Phylogenetic Analysis by RFLP and Sequencing of Mitochondrial DNA in a Korean Population

  • Analysis of molecular nature of mitochondrial DNA (mtDNA) could be powerful marker for anthropological studies of modern populations. While population genetic studies on mtDNA have been reported for several ethnic groups, no such study has been documented for the Korean population. We surveyed mtDNA polymorphisms in the HVS I of noncoding D-loop region and its upstream region from 430 unrelated healthy Korean population by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and direct sequencing analysis. PCR product with 2,790 bp spanning the specific mtDNA region (mt13715-16504) was subjected to RFLP analysis using 6 restriction enzyme (Hinf I, Hae III, Alu I, Dde I, Mbo I, Rsa I). On the PAUP analysis of PCR-RFLP results, 38 mtDNA haplotypes (Hap 1-38) were detected in the Korean populations, which were classified into 11 haplogroups (Grp 1-11) of related haplotypes encompassing all 38 haplotypes. In comparison of sequencing data with Anderson's reference sequence, the transition type was more prevalent than the transversion type. Insertions or deletions were not found. In addition, three of the polymorphic sites (A16240C, A16351G, G16384A) in HVS-I region are determined newly. The polymorphic sites were distributed randomly in the region, though the frequency at each site was variable. Thus, this research might be required for the genealogical study of Orientals.

Cost of Illness of Chronic Disease by Region in Korea (3대 만성질환자의 지역별 의료비 분석)

  • Background: With the recent aging of the population, the transition to a disease structure centered on chronic diseases is accelerating. Moreover, the socio-economic gap and the polarization of the health gap between regions further increase the burden of disease on the country. Accordingly, this study calculated the disease cost of hypertension, diabetes, and hyperlipidemia, which are the three major chronic diseases, to establish an effective health promotion policy strategy for each region, and analyzed the gap in disease cost within the region to determine health determinants at the individual as well as the regional level. Methods: This study utilized data from the 2015 sample cohort of the National Health Insurance Service and calculated the disease cost of patients (diabetes: I10-I15, hypertension: E10-E14, hyperlipidemia: E78) based on the main diagnosis. Results: Based on our analysis, the case of medical use in cities and provinces was higher than in metropolitan cities, with relatively small medical use in Seoul and Gangwon-do. In terms of the disease cost, the cost of chronic diseases in Seoul and Jeju was the highest, but the difference in disease cost between patients in each region was the largest in Seoul and Gangwon-do. Conclusion: The results of this study provide meaningful data for implementing efficient health promotion policies by analyzing the differences in disease cost and identifying health determinants in different regions. Furthermore, in Korea, where socioeconomic differences are clearly revealed, it can be used as a basis for preparing a strategic plan, from a long-term perspective, to improve the health of patients with chronic diseases in the future.

LINC00174 Facilitates Proliferation and Migration of Colorectal Cancer Cells via MiR-3127-5p/ E2F7 Axis

  • The literature indicates that LINC00174 promotes the growth of colorectal cancer (CRC) cells, but its research needs to be enriched. We tried to explore the function and mechanism of LINC00174 in CRC cell proliferation and migration. Bioinformatics analysis predicted the binding relationship and expressions of lncRNA, miRNA and mRNA. Clinical study analyzes the relationship between LINC00174 and clinical data characteristics of CRC patients. The expressions of LINC00174, miR-3127-5p and E2F7 were verified by RT-qPCR, and the combination of the two was verified by dual luciferase analysis and RNA immunoprecipitation as needed. Western blot was used to detect the expression of EMT-related protein and E2F7 protein. Functional experiments were used to evaluate the function of the target gene on CRC cells. LINC00174 was up-regulated in CRC clinical samples and cells and was related to the clinical characteristics of CRC patients. High-expression of LINC00174, contrary to the effect of siLINC00174, promoted cell viability, proliferation, migration and invasion, up-regulated the expressions of N-Cadherin, Vimentin, E2F7, and inhibited the expression of E-Cadherin. MiR-3127-5p was one of the targeted miRNAs of LINC00174 and was down-regulated in CRC samples. In addition, miR-3127-5p mimic partially reversed the malignant phenotype of CRC cells induced by LINC00174. Besides, E2F7 was a target gene of miR-3127-5p, and LINC00174 repressed miR-3127-5p to regulate E2F7. Our research reveals that LINC00174 affected the biological characteristics of CRC cells through regulated miR-3127-5p/ E2F7 axis.

