Optimal condition for efficient DNA transfer in filamentous cyanobacteria by electroporation

  • Filamentous cyanobacteria are an ecologically important group of bacteria because they are able to provide both organic carbon fixed nitrogen that can support the nutritional requirements for other microorganisms. Because of their prokaryotic nature, they can also be used as potentially powerful model systems for the analysis of oxygenic photosynthesis and nitrogen fixation. Gene transfer is an indispensable procedure for genetic analysis of filamentous cyanobacteria. Electroporation was used to introduce foreign DNA into cyanobacterial cells. In experiments designed to optimize the electroporation technique, the effects of the field strength (amplitude of pulse) and time constant (duration of pulse), DNA concentration and host restriction/modification of DNA on the efficiency of electro-transformation were investigated. The results of this research revelaed that a high voltage pulse of short duration was effective for the electro-transformation of Anabaene sp. M131. The maximal number of transformants was obtained at 6 kV/cm with a pulse duration of 5 msec. The efficiency of electro-transformation was also sensitive to concenetration of DNA; even small amounts of DNA (0.01 .mu.g/ml) were able to gie a large number of transformants (1.0 * 10$\^$3/ cfu/ml).

Adventitial Fibroblast Abormality in Thoracic Aortic Aneurysms and Aortic Dissections

  • Background: Development of thoracic aortic aneurysms and aortic dissections (TAAD) is attributed to unbearable wall tension superimposed on defective aortic wall integrity and impaired aortic repair mechanisms. Central to this repair mechanisms are well-balanced and adequately functional cellular components of the aortic wall, including endothelial cells, smooth muscle cells (SMCs), inflammatory cells, and adventitial fibroblasts. Adventitial fibroblasts naturally produce aortic extracellular matrix (ECM), and, when aortic wall is injured, they can be transformed into SMCs, which in turn are involved in aortic remodeling. We postulated the hypothesis that adventitial fibroblasts in patients with TAAD may have defects in ECM production and SMC transformation. Materials and Methods: Adventitial fibroblasts were procured from the adventitial layer of fresh aortic tissues of patients with TAAD (Group I) and of multi-organ donors (Group II), and 4-passage cell culture was performed prior to the experiment. To assess ECM production, cells were treated with TNF-${\alpha}$ (50 pM) and the expression of MMP-2/MMP-3 was analyzed using western blot technique. To assess SMC transformation capacity, cells were treated with TGF-${\beta}1$ and expression of SM ${\alpha}$-actin, SM-MHC, Ki-67 and SM calponin was evaluated using western blot technique. Fibroblasts were then treated with TGF-${\beta}1$ (10 pM) for up to 10 days with TGF-${\beta}1$ supplementation every 2 days, and the proportion of transformed SMC in the cell line was measured using immunofluorescence assay for fibroblast surface antigen every 2 days. Results: MMP-3 expression was significantly lower in group I than in group II. TGF-${\beta}1$-stimulated adventitial fibroblasts in group I expressed less SM ${\alpha}$-actin, SM-MHC, and Ki-67 than in group II. SM-calponin expression was not different between the two groups. Presence of fibroblast was observed on immunofluorescence assay after more than 6 days of TGF-${\beta}1$ treatment in group I, while most fibroblasts were transformed to SMC within 4 days in group II. Conclusion: ECM production and SMC transformation are compromised in adventitial fibroblasts from patients with TAAD. This result suggests that functional restoration of adventitial fibroblasts could well be a novel approach for the prevention and treatment of TAAD.

  • This study, after careful consideration on Piaget's structuralism, showed the relationship between Bourbaki's matrix structure of mathematics and Piaget's structure of mathematical thinking. This, studying the basic characters that structure of knowledge should have, pointed out that 'transformation' and to it, too. Also it revealed that group structure is a 'development' are essential typical one which has very important characters not only of mathematical structure but also general structure, and discussed the problem that learners construct the group structure as a mathematical concept.

