Transgenesis in Fish: Indian Endeavour and Achievement

  • The first Indian transgenic fish was generated in 1991 using borrowed constructs from foreign sources. To construct transformation vectors for the indigenous fishes, growth hormone genes of rohu (r-CH), Labeo rohita and catfish, Heteropneustes fossilis were isolated, cloned and sequenced; their fidelity was confirmed in prokaryotic and eukaryotic systems. A vector was constructed with grass carp b-actin promoter driving the expression of r-GH. Rohu eggs are large. fragile and swell 2~3 times. when fertilized. Hence they were amenable only for electroporated sperm-mediated gene transfer. Accordingly, the sperm electroporation technique was standardized to ensure 25% hatchling survival and 37% Presumptive transgenics without suffering any deformity. Southern analysis confirmed genomic integration in 15% of the tested individuals (Ti) belonging to family lines 2 and 3: another 25% of the Juveniles (Te) were also proved transgenic but with the transgene persisting extrachromosomally for longer than 1 to 2 years. perhaps due to the presence of replicon in the vector. Transgenics belonging to different family lines grew 6~8 times faster than the respective controls. Difference in growth trends of Ti and Te within a family line was not significant. In the Ti family 3 remarkable growth acceleration was sustained for a period longer than 36 weeks but in those of family 2, it gradually decreased. All transgenic fishes including the rohu converted the food at a significantly higher efficiency. Barring the transgenic mudloach, all the other transgenic fishes consumed food at significantly reduced rate.

Design of Fixed Phase Control Circuit of Group Delay Line using Adaptive Vector Control (자동적응 벡터 제어를 이용한 군속도 지연선로의 고정 위상 제어기 설계)

  • The phase characteristic of delay line in feedfarward linearizer has been changed due to variation of operating temperature. In this paper, design method of fixed phase control circuit of group delay line using adaptive vector control is derived. To maintain transfer characteristics of nominal operating temperature, the error correlated signals, which are changed adaptively due to changing of temperature, are added to main signals. The proposed method maintains transfer characteristics under 0.06dB of insertion loss and 0.36$^{\circ}$ of phase variation in case of 1-tone(880 MHz) and under 0.07 dB of insertion loss and 0.35$^{\circ}$ of phase variation in case of 2-tones(877 MHz, 882 MHz) for 10dB input power dynamic range and +/-10$^{\circ}$ phase variation respectively.

  • This paper summarizes a design procedure of radiated noise from engine blocks of marine engines. This air-borne noise is one of the significant noise contributors including the aeroacoustic noise due to intake and exhaust and the re-radiation due to structure-borne noise. Excitation forces by engine operations are evaluated taking into account the power generation mechanism from the burning process to the subsequence motion of internal parts; piston, connecting rod, and crank shaft. The acoustic transfer vector method is incorporated to effectively simulate the radiated noise field under the various operation conditions. A contribution analysis for the various excitations to the radiated noise is conducted. It is found that the firing pressure is the main source of the radiated noise, and so the structure of the cylinder can be modified to significantly reduce the radiated noise from the engine block.

  • The amount of money processed in medicine from the Korea Consumer Agency was studied by the various time series models. The medical data set from the Korea Consumer Agency were consisted of counseling, damage relief and conciliation. For the analysis of time series, autoregressive moving average model, vector autoregressive model and the transfer function model were used. We considered the stationarity and cross correlation function for the identification and fitting. As a result, the transfer function model showed a better prediction. Whereas, the vector autoregressive model also provided good information for the degree and duration of the influence of variables.

  • Infants express their physical and emotional needs to the outside world mainly through crying. However, most of parents find it challenging to understand the reason behind their babies' cries. Failure to correctly understand the cause of a baby' cry and take appropriate actions can affect the cognitive and motor development of newborns undergoing rapid brain development. In this paper, we propose an infant cry recognition system based on deep transfer learning to help parents identify crying babies' needs the same way a specialist would. The proposed system works by transforming the waveform of the cry signal into log-mel spectrogram, then uses the VGGish model pre-trained on AudioSet to extract a 128-dimensional feature vector from the spectrogram. Finally, a softmax function is used to classify the extracted feature vector and recognize the corresponding type of cry. The experimental results show that our method achieves a good performance exceeding 0.96 in precision and recall, and f1-score.