Textbooks and Textiles: Fashion in East Asia, 1920-1945

  • From the 1920s to 1945, East Asia experienced radical social change with the introduction of new fashion styles, and new ways of thinking, from the West. The emergence of a new generation of "new women" educated in modern schools was part of this phenomenon, and functioned as a trend-setting influence in East Asian society. In schools, education in dressmaking, sewing, and home economics were important parts of female education. Adopting a new fashion style is, by necessity, accompanied by the new technology of dressmaking. Given that ready-made clothing was not generally available, dressmaking education also served to introduce a new material culture. In Korea and Taiwan under Japanese colonization, the greater part of school curricula and textbooks mirrored those in Japan, which enabled these countries to develop and adopt transnational styles as well as local styles. This research explores the transition of women's fashion in East Asia in modern and colonial conditions from the 1920s to the 1940s by analysing curricula and textbooks on dressmaking in comparison with the prevailing styles in each region. This is expected to suggest the impact of modernity in East Asia and the transnational styles of fashion in colonial Korea and Taiwan, as well as Japan, developed within the local culture. Colonial conditions are also discussed in terms of their impact and limitations in the transition of styles.

Numerical Prediction of Unsteady Transitional Boundary Layer Flows due to Rotor-Stator Interaction(II)-Characteristics of Unsteady Transitional Boundary Layer Flow- (정익과 동익의 상호작용에 의한 비정상 천이 경계층 유동의 수치해석에 관한 연구 (II))

  • A Navier-Stokes code with a modified low Reynolds number k-.epsilon. turbulence model was used to study the unsteady transitional boundary layer flow due to rotor-stator interaction. The modification, proposed by Launder, to improve prediction of stagnation flows was incorporated to the low Reynolds number k-.epsilon. turbulence model by Fan-Lakshminarayana-Barnett. Numerical solution is shown to capture well the calmed laminar flow as well as the wake induced transitional strip due to rotor-stator interaction and shows improvement, in terms of onset of transition and its length, over previous Euler/boundary layer solution. The turbulent kinetic energy shows local maximum along the upstream rotor wake in the wake induced transitional strip and this characteristics is observed untill the end of transition. The wake induced strip also shown apparent even in the laminar sublayer as the upstream rotor wake penetrates inside the boundary layer.

FLOW CONTROL OF SMART UAV AIRFOIL USING SYNTHETIC JET (Synthetic jet을 이용한 스마트 무인기 익형 주위의 유동 제어)

  • In order to reduce the download around Smart UAV(SUAV) at hovering and transition mode, flow control using synthetic jet has been performed. Many of the complex tilt rotor flow features are captured including wing leading and trailing edge separation, and the large region of separated flow beneath the wing. First, in order to control the trailing edge separation, synthetic jet is located at 30, 95% of flap chord length. The flow control using synthetic jet on flap shows that stall characteristics depending on several mode can be improved through separation vortices resizing. Also, a flap jet and a 0.01c jet which control the separation efficiently are applied at the same time at each test case because controlling the leading edge separation is essential for download reduction. As a result, time averaged download is reduced about 18% comparing with no control case at hovering mode and 48% at transition mode. These research results show that if flow control using leading edge jet and trailing edge jet is used effectively to the SUAV in overall flight mode, flight performance and stability can be improved.

Effects of Freestream Turbulence Intensity on the Flow Past a Circular Cylinder (원형단면 실린더를 지나는 유동에 대한 자유류 난류강도의 영향)

  • In this study, the effects of freestream turbulence intensity on laminar-turbulent transition of separated shear layers in the wake of a circular cylinder are investigated using an immersed boundary method and LES. It is shown that the present numerical results without freestream turbulence for Re=3,900 based on bulk mean velocity and the cylinder diameter are in good agreement with other authors' experimental observations and numerical results, verifying our numerical methodology. Then a 'prescribed power spectrum' method is imposed to generate isotropic turbulence at the inlet of the computational domain at each time step. The principal effects of freestream turbulence intensity on flow statistics are investigated for Re=3,900. Statistical study reveals that the Reynolds stresses in the near-wake region gradually increase, and transition occurs further upstream, as the turbulence intensity increases. On the other hand, the bubble size behind the cylinder decreases as the turbulence intensity increases, which indicates that the freestream turbulence helps mean velocity be quickly recovered.

