Conjugated Linoleic Acid Reduction of Vascular Endothelial Growth Factor Expression in Murine Mammary Tumor Cells through Alteration of Prostaglandin E2

  • Conjugated linoleic acid (CLA) is a group of positional and geometric isomers of linoleic acid that have been used to reduce the incidence, growth and metastasis of breast, colon, prostate and gastric cancer in animals. CLA could reduce tumor growth by altering angiogenesis; a process requiring associated angiogenic factors such as vascular endothelial growth factor (VEGF). In this study, we determined whether CLA could modulate the expression of VEGF in murine mammary tumor cells and adipocytes. The c9, t11-CLA isomer reduced VEGF transcripts and protein when mammary tumor cells were stimulated with PMA. That isomer also reduced VEGF expression in un stimulated mouse 3T3-L1 adipocytes. Since VEGF can be regulated by cyclooxygenase-2 (COX-2), we determined whether CLA could alter COX-2 enzyme expression and $PGE_2$ production. The c9, t11-CLA isomer reduced not only COX-2 enzyme expression but also $PGE_2$ production. Thus, c9, t11-CLA could modulate neovascularization by alteration of VEGF expression from mammary tumor cells and adipocytes by reducing COX-2 metabolites.

메니에르병 환자의 질병 체험 연구 (Qualitative Study on the Experience of Patients with Meniere Disease)

  • Purpose: The purpose of this study was to understand and describe the experiences of patients with Meniere disease. Methods: Data were collected from February 19, 2019, to February 5, 2020, through individual in-depth interviews with 13 Meniere patients. Verbatim transcripts were analyzed using Colaizzi's phenomenological analysis. Results: The experiences of patients with Meniere disease were clustered into the following four themes from 22 meaning units: 1) Complex unfamiliar symptoms that shatter both balance of the body and peace of the heart; 2) A disease that medical treatment and health professionals cannot cure; 3) Suffering hardships that cannot be understood by non-Meniere sufferers; and 4) Making daily efforts to become healthier. Symptoms of Meniere disease are life-shattering and depressing because they are neither visible nor easily curable. Over time, as they accepted the reality of living with the disease, the participants would shift their focus from complete symptomatic cure to leading a healthy and more balanced life. Conclusion: This study shows that Meniere disease has a pervasive impact on all aspects of the patients' lives. Patients are prone to experiencing restrictions in their social functioning and activities. They also experience psychosocial problems due to the unseen nature of their symptoms. This study elucidates the experiences of Meniere patients and the need for nursing intervention to help improve their quality of life and ability to self-manage. Lastly, this study shows the need for a coordinated interdisciplinary approach to raising public awareness of the disease.

Serotonin (5-HT) Receptor Subtypes Mediate Regulation of Neuromodulin Secretion in Rat Hypothalamic Neurons

  • Serotonin (5-HT), the endogenous nonselective 5-HT receptor agonist, activates the inositol-1,4,5-triphosphate/calcium $(InsP3/Ca^{2+})$ signaling pathway and exerts both stimulatory and inhibitory actions on cAMP production and neuromodulin secretion in rat hypothalamic neurons. Specific mRNA transcripts for 5-HT1A, 5-HT2C and 5-HT4 were identified in rat hypothalamic neurons. These experiments were supported by combined techniques such as cAMP and a $Ca^{2+}$ assays in order to elucidate the associated receptors and signaling pathways. The cAMP production and neuromodulin release were profoundly inhibited during the activation of the Gi-coupled 5-HT1A receptor. Treatment with a selective agonist to activate the Gq-coupled 5-HT2C receptor stimulated InsP3 production and caused $Ca^{2+}$ release from the sarcoplasmic reticulum. Selective activation of the Gs-coupled 5-HT4 receptor also stimulated cAMP production, and caused an increase in neuromodulin secretion. These findings demonstrate the ability of 5-HT receptor subtypes expressed in neurons to induce neuromodulin production. This leads to the activation of single or multiple G-proteins which regulate the $InsP3/Ca^{2+}/PLC-{\gamma}$ and adenyl cyclase / cAMP signaling pathways.