Post Trajectory Insertion Performance Analysis of Korea Pathfinder Lunar Orbiter Using SpaceX Falcon 9

  • This paper presents an analysis of the trans-lunar trajectory insertion performance of the Korea Pathfinder Lunar Orbiter (KPLO), the first lunar exploration spacecraft of the Republic of Korea. The successful launch conducted on August 4, 2022 (UTC), utilized the SpaceX Falcon 9 rocket from Cape Canaveral Space Force Station. The trans-lunar trajectory insertion performance plays a crucial role in ensuring the overall mission success by directly influencing the spacecraft's onboard fuel consumption. Following separation from the launch vehicle (LV), a comprehensive analysis of the trajectory insertion performance was performed by the KPLO flight dynamics (FD) team. Both orbit parameter message (OPM) and orbit determination (OD) solutions were employed using deep space network (DSN) tracking measurements. As a result, the KPLO was accurately inserted into the ballistic lunar transfer (BLT) trajectory, satisfying all separation requirements at the target interface point (TIP), including launch injection energy per unit mass (C3), right ascension of the injection orbit apoapsis vector (RAV), and declination of the injection orbit apoapsis vector (DAV). The precise BLT trajectory insertion facilitated the smoother operation of the KPLO's remainder mission phase and enabled the utilization of reserved fuel, consequently significantly enhancing the possibilities of an extended mission.

Effect of Sucrose and Polybrene on the Gene Transfer into Porcine Oocytes using Retroviral Vector (레트로 바이러스 벡터를 이용한 돼지난자에의 유전자 전이에 있어 Sucrose와 Polybrene의 효과)

  • In vitro matured porcine oocytes have very small volume of perivitellinspace (PVS). In these respect, the effect of sucrose and polybrene on the efficiency of gene transfer was investigated. As a gene (hGH) transfer vehicle, vesicular stomatitis virus glycoprotein pseudotyped retroviral vector (VSV-G) was used. Sucrose treatment has no detrimental effect on the rates of cleavage and resulted in the enlargement of PVS for the efficient introduction of retroviral vector stocks. Introduction rates of retrovirus in 0.5, 1, 2, 3 % sucrose treatment group were higher than that of the non-treatment group (39.3, 43.3, 35.7, 40.7 % vs. 8.3 %), respectively. In addition, we observed that sucrose pretreatment during injection procedure significantly reduce the frequency of polyspermy. In general, polybrene is a polycation essential for retrovirus transduction. The groups with the addition of 0.5, 5, 50$\mu\textrm{g}$/$m\ell$ polybrene exhibited a significant effect on gene transfer compared to that of the non-addition group (56.5, 50.0, 57.1 % vs. 34.6 %), respectively But, when the oocytes were co-injected with retrovirus and 50$\mu\textrm{g}$/$m\ell$ polybrene, the rates of cleavage and blastocyst development were 43.3 and 4.6%, respectively. This rates were lower than those of the non-addition group (70.0 and 17.3 %). In conclusion, sucrose pretreatment have increased efficiency of retroviral mediated gene transfer in porcine oocytes with no damage on in vitro fertilization and embryo development. In addition, sucrose pretreatment was beneficial in polyspermy inhibition. Presence of polybrene during microinjection showed a beneficial effect on the gene transfer in porcine oocytes, in low concentration. And these results will provide an useful tool for production of transgenic pigs by retroviral mediated gene transfer.

Configurtion of electron transfer cofactors in photosystem II studied by pulsed EPR

  • The major electron transfer cofactors in photosystem II have been studied by pulsed EPR, pulsed electron electron double resonance (PELDOR) and laser excited spin polarized electron spin echo envelope modulation (ESEEM) methods, in non-oriented and oriented photosystem II membranes. Distances between radical pairs were determined trom the observed dipole interaction constants to be 27.3 A for P680-QA, 30 A, etc. with the error within 1 A. Angles between the distance vector and membrane normal was determined by orientation dependence of oriented membranes with the accuracy of 5˚ The results were compared with the recent structural data by X-ray analysis.

Interference Suppression Using Principal Subspace Modification in Multichannel Wiener Filter and Its Application to Speech Recognition

  • It has been shown that the principal subspace-based multichannel Wiener filter (MWF) provides better performance than the conventional MWF for suppressing interference in the case of a single target source. It can efficiently estimate the target speech component in the principal subspace which estimates the acoustic transfer function up to a scaling factor. However, as the input signal-to-interference ratio (SIR) becomes lower, larger errors are incurred in the estimation of the acoustic transfer function by the principal subspace method, degrading the performance in interference suppression. In order to alleviate this problem, a principal subspace modification method was proposed in previous work. The principal subspace modification reduces the estimation error of the acoustic transfer function vector at low SIRs. In this work, a frequency-band dependent interpolation technique is further employed for the principal subspace modification. The speech recognition test is also conducted using the Sphinx-4 system and demonstrates the practical usefulness of the proposed method as a front processing for the speech recognizer in a distant-talking and interferer-present environment.

  • To achieve a high performance in direct vector control of induction motor, it is essential to correct estimation of rotor flux. The accuracy of flux observers for induction machines inherently depends on parameter sensitivity. This paper presents an analysis method for conventional flux observers using Parameter Sensitivity. The Parameter sensitivity is defined as the ratio of the percentage change in the system transfer function to the percentage change of the parameter variation. We define the ratio between real flux and estimated flux as the transfer function, and analyzed a parameter sensitivity of this transfer function by simulation.