Limited Impact of Music Therapy on Patient Anxiety with the Large Loop Excision of Transformation Zone Procedure - a Randomized Controlled Trial

  • Background: Many studies have pointed to strategies to cope with patient anxiety in colposcopy. Evidence shows that patients experienced considerable distress with the large loop excision of transformation zone (LLETZ) procedure and suitable interventions should be introduced to reduce anxiety. This study aimed to investigate the effects of music therapy in patients undergoing LLETZ. Materials and Methods: A randomized controlled trial was conducted with patients undergoing LLETZ performed under local anesthesia in an out patient setting at Ramathibodi Hospital, Bangkok, Thailand, from February 2015 to January 2016. After informed consent and demographic data were obtained, we assessed the anxiety level using State Anxiety Inventory pre and post procedures. Music group patients listened to classical songs through headphones, while the control group received the standard care. Pain score was evaluated with a visual analog scale (VAS). Statistical analysis was conducted using Pearson Chi-square, Fisher's Exact test and T-Test and p-values less than 0.05 were considered statistically significant. Results: A total of 73 patients were enrolled and randomized, resulting in 36 women in the music group and 37 women in the non-music control group. The preoperative mean anxiety score was higher in the music group (46.8 VS 45.8 points). The postoperative mean anxiety scores in the music and the non-music groups were 38.7 and 41.3 points, respectively. VAS was lower in music group (2.55 VS 3.33). The percent change of anxiety was greater in the music group, although there was no significant difference between two groups. Conclusions: Music therapy did not significantly reduce anxiety in patients undergoing the LLETZ procedure. However, different interventions should be developed to ease the patients' apprehension during this procedure.

  • Ginsenoside Rg3 (G-Rg3) contained only in red ginseng has been found to show various pharmacological effects such as an anticancer, antiangiogenetic, antimetastastic, liver protective, neuroprotective immunomodulating, vasorelaxative, antidiabetic, insulin secretion promoting and antioxidant activities. It is well known that G-Rg3 could be divided into 20(R)-Rg3 and 20(S)-Rg3 according to the hydroxyl group attached to C-20 of aglycone, whose structural characteristics show different pharmacological activities. It has been reported that G-Rg3 is metabolized to G-Rh2 and protopanaxadiol by the conditions of the gastric acid or intestinal bacteria, thereby these metabolites could be absorbed, suggesting its absolute bioavailability (2.63%) to be very low. Therefore, we reviewed the chemical, physical and biological transformation methods for the production on a large scale of G-Rg3 with various pharmacological effects. We also examined the influence of acid and heat treatment-induced potentials on for the preparation method of higher G-Rg3 content in ginseng and ginseng products. Futhermore, the microbial and enzymatic bio-conversion technologies could be more efficient in terms of high selectivity, efficiency and productivity. The present review discusses the available technologies for G-Rg3 production on a large scale using chemical and biological transformation.

The Effects of Cooperative Learning on Children's Understanding of Geometry (협동학습활동이 유아 기하 학습에 미치는 영향)

  • This study was carried out in order to better understand how cooperative learning effects the geometric understanding of young children. The geometry tasks used in the study included the geometric relationship between two dimensional shapes and three dimensional shapes, coordination, symmetry and transformation visualization and spacial reasoning. The subjects were composed of children aged five years and were taken from two kindergartens in a relatively new city close to Seoul. The experimental group of children the comparative learning in geometry. The comparative group of children were enrolled in a kindergarten that uses an the intergrated curriculum. The results indicated that cooperative learning impacted positively on the children's understanding of geometry. The specific results are as follows : The scores that the experimental acquired were higher in terms of p < .001 level. than the scores of the comparative group studying the geometric relationships between two dimensional shapes and three dimensional shapes, coordination, symmetry and transformation visualization & spacial reasoning.

  • The aim of this study was to successfully reflect human body changes on the transformation of the virtual body within 3D virtual fitting spaces. For this purpose, existing problems of shape transformation of the virtual body were analyzed and regression equations which provides useful basic data for transformation of the virtual body that can be applied usefully to the 3D virtual fitting system was developed. Necessary data for the analyses were body measurement and 3D scan data of women with average physical form between the ages of 40 through 49. The reason that we used human body changes of the female subjects in their forties was based on the recognition that fundamental female body changes start to occur from age of forty. Body shapes were largely divided into 3 groups according to obesity which was found to be the biggest factor of shape change. Seven factors were extracted based on factor analysis of 47 body measurement categories and regression equations were created to extract specific measurements for each BMI group based on these seven factors. The major contribution of this paper can be summarized as follows. First, the regression equations to extract specific measurements based on the 7 representative variables remediated existing problem of virtual bodies as it increased the number of body shape transformation areas. Second, the regression equations helped to overcome the problem of current failing to reflecting changes in body cross-section shape based on simple girth measurements based on analysis of cross-section distances.