Differentially Expressed Genes under Cold Acclimation in Physcomitrella patens

  • Cold acclimation improves freezing tolerance in plants. In higher plants, many advances have been made toward identifying the signaling and regulatory pathways that direct the low-temperature stress response; however, similar insights have not yet been gained for simple nonvascular plants, such as bryophytes. To elucidate the pathways that regulate cold acclimation in bryophytes, we used two PCR-based differential screening techniques, cDNA amplified fragment length polymorphism (cDNA-AFLP) and suppression subtractive hybridization (SSH), to isolate 510 ESTs that are differentially expressed during cold acclimation in Physcomitrella patens. We used realtime RT-PCR to further analyze expression of 29 of these transcripts during cold acclimation. Our results show that cold acclimation in the bryophyte Physcomitrella patens is not only largely similar to higher plants but also displays distinct differences, suggests significant alteration during the evolution of land plants.

Detection of Circulating Melanoma Cells by a Two-marker Polymerase Chain Reaction Assay in Relation to Therapy

  • Malignant melanoma is one of the most rapidly increasing cancer types, and patients with metastatic disease have a very poor prognosis. Detection of metastatic melanoma cells in circulation may aid the clinician in assessing tumor progression, metastatic potential, and response to therapy. Tyrosinase is a key enzyme in melanine biosynthesis. The gene is actively expressed in melanocytes and melanoma cells. Melan A is a differentiation antigen that is expressed in melanocytes. The presence of these molecules in blood is considered a marker for circulating melanoma cells. In this study, we analyzed the usefulness of this marker combination I evaluating the response to therapy in the blood of 30 patients with malignant melanoma. Circulating cells were detected by a reverse-transcriptase-polymerase-chain reaction. The tyrosinase expression was observed in 9 (30%) patients and Melan A in 19 (63.3%) patients before therapy. Following treatment, the tyrosinase mRNA was detected in only one patient, while Melan A transcripts were still present in 14 patients. We suggest that this molecular assay can identify circulating melanoma cells that express melanoma-associated antigens and may provide an early indication of therapy effectiveness.

SF3B4 as an early-stage diagnostic marker and driver of hepatocellular carcinoma

  • An accurate diagnostic marker for detecting early-stage hepatocellular carcinoma (eHCC) is clinically important, since early detection of HCC remarkably improves patient survival. From the integrative analysis of the transcriptome and clinicopathologic data of human multi-stage HCC tissues, we were able to identify barrier-to-autointegration factor 1 (BANF1), procollagen-lysine, 2-oxoglutarate 5-dioxygenase 3 (PLOD3) and splicing factor 3b subunit 4 (SF3B4) as early HCC biomarkers which could be detected in precancerous lesions of HCC, with superior capabilities to diagnose eHCC compared to the currently popular HCC diagnostic biomarkers: GPC3, GS, and HSP70. We then showed that SF3B4 knockdown caused G1/S cell cycle arrest by recovering $p27^{kip1}$ and simultaneously suppressing cyclins, and CDKs in liver cancer cells. Notably, we demonstrated that aberrant SF3B4 overexpression altered the progress of splicing progress of the tumor suppressor gene, kruppel like factor 4 (KLF4), and resulted in non-functional skipped exon transcripts. This contributes to liver tumorigenesis via transcriptional inactivation of $p27^{kip1}$ and simultaneous activation of Slug genes. Our results suggest that SF3B4 indicates early-stage HCC in precancerous lesions, and also functions as an early-stage driver in the development of liver cancer.

벼 (Oryza sativa L.)배양세포의 고중력유도성 cDNA의 탐색 (Screening of Gravity Inducible cDNAs in Rice(Oryza sativa L.) Cultured Cell)

  • 벼(Oryza sativa L. cv Nipponbaie)배양세포에 중력 450,000 x g를 처리하여 cDNA library를 만들고, 무처리 및 고중력을 처리한 cDNA 프로브로 스크리닝을 실시하여 고중력에 특이적으로 양성반응을 나타내는 GSC 13 및 GSC124 cDNA를 선발하였다. 선발된 두 유전자 GSC 13 및 GSC 124의 길이는 각각 1.34 및 0.67 kilobase pairs였으며, 배양세포내에서 관련된 transcript의 크기는 각각 2.0 및 1.9 kilobase pairs인 것으로 나타났다. 두 유전자를 프로브로한 Northern hybridization을 실시한 결과 GSC 13, GSC 124 공히 배양세포내에 고중력처리에 의해 특이적으로 축적되는 mRNA가 나타났으며, 중력강도 300,000 x g 에 비해 450,000 x g 처리에서 더욱 강한 축적을 보였고 450,000 x g 4시간 처리에서 최대의 수준을 보였다.