EFFECT Of PYRROLIDONE DERIVATIVES ON MULTILAMELLAR LIPOSOMES OF STRATUM CORNEUM LIPID: A STUDY BY UV SPECTROSCOPY AND DIFFERENTIAL SCANNING CALORIMERY

  • In order to elucidate the mechanism of action of transdermal absorption-enhancing compounds, i.e., pyrrolidone derivatives (2-pyrrolidone, 1-methyl-2-pyrrolidone, 1-ethyl-2-pyrrolidone, 1,5-dimethyl-pyrrolidone and 5-methyl-2-pyrrolidone), multilamellar liposome was prepared from the simulated stratum corneum lipid and employed as a model system for the barrier function of the stratum corneum. The liposomal membrane of the stratum corneum lipid liposome (SCLL) behaves as an osmometer and has an excellent barrier function. In addition, its phase transition temperatures are similar to those of human stratum corneum intercellular lipid region. Therefore, SCLL seems to be a useful skin model. To estimate the barrier function of SCLL, the osmotic behavior of SCLL was measured in the presence of pyrrolidone derivatives and the effect on the phase transition temperature of SCLL was also investigated using differential calorimetry. Above a certain concentration (MLAC), enhances perturb the barrier function of the liposome. The relationship between MLACs and the partition coefficient of the pyrrolidone derivatives was observed; the greater the partition coefficients, the smaller the MLAC. This suggests that the more hydrophobic enhancers penetrate into the lipid layer more easily and reduce the barrier function of membrane more effectively. The results of differential scanning thermograms of the SCLL suggest that the pyrrolidone derivatives had incorporated into the lipid layer in the liposome and increased the fluidity of the lipid layer in the liposome. Such activity might have some correlation with the transdermal absorption-enhancing activity these compounds.

  • The changing process from hovering mode to transition one is of importance to determine a stability of tilt-rotor aircraft, which is utilized in UAV(Unmaned Aerial Vehicle). The analysis on fluid flows and aerodynamic characteristics according to variation of tilting angle of rotor is essential step in development of tilt-rotor. In the present study, the computation domain is divided into the rotating and stationary regions in order to consider the rotating blades. For the convenient realization of various tilting angle as well as application of boundary condition, the whole computation region is constructed into sphere domain. The near farfield boundary condition is adopted. The airfoil used in computation is NACA 0012. The computation results for the hovering mode are validated by comparing with previously conducted experimental results. From the results, the flow fields around rotor blade and the aerodynamic characteristics in transition mode are observed. The computational result will provide the basis for development and performance evaluation of tilt-type aircraft.

A Study on the Axial Velocity Profile of Developing Laminar Flows in a Straight Duct Connected to a Square Curved Duct (정사각단면 곡관덕트에 연결된 직관덕트에서 층류유동의 속도분포)

  • In the present study, characteristics of steady state laminar flows of a straight duct connected to a 180$^{\circ}$ curved duct were examined in the entrance region through experimental and numerical analyses. For the analysis, the governing equations of laminar flows in the Cartesian coordinate system were applied. Flow characteristics such as velocity profiles, and secondary flows were investigated numerically and experimentally in a square cross-sectional straight duct by the PIV system and a CFD code(STAR CD). For the PIV measurement, working fluid produced from mosquito coils smoke. The experimental data were obtained at 9 points dividing the test sections by 400 mm. Experimental and numerical results can be summarized as follows. Critical Reynolds number, Recr which indicates transition from laminar steady flow to transition steady flow was 2,150. As Reynolds number, Re, was increased, dimensionless velocity profiles at the outer wall were increased due to the effect of the centrifugal force and the secondary flows. The intensity of a secondary flow became stronger at the inner wall rather than the outer wall regardless of Reynolds number.
