing of amorphous background upon heat treatment. On further heating, loss of more water from dehydroxylate resulted in the formation of mullite and the characteristic X-ray diffraction patterns of mullite began to appear at about $900{\sim}1,000^{\circ}C$ in kaolins.

This study was conducted in order to estimate the exhaust emissions analysis method of the real driving emission(RDE). The Association for Emissions Control by Catalyst(AECC) has developed a test procedure by using a random cycle method based on the chassis dynamometer. In order to confirm this approach in Korea, Euro 5(DPF), Euro 6(DPF + LNT), and Euro 6(DPF + SCR) were performed on three different vehicles to determine the exhaust gas characteristics of the random cycle, real-road driving test(PEMS), and emission certification driving mode(NEDC). Six different random cycle driving modes were generated by the vehicle specifications(e.g. curb weight, engine power, gear ratio, and maximum acceleration). The NOx emissions were increased in the NEDC, random cycle, and PEMS order in this study regardless of the test vehicles. The random cycle method has the advantage because it utilizes a chassis dynamometer in the laboratories for a repeatable data collection, and it allows any eminent emission improvement checked prior to a real-road driving test with PEMS.

Defatted bovine liver (DBL) is a potential source of protein and minerals. Supercritical carbon dioxide ($SC-CO_2$) and a traditional organic solvent method were used to remove lipid from bovine liver, and the quality characteristics of a control bovine liver (CBL), bovine liver defatted by $SC-CO_2$ ($DBLSC-CO_2$) at different pressures, and bovine liver defatted by organic solvent (DBL-OS) were compared. The $DBLSC-CO_2$ samples had significantly higher (p<0.05) protein, amino acid, carbohydrate, and fiber contents than CBL and DBL-OS. There was a higher yield of lipid from CBL when using $SC-CO_2$ than the organic solvent method. SDS-PAGE analysis demonstrated that the CBL and $DBLSC-CO_2$ had protein bands of a similar intensity and area, whereas DBL-OS appeared extremely poor bands or no bands due to the degradation of proteins, particularly in the 50 to 75 kDa and 20 to 25 kDa molecular weight ranges. In addition, $DBLSC-CO_2$ was shown to have superior functional properties in terms of total soluble content, water and oil absorption, and foaming and emulsification properties. Therefore, $SC-CO_2$ treatment offers a nutritionally and environmentally friendly approach for the removal of lipid from high protein food sources. In addition, $SC-CO_2$ may be a better substitute of traditional organic solvent extraction for producing more stable and high quality foods with high-protein, fat-free, and low calorie contents.
Pectinesterase(PE) has been definitively established as the causative agent for clarification of citrus juice and gelation of frozen concentrates. This enzyme is present in multiple forms in citrus fruit. Although representing a minor fraction of the total PE activity, thermostable pectinesterase(TSPE) accounts for the severe time/temperature processing treatment required to inactivate PE. This study was undertaken to separate and purify thermolabile pectinesterase(TLPE) and TSPE from the crude PE extracts of Valencia orange rag-pulp powder. The approach taken was to carry out the three kind of chromatographies of CM-Sephadex C-50, Phenyl Sepharose CL-4B and CM-Biogel A to the unheated crude PE or the heated crude PE. All of them used could increase the purity of PEs and, specially, Phenyl Sepharose CL-4B chromatography could separate crude PE as the mixture of PEs into two forms of TLPE and TSPE. The purified TLPE had specific activity of 1,005 units/mg, yield of 13.6% and purification of 35 fold, while the TSPE separated from the unheated crude PE showed specific activity of 3,115 units/mg, yield of 1.5% and purification of 100.5 fold, and another TSPE from the heated crude PE was found to be specific activity of 1,803 units/mg, yield of 15.4% and purification of 140 fold.
The Activity Based Costing(ABC) means the process that makes clear how the actions and input resources have changed into service to calculate medical services costs. These days, the number of hospital which is using the ABC system is increasing to make their policy decision making efficient and run the hospitals more resonable. This study analyzes the unbalance in the level of health insurance service fee and the improvement plans based from 8 hospitals(ABC system) and 95 clinics(ABC survey). The cost recovery ratio has shown different levels according to each service type. A surgery service type recorded 76.8% and an evaluation & management service type is 84.6%, a treatment procedure type(85.8%), a function test type(91.6%) and health insurance fee even did not reach to the original cost. Meanwhile, a laboratory test type and imaging test type show high level of cost recovery ratio. they recorded 188.3% and 158.8%. Resultingly now of unbalance in the level of health insurance service fee accelerates supply of every test. so there is a need to make laboratory test type and imaging test type lower to keep balance with the surgery and medical service. These methods should be performed gradually with monitoring the unbalance fee ratio and for this, a panel medical institution have to be established for generalizations of studying result, fairness of selecting researching sample.

Laparoscopic proximal gastrectomy (LPG) is theoretically a superior choice of minimally-invasive surgery and function-preserving surgery for the treatment of proximal early gastric cancer (EGC) over procedures such as laparoscopic total gastrectomy (LTG), open total gastrectomy (OTG) and open proximal gastrectomy (OPG). However, LPG and OPG are not popular surgical options due to three main concerns: the first, oncological safety; the second, functional benefits; and the third, anastomosis-related late complications (reflux symptoms and anastomotic stricture). Numerous recent studies have concluded that OPG and LPG present similar oncological safety profiles and improved functional benefits when compared with OTG and LTG. While OPG with modified esophagogastrostomy does not provide satisfactory results, OPG with modified esophagojejunostomy showed similar rates of anastomosis-related late complications when compared to OTG. At this stage, no standard reconstruction method post-LPG exists in the clinical setting. We recently showed that LPG with double tract reconstruction (DTR) is a superior choice over LTG for proximal EGC in terms of maintaining body weight and preventing anemia. However, as there is no definitive evidence in favor of LPG with DTR, a randomized clinical trial comparing LPG with DTR to LTG was recommended. This trial, the Korean Laparoscopic Gastrointestinal Surgery Study-05 (NCT01433861), is expected to assist surgeons in choice of surgical approach and strategy for patients with proximal EGC.