일제시대 사암침법에 관한 의사학적 고찰 - 문헌중심으로 - (Medical Historical Studies on various parties of Sa-am acupuncture)

  • The Sa-am acupuncture is the acupuncture method created during Chosun Dynasty. It is a unique acupuncture method that was created solely in Korea. Its theories are based on the 69th article of "Classic of Difficult Issues", or Nanjing (難經), which says "tonify its mother when deficiency occurs, and purgate its son when excess occurs" (虛則補其母, 實則瀉其子); the concept of "controlling the viscera which restrains the target" "抑其官" was added to this theory to form the principle of Sa-am acupuncture. It is significant that it turns to the basic principles of the Five Viscera and Six Bowels rather than the 體針 or 阿是針 of the existing acupuncure. Sa-am acupuncture was established in the middle of Chosun Dynasty and was popularized to the practitioners of Korean Medicine through transcripts and printed books. The medical books of at the time of Japanese imperialism that introduced Sa-am acupuncture are "經絡學總論", "舍岩鍼灸訣", "經濟要訣", "靑囊訣", "察病要訣", "濟世寶鑑", "經驗寢具編", and "舍岩 靑囊訣". The magazine that mentions the Sa-am acupuncture is "韓方醫藥". The books on Traditional Medicine that is related to Sa-am acupuncture were of great help in reconstruction of Sa-am acupuncture after the liberation of Korea and explaining the principle of Sa-am acupunture in various angles.

Effects of Acute Metal Exposures on the Viability and mRNA Expression of Metallothionein in Hemibarbus mylodon Fry

  • Transcriptional modulation of metallothionein (MT) during acute metal exposures (cadmium, copper or zinc) was examined in fry of Hemibarbus mylodon, a threatened fish species in Korean peninsula. Viability of H. mylodon fry was most affected by copper exposure (up to 79% of mortality at 1 ppm for 48 hours) and considerably by cadmium exposure (21 to 54% of mortality). On the other hand, Zn showed the least adverse effect on the viability (0 to 13% of mortality) of this species. Based on the semi-quantitative RT-PCR analysis, the stimulation of MT mRNA in response to metal exposures followed generally in a dose-dependent fashion where cadmium was the most potent inducer for the induction of MT transcripts in fry (up to more than 5-fold) while the lowest response was observed in zinc-exposed group (2-fold at maximum). From the exposure using environmentally realistic doses of cadmium (0 to 0.05 ppm for 24 hours), MT expression at mRNA level was also sensitively modulated toward upregulation up to more than 3-fold as relative to non-exposed control. Results from the present study would be a good basis for understanding the adaptive capacity and stress physiology of this endangered fish species during metal pollution.

OrCanome: a Comprehensive Resource for Oral Cancer

  • Oral cancer is one of the most prevalent cancers in India but the underlying mechanisms are minimally unraveled. Cancer research has immensely benefited from genome scale high throughput studies which have contributed to expanding the volume of data. Such datasets also exist for oral cancer genes but there has been no consolidated approach to integrate the data to reveal meaningful biological information. OrCanome is one of the largest and comprehensive, user-friendly databases of oral cancer. It features a compilation of over 900 genes dysregulated in oral cancer and provides detailed annotations of the genes, transcripts and proteins along with additional information encompassing expression, inhibitors, epitopes and pathways. The resource has been envisioned as a one-stop solution for genomic, transcriptomic and proteomic annotation of these genes and the integrated approach will facilitate the identification of potential biomarkers and therapeutic targets.