The Effect of Hyangpaeyangyeongtang and Fritillariae Roylei Bulbus Extracts on the Immune Responses to Epithelioma induced by 3-MCA (3-MCA 유발(誘發) 상피종(上皮腫)에 대(對)한 향패양영탕(香貝養榮湯)과 패모단미제(貝母單味劑)가 면역반응(免疫反應)에 미치는 영향(影響))

  • Hyangpaeyangyeongtang, which was first prescribed in the book of Euijonggumgam (醫宗金鑑), has been widely applied to the treatment of scrofula and carbuncle in the neck and tumor as well in the oriental medicine. The Fritillariae roylei bulbus was also used as an anti-tumor medicine. In order to investigate the effect of Hyangpaeyangyeongtang and Fritillariae roylei bulbus extracts on the immune responses to the epithelioma induced by 3-methylcholanthrene and prostaglandin-$F_2{\alpha}$ in mice, the change of epithelioma size was observed and delayed type hypersensitivity, hemagglutinin titer, hemolysin titer, rosette forming cells, natural killer cell activity, lymphocyte transformation, productivity of interleukin-2 and phagocytic activity were also measured. The results were as follows; 1. The epithelioma size was decreased with statistical significance in the Hyangpaeyangyeongtang group at the 14th day, as compared with the control group. 2. Delayed type hypersensitivity was not shown any decrease in the Hyangpaeyangyeongtang group and the Fritillariae roylei bulbus group, as compared with the control group. 3. Hemagglutinin titer was increased with statistical significance in the Hyangpaeyangyeongtang group, but not in the Fritillariae roylei bulbus group as compared with the control group. 4. Hemolysin titer was increased with statistical significance in the Hyangpaeyangyeongtang group, but not in the Fritillariae roylei bulbus group as compared with the control group. 5. Rosette forming cells were not increased in the Hyangpaeyangyeong tang group, but increased with statistical significance in the Fritillariae roylei bulbus group as compared with the control group. 6. Natural killer cell activity at the ratio 100:1 of effector cells to target cells was increased with statistical significance in the Hyangpaeyayeongngtang group and the Fritillariae roylei bulbus group respectively, and the natural killer cell activity at the ratio of 50:1 and 10:1 of effector cells to target cells Hyangpaeyangyeongtang group was increased with statistical significance as compared with the control group. 7. Lymphocyte transformation was increased in the Hyangpaeyangyeong tang group and the Fritillariae roylei bulbus group respectively, as compared with the control group. 8. Productivity of interleukin-2 was increased in the Hyangpaeyangyecing tang group and the Fritillariae roylei bulbus group respectively, as compared with the control group. 9. Phagocytic activity was not shown any decrease in the Hyangpaeyangyeongtang group and the Fritillariae roylei bulbus group, as compared with the control group.

  • The purpose of this study is to present a digital transformation-based learning model that can be used in universities based on learning digital transformation in order f to be competitive in a rapidly changing environment. Literature review, case study, and focus group interview were conducted and the implications for the learning model from these are as follows. Universities that stand out in related fields are actively using learning analysis to implement dashboards, develop predictive models, and support adaptive learning based on big data, They also have actively introduced advanced edutech to classes. In addition, problems and difficulties faced by other universities and K University when implementing digital transformation were also confirmed. Based on these findings, a digital transformation-based learning model of K University was developed. This model consists of four dimensions: diagnosis, recommendation, learning, and success. It allows students to proceed with learning by diagnosing and recommending various learning processes necessary for individual success, and systematically managing learning outcomes. Finally, academic and practical implications about the research results were discussed.

  • The purpose of this study was to analyze which factor, influenced their change from pre-wage group to family business group. The subject of study classified by wage earner group and family business group. Independent variables effected by transform to family business group are classified by personal variables and workable variables. The major findings of this study are as followings: Sex, age, marriage of personal variables and pre-work time, pre-wage, wholesale - retail business, restaurant business and person service business of pre-industry of work variables had significant effect on transform to family business. Especially, Marriage group and low pre-wage income group significant effected on transfer to family business. By this, personal to transfer to family business can conjugate information of search and choice decision and can be used as a valuable data for future family business study.
